Contriving Multi-Epitope Subunit of Vaccine for COVID-19: Immunoinformatics Approaches.

COVID-19 has recently become the most serious threat to public health, and its prevalence has been increasing at an alarming rate. The incubation period for the virus is ~1-14 days and all age groups may be susceptible to a fatality rate of about 5.9%. COVID-19 is caused by a novel single-stranded, positive (+) sense RNA beta coronavirus. The development of a vaccine for SARS-CoV-2 is an urgent need worldwide. Immunoinformatics approaches are both cost-effective and convenient, as in silico predictions can reduce the number of experiments needed. In this study, with the aid of immunoinformatic

Safety and immunogenicity of a novel quadrivalent subunit influenza vaccine in animal models.

Background: Compared with trivalent influenza vaccines, quadrivalent influenza vaccines are expected to provide wider protection against influenza B virus infections. We developed a novel quadrivalent subunit influenza vaccine which was distinct from the influenza vaccines available on the market in production process. In this research, we evaluated the safety and immunogenicity of the quadrivalent subunit influenza vaccine in animal models. Methods: In toxicity assessment, 40 SD rats were randomly assigned to be intramuscularly injected with 1.0 ml of the tested vaccine (33 μg/ml) or 0.9% sod

Cost-effectiveness of the Adjuvanted Herpes Zoster Subunit Vaccine in Older Adults.

IMPORTANCE: The live attenuated herpes zoster vaccine (ZVL) is recommended for immunocompetent adults 60 years or older, but the efficacy wanes with age and over time. A new adjuvanted herpes zoster subunit vaccine (HZ/su) has higher efficacy but might be more expensive. The choice of vaccines depends on their relative values. OBJECTIVE: To assess the cost-effectiveness of HZ/su. DESIGN, SETTING, AND PARTICIPANTS: Markov decision model with transition probabilities based on the US medical literature. Participants were immunocompetent adults 60 years or older. Data were derived from participa

Development of a Freeze-Dried, Heat-Stable Influenza Subunit Vaccine Formulation.

An influenza pandemic remains a major public health concern. A key strategy to prevent a pandemic is to stockpile and pre-position stable influenza vaccine to allow rapid deployment in response to an outbreak. However, most influenza vaccines today are formulated as liquids that are stable only within a temperature range of 2°C to 8°C and require use of a cold chain, making vaccine transportation, distribution, and storage complicated and expensive, particularly for developing countries. To support the National Strategy for Pandemic Influenza preparedness in the United States and international

Antibody response to inactivated subunit influenza vaccine in patients with Wegener's granulomatosis.

In the present study we investigated the humoral response to inactivated subunit influenza vaccine in patients with Wegener's granulomatosis, who were in clinical and serological remission after immunosuppressive treatment (Group I). The results were compared with patients with Wegener's granulomatosis who were treated immunosuppressively, but were not vaccinated (Group II) and with healthy persons who received the vaccine (Group III). After vaccination, antihemagglutinin and antineuraminidase antibody titers significantly increased in Groups I and Group III subjects when compared with the pre

Stability of an antifertility vaccine consisting of gonadotropin subunits linked to tetanus toxoid.

The shelf life and thermal stability of an antifertility vaccine, in which gonadotropin subunits are linked to carriers such as tetanus toxoid and cholera toxin chain B and has successfully completed phase-I clinical trials at five centres in India, was studied. The vaccine adsorbed on alum was stored at three temperatures, 4 degrees C, room temperature (20-30 degrees C) and at 40 degrees C, for a period of up to 1 year. The human chorionic gonadotropin (hCG) binding capacity of antibodies (peak titres) induced in rodents by the vaccine after 6 months of storage at 40 degrees C and at room tem

Current status of an oral B subunit whole cell cholera vaccine.

Purified B subunit of cholera toxin retains membrane-binding capacity and protective immunogenicity and yet has no toxic activity as tested in animals. These properties suggest that B subunit might be a promising immunogen, particularly as an oral vaccine, for stimulating protective antitoxic immunity against cholera in man. A method has been elaborated which allows preparation of +/- 10 grams of pure B subunit per fermentor culture cycle. As tested in both Swedish and Bangladeshi volunteers purified B subunit alone or in combination with conventional whole cell vaccine gives no side-effects a

Multi-stage subunit vaccines against Mycobacterium tuberculosis: an alternative to the BCG vaccine or a BCG-prime boost?

INTRODUCTION: More than two billion people are latently infected with Mycobacterium tuberculosis. Most tuberculosis (TB)-subunit vaccines currently in various stages of clinical trials are designed for prevention of active TB, but not to prevent reactivation of latent TB-infection. Thus, there is an urgent need for an effective multi-stage vaccine based on early-expressed and latently-expressed antigens that prevents both acute and latent infections. AREAS COVERED: Here, we reviewed the published pre-clinical and clinical studies of multi-stage subunit vaccines against TB, and the protective

Effect of vaccination with a porcine reproductive and respiratory syndrome subunit vaccine on sow reproductive performance in endemic farms.

The objective of this field study was to evaluate the reproductive performance of sows after vaccination with a porcine reproductive and respiratory syndrome (PRRS) subunit vaccine (PRRSFREE PRRS subunit vaccine, Reber Genetics, Taiwan, Republic of China) under field conditions. The study was performed in three farms with endemic infections with both PRRS virus (PRRSV)-1 and PRRSV-2, a situation representative of most Korean farms. Pregnant sows were immunised intramuscularly with 2.0 ml of the PRRS subunit vaccine at 58 and 79 days of gestation (eight and five weeks antepartum) according to t

Future Path Toward TB Vaccine Development: Boosting BCG or Re-educating by a New Subunit Vaccine.

Tuberculosis (TB), an infectious disease caused by Mycobacterium tuberculosis (Mtb), kills 5,000 people per day globally. Rapid development and spread of various multi drug-resistant strains of Mtb emphasize that an effective vaccine is still the most cost-effectives and efficient way of controlling and eradicating TB. Bacillus Calmette-Guerin (BCG), the only licensed TB vaccine, still remains the most widely administered human vaccine, but is inefficient in protecting from pulmonary TB in adults. The protective immunity afforded by BCG is thought to wane with time and considered to last only

[A new influenza subunit vaccine: reactogenicity and antigenicity in comparison to split and whole virus vaccines (author's transl)].

The reactogenicity and immunogenicity of a new influenza subunit vaccine essentially containing only haemagglutinin and neuraminidase was studied in man. The vaccine was compared to commercially available vaccines, an adjuvant containing tween-ether split vaccine (800 IU per dose), and a fluid whole-virus vaccine (2100 IU per dose). Two dosages (700 and 2100 IU) of the fluid subunit vaccine were compared. All vaccines contained the virus strains recommended by the WHO for the 1975/76 season. In a double-blind study 399 volunteers were randomly selected to receive one of the four vaccines. The

Identification of an ideal adjuvant for receptor-binding domain-based subunit vaccines against Middle East respiratory syndrome coronavirus.

Middle East respiratory syndrome (MERS), an emerging infectious disease caused by MERS coronavirus (MERS-CoV), has garnered worldwide attention as a consequence of its continuous spread and pandemic potential, making the development of effective vaccines a high priority. We previously demonstrated that residues 377-588 of MERS-CoV spike (S) protein receptor-binding domain (RBD) is a very promising MERS subunit vaccine candidate, capable of inducing potent neutralization antibody responses. In this study, we sought to identify an adjuvant that optimally enhanced the immunogenicity of S377-588 p

Protection with recombinant Clostridium botulinum C1 and D binding domain subunit (Hc) vaccines against C and D neurotoxins.

Recombinant botulinum Hc (rBoNT Hc) vaccines for serotypes C1 and D were produced in the yeast Pichia pastoris and used to determine protection against four distinct BoNT C and D toxin subtypes. Mice were vaccinated with rBoNT/C1 Hc, rBoNT/D Hc, or with a combination of both vaccines and challenged with BoNT C1, D, C/D, or D/C toxin. Mice receiving monovalent vaccinations were partially or completely protected against homologous toxin and not protected against heterologous toxin. Bivalent vaccine candidates completely survived challenges from all toxins except D/C toxin. These results indicate

Comparison of immunogenicity of mRNA and protein subunit SARS-CoV-2 vaccines in dialysis patients: a multicenter study.

Patients on maintenance hemodialysis (HD) have impaired immune function and are at high risk of severe COVID-19; however, comparative data on immunogenicity and vaccine choice behavior between mRNA and protein subunit SARS-CoV-2 vaccines remain limited. In this observational study, 227 participants (202 patients on HD and 25 non-dialysis individuals) received either an mRNA vaccine (BNT162b2) or a protein subunit vaccine (NVX-CoV2373) in 2024 vaccination season. Anti-SARS-CoV-2 spike antibody levels were assessed at weeks 0, 4, and 24, with week 4 defined as the primary outcome and week 24 as

Selection, Design, and Immunogenicity Studies of ASFV Antigens for Subunit mRNA Cocktail Vaccines with Specific Immune Response Profiles.

Development of safe and effective subunit vaccines for controlling African Swine Fever Virus (ASFV) infection has been hampered by a lack of protective viral antigens, complex virion structures, and multiple mechanisms of infection. Here, we selected ASFV antigens based on their localization on the virion, known functions, and homologies to the subunits of the protective vaccinia virus vaccine. We also engineered viral capsid proteins for inducing optimal antibody responses and designed T cell-directed antigens for inducing broad and robust cellular immunity. The selected antigens in lipid nan

Exploring approaches and advancements in the development and evaluation of multi-epitope subunit vaccines against tick-borne viruses.

Tick-borne viruses (TBVs) pose significant public health challenges worldwide, causing severe diseases such as hemorrhagic fevers and encephalitis. Given the limitations of existing therapeutic and preventive measures, the development of effective vaccines represents a critical strategy for controlling TBV-related infections. This review explores recent advancements in the design and evaluation of multi-epitope subunit vaccines targeting key TBVs, including hemorrhagic fever viruses (e.g., Crimean-Congo hemorrhagic fever virus, Severe fever with thrombocytopenia syndrome virus), encephalitis v

Development of multi epitope subunit vaccines against emerging carp viruses Cyprinid herpesvirus 1 and 3 using immunoinformatics approach.

Cyprinid herpesvirus is a causative agent of a destructive disease in common and koi carp (Cyprinus carpio), which leads to substantial global financial losses in aquaculture industries. Among the strains of C. herpesvirus, C. herpesvirus 1 (CyHV-1) and C. herpesvirus 3 (CyHV-3) are known as highly pathogenic to carp fishes in Europe, Asia, and Africa. To date, no effective vaccine has been developed to combat these viruses. This study aimed to develop unique multi-epitope subunit vaccines targeting the CyHV-1 and CyHV-3 using a reverse vaccinology approach. The study began with a comprehensiv
