Fabrication and evaluation of a chitin whisker/poly(L-lactide) composite scaffold by the direct trisolvent-ink writing method for bone tissue engineering.

Although poly(l-lactide) (PLLA) based porous scaffolds have been widely fabricated through 3D printing, their poor mechanical properties and osteogenic activity still do not meet the needs of bone tissue repair. Herein, chitin whiskers (CHWs), having outstanding mechanical properties, excellent cell affinity, osteogenic activity, etc. were designed to introduce into the PLLA matrix. Moreover, a trisolvent system, including dichloromethane (DCM), 2-butoxyethanlol (2-Bu) and dibutyl phthalate (DBP), instead of a single solvent system of DCM was chosen to prepare CHW/PLLA (CP) composite inks. Barrels by the sea: Barrels XX meeting report.

The 20th annual Barrels meeting brought together researchers who utilize behavioral, physiological, anatomical, and molecular techniques to understand the structure and function of the barrel system. Barrels XX featured talks on the role inhibition has in shaping cortical responses within the barrel system, the molecular cues that influence the development of the whisker-to-barrel system, and the synaptic plasticity that can shape responses within the system. The meeting highlighted why the whisker-to-barrel system is an ideal model to investigate the development of cortical circuitry and how

Plasticity and stability of somatosensory maps in thalamus and cortex.

Work on elucidating the mechanisms of plasticity in somatosensory maps continues apace. Recent work has focused on both the nature of the thalamocortical interactions that determine plasticity, and on the differences between plasticity induced by nerve block or damage versus that induced by experience. Recordings from awake behaving animals have thrown light on the thalamocortical circuit mechanisms that underlie map plasticity; meanwhile, intracellular recordings from cortical slices have thrown light on the precise synaptic mechanisms underlying plasticity.

Population Consequences of Physiological Responses to Stressors: A Road Map for Endocrine Data.

Environmental change causes physiological effects that can last years in long-lived species, yet there is a dearth of methods for quantifying such effects and their population consequences across the necessary decadal timescales. The Population Consequences of Multiple Stressors (PCoMS) framework is intended to link short-term responses to stressors to health, then to vital rates, and from there to population-level effects. To date, implementations of the PCoMS framework have primarily focused on behavioral responses of marine mammals to stressors, enabled by data available from biologging dev

Phenotypic characterization of the KK/HlJ inbred mouse strain.

Detailed histopathological diagnoses of inbred mouse strains are important for interpreting research results and defining novel models of human diseases. The aim of this study was to histologically detect lesions affecting the KK/HlJ inbred strain. Mice were examined at 6, 12, and 20 months of age and near natural death (ie, moribund mice). Histopathological lesions were quantified by percentage of affected mice per age group and sex. Predominant lesions were mineralization, hyperplasia, and fibro-osseous lesions. Mineralization was most frequently found in the connective tissue dermal sheath

Systematic screening for skin, hair, and nail abnormalities in a large-scale knockout mouse program.

The International Knockout Mouse Consortium was formed in 2007 to inactivate ("knockout") all protein-coding genes in the mouse genome in embryonic stem cells. Production and characterization of these mice, now underway, has generated and phenotyped 3,100 strains with knockout alleles. Skin and adnexa diseases are best defined at the gross clinical level and by histopathology. Representative retired breeders had skin collected from the back, abdomen, eyelids, muzzle, ears, tail, and lower limbs including the nails. To date, 169 novel mutant lines were reviewed and of these, only one was found

Effects of neonatal infraorbital lesions upon central trigeminal primary afferent projections in rat and hamster.

Transganglionic and anterograde horseradish peroxidase transport was used to evaluate the central projections of undamaged trigeminal (V) nerve branches in adult rats and hamsters subjected to transection of the infraorbital nerve and to cauterization of the vibrissae follicles at birth. In rats, deafferented regions of the V brainstem nuclear complex did not receive abnormal projections from undamaged mandibular sensory afferents. Undamaged ophthalmic-maxillary fibers also failed to terminate heavily in the region deafferented by the neonatal infraorbital lesions. In the hamster, on the other

Constructing an adult orofacial premotor atlas in Allen mouse CCF.

Premotor circuits in the brainstem project to pools of orofacial motoneurons to execute essential motor action such as licking, chewing, breathing, and in rodent, whisking. Previous transsynaptic tracing studies only mapped orofacial premotor circuits in neonatal mice, but the adult circuits remain unknown as a consequence of technical difficulties. Here, we developed a three-step monosynaptic transsynaptic tracing strategy to identify premotor neurons controlling vibrissa, tongue protrusion, and jaw-closing muscles in the adult mouse. We registered these different groups of premotor neurons o

The clock gene brain and muscle Arnt-like protein-1 (BMAL1) is involved in hair growth.

It is known that baldness caused by androgenetic alopecia is involved with androgen and the androgen receptor. Furthermore, it has been reported that testosterone secretion follows a circadian rhythm. Therefore, we hypothesized that a relationship exists between androgen-induced alopecia and biological rhythm. The mammalian circadian rhythm is controlled by several clock genes. Brain and muscle aryl hydrocarbon receptor nuclear translocator-like protein-1 (BMAL1), one of the clock genes, is a transcription factor that plays central roles in the regulation of circadian rhythms. In this study, w

Stimulation of trigeminal afferents improves motor recovery after facial nerve injury: functional, electrophysiological and morphological proofs.

Recovery of mimic function after facial nerve transection is poor: the successful regrowth of axotomized motoneurons to their targets is compromised by (1) poor axonal navigation and excessive collateral branching, (2) abnormal exchange of nerve impulses between adjacent regrowing axons, and (3) insufficient synaptic input to facial motoneurons. As a result, axotomized motoneurons get hyperexcitable and unable to discharge. Since improvement of growth cone navigation and reduction of the ephaptic cross talk between axons turn out be very difficult, we concentrated our efforts on the third detr

Elevated dietary magnesium prevents connective tissue mineralization in a mouse model of pseudoxanthoma elasticum (Abcc6(-/-)).

Pseudoxanthoma elasticum (PXE) is an autosomal recessive multisystem disorder characterized by ectopic connective tissue mineralization, with clinical manifestations primarily in the skin, eyes, and cardiovascular system. There is considerable, both intra- and interfamilial, variability in the spectrum of phenotypic presentation. Previous studies have suggested that mineral content of the diet may modify the severity of the clinical phenotype in PXE. In this study, we utilized a targeted mutant mouse (Abcc6(-/-)) as a model system for PXE. We examined the effects of changes in dietary phosphat

The expression pattern of the Hmgic gene during development.

The technique of RNA in situ hybridization to mouse embryo sections from different developmental stages was used to perform a detailed analysis of the expression pattern of the gene for the architectural chromatin factor Hmgic. At early stages of fetal development (day 9.5 post conceptionem), Hmgic is expressed at a high rate throughout the whole embryo. In the second half of development, the pattern of expression becomes more restricted. Expression is found in mesenchymal derivatives, which differentiate into cartilage or muscle, in epithelial cell layers of the lung, pancreas, submandibular

Behavioral adaptations after unilateral whisker denervation.

The rodent whisker system provides an excellent model to study experience dependent plasticity in neural morphology, circuitry, and behavior. Rodents use bilateral whisker sensation to gather information about their environment. Unilateral whisker denervation disrupts whisker circuitry but its impact on task specific behavior is largely unknown. Adult mice with unilateral whisker denervation display a preference to using the intact whisker set to inspect objects, but do not have altered open field navigation. An object localization task requiring only the intact whisker set did not detect any

Functional Architecture and Encoding of Tactile Sensorimotor Behavior in Rat Posterior Parietal Cortex.

The posterior parietal cortex (PPC) in rodents is reciprocally connected to primary somatosensory and vibrissal motor cortices. The PPC neuronal circuitry could thus encode and potentially integrate incoming somatosensory information and whisker motor output. However, the information encoded across PPC layers during refined sensorimotor behavior remains largely unknown. To uncover the sensorimotor features represented in PPC during voluntary whisking and object touch, we performed loose-patch single-unit recordings and extracellular recordings of ensemble activity, covering all layers of PPC i

Somatosensory maps.

Somatosensory areas containing topographic maps of the body surface are a major feature of parietal cortex. In primates, parietal cortex contains four somatosensory areas, each with its own map, with the primary cutaneous map in area 3b. Rodents have at least three parietal somatosensory areas. Maps are not isomorphic to the body surface, but magnify behaviorally important skin regions, which include the hands and face in primates, and the whiskers in rodents. Within each map, intracortical circuits process tactile information, mediate spatial integration, and support active sensation. Maps ma

Changes to the somatosensory barrel cortex in C57BL/6J mice at early adulthood (56 days post-natal) following prenatal alcohol exposure.

Children with Fetal Alcohol Spectrum Disorder (FASD) have impaired sensory processing skills as a result of neurodevelopmental anomalies. The somatosensory barrel field of rodent brain is a readily accessible model for studying the effects of alcohol exposure. Within the barrel field, the posterior medial barrel subfield (PMBSF) receives sensory inputs from the large vibrissae on the contralateral face. This study reports on the consequence of prenatal exposure to alcohol on the somatosensory cortices of mice later in life. Two control groups, a sucrose and a non-treated control, were also exa

Hair-Loss Preventing Effect of Grateloupia elliptica.

This study was conducted to evaluate the effect of Grateloupia elliptica, a seaweed native to Jeju Island, Korea, on the prevention of hair loss. When immortalized rat vibrissa dermal papilla cells were treated with extract of G. elliptica, the proliferation of dermal papilla cells significantly increased. In addition, the G. elliptica extract significantly inhibited the activity of 5α-reductase, which converts testosterone to dihydrotestosterone (DHT), a main cause of androgenetic alopecia. On the other hand, the G. elliptica extract promoted PGE2 production in HaCaT cells in a dose-dependent

Distinct laminar origins of sensory-evoked high-gamma and low-frequency ECoG signals revealed by optogenetics.

Electrocorticography (ECoG) provides a high-spatiotemporal-resolution measure of cortical activity (cortical surface electrical potentials, CSEPs) in humans and animals. The CSEP high-gamma band (Hγ, 65 to 170 Hz) correlates with neuronal firing rates at the columnar spatial scale and is widely used as a biomarker of local activity. Whether Hγ reports all stages of columnar processing, intermediate processing in L2/3 (close to the ECoG electrode), or the main columnar output in L5, is unknown. We disentangled the laminar origins of Hγ and other ECoG bands by optogenetically suppressing L2/3 or

High-speed voltage imaging of action potentials in molecular layer interneurons reveals sensory-driven synchrony that augments movement.

Testing whether the synchrony of action potential firing is a cerebellar coding mechanism requires simultaneous recording, with high temporal fidelity, from populations of identified neurons. Here, we used targeted one-photon voltage imaging at 2-4 kHz to record action potentials from groups of ∼10-300 molecular layer interneurons (MLIs) expressing a positively tuned, genetically encoded voltage indicator, FORCE1f or pAce. In awake resting mice, crus I MLIs fired brief (∼1-ms) spikes at 20-60 spikes/s. Sensory stimuli of air puffs to the whiskers evoked short-latency (<10 ms) increases in spik

Structural Bases of Atypical Whisker Responses in a Mouse Model of CDKL5 Deficiency Disorder.

Mutations in the CDKL5 (cyclin-dependent kinase-like 5) gene cause CDKL5 Deficiency Disorder (CDD), a severe neurodevelopmental syndrome where patients exhibit early-onset seizures, intellectual disability, stereotypies, limited or absent speech, autism-like symptoms and sensory impairments. Mounting evidences indicate that disrupted sensory perception and processing represent core signs also in mouse models of CDD; however we have very limited knowledge on their underlying causes. In this study, we investigated how CDKL5 deficiency affects synaptic organization and experience-dependent plasti
