Irreversible binding of chlorinated ethylenes to macromolecules.

Rats have been exposed in a closed system to the chlorinated ethylenes vinyl chloride and trichloroethylene and to carbon tetrachloride as a reference compound. Data of uptake of the compounds, of urinary excretion of metabolites, and of exhalation after exposure show that the chlorinated ethylenes are metabolized much faster than carbon tetrachloride, probably due to their common ethylene structure. To eliminate differences in uptake, calculation of metabolites of the three compounds in tissues was based on the amount actually taken up by the animals. Vinyl chloride, trichloroethylene, and ca…

[Experimental studies on fistula formation after percutaneous trans-hepatic cholangiodrainage].

The process of intra- and extra-hepatic fistula formation after percutaneous transhepatic cholangiodrainage (PTCD) was investigated. Furthermore, effects of such 4 materials of catheter as vinyl chloride, silicon, polyethylene and poly-urethane on this formation process were compared in order to clarify factors influencing the process. Fistula formation began 2 weeks after PTCD, and nearly completed after 4 weeks. A drainage fistula most markedly formed in the use of vinyl chloride, as the fistula wall was thick and its collagen fibers were microscopically observed to be dense and regularly ar…

Evaluation of alternative methods for establishing safe levels of occupational exposure to vinyl halides.

The facts that reduction of occupational vinyl chloride exposures to levels within or below the 0.5-5 ppm range has so far been successful in eliminating vinyl chloride-induced liver angiosarcoma and that humans appear to be less sensitive to the carcinogenic effect of vinyl chloride than rats offered an opportunity to verify or dispute risk assessment extrapolation models used, and proposed, by the U.S. EPA. Safe occupational vinyl chloride exposures were defined as levels associated with an incidence of one angiosarcoma in 100,000 exposed workers, determined from rat bioassay data using defa…

Increased acetone exhalation induced by metabolites of halogenated C1 and C2 compounds.

Rats were exposed, in a closed desiccator jar chamber, to concentrations of various halogenated C1 and C2 compounds at which the metabolizing capacities were saturated (Vmax conditions). Within the exposure period of 50 h concentrations of the xenobiotic and of exhaled acetone were monitored in the gas phase of the system. The quantitative extent of acetone exhalation was dependent on the individual compound examined. Acetone exhalation was stimulated in presence of vinyl chloride, vinyl bromide, vinyl fluoride, vinylidene fluoride, cis- and trans-1,2-dichloroethylene, trichloroethylene, perch…

Reductive dechlorination of high concentrations of tetrachloroethene to ethene by an anaerobic enrichment culture in the absence of methanogenesis.

Tetrachloroethene, also known as perchloroethylene (PCE), is a common groundwater contaminant throughout the United States. The incomplete reductive dechlorination of PCE--resulting in accumulations of trichloroethene, dichloroethene isomers, and/or vinyl chloride--has been observed by many investigators in a wide variety of methanogenic environments. Previous mixed-culture studies have demonstrated that complete dechlorination to ethene is possible, although the final dechlorination step from vinyl chloride to ethene is rate limiting, with significant levels of vinyl chloride typically persis…

Chloroethene biodegradation in sediments at 4 degrees C.

Microbial reductive dechlorination of [1,2-14C]trichloroethene to [14C]cis-dichloroethene and [14C]vinyl chloride was observed at 4 degrees C in anoxic microcosms prepared with cold temperature-adapted aquifer and river sediments from Alaska. Microbial anaerobic oxidation of [1,2-14C]cis-dichloroethene and [1,2-14C]vinyl chloride to 14CO2 also was observed under these conditions.

A new syndrome with pigmentation, scleroderma, gynaecomastia, Raynaud's phenomenon and peripheral neuropathy.

A middle-aged man developed impotence, gynaecomastia, peripheral neuropathy, pigmentation lymphadenopathy, scleroderma, malabsorption and Raynaud's phenomenon following prolonged exposure to trichlorethylene. Somes, but not all, of these features have previously been reported in vinyl chloride disease, and trichlorethylene is related chemically to vinyl chloride. Seven patients with a similar but not identical syndrome have been described in Japan and U.S.A., apparently unrelated to exposure to industrial toxins.

PVC: health implications and production trends.

Poly(vinyl chloride) (PVC) is a complex plastic system. Individual components of the PVC system, including residual vinyl chloride monomer (RVCM) and certain additives, may pose risks of harm to human health. There have been significant reductions in the RVCM content of PVC resin since 1974, reducing the cancer risk of workers in PVC fabrication plants and consumers of PVC products. A "no-effect" level for vinyl chloride monomer (VCM)-induced carcinogenesis has not been found to date; therefore, the significance of human exposure to low levels of RVCM remains to be determined. Exposure to PVC …

Epidemiology of hepatic angiosarcoma in the United States: 1964-1974.

A nationwide survey of hepatic angiosarcoma (HAS) in the United States during the years 1964 through 1974 identified 168 cases. Of these, 42 cases (25%) were associated with known etiologic factors, such as vinyl chloride monomer exposure during preparation of poly(vinyl chloride), use of Thorotrast in angiography, exposure to inorganic arsenic, and treatment with androgenic-anabolic steroids; 126 cases (75%) are of uncertain etiology. HAS most often affects males (ratio of approximately 3:1), peaks in the sixth and seventh decades of life (somewhat earlier than other sarcomas of the liver) an…

Covalent binding to apoprotein is a major fate of heme in a variety of reactions in which cytochrome P-450 is destroyed.

The heme in rat liver microsomal cytochrome P-450 was labeled with 14C or 3H and the microsomes were fractionated after in vitro incubations with a variety of agents known to destroy cytochrome P-450 heme. A major fraction of the heme label was irreversibly bound to apoprotein in all cases, including incubations with fluroxene, 1-octene, vinyl bromide, trichloroethylene, vinyl chloride, parathion, cumene hydroperoxide, NaN3, or iron-ADP complex. Label was also extensively bound to apoprotein when purified and reconstituted cytochrome P-450 was incubated with NADPH and vinyl chloride. This proc…

[Acroosteolysis in PVC autoclave cleaners: history of an occupational disease].

OBJECTIVES: This paper examines the history of an occupational disease which has now disappeared: acroosteolysis of manual tank cleaners in the production of polyvinyl chloride (PVC), which is a rare disease characterized by destructive alterations of the distal phalanges of the hands. METHODS: All the available literature on this disease was examined. The history of acroosteolysis was studied within the general framework of the history of the discovery of adverse health effects of exposure to vinyl chloride, and this history was studied up to the end of the 1960's. RESULTS: The disease was …

Research on pollution identification and safety thresholds based on the olfactory effect on a halogenated hydrocarbon contaminated site.

To effectively address odor control issues at sites contaminated with halogenated hydrocarbons, it is essential to establish an odor risk pr … r soil and groundwater based on olfactory effects. The findings indicate that 1,1-dichloroethylene, vinyl chloride, chloroform, and 1,1-dichloroethane are prevalent throughout the contaminated site, displaying elevated concentration levels and substantially influencing the overall contamination extent. Metabolic activation of chlorinated ethylenes: dependence of mutagenic effect on electrophilic reactivity of the metabolically formed epoxides.

In chlorinated ethylenes, the chlorine substitution exerts, by its electron withdrawal effect, a stabilization of the molecule which increas … out: mutagenic potential is bound to unsymmetric substitution and high chemical reactivity (as with vinyl chloride, vinylidene chloride, and trichloroethylene), symmetric substitution results in lower chemical reactivity and non-mutagenicity. So far, the rule is substantiated by positive carcinogenic effects in animal experiments with vinyl chloride, vinylidene …

[Liver disease in polyvinyl chloride production workers - Clinical and peritoneoscopic aspects (author's transl)].

Peculiar clinical symptoms observed in polyvinyl chloride production workers of a nearby chemical plant arose the suspicion of liver and spleen disease in these workers before first reports on vinyl chloride-induced angiosarcoma of the liver became known. Starting at the end of 1972, 44 workers were followed up by biochemical investigations, peritoneoscopy and liver biopsy. As early as 1973 it could be ascertained, that advanced vinyl chloride-induced liver damage presents as hepatic fibrosis with splenomegaly, portal hypertension, and thrombocytopenia without conspicuous deterioration of hepa…

Mutagenic activity of vinyl compounds and derived epoxides.

Many vinyl compounds, such as vinyl chloride and some inhalational anesthetics, are known to be mutagens. In the present study, 10 vinyl compounds or derived epoxides, widely used in industry, were assayed in the Salmonella typhimurium/mammalian microsome system. 3 strains of histidine-dependent S. typhimurium, TA1535, TA98 and TA100 were used. Of the 10 compounds, 4 were mutagens. They were 9-vinylanthracene, vinylcarbazole, 3-vinyl-7-oxabicyclo[4.1.0]heptane and 3-epoxyethyl-7-oxabicyclo[4.1.0]-heptane. The study confirmed the overall genotoxicity of vinyl compounds and epoxides and the need…

First-row transition-metal chloride complexes of the wide bite-angle diphosphine (iPr)DPDBFphos and reactivity studies of monovalent nickel.

The diphosphine 4,6-bis(3-diisopropylphosphinophenyl)dibenzofuran (abbreviated as (iPr)DPDBFphos) has been metalated with transition metal d … vely. The monovalent nickel complex reacts with substrates containing C-X bonds; and in the case of vinyl chloride, a Ni(II) vinyl species ((iPr)DPDBFphos)Ni(CH═CH(2))Cl is generated along with the Ni(II) dichloride complex. The monovalent Ni(I) chloride is an active catalyst in the Kumada cross-coupling reaction of vinyl chloride and phenyl Grignard reagent.

Characteristics of haloethylene-induced acetonemia in rats.

A series of halogenated ethylenes (vinyl chloride, vinylidene fluoride, cis- and trans-1,2-dichloroethylene, perchloroethylene) induces increased acetone exhalation in rats. Exposures of differently pre-treated rats to vinylidene fluoride suggest that a metabolite of the haloethylene must be envolved in eliciting this formation of acetone. This conclusion is based on (a)dependence of acetone exhalation on the concentration of vinylidene fluoride, (b)effect of inducing agents, (c)effect of pyrazol, a metabolic inhibitor, (d)effect of cysteine, (e)effect of hypoxia and (f)the time course of acet…

Angiosarcoma of the spleen: a report of two cases and review of the literature.

Results of the ultrastructural study of one of two cases of splenic angiosarcoma established the blood vessel origin of this tumor. Fifty-three previously reported cases were reviewed. None of the 55 patients had a history of exposure to thorium dioxide, vinyl chloride, or arsenic, which are known to be associated with hepatic angiosarcoma and other tumors. A comparison of the splenic and hepatic angiosarcomas showed that tumors not associated with exogenous material frequently involve the spleen and liver simultaneously, and that tumors associated with thorium dioxide, vinyl chloride, or arse…

Mutagenic and carcinogenic risks associated with halogenated olefins.

Recent experimental evidence indicates that structural analogs of vinyl chloride namely, vinylidene chloride and trichloroethylene, are mutagenic. Carcinogenic response also has been observed in experimental animals following exposure to vinylidene chloride, trichloroethylene, and perchloroethylene. More recent observations demonstrate low-level vinyl chloride-induced mammary carcinoma. An additional chlorinated olefin, chloroprene, has demonstrated a mutagenic response in several test systems. Likewise, several studies have indicated significant excesses of chromosomal aberrations as well as …
