Neuropathological stages of neuronal, astrocytic and oligodendrocytic alpha-synuclein pathology in Parkinson's disease.

Alpha-synucleinopathies are neurodegenerative diseases characterized by the spread of alpha-synuclein (α-syn) aggregates throughout the central nervous system in a stereotypical manner. These diseases include Lewy body disease (LBD), which encompass Dementia with Lewy bodies (DLB), Parkinson's Disease (PD), and Parkinson's Disease Dementia (PDD), and Multiple System Atrophy (MSA). LBD and MSA chiefly contain α-syn aggregates in neurons and oligodendrocytes, respectively, although glial α-syn pathology in LBD is increasingly being recognized. Semi-quantitative and machine learning-based quantif

Astrocytic gamma-aminobutyric acid dysregulation as a therapeutic target for posttraumatic stress disorder.

Post-traumatic stress disorder (PTSD) remains a debilitating psychiatric condition with limited pharmacological treatment options. Identifying novel therapeutic targets is critical for addressing its unmet clinical needs. Through our comprehensive human clinical research, including both cross-sectional and longitudinal studies, we revealed a compelling link between dysregulated prefrontal gamma-aminobutyric acid (GABA) levels and PTSD symptoms. Notably, elevated prefrontal GABA levels in PTSD patients are associated with impaired cerebral blood flow (CBF) and symptom severity, normalizing with

Compartment-specific analysis reveals disrupted astrocytic calcium transients in Alzheimer's mice.

Alzheimer's disease (AD) is characterized by presence of extracellular amyloid plaques, intracellular tau tangles, and extensive neuronal cell death. In addition to neurons, astrocytes modulate neuronal network activity through tripartite synapses and are increasingly recognized for their involvement in AD pathology. Astrocytic calcium signaling has been implicated in AD pathological processes, including disrupted synaptic transmission, dysregulated glutamate homeostasis, and impaired vascular function via astrocytic endfeet. However, a systematic analysis of calcium dynamics within specific a…

Astrocyte Loss Augments Body Weight Through Reduction in Adipose Sympathetic Outflows.

Emerging evidence indicates that astrocytes modulate energy metabolism and homeostasis. However, one important but poorly understood element is the necessity of astrocytes in the control of body weight. Here, we apply viral vector-assisted brain-region selective loss of astrocytes to define physiological roles played by astrocytes in the arcuate nucleus of the hypothalamus (ARH) and to elucidate the involved mechanism. We find that astrocyte loss potently augments body weight in adult mice fed chow or high-fat diet. Mechanistically, we find that the loss of astrocytes reduces adipose tissue no…

MTOR Promotes Astrocyte Activation and Participates in Neuropathic Pain through an Upregulation of RIP3.

Neuropathic pain (NP), a chronic pain condition, is the result of abnormalities in both central and peripheral pain conduction pathways. Here, we investigated the underlying mechanisms associated with this effect. We found that following chronic constriction injury (CCI) surgery, there was an increase of mTOR in astrocytes and an activation of astrocytes within the spinal cord. Pharmacological inhibition of mTOR reversed CCI-induced hyperalgesia and neuroinflammation. Moreover, knockdown of astrocytic mTOR rescued the downregulation of spinal glutamate metabolism-related protein expression, un…

Investigating cocaine- and abstinence-induced effects on astrocyte gene expression in the nucleus accumbens.

In recent years, astrocytes have been increasingly implicated in cellular mechanisms of substance use disorders (SUD). Astrocytes are structurally altered following exposure to drugs of abuse; specifically, astrocytes within the nucleus accumbens (NAc) exhibit significantly decreased surface area, volume, and synaptic colocalization after operant self-administration of cocaine and extinction or protracted abstinence (45 days). However, the mechanisms that elicit these morphological modifications are unknown. The current study aims to elucidate the molecular modifications that lead to observed …

Atypical antipsychotics alter microglial functions via astrocyte-derived extracellular vesicles.

A limited understanding of the underlying molecular mechanisms of atypical antipsychotics has hindered efforts to develop the next generation of treatments for schizophrenia. In particular, there has been little investigation of how medications like clozapine and olanzapine modulate human non-neuronal cells, including astrocytes and microglia. Recent postmortem and serum-based studies suggest that schizophrenia etiology involves dysregulated cellular communication through extracellular vesicles (EVs). Astrocytes are a major source of these EVs and are strongly implicated in the etiology of sch…

Astrocytic GABA controls fidelity of temporal cortical processing in Fragile X Syndrome.

Astrocytes control neural communications by influencing GABAergic transmission through uptake and synthesis of GABA. Impaired GABAergic signaling is thought to underlie cortical hyperexcitability in autism. Here we show that dysregulation of astrocyte GABA transport in Fragile X syndrome (FXS), a leading genetic cause of autism, contributes to circuit hyperexcitability. Human FXS astrocytes derived from patient-specific induced pluripotent stem cells and mouse Fmr1 knockout (KO) astrocytes display a significant increase in levels of GABA and GABA-synthesizing enzyme GAD65/67. Our astrocyte-spe…

Functional contribution of astrocytic Kir4.1 channels to spasticity after spinal cord injury.

Spasticity, a prevalent motor issue characterized by network hyperexcitability, causes pain and discomfort, with existing treatments offering limited relief. While past research has focused on neuronal factors, the role of astrocytes in spasticity has been overlooked. This study explores the potential of restoring astrocytic K+ uptake to reduce spasticity following spinal cord injury (SCI). Astrocytes buffer extracellular K+ via Kir4.1 channels, preventing neuronal hyperexcitability. Following SCI, Kir4.1 levels decrease at the injury site, though the consequences and mechanisms of this reduct…

Astrocytic PERK Deficiency Drives Prefrontal Circuit Dysfunction and Depressive-Like Behaviors.

Major depressive disorder (MDD) is associated with dysfunction in prefrontal cortex (PFC) circuits, yet the glial mechanisms underlying these abnormalities remain unclear. Here, downregulation of the endoplasmic reticulum (ER) stress sensor PERK in PFC astrocytes is identified as a mechanistic contributor to depression-related phenotypes. PERK expression is markedly reduced in PFC astrocytes from individuals with MDD and in two chronic-stress mouse models. Astrocyte-specific PERK deletion in stress-naïve mice is sufficient to induce robust depressive-like behaviors and widespread PFC circuit p…

The Impact of Small Molecule on the Astrocyte's Viability Derived From Epileptic Brain Tissues.

INTRODUCTION: Astrocyte dysfunction plays a crucial role in epileptogenesis by impacting neuronal excitability and synaptic transmission. This study investigates how specific small molecules (SMs) affect the survival of astrocytes derived from various brain regions (the neocortex, hippocampus, or amygdala) of patients with medically refractory epilepsy. METHODS: This study evaluated astrocyte responses to three distinct SMs (valproate, forskolin, and GSK3 inhibitor/WNT activator CHIR99021), individually or in combined forms, to determine their differential effects on astrocyte survival. RESU…

Regional Position and Axonal Environment Shape Astrocyte Morphology in the Mouse Optic Projection.

Optic neuropathies encompass a diverse group of disorders that result in damage to the optic nerve and lead to vision loss. Despite the initial site of damage often being injury specific, we have an incomplete understanding of how differential regional susceptibility arises. Recent studies have implicated astrocytes in this phenomenon in mice, showing regional transcriptional heterogeneity in optic nerve astrocytes in response to glaucoma and optic nerve crush. Because astrocytes provide structural support and protection from tissue deformation, we examined the length of the optic nerve for as…

Maternal dietary folate imbalance alters cerebellar astrocyte morphology and density in offspring.

BACKGROUND: Maternal folate usage is essential for neurodevelopment, but its effects on cerebellar structure are unclear. Cerebellum undergoes a protracted period of development, making it sensitive to maternal nutritional imbalances. Astrocytes are necessary for cerebellar cortex structure and function. This study examined the impact of varying maternal dietary folate levels on the morphology and density of cerebellar astrocytes in rat offspring. MATERIALS AND METHODS: Twelve adult female rats (Rattus norvegicus) were randomly allocated to one of four premixed food groups: standard (2 mg/kg)…

Astrocyte-neuron communication through the complement C3-C3aR pathway in Parkinson's disease.

Neuroinflammation and autoimmunity are pivotal in the pathogenesis of neurodegenerative diseases. Complement activation and involvement of astrocyte-neuron C3/C3aR pathway have been observed, yet the mechanisms influencing α-synuclein (α-syn) pathology and neurodegeneration remain unclear. In this study, elevated levels of complement C3 were detected in the plasma of α-syn PFF-induced mice and the substantia nigra of A53T transgenic mice. Colocalization of complement C3 with astrocytes was also observed. Overexpression of complement C3 exacerbated motor dysfunction, dopaminergic neuron loss, a…

Astrocytic centrin-2 expression in entorhinal cortex correlates with Alzheimer's disease severity.

Astrogliosis is a condition shared by acute and chronic neurological diseases and includes morphological, proteomic, and functional rearrangements of astroglia. In Alzheimer's disease (AD), reactive astrocytes frame amyloid deposits and exhibit structural changes associated with the overexpression of specific proteins, mostly belonging to intermediate filaments. At a functional level, amyloid beta triggers dysfunctional calcium signaling in astrocytes, which contributes to the maintenance of chronic neuroinflammation. Therefore, the identification of intracellular players that participate in a…

Astrocyte tau deposition in progressive supranuclear palsy is associated with dysregulation of MAPT transcription.

Progressive supranuclear palsy (PSP) is a neurodegenerative disease characterized by 4R tau deposition in neurons as well as in astrocytes and oligodendrocytes. While astrocytic tau deposits are rarely observed in normal aging (so-called aging-related tau astrogliopathy, ARTAG) and Alzheimer's disease (AD), astrocytic tau in the form of tufted astrocytes is a pathognomonic hallmark of PSP. Classical biochemical experiments emphasized tau synthesis in neurons in the central nervous system, suggesting that astrocytic tau inclusions might be derived from uptake of extracellular neuronal-derived t…

Impairment of hippocampal astrocyte-mediated striatal dopamine release and locomotion in Alzheimer's disease.

BACKGROUND: Clinical and translational research has identified deficits in the dopaminergic neurotransmission in the striatum in Alzheimer's disease (AD) and this could be related to the pathophysiology of psychiatric symptoms appearing even at early stages of the pathology. HYPOTHESIS: We hypothesized that AD pathology in the hippocampus may influence dopaminergic neurotransmission even in the absence of AD-related lesion in the mesostriatal circuit. METHODS: We chemogenetically manipulated the activity of hippocampal neurons and astrocytes in wild-type and hemizygous TgF344-AD (Tg) rats, a…
