Interaction of estrogen mimics, singly and in combination, with plasma sex steroid-binding proteins in rainbow trout (Oncorhynchus mykiss).

The plasma sex steroid-binding protein (SBP) is believed to be involved in regulating circulating endogenous sex steroids as well as cellular signal transduction to nuclear steroid receptors in sex steroid-sensitive tissues. In this study, a variety of estrogen mimics (EMs), which may contribute to the endocrine disrupting effects observed in fish, were tested for the ability to interact with the rainbow trout plasma sex steroid-binding protein (rtSBP) either singly or in binary combinations. The EMs ethynylestradiol, diethylstilbestrol, 4-hydroxytamoxifen, genistein, zearalenone, 4-t-octylphe

Exposure to Endocrine-Disrupting Chemicals during Pregnancy and Weight at 7 Years of Age: A Multi-pollutant Approach.

BACKGROUND: Prenatal exposure to endocrine-disrupting chemicals (EDCs) may induce weight gain and obesity in children, but the obesogenic effects of mixtures have not been studied. OBJECTIVE: We evaluated the associations between pre- and perinatal biomarker concentrations of 27 EDCs and child weight status at 7 years of age. METHODS: In pregnant women enrolled in a Spanish birth cohort study between 2004 and 2006, we measured the concentrations of 10 phthalate metabolites, bisphenol A, cadmium, arsenic, and lead in two maternal pregnancy urine samples; 6 organochlorine compounds in maternal

Degradation and partial oxidation of waste plastic express packaging bags in supercritical water: Resources transformation and pollutants removal.

Millions of waste plastic express packaging bags (PEPBs) were generated with the rapid development of the express delivery industry due to the boom of electronic commerce. Waste PEPBs contain polyethylene (PE) material and large number of pollutants such as plasticizers and flame retardants. In this study, two effective and environmental-friendly methods were proposed to produce valuable products and remove pollutants from waste PEPBs by supercritical water degradation (SCWD) and supercritical water partial oxidation (SCWPO) treatments. Both SCWD and SCWPO treatments could effectively obtain v

Induced aging, structural change, and adsorption behavior modifications of microplastics by microalgae.

The effects of microalgal biofouling on microplastic (MP) may differ from bacterial biofouling. In this study, the influence of microalgae on MP surface alteration, structural change, and adsorption of organic micropollutants was evaluated. Virgin polyethylene (PE), polyvinyl chloride (PVC), and polyamide (PA) were each immersed in algal photobioreactor and river freshwater for 30 days to simulate algal and river microbe biofouling respectively. Consequently, their physicochemical changes and adsorption potential of a mixture of six bisphenol analogues (BPA, BPS, BPE, BPB, BPF, BPAF) and two p

Dental microplastics as emerging neurotoxicants: a systematic review on human data.

BACKGROUND: Microplastics and compounds linked to plastic have recently emerged as potential contaminants that might affect brain function; nevertheless, results from these investigations have been inconsistent across epidemiological, clinical, and mechanistic research. Whether in people or in vitro models, this systematic review sought to compile the most recent data on the link between exposures to microplastics and neurological effects. METHODS: A comprehensive search of PubMed, Scopus, MEDLINE, EBSCO, and ScienceDirect was performed to identify studies published from January 2015 to Decem

Bisphenol A accelerates the vascular complications in patients with Type 2 diabetes mellitus through vascular calcification-a molecular approach.

PURPOSE: Environmental pollutant Bisphenol A (BPA) strongly interacts with insulin resistance, which leads to type 2 diabetes mellitus (T2DM). Uncontrolled glucose levels in both blood and urine develops vascular complications in T2DM patients. However, glucose-controlled diabetic patients are also affected by vascular complications due to vascular calcification, and there is a lack of clinically relevant data on BPA levels available in patients with T2DM-associated vascular complications due to vascular calcification. Therefore, we measured BPA levels in T2DM-associated vascular complications

Toughening and polymerization stress control in composites using thiourethane-treated fillers.

Filler particle functionalization with thiourethane oligomers has been shown to increase fracture toughness and decrease polymerization stress in dental composites, though the mechanism is poorly understood. The aim of this study was to systematically characterize the effect of the type of filler surface functionalization on the physicochemical properties of experimental resin composites containing fillers of different size and volume fraction. Barium glass fillers (1, 3 and 10 µm) were functionalized with 2 wt% thiourethane-silane (TU-Sil) synthesized de novo and characterized by thermogravim

Prenatal exposure to bisphenol A disrupts RNA splicing in the prefrontal cortex and promotes behaviors related to autism in offspring.

Prenatal exposure to bisphenol A (BPA), a common endocrine disruptor, has been increasingly implicated in neurodevelopmental disorders, including autism spectrum disorder. This study explores the molecular mechanisms by which prenatal BPA exposure affects alternative RNA splicing in the prefrontal cortex and investigates the potential link between alternative RNA splicing and autism-related behaviors in rat offspring. Using RNA sequencing and high-resolution melting real-time PCR, we identified differentially alternative splicing events associated with autism candidate genes. Gene ontology and

Combinational exposure to Bisphenol A and a high-fat diet causes trans-generational Malfunction of the female reproductive system in mice.

Bisphenol A (BPA) is a common endocrine disruptor and a high-fat diet (HFD) also affects fertility. However, little is known about the long-term consequences of simultaneous exposure to BPA and a HFD on reproductive health. Herein, we assessed the effects of maternal exposure to BPA in combination with a HFD on reproductive function in subsequent generations of female mice and evaluated its effects on the hypothalamic-pituitary-gonadal axis. We found that the combination of maternal exposure to BPA and a HFD led to increased urine BPA levels, precocious puberty, altered estrous cyclicity, decr

Association Between Gestational Exposure to Toxicants and Autistic Behaviors Using Bayesian Quantile Regression.

Autism spectrum disorder, which is characterized by impaired social communication and stereotypic behaviors, affects 1%-2% of children. Although prenatal exposure to toxicants has been associated with autistic behaviors, most studies have been focused on shifts in mean behavior scores. We used Bayesian quantile regression to assess the associations between log2-transformed toxicant concentrations and autistic behaviors across the distribution of behaviors. We used data from the Maternal-Infant Research on Environmental Chemicals study, a pan-Canadian cohort (2008-2011). We measured metal, pest

Bisphenol A-induced oxidative stress, hepatotoxicity and altered estrogen receptor expression in Labeo bata: impact on metabolic homeostasis and inflammatory response.

Bisphenol A (BPA), a weak estrogenic endocrine disruptor and a well-known plasticizer, has the potential to perturb diverse physiological functions; however, its impact on immune and metabolic function in aquatic vertebrates is relatively less understood. The present study aims to investigate the impact of BPA on hepatotoxicity, metabolic and immune parameters vis-à-vis estrogen receptor expression modulation in a freshwater teleost, Labeo bata (Cyprinidae, Cypriniformes). The 96-h median lethal concentration of BPA in L. bata has been determined as 4.79 mg/L. Our data demonstrate that congrue

Impact of exposure to phenols during early pregnancy on birth weight in two Canadian cohort studies subject to measurement errors.

BACKGROUND: It is of interest to know whether early pregnancy exposure to phenols such as bisphenol-A (BPA) or triclosan (TCS) negatively impacts birth weight outcomes. Exposure to these chemicals is widespread in the Canadian population but obtaining accurate measurements of average exposure is difficult because these chemicals are rapidly excreted from the body, causing body levels to fluctuate both within and between days, as observed in a recent Canadian study (P4). This measurement error can attenuate the estimated effects of exposures. METHODS: Data from two Canadian cohort studies, the

Endocrine disruptors and polycystic ovary syndrome (PCOS): elevated serum levels of bisphenol A in women with PCOS.

CONTEXT: Bisphenol A (BPA) is a widespread industrial compound used in the synthesis of polycarbonate plastics. In experimental animals, neonatal exposure to BPA results in a polycystic ovary-like syndrome (PCOS) in adulthood. A bidirectional interaction between androgens and BPA levels has been disclosed. OBJECTIVE: To determine BPA levels in PCOS women as well as the association between BPA and hormonal/metabolic parameters compared to a control group. DESIGN, SETTING, AND PARTICIPANTS: Cross-sectional study of 71 PCOS (National Institutes of Health criteria) and 100 normal women, age- and

Assessment of human contamination of estrogenic endocrine-disrupting chemicals and their risk for human reproduction.

There is broad human exposure to estrogenic endocrine-disrupting chemicals (EDCs), but the data sets that exist are primarily for various environmental media such as food and water rather than the most relevant internal exposure. We have detected various kind of EDC contamination in humans including dioxin and bisphenol A (BPA) widely used for the production of plastic products. BPA was present in serum and follicular fluid at approximately 1-2 ng/ml, as well as in fetal serum and full-term amniotic fluid, confirming passage through the placenta. An approximately five-fold higher concentration

BPAF-induced inflammatory response via estrogen signaling: insights from in vivo and in vitro evidence.

This study investigated the effects of BPAF on the indicators related to inflammatory response and explored the roles of G protein estrogen membrane receptor 1 (GPER1) in this process. The results found that BPAF promoted the secretion of pro-inflammatory cytokines such as IL-1β, IL-8 and TNF-α, increased the protein expression of IL-1β and IL-6, and upregulated the relative mRNA levels of inflammatory related genes in Jurkat cells. In vivo SD rat experiments also found that BPAF exposure significantly upregulated the levels of serum inflammatory cytokines, elevated the relative mRNA expressio

Maternal exposure to bisphenol A has transgenerational effects on the development of experimental asthma through bromodomain-containing protein 4-zinc finger DHHC-type containing 1-stimulators of interferon genes axis.

Perinatal exposures to the environmental estrogen bisphenol A (BPA) are associated with increased asthma prevalence. We tested the hypothesis that perinatal BPA exposure transgenerationally enhances allergic asthma development through the bromodomain-containing protein 4 (BRD4) - zinc finger HHC-1 (ZDHHC1) - stimulators of IFN genes (STING) axis. Female BALB/c mice (F0) were exposed to 10 μg/mL BPA in their drinking water during pregnancy until F1 pups were weaned. Pups were sensitized with low doses of ovalbumin (OVA) on postnatal day 4 (PND 4) and 1% OVA inhaler on PND 18-20. Asthma phenotyp

Analysis of chemical exposures in racial populations in Canada: An investigation based on the Canadian health measures survey.

Despite demonstrated disparities in environmental chemical exposures by racial identity, no Canadian study has systematically assessed the feasibility of using a nationally representative dataset to examine differences in chemical concentrations by race. We assessed the feasibility and constraints of analysing chemical exposures in racial populations, including visible minorities and populations of Indigenous identity, using biomonitoring data collected through the Canadian Health Measures Survey (CHMS). Our primary objectives were to assess the ability to 1) generate geometric means and perce

Alteration of Extracellular Matrix Components in the Anterior Pituitary Gland of Neonatal Rats Induced by a Maternal Bisphenol A Diet during Pregnancy.

The extracellular matrix (ECM) plays crucial roles in the anterior pituitary gland via the mechanism of cell-ECM interaction. Since bisphenol A (BPA), a well-known endocrine disruptor, can cross through the placenta from mother to fetus and bind with estrogen receptors, cell populations in the neonatal anterior pituitary gland could be the target cells affected by this chemical. The present study treated maternal rats with 5000 µg/kg body weight of BPA daily throughout the pregnancy period and then investigated the changes in ECM-producing cells, i.e., pericytes and folliculostellate (FS) cell
