Protocol to generate microglia-containing cerebral organoids to model HIV neuroinflammation.

Cerebral organoids (COs) are valuable for studying neurodegenerative diseases and pathogens, but their limited microglia pose challenges. Here, we present a protocol to generate organoids with physiologically relevant microglia (CO-iMs). We describe steps for differentiating induced pluripotent stem cells (iPSCs) into mesoderm, seeding embryoid bodies (EBs), and harvesting hematopoietic progenitor cells (HPCs). We detail procedures for co-culturing HPCs and iPSCs to generate CO-iMs, followed by their infection and treatment. The model provides a platform to study viral infections and neuroinfl

Serum sPD-1 as a marker of T cell exhaustion in ART-naïve, ART-experienced, and intestinal parasite co-infected HIV-positive adults at the university of Gondar comprehensive specialized hospital, Northwest Ethiopia, 2024.

BACKGROUND: Human Immunodeficiency Virus (HIV) remains a significant global health challenge despite advancements in prevention and treatment. It impairs immune function and leads to T-cell exhaustion, driven by sustained expression of the immune checkpoint receptor PD-1, which exists in membrane-bound and soluble forms. While serum soluble programmed death protein 1 (sPD-1) may serve as a marker for immune exhaustion, its role in antiretroviral therapy (ART) and intestinal parasite (IP) co-infections remains underexplored, especially in developing countries like Ethiopia with high HIV and IP

A retrospective cross-sectional study of FGFR2b alterations in Chinese patients with advanced squamous non-small cell lung cancer: prevalence, programmed death-ligand 1 co-expression patterns, and association with clinical outcomes.

BACKGROUND: Current standard of care for squamous non-small cell lung cancer (sqNSCLC) is not very effective, and FGFR2b-targeted therapy may be a new option for patients with sqNSCLC. The prevalence and clinical implications of fibroblast growth factor receptor 2b (FGFR2b) expression in Chinese patients with advanced sqNSCLC remain poorly characterized. This study evaluated FGFR2b expression patterns, their association with programmed death-ligand 1 (PD-L1) expression, and clinical outcomes in this population. METHODS: This retrospective cross-sectional analysis screened three hundred newly

Low Interfacial Resistance and Superior Suppression to Li-Dendrite Penetration Facilitated by Air-Stable and Mechanically Robust Al/Mg-Co-Doped Li-La-Zirconate as Electrolyte for Li-Based Solid-State Cells.

In the context of usage as a solid electrolyte (SE) for Li-based solid-state cells, the interfacial characteristics of Li-La-zirconate (LLZO) with the electrodes and the mechanical properties of LLZO influence the overall impedance and stability. In this regard, the newly developed air-stable Al/Mg-co-doped LLZO has been found to possess greater resistance to crack propagation (by ∼31%) and fracture stress (by ∼52%), along with elevated hardness and stiffness, as compared to simply Al-doped LLZO. Furthermore, as directly visualized via cross-section electron microscopy at the Li/LLZO interface

Pre-Human Immunodeficiency Virus (HIV) infection Th17 CD4+ T cells as predictors of early HIV disease progression.

Interleukin-17-producing T helper (Th17) CD4+ T cells are highly susceptible to HIV infection and are depleted early in people living with HIV. Here, we investigated whether systemic Th17 cell levels prior to HIV infection are associated with subsequent HIV disease progression. We analyzed archived cryopreserved peripheral blood mononuclear cells (PBMCs) collected within one year prior to HIV acquisition from participants enrolled in a South African cohort (HIV Vaccine Trials Network [HVTN] 503; n = 35) and an East African cohort (Partners Pre-exposure Prophylaxis/Couples' Observational Study

PLK2 modulation of enriched TAp73 affects osteogenic differentiation and prognosis in human osteosarcoma.

There are three subtypes of undifferentiated human conventional osteosarcoma (HCOS): osteoblastic osteosarcoma (OOS), chondroblastic osteosarcoma (COS), and fibroblastic osteosarcoma (FOS). HCOS also exhibits heterogeneous pathological maldifferentiation in individual patients. Currently, the mechanism regulating HCOS differentiation remains unclear, and therapies are ineffective. Osteopontin (OPN) and osteocalcin (OCN) are markers of osteoblast maturation, and their expression is inhibited in HCOS. A previous study found that PLK2 inhibited TAp73 phosphorylation and consequent anti-OS functio

Primary structure, ligand binding, and localization of the human type 3 inositol 1,4,5-trisphosphate receptor expressed in intestinal epithelium.

The second messenger, inositol 1,4,5-trisphosphate (InsP3) transduces many hormonal signals which regulate Ca(2+)-dependent processes in the intestinal epithelium. To study the receptors for InsP3 (InsP3Rs), which function as intracellular Ca2+ channels, cDNA clones encoding InsP3Rs were isolated from a human colon adenocarcinoma cell line, HT29. The majority of clones encoded the type 3 InsP3R, the product of the ITPR3 gene on chromosome 6, for which only a 147-amino-acid fragment was known previously (Ozcelik, T., Sudhof, T. A biomimetic folding paper co-culture system for analyzing cellular crosstalk in bone healing: Interactions of bone morphogenetic protein-2, Wnt family member 1, periostin, and platelet-derived growth factor subunit A between periosteum-derived progenitor cells and adipose-derived stem cells.

Bone defects, commonly associated with osteoporosis, result in fragile bones prone to fractures in both men and women. Osteoporotic fractures often lead to prolonged healing due to impaired cell differentiation. Understanding cellular crosstalk during bone regeneration is crucial for developing effective treatments. Periosteum-derived progenitor cells (PDPCs) and adipose-derived stem cells (ADSCs) play essential roles in bone formation because ADSCs secret key growth factors, such as bone morphogenetic protein-2, Wnt family member 1, Periostin, and platelet-derived growth factor subunit A, tha

T CD4+ cells count among patients co-infected with human immunodeficiency virus type 1 (HIV-1) and human T-cell leukemia virus type 1 (HTLV-1): high prevalence of tropical spastic paraparesis/HTLV-1-associated myelopathy (TSP/HAM).

INTRODUCTION: HIV positive patients co-infected with HTLV-1 may have an increase in their T CD4+ cell counts, thus rendering this parameter useless as an AIDS-defining event. OBJECTIVE: To study the effects induced by the co-infection of HIV-1 and HTLV-1 upon CD4+ cells. MATERIAL AND METHODS: Since 1997, our group has been following a cohort of HTLV-1-infected patients, in order to study the interaction of HTLV-1 with HIV and/or with hepatitis C virus (HCV), as well as HTLV-1-only infected asymptomatic carriers and those with tropical spastic paraparesis/HTLV-1 associated myelopathy (TSP/HAM

Intracellular distribution and late endosomal effects of the ocular albinism type 1 gene product: consequences of disease-causing mutations and implications for melanosome biogenesis.

To investigate the function of ocular albinism type 1 (OA1), the gene responsible for X-linked ocular albinism, we employed a construct cont … d the ER and trafficked to endolysosomal compartments. We observed that expression of WT Oa1-GFP in COS cells caused an apparent enlargement of late endosomes and a redistribution of the mannose-6-phosphate receptor (M6PR). None of the mutants displayed the full range of effects on the redistribution of M6PR exhibited by WT Oa1. The effects of Oa1 on late e …

A natural synergy-based nanosystem co-delivering siRNA and paclitaxel for full-stage apoptosis promotion in melanoma.

Paclitaxel (PTX) kills tumor cells by stabilizing microtubules to induce apoptosis, but its efficacy is limited by resistance mediated by the anti-apoptotic protein survivin. Targeted inhibition of survivin with siRNA could synergistically enhance PTX-induced apoptosis; however, nucleic acid-based therapeutics, such as siRNA, exhibit high instability and susceptibility to degradation, making their efficacy highly dependent on specialized delivery systems. Thus, co-delivery systems for PTX and siRNA are critical to achieving synergistic antitumor activity. Natural products present several advan

Gene analysis of inherited antithrombin deficiency and functional analysis of abnormal antithrombin protein (N87D).

Inherited antithrombin (AT) deficiency is one of the most clinically significant forms of congenital thrombophilia and follows an autosomal dominant mode of inheritance. We analyzed SERPINC1 in a patient who developed deep-vein thrombosis and low AT activity during pregnancy, and identified a novel missense mutation c.259A>G (p.Asn87Asp; N87D). Surprisingly, analysis of the parents' DNA showed that they did not possess this mutant, and thus, it may have been due to a de novo mutation. We also expressed this mutant AT protein in COS-1 cells and compared its intracellular localization and intrac

Role of P450IID6, the target of the sparteine-debrisoquin oxidation polymorphism, in the metabolism of imipramine.

The formation of three oxidative metabolites of imipramine, N-desmethylimipramine (desipramine), 2-hydroxyimipramine, and 10-hydroxyimipramine was studied in microsomes of an extensive metabolizer liver (KDL 26) and of a poor metabolizer liver (KDL 31) and in a homogenate of COS-1 cells in which the P450IID6 complementary deoxyribonucleic acid had been expressed. The following data support the role of P450IID6 in the 2-hydroxylation of imipramine: (1) The formation of 2-hydroxyimipramine was reduced to less than 20% of the control value when microsomes were incubated with serum containing inhi

3D organoid cultivation improves the maturation and functional differentiation of cholangiocytes from human pluripotent stem cells.

Idiopathic cholangiopathies are diseases that affect cholangiocytes, and they have unknown etiologies. Currently, orthotopic liver transplantation is the only treatment available for end-stage liver disease. Limited access to the bile duct makes it difficult to model cholangiocyte diseases. In this study, by mimicking the embryonic development of cholangiocytes and using a robust, feeder- and serum-free protocol, we first demonstrate the generation of unique functional 3D organoids consisting of small and large cholangiocytes derived from human pluripotent stem cells (PSCs), as opposed to trad

Cloning and functional expression of a human neuroendocrine vasoactive intestinal peptide receptor.

Vasoactive intestinal peptide (VIP) is a potent mediator of gastrointestinal, nervous, pulmonary, vascular and immune functions. A cDNA was obtained from human HT29 intestinal epithelial cells and found to encode a 457 amino acid, 52 kDa VIP receptor. Transfection of the cDNA into COS-7 cells and 293 cells resulted in expression of specific saturable binding of VIP with a Kd of 0.8 nM, and induction of increases in intracellular cAMP by VIP with an EC50 of 1 nM. The human VIP receptor is homologous to other G protein-coupled receptors of the secretin-parathyroid hormone receptor family. A 2.8

Pathogenesis of Focal Segmental Glomerulosclerosis Caused by a Leu754Val Mutation in ARHGAP32.

Focal segmental glomerulosclerosis (FSGS) shows a poor response to hormones and other treatment schemes and rapidly progresses to end-stage renal disease. Genetic factors are important causes of FSGS. We recently identified a new candidate pathogenic ARHGAP32 mutation in a family affected by FSGS and further investigated its functional impact through in vivo and in vitro studies. We established in vitro models of ARHGAP32 overexpression in podocytes and COS-7 kidney cells by plasmid transfection. Mice with the point mutation were established using CRISPR/Cas9 technology, followed by the establ

A novel type 2A (Group II) von Willebrand disease mutation (L1503Q) associated with loss of the highest molecular weight von Willebrand factor multimers.

Type 2A von Willebrand disease (VWD) is characterized by decreased platelet-dependent function of von Willebrand factor (VWF) associated with an absence of high-molecular-weight multimers. In this study, sequence analysis of the VWF gene from a Type 2A VWD patient showed a novel, heterozygous T-->A transversion at nucleotide 4510, resulting in the non-conservative substitution of L1503Q in the mature VWF subunit. This substitution, which was not found in 55 unrelated normal individuals, was reproduced by in vitro site directed mutagenesis of a full-length VWF cDNA and was subsequently expresse
