Dorsomedial Striatum Calcium Permeable AMPA Receptors in the Development of Aversion-Resistant Alcohol Drinking.

RATIONALE: Alcohol use disorder is defined by drinking alcohol despite knowledge of negative consequences, often referred to as aversion-resistant drinking (ARD). The dorsomedial (DMS) and dorsolateral striatum (DLS) are necessary for goal-directed and habitual action selection, respectively. Leading hypotheses posit that once drug use becomes compulsive, DMS dependence degrades while DLS dependence increases. This shift may be mediated by changes in synaptic weights from glutamatergic inputs. OBJECTIVES: Using a combination of western-blot, micro-injections, and ex-vivo electrophysiology, we…

Automated Operant Intermittent Alcohol Drinking In Socially-Housed Mice via The Intellicage Testing System.

Despite known confounds of social isolation on rodent behavior, many investigators rely on singly housing mice to assess alcohol preference in preclinical models of alcohol use disorder (AUD). This protocol describes a social alcohol drinking task that allows for high-throughput, automated assessment of alcohol drinking behaviors in mice without the confounding impacts of stress induced by social isolation. The IntelliCage testing system enables investigators to simultaneously assess operant alcohol drinking behavior in up to 16 same-sex mice, each uniquely identified with a subcutaneously imp…

Drinking water nitrate, disinfection byproducts, and prostate cancer incidence in the Agricultural Health Study.

BACKGROUND: Drinking water can be an important source of exposure to nitrate and disinfection by-products, including trihalomethanes (THMs) and haloacetic acids (HAAs). N-nitroso compounds formed endogenously after nitrate ingestion are animal carcinogens, and THM and HAA exposures increase the risk of some cancers. Our objectives were to evaluate associations of drinking water nitrate and disinfection byproducts with total and aggressive (distant stage, poorly differentiated grade, fatal, or Gleason score ≥7) prostate cancer in the Agricultural Health Study cohort. METHODS: Male participants…

Alpha1 adrenergic receptors integrate across multiple separable heart rate variability metrics during alcohol drinking.

Alcohol Use Disorder remains a major challenge and is now a leading cause of death in adults. Biomarkers like heart rate variability (HRV) could give important and novel insights into how pharmacotherapies counteract excessive drive for alcohol, including those promoted by adrenergic receptors (AR). Indeed, α1AR inhibition helps heavy drinkers with greater withdrawal-related mood disruption and arousal, while α1ARs more generally are implicated in cognitive flexibility. To uncover how α1ARs might regulate drinking-related arousal, we used cardiovascular telemetry in adult male and female rats.…

Sex differences in insular cortex function in persistent alcohol drinking despite aversion in mice.

One major hallmark of alcohol use disorder (AUD) is the persistence of drinking despite negative consequences. Among indicators of AUD vulnerability, binge drinking has been identified as one of the strongest risk factors. Although the lifetime prevalence of both binge drinking and AUD has historically been higher in men than women, this gap has dramatically narrowed in the last decade. Additionally, sex differences in AUD and binge drinking have been found in clinical and preclinical studies. At the neurobiological level, the insular cortex plays an important role in AUD, with the anterior (a…

Exceeding Intended Intoxication Predicts Lower Drinking at the Next Event Through Reduced Intoxication Intentions.

OBJECTIVE: Social learning theory posits that direct experience shapes future cognition and behavior. Yet, it remains unclear how drinking experiences from one event influence intentions and behavior of the next drinking event. Exceeding intended levels of intoxication may motivate self-correction or adjustment of future intentions and behavior. However, this process may depend on positive or negative consequences experienced. Therefore, the current study examined whether exceeding intended intoxication and its associated consequences during a given drinking episode predicted intoxication inte…

Investigating Chlorate Occurrence in Hungarian Drinking Water: A Survey Analysis of Treatment Plant Practices.

The revised EU Drinking Water Directive, which designates chlorate as a regulated parameter, presents a significant challenge for Hungary, where breakpoint chlorination with sodium hypochlorite is widely used for ammonium removal. Because hypochlorite decomposes during storage, half of the surveyed plants reported chlorate concentrations exceeding the national limit of 0.25 mg/L. A survey conducted on 208 drinking water treatment plants evaluated current operational practices and mitigation strategies. Chlorate occurrence was primarily associated with systems using sodium hypochlorite as the m…

Longitudinal Antimicrobial Susceptibility Shifts in Environmental Legionella pneumophila from a Drinking Water Distribution System.

Legionella pneumophila (L. pneumophila) persists in engineered water systems and is the causative agent of Legionnaires' disease. Despite its clinical importance, little is known about whether environmental populations are undergoing gradual changes in their antimicrobial susceptibility. Here, we compared the antimicrobial susceptibility of 95 environmental L. pneumophila isolates collected in 2013-2014 with those obtained in 2024-2025 from the same sites in a drinking water distribution system (DWDS) using Epsilometer (E-test). The analyses showed both temporal and spatial variation: Ciproflo…

Enhancing gastric ultrasound training with standardized patient-based drinking simulation: a self-controlled study.

BACKGROUND: Gastric cancer remains a major health burden in China, with low early screening rates and limited access to endoscopic services. Gastric ultrasound offers a noninvasive, low-cost alternative but is underutilized due to the lack of effective training models. This study aimed to evaluate a novel standardized patient (SP)-based drinking simulation for teaching gastric ultrasound to residents. METHODS: In a self-controlled design, 32 ultrasound residents underwent a structured training program comprising a 90-minute theoretical lecture and a 2-hour hands-on workshop. Trainees acted as …

Assessment of PFAS levels in drinking water: A case study from Poznań County (Poland).

Exposure to per- and polyfluoroalkyl substances (PFAS) through drinking water has emerged as a significant public health concern due to their persistent, bioaccumulative nature and adverse health effects. Drinking water is the primary non-occupational source of PFAS exposure. Recently, investigative journalism has alerted about the presumptive contamination of drinking water at various European sites, including Poland. However, direct data on PFAS concentrations in raw and treated drinking water in Poland remain scarce. Therefore, this study analyzed the concentrations of 20 PFAS in 18 drinkin…

Transmissible antimicrobial resistance in Escherichia coli isolated from household drinking water in Ibadan, Nigeria.

Contaminated household water in peri-urban communities is a reservoir for virulent Escherichia coli, but its role in the environmental transmission of antibiotic resistance genes (ARGs) remains poorly understood. This study characterized E. coli from household water and additionally aimed to investigate the transmissibility of ARGs from drug-resistant isolates. Twenty-five E. coli isolates from thirteen household well water sources were tested for resistance to 14 antibiotics by disc diffusion and whole-genome sequenced using the Illumina platform. The ARGs and plasmid replicon types were resp

Drinking water quality assessment through multivariate statistical approach and formulated PCA-based contamination index.

Ensuring safe drinking water remains a major public health and sustainability challenge in rapidly urbanizing, groundwater-stressed and semi-arid regions. Conventional monitoring lacks statistical integration and holistic assessment. This study evaluated 167 drinking water samples from surface (PHED supply from Bisalpur Dam) and groundwater sources, assessing parameters including physicochemical, heavy metals, bacteriological indicators and antibiotic resistance profiles of 141 E. coli-like isolates. To enhance interpretability and risk stratification, an advanced multivariate statistical fram…

Work-family-self-care and day drinking among women during the COVID-19 pandemic.

BACKGROUND: The term "day drinking" is used colloquially to refer to drinking alcohol during daytime hours and has become part of the vernacular surrounding alcohol use. Anecdotal evidence is suggestive of increased day drinking among women during the COVID-19 pandemic, but empirical studies are few. This study assessed changes in the time of day women consumed alcoholic beverages during the pandemic and investigated whether their difficulty balancing work, family, and self-care was associated with the initiation of daytime drinking. METHODS: In June of 2020, 546 women age 25 and older comple…

Factors that increase risk for alcohol-induced blackouts in high-intensity drinking young adults.

BACKGROUND: Alcohol-induced blackouts are prospectively associated with negative drinking outcomes. While typically requiring heavy drinking, blackouts are not reported on all heavy drinking events or by all individuals who drink heavily. This study extends previous research by identifying the young adults most likely to experience blackouts assessed prospectively. Hypotheses focused on previously supported (female sex, White race, younger age, family history of alcohol problems, lower subjective response to alcohol, and higher tolerance) and novel predictors (possible traumatic brain injury; …

Predictors of high- and low-risk drinking after group treatment for alcohol use disorder.

BACKGROUND: Understanding the factors influencing alcohol use disorder (AUD) treatment outcomes is essential. More knowledge about patient characteristics that predict treatment outcomes can help personalise interventions, improve treatment planning and address the needs of specific subgroups. The frequency of treatment attendance may also affect drinking outcomes after treatment. Despite research efforts, uncertainty remains about how patient factors and treatment attendance influence treatment outcomes. AIMS: To examine how patient factors and treatment attendance predict high- or low-risk …

Early life stress modulates behavioral sensitivity to alcohol and promotes escalation of alcohol drinking.

Adverse childhood experiences significantly increase the risk of developing alcohol use disorder (AUD) in adulthood. We used a model of combined limited bedding/nesting and maternal separation (LBN+MS) in C57BL/6J background mice to investigate how early life stress (ELS) modulates behavioral sensitivity to alcohol, long-term alcohol drinking patterns, and the effects of alcohol on social behaviors. Our findings reveal that ELS increased sensitivity to the stimulatory locomotor effects of alcohol (1.75 g/kg) selectively in females and reduced sensitivity to the sedative effects of alcohol (4.0…

Compliance with event-contingent ecological momentary assessments and subjective follow-ups during drinking episodes.

One of the most ecologically valid methods of assessing alcohol use is event-contingent ecological momentary assessments, being self-initiated ecological momentary assessments when drinking commences. However, studies of compliance with event-contingent drinking reports, and subjective momentary follow-ups thereafter, are scant. These reports are particularly important in alcohol research, as they glean subjective assessments during acute drinking moments. This study tested patterns and predictors of compliance with event-contingent reports and subsequent subjective follow-ups during drinking …

Impact of alcohol strength on attitudes and decisions concerning special occasion drinking during pregnancy.

BACKGROUND: Special occasions are a risk factor for drinking during pregnancy. This study determined the impact of alcohol strength on attitudes around, and perceived harm of, drinking in pregnancy. If perceived harm decreases with lower strength alcohol, this may promote drinking when abstinence is recommended. METHODS: Two online 'special occasion' vignette studies randomised female participants to one of three drink strength conditions (11%, 7.5%, 0% alcohol beverage volume [ABV]). In the study (N = 1128), participants were asked to imagine themselves or someone else choosing to consume th…

Intensity Matters: Heterogeneous impact of cigarette tax reform on drinking behaviors by smoking intensity.

This paper investigates the relationship between smoking and drinking behaviors across different smoking intensity groups, using a unique policy shock that increased cigarette taxes in South Korea. Using the Korean Health Panel, we employ a difference-in-differences approach to estimate the causal effect of the cigarette price increase on alcohol consumption among heavy, average, and light smokers. We find that an increase in cigarette prices reduces drinking behavior only among heavy smokers, while cigarette consumption is reduced across all smokers. Subgroup analysis shows that the reduction…
