Exploitation of actinomycetes from coastal ecosystems of İzmir Peninsula for antimicrobial activity via strain identification, untargeted metabolomics and drug combination studies.

BACKGROUND: Antimicrobial resistance poses a major global health threat, highlighting the urgent need for novel bioactive compounds. Actinomycetes in the phylum Actinomycetota are prolific producers of antimicrobial agents; however, marine-derived strains remain underexplored, despite their ecological diversity and biosynthetic potential. In this study, we investigated the antimicrobial properties of Actinomycetes isolated from coastal sediments of İzmir Peninsula, situated on the Aegean Sea. RESULTS: Our extensive isolation efforts yielded 945 strains from 46 sediment samples. Overall, 38% of

Topoisomerase I inhibitor antibody-drug conjugates in breast cancer: the relevance of the DNA damage response to resistance, response, and combination treatment strategies.

INTRODUCTION: Antibody drug conjugates (ADCs) have revolutionized cancer treatment. ADCs that contain topoisomerase I inhibitors have been especially important in breast cancer treatment. Despite the substantial progress brought about by these ADCs, there remains a critical need to explore combination treatment strategies to overcome resistance and enhance the efficacy of these agents. Topoisomerase I inhibitors induce DNA damage and thus activate the DNA damage response (DDR). DDR elements have been examined in terms of their role in resistance and response to topoisomerase I inhibitors, and

Ginsenosides Rh2 and Rg3 in first-line combination therapy: Prospects for rapid clinical translation in drug-resistant triple-negative breast cancer.

Triple-negative breast cancer (TNBC), defined by the absence of estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor-2 (HER2), remains one of the most aggressive breast cancer subtypes, characterized by high metastatic potential, poor prognosis, and limited therapeutic options. Despite extensive clinical efforts, durable improvements in survival have been limited due to the intrinsic heterogeneity of TNBC and the rapid emergence of multidrug resistance (MDR), primarily mediated by P-glycoprotein (P-gp) overexpression. P-gp-enriched cancer stem cells fr

Enhanced combination therapy through tumor microenvironment-activated cellular uptake and ROS-sensitive drug release using a dual-sensitive nanogel.

Although the co-delivery of chemotherapeutic and photodynamic agents has been studied for years, developing a simple and efficient nanoplatform for high co-delivery efficiency remains a challenge for clinical applications. In this study, we prepared a reactive oxygen species (ROS) and pH dual-sensitive nanogel for the co-encapsulation of doxorubicin (DOX) and indocyanine green (ICG)-conjugated bovine serum albumin (BSA) via a simple inverse miniemulsion polymerization process. This was followed by modification with pegylated cell-penetrating peptides (CPPs) containing citraconic anhydride (CDM

Does Metamizole Cause Less Acute Kidney Injury than Non-Steroidal Anti-Inflammatory Drugs When Combined with Diuretics and Antihypertensives?

The concurrent use of (a) diuretics, (b) renin-angiotensin-aldosterone system inhibitors (RAASIs), and (c) non-steroidal anti-inflammatory drugs (NSAIDs) or metamizole, known as the triple whammy (TW) combination, increases the risk of acute kidney injury (AKI). This study compared TWs including metamizole versus NSAIDs regarding hospitalisation for AKI, need for renal replacement therapy (RRT), and all-cause mortality during hospitalisation. Serum creatinine (sCr) and estimated glomerular filtration rate (eGFR) changes in the first year after TW initiation were also assessed. A nested case-co
