Gestodene Accelerates Cutaneous Wound Healing via PAR1-Selective Positive Allosteric Modulation.

Protease-activated receptor 1 (PAR1), a G protein-coupled receptor, plays a central role in coordinating multiple phases of cutaneous wound healing, including hemostasis, cell proliferation, migration, and extracellular matrix remodeling. Despite its therapeutic potential, PAR1-selective positive allosteric modulators (PAMs) remain limited. Here, we characterized the wound healing efficacy of gestodene, a third-generation progestin previously identified as a selective PAM of PAR1. Gestodene exhibited no intrinsic agonist activity but selectively potentiated PAR1-activating peptide (PAR1-AP)-in

The Impact of Calcination on the Structure, Bioactivity, and Biocompatibility of Sol-Gel-Derived Glasses.

Bioactive glasses (BG) are promising materials for bone tissue engineering due to their bonding ability to biological tissues. In this study, a BG was synthesized via a modified sol-gel method that incorporated fumed silica. Two thermal treatments were applied: stabilization at 36.5 °C and calcination at 700 °C. X-ray diffraction analysis revealed the amorphous nature of BG stabilized at 36.5 °C, while calcination induced the formation of crystalline calcite. Nitrogen adsorption-desorption analysis revealed a specific surface area of 62 m2 g-1 for the stabilized BG, which decreased to 32 m2 g-

Human keratinocytes exposed to a clinical strain of Leishmania tropica can result in parasite internalization.

Cutaneous Leishmaniasis (CL) due to Leishmania tropica is a public health burden in Morocco. The increasing clinical polymorphism challenges its proper diagnosis and treatment. Although the immunopathogenesis of CL due to L. tropica has been documented, the role of keratinocytes, a critical cell type in skin immunity, has never been investigated in this pathology. Overall, keratinocytes play a crucial role in the recognition and early immune response to Leishmania parasites upon skin inoculation, influencing the local immune response by producing early cytokines that shape anti-leishmanial imm

Hyaluronic acid engineered gallic acid embedded chitosan nanoparticle as an effective delivery system for treatment of psoriasis.

Psoriasis is a deleterious auto-immune disorder which seriously harms the patients physical and mental health. CD44 are found to be over-expressed on psoriatic lesions which are highly responsible for epidermal hyperproliferation and inflammation. Gallic acid (GA), a phenolic acid natural compound has potential inhibitory impact on pro-inflammatory transcription factors. However, the penetration across skin and availability is low when applied topically, making the treatment extremely challenging. Considering such factors, we developed GA loaded chitosan nanoparticles and modified with hyaluro

Kynureninase contributes to the pathogenesis of psoriasis through pro-inflammatory effect.

Kynureninase (KYNU) is a key enzyme in the tryptophan metabolism pathway with elevated expression in psoriatic lesions relative to normal skin. However, whether KYNU contributes to the pathogenesis of psoriasis remains unknown. We sought to investigate the role of KYNU in psoriasis and its possible regulation mechanism. In the results, KYNU is upregulated in psoriatic skin samples from patients or animal models compared with normal skin control which was assayed in psoriatic patient samples, IMQ-induced psoriasis-like skin inflammation in BABL/c mice and M5-stimulated keratinocyte cell lines b

In Vitro and in Vivo Wound Healing Properties of Plasma and Serum from Crocodylus siamensis Blood.

The plasma and serum of Crocodylus siamensis have previously been reported to exhibit potent antimicrobial, antioxidant, and anti-inflammatory activities. During wound healing, these biological properties play a crucial role for supporting the formation of new tissue around the injured skin in the recovery process. Thus, this study aimed to evaluate the wound healing properties of C. siamensis plasma and serum. The collected data demonstrate that crocodile plasma and serum were able to activate in vitro proliferation and migration of HaCaT, a human keratinocyte cell line, which represents an e

Suppressor of Cytokine Signaling 3 Expression Induced by Varicella-Zoster Virus Infection Results in the Modulation of Virus Replication.

Varicella-zoster virus (VZV) is an important viral pathogen that is responsible for causing varicella (chickenpox) and herpes zoster (shingles). VZV has been shown to suppress early anti-viral innate immune responses, but the exact mechanisms are not yet well understood. Here we demonstrate that host control of VZV is impaired by the expression of suppressor of cytokine signaling (SOCS)3. We used three different cell types to characterize VZV-induced anti-viral and inflammatory responses. Infection of human fibroblasts (MRC-5) and human macrophages (THP-1) with VZV triggered upregulation of an

Performance of a novel keratinocyte-based reporter cell line to screen skin sensitizers in vitro.

In vitro tests are needed to replace animal tests to screen for the skin sensitization potential of chemicals. Skin sensitizers are electrophilic molecules and the Nrf2-electrophile-sensing pathway comprising the repressor protein Keap1, the transcription factor Nrf2 and the antioxidant response element (ARE) is emerging as a toxicity pathway induced by skin sensitizers. Previously, we screened a large set of chemicals in the reporter cell line AREc32, which contains an eight-fold repeat of the rat GSTA2 ARE-sequence upstream of a luciferase reporter gene in the human breast cancer cell line M

IL-19 promotes psoriasis progression by inhibiting keratinocyte apoptosis via the Bcl-2/Caspase-3 axis.

BACKGROUND: Keratinocyte apoptosis dysregulation is central to psoriasis pathogenesis. Interleukin-19 (IL-19) is upregulated in psoriatic lesions, but its functional role in apoptotic signaling remains unclear. This study investigated the impact of IL-19 on keratinocyte apoptosis and its underlying mechanism via the Bcl-2/Caspase-3 axis. METHODS: Clinical samples from psoriasis vulgaris patients (n = 34) and healthy controls (n = 32) were analyzed by immunohistochemistry. In vitro experiments employed M5-stimulated HaCaT cells with IL-19 knockdown/overexpression, assessed by CCK-8, flow cytom…

Lipofuscin in keratinocytes: Production, properties, and consequences of the photosensitization with visible light.

A dysfunction in the mitochondrial-lysosomal axis of cellular homeostasis is proposed to cause cells to age quicker and to accumulate lipofuscin. Typical protocols to mediate lipofuscinogenesis are based on the induction of the senescent phenotype either by allowing many consecutive cycles of cell division or by treating cells with physical/chemical agents such as ultraviolet (UV) light or hydrogen peroxide. Due to a direct connection with the physiopathology of age-related macular degeneration, lipofuscin that accumulates in retinal pigment epithelium (RPE) cells have been extensively studied

Comparative characterization of mitochondrial components across size-defined extracellular vesicle populations derived from HaCaT keratinocytes.

The transfer of mitochondrial cargo through extracellular vesicles (EVs) has been reported in several model systems and cell types, yet a detailed characterization of mitochondrial components across different EV populations remains limited. Here, we characterized the mitochondrial cargo of small EVs (sEVs) and large EVs (lEVs) released by human HaCaT keratinocytes and examined the impact of oxidative stress on their content composition. EVs were isolated from keratinocyte-conditioned medium and characterized using complementary analytical approaches. Their mitochondrial cargo was assessed usin

Modulation of the Toxicity of Three Red Tattoo Pigments by Artificial Aging.

Pigments used in tattoos represent a unique case of voluntary, long-term exposure to particles trapped in the skin. Evidence is accumulating that these compounds, and in particular red pigments, are associated with skin disorders and possibly cutaneous cancers. In the present work, we tested the hypothesis of degradation of pigments in the dermis that would lead to the release of diffusible nanoparticles or soluble degradation products. These species could then reach the epidermis and trigger physiological responses by impacting keratinocytes. Two degradation pathways of tattoo pigments were i
