Involvement of delta-opioid receptors in pentylenetetrazol kindling development and kindling-related processes in rats.

The role of delta-opioid receptors on the development of kindling induced by the convulsant pentylenetetrazol (37.5 mg kg(-1) i.p.) was investigated in rats. Besides the seizure development, the kindling induced enhancement of glutamate binding and the kindling-induced learning deficit were examined. A clear depression of kindling development by blocking of delta-opioid receptors by intracerebroventricular administration of naltrindole (10 nmol/5 microl) was found. In an acute convulsion test performed 8 days after kindling completion, animals pretreated with naltrindole during kindling induct

Effect of intracerebroventricular injection of atropine on metabolic responses during exercise in untrained rats.

To investigate the role of the central cholinergic system in the regulation of metabolism during exercise, we injected atropine (5 x 10(-7) mol) into the lateral cerebral ventricle of normal and adrenodemedullated (ADM) untrained rats submitted to exercise on a treadmill (15 m min(-1), 5% grade) until exhaustion. Concentrations of blood glucose, plasma free fatty acids (FFA), and lactate were measured before and every 10 min after the start of exercise for a period of 60 min. Adrenomedullectomy had no effect on the maximal capacity of exercise (MCE), but atropine administered intracerebroventr

Peptide N- and P/Q-type Ca2+ blockers inhibit stimulant-induced hyperactivity in mice.

omega-Conotoxin GVIA and omega-agatoxin IVA are specific peptide blockers of N- and P/Q-type calcium channel, respectively. Effects of their intracerebroventricular injection (1-3 pmol/mouse) on psychostimulant-induced hyperactivity were investigated in mice. omega-Conotoxin GVIA antagonized methylphenidate-, methamphetamine- and phencyclidine-induced hyperactivity in a dose-dependent manner. omega-Agatoxin IVA blocked methylphenidate-induced but not methamphetamine- or phencyclidine-induced hyperactivity. Neither peptides showed any effect on apomorphine-induced hyperactivity or spontaneous a

Inhibition of dopamine and choline acetyltransferase concentrations in rat CNS neurons by rat alpha 1- and alpha 2-macroglobulins.

Previous studies have implicated human alpha-2-macroglobulin (alpha2M) as a potential regulator of neuronal development and function. Rat alpha-1-macroglobulin (alpha1M) and acute-phase alpha-2-macroglobulin (alpha2M) are murine homologues of human alpha2M. In this report, we tested the effect of intracranially infused serotonin-activated rat alpha1M (5HT-alpha1M) on the concentration of dopamine (DA) in the corpus striatum in vivo and the effect of 5HT-activated rat alpha1M and alpha2M on the choline acetyltransferase (ChAT) activity upon embryonic basal forebrain neurons in culture. Intracerebroventricular secretin enhances pancreatic volume and bicarbonate response in rats.

BACKGROUND: Although secretin has been found within the brain, its central role in pancreatic exocrine function has not been previously addressed. The hypothesis that intracerebroventricular secretin enhances pancreatic volume and bicarbonate output at doses that have no effect when given intravenously was tested. METHODS: Sprague-Dawley rats had a cannula stereotactically placed into the left lateral cerebral ventricle 24 hours before study. At laparotomy the bile and pancreatic ducts were separately cannulated and excluded for tared collections and bicarbonate assay. RESULTS: Increasing do

Effect of receptor blockers on brain natriuretic peptide and C-type natriuretic peptide caused anxiolytic state in rats.

Effect of different doses of centrally administered brain natriuretic peptide (BNP) and C-type natriuretic peptide (CNP) were examined in rats with respect to anxiolytic properties in an elevated plus-maze model. BNP in doses of 100, 200 and 400 ng, and CNP in doses of 100 and 200 ng abolished the normal preference for the closed arms of the maze, and increased the percentage time spent in the open arms; this is consistent with an 'anxiolytic-like' effect. Doses of 50 and 1000 ng BNP, and of 25, 50, 400 and 1000 ng CNP produced no behavioural effects in the elevated plus-maze model. Pretreatme

SNF9007: a novel analgesic that acts simultaneously at delta 1, delta 2 and mu opioid receptors.

Intracerebroventricular administration of the synthetic cholecystokinin analog SNF9007 (Asp-Tyr-D-Phe-Gly-Trp-[NMe]-Nle-Asp-Phe-NH2) produced antinociception in the mouse hot-plate and warm water tail-flick tests. The mechanisms of its analgesic actions were assessed by administering antagonists selective for CCK (cholecystokinin octapeptide, sulfated)-A and CCK-B receptors, as well as specific antagonists for the mu opioid receptor (D-Phe-Cys-Tyr-D-Trp-Arg-Thr-Pen-Thr-NH2, 1 microgram i.c.v.), the delta-1 opioid receptor [D-Ala2-Leu5,Cys6]enkephalin, 4.57 nmol i.c.v., 24 hr pretreatment), the

Blood-borne interleukin-1 alpha is transported across the endothelial blood-spinal cord barrier of mice.

1. Previous work has shown that one mechanism by which blood-borne interleukin-1 alpha (IL-1) may be able to affect the central nervous system (CNS) is by direct transport into the brain across the blood-brain barrier (BBB). The BBB of the brain consists of endothelial (between blood and interstitial fluid) and ependymal (between blood and cerebrospinal fluid) barriers. Which of these barriers IL-1 can cross has not previously been investigated. At the spinal cord, which could be the site of action for some of the effects of IL-1 such as analgesia, the BBB consists only of the endothelial barr

[Pulsatile secretion of gonadotropins and prolactin and response to luliberin in female rats after injection of galanin into the third cerebral ventricle].

Galanin injection in a dose 200 ng/2 microliters into the third brain ventricle of ovariectomized Sprague-Dawley rats increased pulsatile release of LH and prolactin but did not influence FSH release. Gonadotropin response to LHRH was unchanged. In ovariectomized estrogen-primed rats (50 micrograms estradiol benzoate 73 h prior to the experiment) galanin stimulating effect on LH secretion was slightly manifest and prolactin and FSH release did not change at all.

Toxic effects of somatostatin in the cerebellum and vestibular nuclei: multiple sites of action.

This study demonstrates that somatostatin (SRIF), an endogenous peptide in vestibular nuclei and cerebellum, can produce both a dose-dependent death of Purkinje cells in distinct sagittal regions of cerebellar cortex and vascular infarcts centered selectively in the inferior vestibular nucleus. Alert, adult male rats were given a 5 microliters intracerebroventricular (i.c.v.) bolus of either SRIF alone (20 or 40 micrograms) or a combined dose of SRIF plus either arginine-vasopressin (AVP, 1 micrograms) or an AVP V1 antagonist, (1-(beta-mercapto-beta,beta-cyclopentamethylene propionic acid), 2-

Use of amfonelic acid to discriminate between classical and atypical neuroleptics and neurotensin: an in vivo voltammetric study.

Previous ex vivo studies have shown that the non-amphetamine stimulant amfonelic acid potentiates the increase in DOPAC induced by classical but not by atypical neuroleptics. In the present study, we have demonstrated that this neurochemical model can be used to discriminate typical from atypical neuroleptics in vivo using differential pulse voltammetry with carbon fibre electrodes. The study also compared the effect of intracerebroventricular (i.c.v.) administration of neurotensin, on extracellular striatal DOPAC following amfonelic acid, with the effects of both classical and atypical neurol

Acute neurotoxicity of sodium azide and nitric oxide.

Sodium azide is a chemical of rapidly growing commercial importance with a high acute toxicity and an unknown mechanism of action. Although it has some chemical properties and biological effects in common with cyanide, its lethality does not appear to be due to inhibition of cytochrome oxidase. Unlike cyanide it is a potent vasodilator and inhibitor of platelet aggregation presumably by virtue of its conversion to nitric oxide in vivo and in isolated preparations of blood vessels and thrombocytes. It is not clear whether the high toxicity of azide is due to nitric oxide or to the parent anion.

Effects of 5,7-dihydroxytryptamine and 6-hydroxydopamine on head-twitch response induced by serotonin, p-chloroamphetamine, and tryptamine in mice.

Head-twitch response (HTR) in mice was induced by intracerebroventricular injection of tryptamine (TRA) as well as serotonin (5-HT) and p-chloroamphetamine (PCA). Pretreatment with 5,7-dihydroxytryptamine enhanced both the 5-HT-induced and the TRA-induced HTR. The PCA-induced HTR, however, was attenuated by the drug. On the other hand, pretreatment with 6-hydroxydopamine did not alter the 5-HT response but enhanced both the PCA- and the TRA-induced response. These results suggest that 5-HT may directly stimulate the post-synaptic receptors, while the PCA response may be based on the release of

Effects of delta-9-tetrahydrocannabinol on the hypothalamic-pituitary control of luteinizing hormone and follicle-stimulating hormone secretion in adult male rats.

The main psychoactive component of marihuana, delta-9-tetrahydrocannabinol (THC) was injected into the 3rd cerebral ventricle. A single dose of THC (2 microliter of 10(-6) M) decreased serum LH temporarily but did not alter serum follicle-stimulating hormone (FSH) levels. The mediobasal hypothalamic (MBH) luteinizing hormone-releasing hormone (LHRH) content was elevated by 30 min after the injection. The elevation persisted for 1 h. Then, the LHRH content returned towards the preinjection level. In contrast, the LHRH in the organum vasculosum of the lamina terminalis did not change after a sin

Involvement of vasopressin in the cardiovascular effects of intracerebroventricularly administered alpha 1-adrenoceptor agonists in the conscious rat.

To determine the interaction between central adrenergic and vasopressinergic mechanisms in the regulation of cardiovascular function, the effects of intracerebroventricular (i.c.v.) administration of the alpha 1-agonists, methoxamine and phenylephrine, in conscious Long-Evans (LE) rats were compared with those in Brattleboro rats with hereditary hypothalamic diabetes insipidus (DI). In LE rats, both i.c.v. methoxamine and phenylephrine increased mean arterial pressure (MAP) and decreased heart rate (HR) in a dose-related manner, while they had no effect on MAP and HR in DI rats within the dose

Study of central regulation of intraocular pressure using ventriculocisternal perfusion.

The ability of hypoosmotic solution, prostaglandin E1 (PGE1) and clonidine to influence intraocular pressure (IOP) by a central mechanism was studied using the technique of ventriculocisternal perfusion in conscious rabbits. IOP remained unchanged during the perfusion of 150 mOsm artificial cerebrospinal fluid. IOP rapidly increased by 15 mmHg during the perfusion of PGE1 at the dose of 1 or 3 micrograms/min. However, when PGE1 was perfused intravenously at the dose of 1 microgram/min, a similar IOP response was observed. Furthermore, during the ventriculocisternal perfusion of PGE1 a signific

Age and strain differences in some behavioral effects of intracranial substance P.

Intracerebroventricular (ICV) injections of substance P (SP) induce a vigorous reciprocal hindlimb scratching (RHS) syndrome, accompanied by extensive grooming behavior. There is a significant (approximately 1000X) difference in responsiveness to SP, as measured by RHS and grooming, in mice as a function of genetic strain (Swiss/Webster, C57 or DBA) and age. There was considerable specificity in the ability of drugs to increase responsiveness in the least responsive type of mouse (aged DBA/2J). Responding in old DBAs was enhanced by high doses of naloxone, suggesting the involvement of opioid

Effect of intrahypothalamic injection of [D-Ala2,D-Leu5]enkephalin on feeding and temperature in the rat.

The effectiveness of the delta-opioid agonist [D-Ala2,D-Leu5]enkephalin (DADLE) in eliciting alterations in feeding and core temperature in rats was compared to morphine. When injected into the ventromedial hypothalamus this peptide caused a dose-related increase in feeding which was rapid in onset and of short duration, and a short-lived increase in temperature. Neither of these effects was blocked by the intrahypothalamic injection of naloxone. The alpha-adrenoreceptor antagonist phentolamine significantly reduced DADLE-stimulated feeding, although it did not counteract the hyperthermia. Sin
