Subchronic toxic effects of Fusarium moniliforme and fumonisin B1 in rats and mice.

Fumonisins are mycotoxins produced by the fungi Fusarium moniliforme, F. proliferatum, and other Fusarium species. Fumonisin B1, the most commonly found fumonisin, causes the fatal diseases equine leukoencephalomalacia and porcine pulmonary edema. Fumonisins are suspected human carcinogens because of the extraordinarily high incidences of esophageal cancer coincidentally found in areas of southern Africa and China where F. moniliforme-contaminated corn is consumed as a dietary staple. The subchronic (up to 90 days) effects of F. moniliforme-contaminated corn, corn cultures of this fungus, and …

Emerging mycotoxic syndromes in France: Epidemiological trends and public health implications (2015-2022).

Mushroom poisoning is a common cause of toxic exposure, and several "new mycotoxic syndromes" have been recognized since the 1990s. We conducted a retrospective multicenter study using the French Poison Control Centers cases database to describe their epidemiology in mainland France between 2015 and 2022. Cases involving species known to cause one of the seven « new » syndromes were included. Overall, 275 cases were identified, accounting for 1.7 % of the 2000 mushroom poisonings reported annually. Four syndromes were observed: neurotoxic morel syndrome (n = 159, 57.8 %), flagellate dermatitis

Development of a model for investigation of perennial ryegrass toxicosis in sheep.

AIMS To develop a clinical model of perennial ryegrass toxicosis (PRGT) based on feeding a known dose of lolitrem B and ergotamine, and to produce a consistent clinical presentation for assessment of disease pathophysiology, neurological changes and neurohistopathology. METHODS Male lambs, aged between 10-12 months, were randomly assigned to either Treatment (n=9) or Control (n=9) groups. Lambs in the Treatment group received feed containing a novel endophyte-infested perennial ryegrass seed, commencing on Day 0 of the Feeding phase with a low induction dose, then increasing after 3 days to pr

Penicillium species present in Uruguayan salami.

The surface coverage of certain dry fermented sausages such as Italian salami by some species of Penicillium provides their characteristic flavor and other beneficial properties. One of them is the protective effect by means of a uniform film of white mold against undesirable microorganisms. The aim of this work was to identify and to isolate the fungal species present in mature Italian type of salami and to evaluate if it is possible to obtain some of them as starters. In addition, the effects of temperature (14 °C and 25 °C), water activity (a w) (0.90, 0.95 and 0.995) and 2.5 % sodium chlor

The effect of cyclopiazonic acid on the development of pale, soft, and exudative pork from pigs of defined malignant hyperthermia genotype.

Malignant hyperthermia (MH) and the mycotoxin cyclopiazonic acid (CPA) are each associated with abnormal calcium homeostasis in skeletal muscle, a key underlying factor in the development of pale, soft, and exudative (PSE) pork. To determine whether the natural presence of CPA in livestock feed ingredients contributes to the varying incidence of PSE in the pork industry, various levels of CPA (.1 to 50 mg/kg of diet) were included in the diets of market weight hogs (n = 52) of defined malignant hyperthermia genotype (NN = normal, Nn = a MH carrier, and nn = MH-positive). Animals with two copie

Enhanced electrochemiluminescence of RuSi nanoparticles for ultrasensitive detection of ochratoxin A by energy transfer with CdTe quantum dots.

This paper develops a new approach to enhance the electrochemiluminescence (ECL) emission of the Ru(bpy)3(2+)-tripropyl amine (TPrA) system for ultrasensitive determination of ochratoxin A (OTA). Ru(bpy)3(2+)-doped silica nanoparticles (RuSi NPs) act as ECL materials, which are immobilized on the surface of electrode by chitosan to fabricate a solid-state ECL sensor. CdTe quantum dots (QDs) can enhance the ECL emission of the Ru(bpy)3(2+)-TPrA ECL system by energy transfer. This strategy can improve the sensitivity of the sensor. In this assay, we combine the ECL with molecular imprinting tech

Neurologic diseases in llamas and alpacas.

Llamas and alpacas are affected by a wide range of neurologic diseases, including listeriosis, polioencephalomalacia, encephalitis caused by West Nile virus and equine herpesvirus, and meningeal worm. Cervical luxations and subluxations appear to be more common in camelids than in other species. Camelids that have otitis or discospondylitis may not present with clinical signs until the underlying pathology is severe. Meningitis and brain abscesses are an important complication of failure of passive transfer of immunity in crias. The cerebrospinal fluid tap is especially useful for diagnosing m

Culture filtrate of Lasiodiplodia theobromae restricts the development of natural resistance in Brassica nigra plants.

Culture filtrate of Lasiodiplodia theobromae increased respiration rate, phenylalanine ammonia lyase activity, and levels of hydrogen peroxide, lipid peroxides and salicylic acid in B. nigra plants. Salicylic acid (SA) level increased for 1 hr of interaction and reduced later. Development of systemic acquired resistance (SAR) was found restricted in plants infected with L. theobromae due to deficiency of SA, which is a major signal for development of SAR. Exogenously supplied SA did develop resistance and plant death was delayed. It was hypothesized that deficiency of SA could be due to jasmon

Experimental equine leukoencephalomalacia, toxic hepatosis, and encephalopathy caused by corn naturally contaminated with fumonisins.

A study to evaluate the effects of dietary fumonisin B1 was conducted using 6 ponies (4 test and 2 control). A ration naturally contaminated with fumonisin B1 was fed in 3 phases: 1) 44 ppm fumonisin B1, 2) less than 1 ppm fumonisin B1, and 3) 88 ppm fumonisin B1. All ponies were monitored daily, weighed weekly, and limit fed at a rate of 0.8% body weight plus hay. Feed intake was measured daily, and a serum chemistry panel was completed once or twice weekly. Four to 7 days after initiation of the trial (Phase 1), all 4 test ponies had decreased feed consumption, and selected serum chemistry p

Evaluation of the Necessity of a Cleanup Step After Microwave-Assisted Extraction (MAE) of Aflatoxins in Pea Flour.

Aflatoxins (AFs) are among the most hazardous mycotoxins in food, yet their occurrence in legume-based products remains poorly investigated, and official standardized analytical methods validated by international regulatory bodies are still limited for these specific matrices. This study evaluates the effectiveness of microwave-assisted extraction (MAE) coupled with HPLC-FLD for the determination of AFB1, AFB2, AFG1, and AFG2 in pea flour (Pisum sativum), with a particular focus on assessing the necessity of a solid-phase extraction (SPE) cleanup step. Two analytical workflows were compared: d…

Rational Engineering of AKR13B3 from Devosia A6-243 for Enhanced Aflatoxin B1 Degradation: A Dual Mechanism of Substrate Polarization and Tunnel Remodeling.

Aflatoxin B1 (AFB1) is one of the most toxic mycotoxins, widely contaminating agricultural products and posing a serious threat to food safety and human health. Enzymatic degradation is considered a promising detoxification strategy due to its high efficiency, strong specificity, and lack of secondary pollution. AKR13B3, a member of the aldo-keto reductase family, possesses intrinsic catalytic activity for AFB1 degradation; however, its low natural activity severely limits practical application. In this study, the binding mode of the AKR13B3-NADPH complex with AFB1 was first determined using A…

Ginkgo biloba sarcotesta extract suppresses Alternaria alternata via membrane disruption and oxidative damage: a dual-mode antifungal mechanism.

Alternaria alternata is a major postharvest pathogen of apples, capable of producing multiple mycotoxins that pose significant health and safety risks. This study evaluated the antifungal activity and underlying mechanisms of Ginkgo biloba sarcotesta extract (GFSE) against A. alternata in vitro and in vivo. Among three types of tested extracts, GFSE exhibited the strongest inhibitory effect on mycelial growth, with a calculated IC50 of approximately 477 mg/L. Mechanistic assays revealed that GFSE disrupted cell membrane integrity, as evidenced by increased electrolyte leakage and enhanced effl…

Pulsed light induces antifungal activity, oxidative stress, and transcriptional reprogramming in Alternaria alternata isolated from cheese.

Alternaria alternata is a major spoilage fungus of dairy products and a potential producer of mycotoxins. This study evaluated Pulsed Light (PL) as a non-thermal strategy to control A. alternata FCS26 strain isolated from Italian Caciocavallo cheese and investigated its physiological and transcriptional stress responses. To the best of our knowledge this is the first report on the anti-fungal mechanism of PL against A. alternata. In vitro, treatments with 30 PL (11.46 J cm-2) and 120 PL (45.84 J cm-2) differentially inhibited fungal structures: spore germination was reduced by 77% and 83%, res…

T-2 toxin-induced splenic injury by disrupting the gut microbiota-spleen axis via promoting IL-6/JAK/STAT1 signaling-mediated inflammation and apoptosis and its mitigation by elemental nano-selenium.

T-2 toxin is one of the most toxic A trichothecene mycotoxins prevalent in the environment and food chain, which brings severe health hazards to both animals and humans and it can significantly damage immune function. In this study, we comprehensively explained the impact of T-2 toxin on the spleen through gut microbiota-spleen axis by integrating the transcriptome and microbiome. Our results revealed that dietary T-2 toxin ≥ 1.0 mg/kg exposure significantly inhibited the growth performance and caused spleen injury in broilers chicks, accompanied by oxidative stress and histopathological damag…
