BACKGROUND: Although prolactin (PRL) is produced predominantly by the anterior pituitary gland, recent studies have found PRL in normal brain tissues and in various neoplasms. Many of these studies have used immunohistochemical methods to detect PRL production, so the distinction between de novo synthesis and uptake by tissues with PRL receptors was not possible with these approaches. Puberty was found to be delayed in female rats who had not obtained necessary amount of prolactin with the mother's milk. These rats developed hyperprolactinemia in reproductive age associated with a decrease in the hypothalamus' dopaminergic tonus as well as a decrease of the sexual steroids level. The present study describes the effect on plasma prolactin values and tumour size of bromocriptine withdrawal in 12 patients who had been treated for macroprolactinomas for a period of 3.5-7 (mean 4.9) years. Pretreatment plasma prolactin values ranged from 12,000 to 210,000 (mean: 66,000) mU/l. We studied a young woman with surgical hypoparathyroidism who, on her usual maintenance dose of calcitriol, developed hypercalcaemia 9 d postpartum when lactation was established. Serum values of 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) values were very high (127 pg/ml). We studied prospectively eight healthy postpartum breast-feeding women for 6 months during early, middle, and late lactation. Blood for measurement of oxytocin (OT), PRL, and arginine vasopressin was drawn before and every 3 min from each women during 15 min of infant suckling for two consecutive feedings during each stage of lactation.
