Chitosan-Based Nanoparticles for Targeted Nasal Galantamine Delivery as a Promising Tool in Alzheimer's Disease Therapy.

Natural alkaloid galantamine is widely used for the treatment of mild to moderate Alzheimer's dementia. Galantamine hydrobromide (GH) is available as fast-release tablets, extended-release capsules, and oral solutions. However, its oral delivery can cause some unwanted side effects, such as gastrointestinal disturbances, nausea, and vomiting. Intranasal administration is one possible way to avoid such unwanted effects. In this work, chitosan-based nanoparticles (NPs) were studied as potential GH delivery vehicles for nasal application. The NPs were synthesized via ionic gelation and studied us

Preclinical development of the Islet Sheet.

The Islet Sheet is a thin planar bioartificial endocrine pancreas fabricated by gelling highly purified alginate and islets of Langerhans. Acellular alginate layers form a uniform immunoprotective barrier to host rejection of the encapsulated cells, with the tissue nourished by passive diffusion from adjacent host tissue. The overall thickness of the Islet Sheet, 250 microm, is chosen to maximize nutrient diffusion. In this paper we describe the early development of the Islet Sheet, including purification and fractionation of the alginates used, difficulties in maintaining sheet planarity, and…

Recent Advances in the Pharmacological Management of Gastroesophageal Reflux Disease.

The management of proton pump inhibitor-refractory GERD (rGERD) is a challenge in clinical practice. Since up to one-third of patients with typical GERD symptoms (heartburn and/or acid regurgitation) are not satisfied with proton pump inhibitor (PPI) therapy, new drug development targeting different pathophysiologies of GERD is imperative. At present, no other drugs serve as a more potent acid suppression agent than PPIs. As an add-on therapy, histamine type-2 receptor antagonists, alginates, prokinetics and transient lower esophageal sphincter relaxation inhibitors have some impact on the sub…

Cell adhesion behavior in 3D hydrogel scaffolds functionalized with D- or L-aminoacids.

Alginate hydrogels functionalized with D-, or L-penicillamine (D-, L-PEN-Alg) are used as new 3D scaffolds for cell adhesion studies. The cells recognize and show different adhesion properties in the respective 3D hydrogel scaffolds. C-6-glioma and endothelial cells show higher affinity to the D-PEN than to the L-PEN functionalized 3D alginate hydrogel scaffold. The cultivated cells are harvested from the hydrogel and are reused, for example, for cell growth experiments on 2D surfaces to prove their viability as well.

A simple and low-cost technic for immobilization of mandible during head and heck radiotherapy: a technical note.

UNLABELLED: Immobilization protocols for head and neck radiotherapy (HNR) are needed to reduce radiation dispersion in non-tumoral tissues and to reduce damage to noble structures; however, involuntary mandibular movements require additional adaptations of thermoplastic masks. PURPOSE: Our objective was to describe an easy and low-cost disposition to help thermoplastic masks immobilize the mandible during HNR. MATERIALS AND METHODS: We created Styrofoam models to stabilize the mandible which was used to make adaptation to the thermoplastic masks. The Styrofoam model was inserted into alginat

Aqueous Two-Phase Droplet-Templated Colloidosomes Composed of Self-Formed Particles via Spatial Confined Biomineralization.

A facile and green approach is developed for fabricating colloidosomes with well-controlled size and structure from the microfluidic-generated aqueous two-phase system (ATPS) emulsion droplet. Unlike other methods that rely on self-assembly of externally added colloidal particles at the emulsion interface, urease-mediated biomineralization induced by "drainage" is introduced to form CaCO3 particles at the alginate emulsion interface for preparing Ca-alg@CaCO3 colloidosomes. Two types of bioactive molecules (bovine serum albumin and catalase) can be encapsulated with high efficiency (>85%) beca

Evaluation of topical external medicine for 5-fluorouracil-induced oral mucositis in hamsters.

Oral ulcerative mucositis is a common and painful toxicity associated with chemotherapy for cancer. Current treatment for chemotherapy-induced oral mucositis is largely palliative, and no adequate treatment with conclusive evidence exists. The purpose of this study was to evaluate the potential effectiveness of the topical external medicines used in clinical settings, and the authors investigated the effects of 1% azulene ointment, 0.12% dexamethasone ointment, and polaprezinc-sodium alginate suspension on an animal model for oral mucositis induced by chemotherapy. Oral mucositis was induced i

High precision acoustofluidic synthesis of stable, biocompatible water-in-water emulsions.

Water-in-water (w/w) emulsions, comprising aqueous droplets within another continuous aqueous phase, rely on a low interfacial tension for stability. Thus far, it has been challenging to control their size and stability without the use of stabilizers. In this study, we introduce a microfluidic technique that addresses these challenges, producing stable w/w emulsions with precisely controlled size and uniformity. Results shows that using an acoustically actuated microfluidic mixer, PEG, Dextran, and alginate solutions (84.66 mPa.s viscosity difference) were homogenized rapidly, forming uniforml

Hydrogel-based co-culture and application of two types of islet cells.

This study aimed to develop a hydrogel-based co-culture system for pancreatic α- and β-cells to mimic native islet composition and achieve bidirectional blood glucose regulation. Monodisperse sodium alginate microsphere encapsulating α-TC6 and β-TC6 cells in a 2:8 ratio were fabricated using a microfluidic electrostatic spray platform. Key processing parameters (voltage, collection distance, flow rate, and alginate concentration) were optimized to precisely control microsphere diameter, yielding highly uniform spheres with excellent monodispersity. The alginate hydrogel exhibited favorable swe

Effectiveness of exosome mediated miR-126 and miR-146a delivery on cardiac tissue regeneration.

Despite advances in the treatment of acute myocardial infarction, due to the non-proliferative nature of adult cardiomyocytes, the injured myocardium is mainly replaced by fibrotic tissue, which ultimately causes heart failure. To prevent heart failure, particularly after myocardial infarction, exosome-based therapy has emerged as one of the most promising strategies to regenerate cardiac function. Exosomes can carry microRNAs in support of neovascularization, anti-inflammatory, and endogenous cardiac regeneration. This study demonstrated that animal rat models' combination treatment with micr

A new strategy based on SmRho protein loaded chitosan nanoparticles as a candidate oral vaccine against schistosomiasis.

BACKGROUND: Schistosomiasis is one of the most important neglected tropical diseases and an effective control is unlikely in the absence of improved sanitation and vaccination. A new approach of oral vaccination with alginate coated chitosan nanoparticles appears interesting because their great stability and the ease of target accessibility, besides of chitosan and alginate immunostimulatory properties. Here we propose a candidate vaccine based on the combination of chitosan-based nanoparticles containing the antigen SmRho and coated with sodium alginate. METHODS AND FINDINGS: Our results sho

Shape retaining injectable hydrogels for minimally invasive bulking.

PURPOSE: Particle migration, poor shape definition and/or rapid resorption limit the success of current urethral bulking agents. We propose that shape defining porous scaffolds that allow cell infiltration and anchoring, and may be delivered in a minimally invasive manner may provide many advantageous features. MATERIALS AND METHODS: Alginate hydrogels were prepared with varying degrees of covalent cross-linking and different pore characteristics. Dehydrated scaffolds were compressed into smaller, temporary forms, introduced into the dorsal subcutaneous space of CD-1 mice by minimally invasiv

Management of pediatric skin-graft donor sites: a randomized controlled trial of three wound care products.

Skin grafts are used to treat many types of skin defects in children, including burns, traumatic wounds, and revision of scars. The objective of this prospective randomized controlled trial was to compare the effectiveness of three dressing types for pediatric donor sites: foam, hydrofiber, and calcium alginate. Children attending a pediatric Burns & Plastics Service from October 2010 to March 2013, who required a split-skin graft, were recruited to the trial. Patients were randomly assigned to the two experimental groups, foam or hydrofiber, and to the control group, calcium alginate. Data we

Wound care-Part I: A practical approach to local wound care and dressing selection.

Dermatologists have expertise in diagnosing and managing many ulcerative conditions. However, dermatology training often lacks substantial teaching or hands-on experience in wound care. Proper wound management begins with accurately identifying the underlying cause of a wound, along with any patient-specific factors that may impede healing. These factors help guide the selection of appropriate wound care techniques and dressings. The main categories of dressings include films, foams, alginates, hydrogels, hydrocolloids, gelling fibers, and super-absorbents. For critically colonized wounds, or …

Evaluation in vivo and in vitro of the performance of interactive dressings in the management of animal soft tissue injuries.

Over the last 35 years there has been extensive research into human wounds and consequent evolutionary changes in their management. Products such as vapour-permeable films, alginates, hydro-colloids, hydrogels and foams have been developed, all of which contribute in different ways to the enhance-ment of the healing cascade. The first part of this study assessed the influence on animal wound healing of some of these contemporary wound-management products  With animal wounds the major problem is their isolation from the aggression of both the patient and the environment. The second part of this…

Construction of anisotropic cellulose roll capsules via directional freeze casting for pH-responsive probiotic encapsulation and sustained intestinal release.

Probiotics offer diverse health benefits, but their efficacy is compromised by gastrointestinal stress and burst release in conventional encapsulation systems, limiting the efficacy of probiotics in functional foods. Herein, an alginate-coated cellulose roll capsule with anisotropic porous structures and sustained release property was developed via a combined strategy of directional freeze casting and ethanol coagulation bath, which avoids the need for energy-intensive freeze-drying. The pH-responsive alginate shell and layered cellulose roll core synergistically blocked acidic penetration and
