BDNF and VEGF in the pathogenesis of stress-induced affective diseases: an insight from experimental studies.

Stress is known to play an important role in etiology, development and progression of affective diseases. Especially, chronic stress, by initiating changes in the hypothalamic-pituitary-adrenal axis (HPA), neurotransmission and the immune system, acts as a trigger for affective diseases. It has been reported that the rise in the concentration of pro-inflammatory cytokines and persistent up-regulation of glucocorticoid expression in the brain and periphery increases the excitotoxic effect on CA3 pyramidal neurons in the hippocampus resulting in dendritic atrophy, apoptosis of neurons and possib

Repeated methylphenidate administration during lactation reduces maternal behavior, induces maternal tolerance, and increases anxiety-like behavior in pups in adulthood.

Methylphenidate (MPD) is a dopamine uptake inhibitor and the most commonly prescribed drug for the treatment of attention-deficit/hyperactivity disorder in children. Several studies have shown that such stimulants as cocaine and amphetamine that are administered during gestation and lactation may disrupt maternal behavior. Also, MPD is used in lactation. Repeated MPD administration can induce either sensitization or tolerance. The aim of the present study was to investigate whether repeated MPD administration alters maternal behavior and promotes tolerance or sensitization in these females. Th

Attention deficit hyperactivity disorder in children and adolescents following traumatic brain injury.

To better characterize pediatric psychopathology after neurological insult, secondary attention deficit hyperactivity disorder (SADHD)-or ADHD that develops after traumatic brain injury (TBI)-and its clinical and neuroimaging correlates were investigated. Outcome data were available for 118 children, ages 5 through 14 at the time of hospitalization following TBI (severe TBI n = 37; mild-moderate TBI n = 57) and orthopedic injury (n = 24). Standardized psychiatric, adaptive functioning, cognitive functioning, family functioning, and family psychiatric history assessments were conducted on all p

Cognitive-Functional Intervention for Adults With ADHD (Cog-Fun-A): An Extreme Case Series Analysis.

Adults with attention-deficit/hyperactivity disorder (ADHD) often experience executive dysfunction that disrupts daily routines, occupational performance, and quality of life. Occupation-based interventions such as Cognitive-Functional Intervention for Adults (Cog-Fun-A) aim to address these challenges, yet little is known about variability in engagement and outcomes. This brief report describes three contrasting cases drawn from a Cog-Fun-A pilot study. Participants completed pre-post assessments including the Behavior Rating Inventory of Executive Function-Adult Version, Canadian Occupationa

Efficacy of mindfulness-based interventions for children and adolescents with attention deficit hyperactivity disorder: a systematic review and meta-analysis.

BACKGROUND AND OBJECTIVE: Attention deficit hyperactivity disorder (ADHD) is a common neurodevelopmental disorder in childhood. Although mindfulness-based interventions (MBIs) have been proposed as promising non-pharmacological treatments, evidence regarding their efficacy in children and adolescents with ADHD remains inconsistent. METHODS: A systematic search of PubMed, Embase, the Cochrane Library, Scopus, and Web of Science was conducted up to December 31, 2025, supplemented by manual screening of reference lists. Eligible studies were randomized controlled trials (RCTs) in which mindfulne

Modulation of prepulse inhibition and stereotypies in rodents: no evidence for antipsychotic-like properties of histamine H3-receptor inverse agonists.

RATIONALE: H(3)-receptor inverse agonists raise a great interest as innovative therapeutics in several central disorders. Whereas their procognitive properties are well established, their antipsychotic-like properties are still debated. OBJECTIVES: We further explored the effect of maximal doses (3-10 mg/kg) of ciproxifan, BF2.649, and ABT-239, three selective H(3)-receptor inverse agonists, on deficits of prepulse inhibition (PPI) induced by apomorphine, MK-801, and phencyclidine (PCP). Their effect was also investigated on stereotypies induced by apomorphine and methamphetamine. RESULTS: C

Depressive symptoms as a side effect of the sustained release form of methylphenidate in a 7-year-old boy with attention-deficit hyperactivity disorder.

INTRODUCTION: Hyperkinetic disorder or attention-deficit hyperactivity disorder (ADHD) is a clinical entity consisting of a cluster of symptoms including hyperactivity, attention disorder and impulse control disorder group. In the context of ADHD etiology we may say that genetic, clinical and imaging studies point out a disruption of the brain dopamine system, which is corroborated by the clinical effectiveness of stimulant drugs, which increase extracellular dopamine in the brain. Basically, it is a biological and not psychological disorder, which is important both for the comprehension and t

ADHD and ADHD-related neural networks in benign epilepsy with centrotemporal spikes: A systematic review.

BACKGROUND: Attention-deficit/hyperactivity disorder (ADHD) and benign epilepsy with centrotemporal spikes (BECTS or rolandic epilepsy) present with a very high level of comorbidity. We aimed to review the existing literature focusing on two aspects: the possible role of epileptic activity in the damage of ADHD-related neural networks and the clinical approach to patients presenting with both conditions. MATERIAL AND METHODS: A systematic review was performed using Sapienza Library System and PubMed. The following search terms have been considered: attention networks, ADHD, attention systems,

Loss of Zmiz1 in Mice Leads to Impaired Cortical Development and Autistic-Like Behaviors.

BACKGROUND: De novo mutations in transcriptional regulators are emerging as key risk factors contributing to the etiology of neurodevelopmental disorders. Human genetic studies have recently identified ZMIZ1 and its de novo mutations as a cause of a neurodevelopmental syndrome strongly associated with intellectual disability, autism, attention-deficit/hyperactivity disorder, microcephaly, and other developmental anomalies. However, the role of ZMIZ1 in brain development or how ZMIZ1 mutations cause neurological phenotypes is unknown. METHODS: We generated forebrain-specific Zmiz1 mutant mice

Sensitivity and Bias in Face-Emotion Labeling: Replication and Extension to Youth With Irritability and Anxiety.

OBJECTIVE: Pediatric anxiety and irritability are common, impairing, co-occurring symptoms. Biases in interpreting ambiguous face-emotions have been linked to both phenotypes. Here, we assessed whether biases represent a shared cognitive and neural profile. In addition, we attempted replication of prior age associations and group behavioral differences in face-emotion labeling. METHOD: In a cross-sectional functional magnetic resonance imaging study at a research facility at the National Institute of Mental Health, we used a variant of a drift diffusion model to decompose perceptual and cogni

Sex-Specific ADHD-like Behaviour, Altered Metabolic Functions, and Altered EEG Activity in Sialyltransferase ST3GAL5-Deficient Mice.

A deficiency in GM3-derived gangliosides, resulting from a lack of lactosylceramide-alpha-2,3-sialyltransferase (ST3GAL5), leads to severe neuropathology, including epilepsy and metabolic abnormalities. Disruption of ganglioside production by this enzyme may also have a role in the development of neuropsychiatric disorders. ST3Gal5 knock-out (St3gal5-/-) mice lack a-, b-, and c-series gangliosides, but exhibit no overt neuropathology, possibly owing to the production of compensatory 0-series glycosphingolipids. Here, we sought to investigate the possibility that St3gal5-/- mice might exhibit a

Blockade of NMDA GluN2B receptors selectively impairs behavioral flexibility but not initial discrimination learning.

RATIONALE: Behavioral flexibility is the ability to adjust behavior when environmental contingencies change and is compromised in disease states such as schizophrenia, attention deficit hyperactivity disorder, and following damage to the prefrontal cortex. METHODS: The present study investigated the contribution of N-methyl-D-aspartate GluN2B receptor subunits in the initial learning and in shifting between or within discrimination strategies (i.e., strategy set-shifting and reversal learning), using tasks conducted in operant chambers. Strategy set-shifting required rats initially to learn a

The shared genetic etiology of antisocial behavior and psychiatric disorders: Insights from pleiotropy and causality analysis.

BACKGROUND: Antisocial behavior (ASB) infringes on the rights of others and significantly disrupts social order. Studies have shown that ASB is phenotypically associated with various psychiatric disorders. However, these studies often neglected the importance of genetic foundations. METHODS: This study utilized genome-wide association studies and pleiotropy analysis to explore the genetic correlation between ASB and psychiatric disorders. Linkage disequilibrium score regression (LDSC) and high-definition likelihood (HDL) methods were employed to assess genetic correlations, and the PLACO meth
