Bisphenol A induces human uterine leiomyoma cell proliferation through membrane-associated ERα36 via nongenomic signaling pathways.

The role of ERα36 in regulating BPA's effects and its potential as a risk factor for human uterine fibroids were evaluated. BPA at low concentrations (10-6 μM - 10 μM) increased proliferation by facilitating progression of hormonally regulated, immortalized human uterine leiomyoma (ht-UtLM; fibroid) cells from G0-G1 into S phase of the cell cycle; whereas, higher concentrations (100 μM-200 μM) decreased growth. BPA upregulated ERα36 gene and protein expression, and induced increased SOS1 and Grb2 protein expression, both of which are mediators of the MAPKp44/42/ERK1/2 pathway. EGFR (pEGFR), Ra

Long-term oral exposure to bisphenol A induces glucose intolerance and insulin resistance.

Bisphenol A (BPA) is a widely used endocrine disruptor. Recent epidemiologic results have suggested an association between exposure to BPA and cardiovascular disease, type 2 diabetes, and obesity. We investigated the in vivo effects of long-term oral exposure to BPA on insulin resistance and glucose intolerance. In the present study, 4- to 6-week-old male mice on a high-fat diet (HFD) were treated with 50 μg/kg body weight per day of BPA orally for 12 weeks. Long-term oral exposure to BPA along with an HFD for 12 weeks induced glucose intolerance in growing male mice. Intraperitoneal glucose t

The plausibility of maternal toxicant exposure and nutritional status as contributing factors to the risk of autism spectrum disorders.

UNLABELLED: Recent research suggests the maternal environment may be especially important for the risk of developing autism spectrum disorders (ASD). In particular maternal infections, micronutrient deficiencies, obesity, and toxicant exposures are likely to interact with genetic risk factors to disrupt fetal brain development. OBJECTIVES: The goal of this paper is to investigate the plausibility of maternal toxicant exposure and nutritional status as causal factors in the development of ASD. METHODS: This paper reviews current research investigating the hypothesis that maternal toxicant exp

Exposure to phthalates, bisphenol A and metals in pregnancy and the association with impaired glucose tolerance and gestational diabetes mellitus: The MIREC study.

BACKGROUND: Studies from several countries report increases in rates of gestational diabetes mellitus (GDM) over recent decades. Exposure to environmental chemicals could contribute to this trend. OBJECTIVES: To determine the associations between plasticisers and metals measured in early pregnancy with impaired glucose tolerance (IGT) and GDM in a Canadian pregnancy cohort. METHODS: Women enrolled in the Maternal-Infant Research on Environmental Chemicals (MIREC) Study were included if they had a singleton delivery and did not have pre-existing diabetes. Eleven phthalate metabolites and tota

Cloning of circadian rhythmic pathway genes and perturbation of oscillation patterns in endocrine disrupting chemicals (EDCs)-exposed mangrove killifish Kryptolebias marmoratus.

To investigate the effect of endocrine disrupting chemicals (EDCs) on the circadian rhythm pathway, we cloned clock and circadian rhythmic pathway-associated genes (e.g. Per2, Cry1, Cry2, and BMAL1) in the self-fertilizing mangrove killifish Kryptolebias marmoratus. The promoter region of Km-clock had 1 aryl hydrocarbon receptor element (AhRE, GTGCGTGACA) and 8 estrogen receptor (ER) half-sites, indicating that the AhRE and ER half sites would likely be associated with regulation of clock protein activity during EDCs-induced cellular stress. The Km-clock protein domains (bHLH, PAS1, PAS2) were

Toxicokinetics of bisphenol A in pregnant DA/Han rats after single i.v. application.

Bisphenol A (BPA) is an important chemical in the production of epoxy resins and polycarbonate plastics, and basic monomers which are used for a variety of applications. Consumer exposure to BPA may be possible from migration of BPA from dental sealants or from polycarbonate or epoxy-lined food and drink containers. BPA is known to act as weak estrogen mimic in rodents, and there is a concern of adverse endocrine effects, especially from prenatal exposure to this potential 'endocrine disruptor'. To address this concern, we have studied the disposition and transplacental transfer of BPA in preg

Bisphenol S exposure before sexual maturity impairs follicular development via Cathepsin L-mediated granulosa cell death.

Bisphenol S (BPS) poses potential risks to the reproductive system, but its effects during pre-sexual maturity on follicle development and its underlying mechanisms remain unclear. This study aimed to investigate the association between pre-sexual BPS exposure and follicular dysplasia, as well as to decipher the molecular mechanisms involved. Urine samples from females aged 3-16 years were first analyzed, revealing a 95.7% detection rate of BPS, with a geometric mean concentration of 51.74 ng/L. Furthermore, the pre-sexual rat model revealed that BPS (0.05, 5, and 250 mg/kg body weight/day) re

Bisphenol S induces hepatic steatosis and insulin resistance through AMPK inhibition and endoplasmic reticulum stress.

Bisphenol S (BPS), a common substitute for bisphenol A, has emerged as a widespread environmental contaminant, yet its metabolic effects remain poorly understood. Increasing evidence links BPS exposure to metabolic dysfunction-associated steatotic liver disease (MASLD), although the underlying mechanisms are unclear. Here, we investigated the metabolic effects of chronic BPS exposure (4, 25, and 50 μg/kg/day for 12 weeks) in C57BL/6 mice and evaluated its impact on human Huh-7 hepatocytes. In vivo, exposure to 25 μg/kg/day exhibited obesogenic potential, while all doses induced insulin resista

Adiponectin Enhances Quiescence Exit of Murine Hematopoietic Stem Cells and Hematopoietic Recovery Through mTORC1 Potentiation.

Myelotoxic injury, such as chemotherapeutic agents and ionizing radiation, unlocks the vigorous power of hematopoietic stem cells (HSCs) to replenish the hematopoietic system, making quiescent HSCs enter the cell cycle. Considering that both HSC-intrinsic and -extrinsic mechanisms enforce quiescence of HSCs, the drastic change in bone marrow (BM) environment after injury, represented by massive expansion of BM adipocytes, might trigger HSC activation. BM adipocytes, the major cellular component in the ablated marrow, however, reportedly suppress proliferation of hematopoietic cells, which may

Antidiabetic thiazolidinediones inhibit collagen synthesis and hepatic stellate cell activation in vivo and in vitro.

BACKGROUND & AIMS: The ligand-dependent transcription factor peroxisome proliferator-activated receptor gamma (PPARgamma) is expressed in hepatic stellate cells (HSC), and its transcriptional activity is reduced during cell transdifferentiation in culture. PPARgamma transcriptional activation decreases platelet-derived growth factor-induced proliferation and inhibits alpha-smooth muscle actin expression in cultured HSC. The aim of our study was to evaluate whether oral administration of synthetic PPARgamma ligands, thiazolidinediones (TZD), might affect collagen deposition in animal models of

CHRONIC EXPOSURE TO BISPHENOL A INDUCED TESTICULAR DYSFUNCTION IN GERBIL: REPRO-PROTECTIVE EFFECT OF JUJUBE HONEY.

OBJECTIVE: Infertility affects an estimated 17.5% of couples worldwide, according to the last report published in 2023 by WHO. A significant cause of this global fertility decline is due to environmental endocrine-disrupting chemicals exposure, which have the ability to interfere with the endocrine system. Bisphenol A (BPA), a synthetic chemical product leads to health concerns for its endocrine-disrupting properties. Jujube honey contains antioxidants and active compounds, has various therapeutic properties. This study aims to determine testicular dysfunction and endocrine imbalance induced b

Removal of emerging pollutants by a 3-step system: Hybrid digester, vertical flow constructed wetland and photodegradation post-treatments.

The removal of emerging pollutants from municipal wastewater was studied for the first time using a three-step pilot-scale system: 1) hybrid digester (HD) as first step, 2) subsurface vertical flow constructed wetland (VF) as second step, and 3) photodegradation (PD) unit as third step or post-treatment. The HD and VF units were built and operated in series with effluent recirculation at pilot scale. For the PD post-treatment, three alternatives were studied at lab-scale, i) UVC irradiation at 254 nm (0.5 h exposure time), ii) UVA irradiation at 365 nm using a TiO2-based photocatalyst and iii)

Bisphenol A exposure disrupts the circadian clock system and impairs glycolipid metabolic function in mice liver.

Bisphenol A (BPA) is a widely existing endocrine-disrupting chemical that poses potential threats to human and animal health, including inducing metabolic disorders. Although previous studies have reported the adverse effects of BPA on liver glucose and lipid metabolism, the underlying molecular mechanisms remain incompletely elucidated. This study systematically evaluated the effects of BPA exposure on liver glucose and lipid metabolism in mouse hepatocytes (AML12) and mouse models, and detected its association with circadian clock disruption. In vitro models, BPA exposure for 24 h significan

Sublethal bisphenol A compromises BV-2 microglial immunity by disrupting TLR4 signaling through the ROS/Nrf2-Keap1/HO-1 axis.

Bisphenol A (BPA), a ubiquitous environmental contaminant, is increasingly recognized as a neurotoxicant. Epidemiological and experimental evidence links BPA exposure with neurodegenerative diseases and abnormal neurobehavior, implicating it as a risk factor for neuronal dysfunction and cognitive impairment. Given the critical role of microglia in maintaining brain immune homeostasis, we investigated whether BPA interfered with microglial immune reactivity, which may be one of the reasons why BPA causes neuropathological alterations. Using an in vitro BV-2 microglial model, sublethal BPA expos

Prenatal exposure to low doses of bisphenol A increases pituitary proliferation and gonadotroph number in female mice offspring at birth.

The pituitary gland is composed of hormone-producing cells essential for homeostasis and reproduction. Pituitary cells are sensitive to endocrine feedback in the adult and can have altered hormonal secretion from exposure to the endocrine disruptor bisphenol A (BPA). BPA is a prevalent plasticizer used in food and beverage containers, leading to widespread human exposure. Although prenatal exposure to BPA can impact reproductive function in the adult, the effects of BPA on the developing pituitary are unknown. We hypothesized that prenatal exposure to low doses of BPA impacts gonadotroph cell

Visible-light-induced photocatalytic reduction of Cr(VI) with coupled Bi2O3/TiO2 photocatalyst and the synergistic bisphenol A oxidation.

Coupled Bi2O3/TiO2 photocatalysts were fabricated by sol-gel and hydrothermal methods and characterized using various spectroscopy techniques. Photocatalytic reduction of Cr(VI) in aqueous solution, together with the synergistic effect of photodegradation of bisphenol A (BPA), was investigated using these coupled Bi2O3/TiO2 under visible-light irradiation. Coupling of Bi2O3 inhibited the phase transformation from anatase to rutile and extended absorption region to visible light. Bi ions did not enter TiO2 lattice and were more likely to bond with oxygen atoms to form Bi2O3 on the surface of Ti

Identification of novel low-dose bisphenol a targets in human foreskin fibroblast cells derived from hypospadias patients.

BACKGROUND/PURPOSE: The effect of low-dose bisphenol A (BPA) exposure on human reproductive health is still controversial. To better understand the molecular basis of the effect of BPA on human reproductive health, a genome-wide screen was performed using human foreskin fibroblast cells (hFFCs) derived from child hypospadias (HS) patients to identify novel targets of low-dose BPA exposure. METHODOLOGY/PRINCIPAL FINDINGS: Gene expression profiles of hFFCs were measured after exposure to 10 nM BPA, 0.01 nM 17β-estradiol (E2) or 1 nM 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) for 24 h. Reducing hazardous chemical exposures in the neonatal intensive care unit: a new role for nurses.

A wide range of toxic chemicals have been found in the umbilical cord blood of newborns, indicating the potential for health risks from chemical exposure that begin in utero. The neonatal intensive care unit (NICU) setting may also have potential chemical exposures that create health risks. Given the extreme vulnerability of this patient population, it is critical to minimize unnecessary hazardous chemicals. Neonatal intensive care unit nurses have an important role to play in making the NICU as safe as possible for their patients and themselves. This article will focus on the human health eff

Bisphenol-A (BPA) affects reproductive formation across generations in mice.

To understand effects of Bisphenol-A (BPA) exposure on the reproductive organ across generations, we analyzed morphology of the uterus and ovary, and the methylation pattern of HOXA10 gene of the 2(nd) generation. Pregnant mice (F0) were treated with sc injection of BPA in sesame oil at various doses of 0-1,000 mg/kg Bwt on days 12-16 of gestation. Their offspring (F1) were bred by foster mice, and the offspring (F2) from F1 mice were prepared. That is, F1 mice experienced in utero BPA exposure during the developmental period of reproductive organs, while F2 mice did not at all. Using these F2
