INTRODUCTION: In the parathyroid gland, the calcium sensing receptor responds to small changes in circulating levels of Ca(2+), and consequently stimulates or inhibits the secretion of parathyroid hormone (PTH). Thus, ligands potentiating the action of calcium (calcimimetics) lead to decreased PTH secretion and can thus be useful for the treatment of hyperparathyroidism. On the other hand, ligands which antagonize the action of calcium (calcilytics) stimulate PTH secretion, favoring bone tissue regeneration.
AREAS COVERED: This review first discusses the rapid development of calcimimetics (on…

Previous research, in which the bonding effectiveness of the 3 self-etch monomers HAEPA, EAEPA, and MAEPA was determined, showed that MAEPA was most effective. In this study, the molecular interactions of these monomers with hydroxyapatite and dentin were investigated by combining x-ray diffraction, infrared spectroscopy, and scanning electron microscopy. We tested the null hypothesis that the bonding performance of these monomers does not correlate to the formation of monomer-calcium salts and to hydrolytic stability of these monomer-calcium complexes. Monomer/ethanol/water solutions were pre…

Calcinosis cutis is the deposition of insoluble calcium in the cutaneous tissue. Calcinosis cutis can be classified as metastatic, dystrophic, idiopathic or exogenous. We report a 48-year-old white man who was dismantling a portable ice skating rink when calcium chloride solution from the pipes spilt onto his clothing. Several days later, he started to develop mildly pruritic erythematous papules, some studded with white deposits and some with umbilication over the exposed areas corresponding to the spillage of the calcium chloride solution. Histological features revealed interstitial fibrohis…

Since cloning of the T-type or Ca(V)3.n calcium channel family in 1998-1999 much progress was made in investigation of their regulation. Most effective metal Ca(V)3 channel blockers are trivalent cations from lanthanide group together with transition metals La(3+) and Y(3+). Divalent cations Zn(2+), Cu(2+) and Ni(2+) inhibit Ca(V)3.2 channels more efficiently than Ca(V)3.1 and Ca(V)3.3 channels via second high-affinity binding site including histidine H191 specific for the Ca(V)3.2 channel. Dihydropyridines and phenylalkylamines in addition to block of L-type calcium channel can inhibit Ca(V)3…

Batch treatability studies for a Cr(VI)-contaminated glacial soil from a Cr plating facility were conducted using 1X and 2X the stoichiometric ratio of calcium polysulfide (CPS). The pH of the treated soil increased from 6 to 11 upon CPS addition, but progressively returned to 8-8.5 over the course of 1 year. The 1X dosage maintained a highly reducing environment up to 21 days of monitoring with the samples exposed to atmospheric oxygen, while 2X was reducing up to 180 days of curing. The EPA regulatory method for solid Cr(VI) could not reliably predict Cr(VI) in the treated solid due to ongoi…

Ionic calcium has been known as an important intracellular second messenger for many decades. In addition, a whole series of experimental and clinical studies from the past fifteen years have provided evidence that extracellular ionic calcium itself is also a first messenger, since it is the ligand of a cell surface G-protein coupled receptor called calcium-sensing receptor. This review summarizes the current knowledge on the role of calcium-sensing receptor in the maintenance of calcium homeostasis, its functions in various tissues and some of the most important disorders characterized by def…

Effects of haloperidol on L-type CaV1.2 channel were studied. Calcium current was measured in whole cell patch-clamp using calcium as a charge carrier. Inhibition by haloperidol was investigated in CaV1.2 channel natively expressed in rat cardiac myocytes and recombinant cardiac (CaV1.2a) and vascular (CaV1.2b) splice variants of the channel expressed in HEK 293 cells. Haloperidol inhibited L-type calcium current in a concentration-dependent manner with a threshold of 1 nmol/l. 1 micromol/l haloperidol inhibited 20.6 +/- 3.6% of calcium current amplitude in cardiomyocytes, 25.4 +/- 2.6% of cur…

A 73-year-old woman underwent an uncomplicated focused parathyroidectomy for an adenoma. Immediately after extubation she developed respiratory distress and her airway became compromised. No obvious cause could be found for the stridor and no response was obtained from nebulised adrenaline (norepinephrine). An intravenous bolus of calcium provided immediate but temporary relief. Lasting relief was obtained following the administration of an intravenous calcium infusion. Serology later confirmed a small decrease in calcium concentrations but at the time of the respiratory compromise the values …

Voltage-dependent calcium channels are essential in neuronal signaling and synaptic transmission, and their functional alterations underlie numerous human disorders whether monogenic (e.g., ataxia, migraine, etc.) or autoimmune. We review recent work on Ca(V)2.1 or P/Q channelopathies, mostly using neuromuscular junction preparations, and focus specially on the functional hierarchy among the calcium channels recruited to mediate neurotransmitter release when Ca(V)2.1 channels are mutated or depleted. In either case, synaptic transmission is greatly compromised; evidently, none of the reported …

High-resolution sonography is a rapidly evolving technique that is gaining an increasing success in the assessment of crystalline arthropathies. In calcium pyrophosphate dihydrate crystal deposition disease, the sonographic features of crystal deposition include hyperchoic spots within hyaline cartilage and/or fibrocartilage and soft tissue calcifications. The aim of this pictorial essay was to present the main findings evocative of crystal deposition in patients with pyrophosphate arthropathy.

The correlation between an increased production of reactive oxygen species (ROS) and an enhanced calcium entry in primed neutrophils stimulated with fMLP suggests that endogenous ROS could serve as an agonist to reinforce calcium signaling by positive feedback. This work shows that exogenous H2O2 produced a rapid influx of Mn2+ and an increase of intracellular calcium. The H2O2 was insufficient to produce significant changes in the absence of extracellular calcium but addition of Ca2+ to H2O2-treated cells suspended in a free Ca2+/EGTA buffer resulted in a great increase in [Ca2+]i reflecting …

INTRODUCTION: Several studies have noted that, despite beneficial correction of abnormalities of mineral metabolism after successful renal transplantation, renal functional recovery is incomplete. Also, persistence of hyperparathyroidism and metabolic acidosis among patients with chronic impairment of graft function together with the use of loop diuretics and immunosuppressive drugs with adverse effects may alter mineral metabolism. We determined calcium and phosphorus levels in recipients.
METHODS: This cross-sectional study enrolled 398 recipients in 2 medical centers in Iran from 1988 to 2…

In this study we tested the hypothesis that calcium due to atherosclerosis in the renal arteries would be significantly associated with calcium in multiple other vascular beds, independent of traditional risk factors for cardiovascular disease (CVD). Electron beam computed tomography was conducted in 1461 consecutive asymptomatic patients free of clinical CVD to determine the presence and extent of calcium in the renal arteries, coronary and non-coronary vasculature and the aortic and mitral annuli. The overall prevalence for calcium in either renal artery was 18.0%, with men having a signific…

Lercanidipine is a new highly lipophylic dihydropyrdine derivative of the third generation with equal efficacy but an improved tolerability profile. Comparative therapeutic trials have shown that it is as effective as other dihydropyridines, in particular amlodipine, beta-blockers, and angiotensin-converting enzyme inhibitors. Lercanidipine is well tolerated, with most treatment-emergent events related to vasodilation. Lercanidipine produces less reflex tachycardia and peripheral oedema. Common adverse events included headache and flushing. Because of its efficacy and favorable safety profile,

Voltage-gated calcium channels are key sources of calcium entry into the cytosol. Mutations in calcium channels have been implicated in numerous disorders such as migraine, incomplete congenital X-linked stationary night blindness, epilepsy, and ataxia, and they are important therapeutic targets for the treatment of pain, stroke, hypertension, and epilepsy. Calcium channel antagonists can be broadly classified into three groups. 1) Inorganic ions typically nonselectively block the pore of most calcium channel subtypes, and in some cases, alter gating kinetics. 2) Peptides isolated from arachni…

High dietary protein intakes are known to increase urinary calcium excretion and, if maintained, will result in sustained hypercalciuria. To date, the majority of calcium balance studies in humans have not detected an effect of dietary protein on intestinal calcium absorption or serum parathyroid hormone. Therefore, it is commonly concluded that the source of the excess urinary calcium is increased bone resorption. Recent studies from our laboratory indicate that alterations in dietary protein can, in fact, profoundly affect intestinal calcium absorption. In short-term dietary trials in health…
