Inflammation, vascularization and goblet cell differences in LSCD: Validating animal models of corneal alkali burns.

Limbal stem cell deficiency (LSCD) is one of the serious cause of visual impairment and blindness with loss of corneal clarity and vascularization. Factors such as ocular burns (acids, lime, thermal), genetic disorders or infections results in the loss of limbal stem cells leading to LSCD. Reliable animal models of LSCD are useful for understanding the pathophysiology and developing novel therapeutic approaches. The purpose of the present study was to validate small and large animal models of LSCD by immunohistochemcal, clinical and histopathological comparison with human. The animal models of

Epigenetic regulatory networks in the pathogenesis and treatment of osteoporosis subtypes.

Osteoporosis is a systemic skeletal disorder characterized by reduced bone mineral density and heightened bone fragility, encompassing primary (senile and postmenopausal) and secondary (glucocorticoid-induced and diabetic) subtypes. Maintaining skeletal health relies on the dynamic balance between osteoblast-mediated bone formation and osteoclast-mediated bone resorption. Among the multitude of factors orchestrating this bone remodeling process, epigenetics, which regulates gene expression via reversible chemical modifications without altering the primary DNA sequence, plays an essential role

Developmental Exposure to Endocrine Disruptors and Persistent Pollutants Heightens Addiction Risk via Toxicological Mechanisms.

Endocrine-disrupting chemicals (EDCs) and persistent organic pollutants (POPs) cross the placenta and accumulate during gestation and early postnatal life, periods of heightened hormonal and neurodevelopmental plasticity. Exposure to contaminants such as bisphenol A (BPA), phthalates, polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) during these critical windows can reprogram endocrine and neural circuits, resulting in persistent behavioral alterations. This review synthesizes mechanistic evidence from animal models and epidemiological studies linking developmental E

Unraveling the allosteric inhibition mechanism of TLR2 by CU-CPT22: A combined DFT and enhanced sampling MD study.

Aberrant activation of Toll-like receptors 1/2 (TLR1/2) is critically implicated in the pathogenesis of numerous inflammatory and immune-mediated disorders, positioning this receptor complex as a high-value therapeutic target. CU-CPT22, a well-established small-molecule TLR1/2 inhibitor, exhibits potent and selective inhibition of TLR1/2 signaling. However, the atomistic details governing its molecular recognition and allosteric modulation remain elusive. Herein, we present an integrated computational framework that synergistically combines enhanced-sampling molecular dynamics (MD) simulations

Discovery of pyridine amide derivatives exhibiting vasodilatory activity.

Cardiovascular disease (CVD) has long been recognized as the leading cause of global mortality. Vasodilator agents are widely used in the clinical management of cardiovascular disorders. In recent years, pyridine amide derivatives have attracted considerable research interest owing to their distinctive chemical structures and promising biological profiles. Emerging evidence suggests that certain pyridine amide derivatives can synergistically enhance nitric oxide (NO) release and activate the cyclooxygenase-1 (COX-1)-mediated prostacyclin (PGI2) signaling pathway, thereby promoting vascular rel

Rapid Lipidomics Analysis of Oxidised Phosphatidylcholines in Carbon Tetrachloride-Induced Acute Hepatic Injury in Mice and Their Regulation by Rhodiola crenulata Oligomeric Proanthocyanidins.

RATIONALE: Acute hepatic injury (AHI) is a severe liver disorder that can be aggravated by oxidative stress and inflammatory responses. Oxidative stress initiates lipid peroxidation, generating a spectrum of oxidised phosphatidylcholines (oxPCs). OxPCs are commonly identified by their positional and isomeric variations. However, their structural diversity and chemical reactivity make their identification challenging. Oligomeric proanthocyanidins (OPCs) are bioactive metabolites of Rhodiola crenulata, well-known for their potential therapeutic antioxidant and anti-inflammatory effects. However,

Targeting 5-HT7 Receptor with Biased Ligands to Alleviate Pain and Spinal Neuroinflammation.

The serotonin 5-HT7 receptor (5-HT7R), a member of the rhodopsin-like family of G protein-coupled receptors (GPCRs), is highly expressed in the central nervous system (CNS) and represents a promising target for treating CNS disorders such as sleep disturbances, migraine, neuropsychiatric conditions, and neuropathic pain. Because of its therapeutic potential, extensive efforts have focused on developing selective 5-HT7R ligands. In the last decades, biased signaling has emerged as a key concept in GPCR pharmacology as biased ligands can stabilize specific active states of the receptor and trigg

Opto-biochemical point-of-care device for last-mile sickle cell anemia diagnosis.

Sickle Cell Anemia (SCA), a genetic disorder causing abnormal hemoglobin production, affects ∼300,000 newborns every year globally. With limited therapeutic options, efficient screening is crucial to curb its hereditary transmission. Reliable laboratory-based diagnostic methods (HPLC, Electrophoresis, Mass Spectrometry, RT-PCR) lack portability and are skill-intensive, and affordable Point-of-Care (PoC) screening technologies such as Solubility Test and Lateral Flow Assay, while suitable for mass screening, are qualitative and susceptible to misclassification due to subjective interpretations.

Reduction of colitis in mice by chemically programmed supramolecular nanoassemblies of vitamin-lipid conjugates.

Inflammatory bowel disease (IBD) is a relapsing disorder characterized by uncontrolled chronic inflammation of the gastrointestinal tract, posing a significant therapeutic challenge owing to the limited efficacy and undesirable side effects of current therapeutic options. A key pathological hallmark of IBD is the excessive production of reactive oxygen species (ROS). Hence, therapeutic strategies aimed at reducing ROS levels are promising for relieving these inflammatory conditions. Vitamin C-a natural nutrient for the human body-is well known for its potent antioxidant effects. However, the c

A Non-Pharmacological Paradigm Captures the Complexity in the Mechanism of Action of Poliprotect Against Gastroesophageal Reflux Disease and Dyspepsia.

When the protective mechanisms of the gastroesophageal mucosa are overwhelmed by injurious factors, the structural and functional mucosal integrity is compromised, resulting in a wide spectrum of disorders. Poliprotect has recently been shown to be non-inferior to standard-dose omeprazole for the treatment of endoscopy-negative patients with heartburn and/or epigastric pain or burning. Here, we provide preclinical data describing the mechanism of action of the Poliprotect formulation, a 100% natural, biodegradable, and environmental friendly medical device according to EU 2017/745 and containi

Generation and phenotypic characterization of a sigma-1 receptor knockout rat.

The sigma-1 receptor (σ1R) is a chaperone involved in multiple physiological and pathological processes, including pain modulation, neuroprotection, and neurodegenerative diseases. Despite its functional significance, its precise roles remain unclear due to the lack of suitable models for detailed mechanistic studies. In this work, we describe the generation and phenotypic characterization of a novel σ1R knockout (σ1R KO) rat model. Using CRISPR/Cas9 technology, we introduced a specific 218-base-pair deletion into the σ1R gene, resulting in a complete loss of receptor expression, as confirmed
