Acquired rod-body myopathy associated with human immunodeficiency virus infection.

A 35 year old homosexual man showed clinical features of myopathy, with progressive muscular weakness of proximal muscles. EMG demonstrated a myopathic pattern; serum CPK was mildly elevated and CSF examination revealed antibodies to HIV and a blood-brain barrier damage. An open biopsy of the quadriceps femoris muscle showed myopathic changes with inflammatory features including a marked variation in fiber size, necrotic fibers and phagocytosis, a profusion of internal nuclei. Fiber type analysis with myosin ATPase reaction revealed that myopathic changes involved both fiber types. Changes in

Ubiquitin is associated with abnormal cytoplasmic filaments characteristic of neurodegenerative diseases.

Several degenerative diseases of the nervous system are characterized by the presence of neuronal inclusions. Most of these inclusions are made of abnormal filaments and share epitopes with cytoskeletal proteins. One of these inclusions, the neurofibrillary tangle of Alzheimer disease, has recently been shown to contain ubiquitin, a regulatory protein thought to play a role in the degradation of abnormal proteins. We carried out light and electron microscopic immunocytochemistry with several polyclonal and monoclonal antibodies to investigate the presence of ubiquitin in neuronal inclusions of

Studies on the mechanisms of neurulation in the chick: morphometric analysis of the relationship between regional variations in cell shape and sites of motive force generation.

Microfilaments, which are organized into bundles in the apical ends of neuroepithelial cells, are generally thought to play a major role in generating the driving forces for neural tube closure. Because of their proximity to the luminal surface, the contractile activity of these microfilament bundles results in conspicuous changes in the overall shape of neuroepithelial cells, most notably apical constriction and apical surface folding. In the present study, we have used morphometric methods and computer-assisted image analysis to reveal the distribution of microfilament-mediated forces in the

Pick's disease: an immunocytochemical study of neuronal changes. Monoclonal antibodies show that Pick bodies share antigenic determinants with neurofibrillary tangles and neurofilaments.

We used rabbit antisera to the 210,000; 155,000; and 70,000 mol. wt. neurofilament - polypeptides and monoclonal antibodies (BF 10; RT97) known to react with human neurofilaments in an immunohistochemical study of neuronal changes in Pick's disease and in senile dementia of the Alzheimer type. Pick bodies as well as neurofibrillary tangles and neurites showed strong reactivity with the monoclonal antibodies but remained unlabeled when treated with the rabbit polyclonal antisera. Our results indicate that the stained material in Pick bodies share antigenic determinants with neurofibrillary tang

Metastatic malignant melanoma. A case study with unusual ultrastructure.

The unusual ultrastructure of a metastatic malignant melanoma is discussed. Light microscopy revealed most tumor cells to be nonpigmented, multinucleated giant cells. Electron microscopy demonstrated bizarre, cytoplasmic filaments void of melanin deposition. The internal structure of these filaments exhibited distinct periodicity similar to that seen in normal melanosomes.

Dual Roles of p53-Related Protein Kinase in Cytoskeletal Remodeling and Translational Control.

P53-related protein kinase (PRPK; TP53RK) is an evolutionarily conserved atypical kinase whose biology reveals an unexpected integration of translational control, cytoskeletal regulation, and cell-type-specific stress responses. As a core component of the TCTC complex (Threonyl-Carbamoyl Transferase Complex), PRPK participates in the universally conserved synthesis of the tRNA modification t6A, an essential process for translational fidelity, proteostasis, and cellular viability. Structural and biochemical studies from yeast to humans highlight a deeply conserved ATPase-driven mechanism in whi

Irregular curvature at focal adhesions modulates Piezo1 activity and low-frequency ultrasound-induced apoptosis in cancer cells.

Low-frequency, low intensity ultrasound (LIUS) has emerged as a promising physical modality capable of inducing selective apoptosis of cancer cells, while sparing healthy epithelial cells and fibroblasts. Hitherto, the mechanism underlying this selectivity has been unclear, but we now propose and develop a theoretical framework linking the distinct mechanical behaviours of cancer versus healthy cells to their differential responses to LIUS. We point out that cancer cells exhibit inhomogeneous ventral stress-fiber networks, which can produce irregular focal adhesion geometry and inward membrane

Changes in nuclear and actin mechanics from G1 to G2 affect nuclear integrity.

The structural integrity of the nucleus is dependent on nuclear mechanical elements of chromatin and lamins resisting antagonistic actin cytoskeleton forces. Force imbalance results in nuclear blebbing, rupture and cellular dysfunction found in many human diseases. Here, we used the fluorescent ubiquitin cell cycle indicator (FUCCI) cells to determine how cell cycle changes affect the nucleus and actin force balance. Whereas nuclear blebs were present equally throughout interphase, nuclear blebs formed predominantly in G1 and then persisted into G2. Actin-based nuclear confinement and focal ad…

Nuclear mechanobiology in confined cell migration.

Nuclear deformation is a central challenge for migration of cells through confined spaces in the tissue interstitium. In this paper, we review studies on the mechanical roles of the nucleus in confined cell migration. We focus on mechanical force generation by the cytoskeleton on the nuclear surface, the properties of sub-nuclear structures in the process, and functional responses of the nucleus in response to mechanical forces, all in the context of confined cell migration. An emerging theme is that the nucleus acts not only as a barrier for confined migration, but also as a mechanoresponsive…

Emerging Roles of Cytoneme-Mediated Signaling in Cancer.

Intercellular communication across cancer cells and the tumor microenvironment (TME) is essential for tumor growth, invasion, metastasis, and therapeutic resistance. Traditionally, these interactions have been viewed through the lens of diffusible signaling molecules and extracellular vesicles. However, growing evidence supports an additional paradigm in which specialized cytoskeleton-based membrane extensions, like tunneling nanotubes (TNTs), tumor microtubes (TMs), and cytonemes, mediate direct, contact-dependent communication between cells. This review examines the emerging roles of these c…

Cytoskeletal Remodeling by Oncoviruses: A Key Factor in Tumor Invasion and Metastasisa.

Tumor invasion and metastasis, the leading causes of cancer-related mortality, are critically driven by cytoskeletal remodeling, a process extensively hijacked by oncoviruses to promote malignant progression. This review comprehensively examines how oncoviruses-including Epstein-Barr virus (EBV), Kaposi's sarcoma herpesvirus (KSHV), hepatitis B virus (HBV), hepatitis C virus (HCV), human papillomavirus (HPV), and others-remodel key cytoskeletal components such as actin microfilaments, intermediate filaments (IFs), and microtubules (MTs). These virus-induced alterations facilitate epithelial-me

Pulling Forces Dampen Torsional Fluctuations of Actin Filaments and Reduce Cooperative Cofilin Binding.

A variety of potential biological roles of mechanical forces have been proposed in the field of cell biology. In particular, mechanical forces alter the mechanical conditions within cells and their environment, exerting a strong effect on the reorganization of the actin cytoskeleton. Single-molecule imaging studies have provided evidence that an actin filament may act as a mechanosensor. An increase in the tension within actin filaments causes changes in their conformation and affinity to their regulatory proteins. However, our current understanding of the molecular mechanisms of the tension s…

Inhibitory CCK+ basket synapse defects in mouse models of dystroglycanopathy.

Dystroglycan (Dag1) is a transmembrane glycoprotein that links the extracellular matrix to the actin cytoskeleton. Mutations in Dag1 or the genes required for its glycosylation result in dystroglycanopathy, a type of congenital muscular dystrophy characterized by a wide range of phenotypes including muscle weakness, brain defects, and cognitive impairment. We investigated interneuron (IN) development, synaptic function, and associated seizure susceptibility in multiple mouse models that reflect the wide phenotypic range of dystroglycanopathy neuropathology. Mice that model severe dystroglycano…

The completing of the second meiotic division by MII mouse oocytes correlates with the positioning of F-actin and mitochondria in the ooplasm.

Actin filaments play an essential role in the process of oocyte maturation and completion of meiosis. However, whether the localization of F … the pronuclei. Changes in colocalization of F-actin and mitochondria likely resulted from disturbed cytoskeleton architecture. The perturbations in the amount of F-actin and its distribution largely coincide with mitochondrial redistribution. Based on these data, we suggest actin microfilament's participation in redistribution of mitochondria during MII oocyte …

Current landscape of monogenic autoinflammatory actinopathies: A literature review.

Autoinflammatory diseases (AID) are conditions leading to a hyperactivation of innate immunity without any underlying infection, and may be poly- (e.g. Still's disease) or monogenic. The number of monogenic AID is continuously expanding, with the discovery of novel pathologies and pathophysiological mechanisms, facilitated in part by easier access to pangenomic sequencing. Actinopathies with autoinflammatory manifestations represent a newly emerging subgroup of AID, associated with defects in the regulation of actin cytoskeleton dynamics. These diseases typically manifest in the neonatal perio…

Impaired tectorial membrane and ribbon synapse maturation in the cochlea of mice with congenital hypothyroidism.

Thyroid hormone deficiency can lead to abnormal auditory development of varying severity. Retardation of morphological development, including delays in degeneration of Kölliker's organ and subsequent delayed formation of the inner sulcus, along with delayed opening of the tunnel of Corti and malformation of the tectorial membrane, was consistently observed in an antithyroid drug-induced congenital hypothyroidism rodent model. Abnormal morphological development could partly explain impaired adult auditory function. However, whether the development of inner hair cell ribbon synapses is influence

Calcium-Binding Proteins in the Nervous System during Hibernation: Neuroprotective Strategies in Hypometabolic Conditions?

Calcium-binding proteins (CBPs) can influence and react to Ca2+ transients and modulate the activity of proteins involved in both maintaining homeostatic conditions and protecting cells in harsh environmental conditions. Hibernation is a strategy that evolved in vertebrate and invertebrate species to survive in cold environments; it relies on molecular, cellular, and behavioral adaptations guided by the neuroendocrine system that together ensure unmatched tolerance to hypothermia, hypometabolism, and hypoxia. Therefore, hibernation is a useful model to study molecular neuroprotective adaptatio

Tracing Actin Filament Bundles in Three-Dimensional Electron Tomography Density Maps of Hair Cell Stereocilia.

Cryo-electron tomography (cryo-ET) is a powerful method of visualizing the three-dimensional organization of supramolecular complexes, such as the cytoskeleton, in their native cell and tissue contexts. Due to its minimal electron dose and reconstruction artifacts arising from the missing wedge during data collection, cryo-ET typically results in noisy density maps that display anisotropic XY versus Z resolution. Molecular crowding further exacerbates the challenge of automatically detecting supramolecular complexes, such as the actin bundle in hair cell stereocilia. Stereocilia are pivotal to…
