Unsuccessful chimeraplast strategy for the correction of a mutation causing Gaucher disease.

A number of gene therapy approaches have been developed for the treatment of genetic diseases, most of them based on the use of viral vectors. However, in general, they have not been successful and some complications, such as immune reactions induced by adenoviral vectors or random integration of retroviral vectors, have been reported frequently. To overcome these limitations, novel strategies have recently emerged. One of them is chimeraplasty, based on the correction of single-base mutations by mismatch repair mechanisms using chimeric RNA/DNA oligonucleotides, named chimeraplasts. Several p

Exclusion of breast cancer as an integral tumor of hereditary nonpolyposis colorectal cancer.

Hereditary nonpolyposis colorectal cancer (HNPCC) is an autosomal dominant genetic predisposition syndrome that accounts for 2-7% of all colorectal cancers. Diagnosis of HNPCC is based on family history (defined by Amsterdam or Bethesda Criteria), which often includes a history of multiple synchronous or metachronous cancers. The majority of HNPCC results from germ-line mutations in the DNA mismatch repair (MMR) genes hMSH2 and hMLH1 with rare alterations in hMSH6 and hPMS2 in atypical families. Both HNPCC and sporadic MMR-deficient tumors invariably display high microsatellite instability (MS…

Survey of genetic alterations in gastrinomas.

Gastrinomas are rare gastrin-secreting endocrine tumors that usually arise in the duodenum or pancreas and, if untreated, can cause severe peptic ulcers or metastatic disease. Although most tumors are sporadic they are especially common in patients with multiple endocrine neoplasia type 1 (MEN1), and most studies of these tumors have focused on the role of the MEN1 gene. Although the gene is commonly altered in sporadic tumors, this finding is not universal, and it is highly likely that other genetic defects play a significant role. In the present study, an in-depth analysis of the DNA of eigh

Current Strategies and Emerging Paradigms of Immunotherapy in Microsatellite Stable Colorectal Cancer: A Review Article.

Immunotherapy, especially through the innovative use of immune checkpoint inhibitors (ICIs), has rapidly become an essential component in both first-line and subsequent treatment strategies for cancers that exhibit DNA mismatch repair defects or display high microsatellite instability (dMMR/MSI-H). These therapies harness the power of the immune system to target and destroy cancer cells more effectively. However, the application of ICIs in patients diagnosed with microsatellite stable (MSS) colorectal cancer presents considerable hurdles. These challenges largely arise from the inherent tumor …

Evolutionary perspectives on endometrial cancer: antagonistic pleiotropy.

The incidence of endometrial cancer is increasing worldwide, with a particularly marked rise in early-onset cases. Modern environmental and lifestyle factors, including low parity, delayed childbirth, obesity, insulin resistance, and chronic inflammation, are thought to contribute to this trend. Although TCGA-based molecular classification has revealed substantial heterogeneity-such as defects in DNA repair, constitutive activation of the PI3K-AKT-mTOR pathway, and disruption of the p53 pathway-the reasons these abnormalities preferentially accumulate in endometrial cancer remain incompletely

Lynch Syndrome: From Multidisciplinary Management to Precision Prevention.

BACKGROUND AND AIMS: Lynch syndrome (LS) is currently one of the most prevalent hereditary cancer conditions, accounting for 3% of all colorectal cancers and for up to 15% of those with DNA mismatch repair (MMR) deficiency, and it was one of the first historically identified. The understanding of the molecular carcinogenesis of LS tumors has progressed significantly in recent years. We aim to review the most recent advances in LS research and explore genotype-based approaches in surveillance, personalized cancer prevention, and treatment strategies. METHODS: PubMed was searched to identify re…

Screening of candidate genes for primary open angle glaucoma.

PURPOSE: Primary open-angle glaucoma (POAG) is one of the leading causes of irreversible blindness in the world. To make progress in understanding POAG, it is necessary to identify more POAG-causing genes. METHODS: Using haplotype analysis, we found that mutational region is located on chromosome 2 in two families. Furthermore, we screened 11 candidate genes on chromosome 2 by protein-protein interaction (PPI) analysis, including mutS homolog 6 (MSH6), mutS homolog 2 (MSH2), v-rel reticuloendotheliosis viral oncogene homolog (REL), endothelial PAS domain protein 1 (EPAS1), vaccinia related ki

Evaluation of predictive models in daily practice for the identification of patients with Lynch syndrome.

The optimal strategy for identifying patients with Lynch syndrome among patients with newly diagnosed colorectal cancer (CRC) is still debated. Several predictive models (e.g., MMRpredict, PREMM1,2 and MMRpro) combining personal and familial data have recently been developed to quantify the risk that a given patient with CRC carries a Lynch syndrome-causing mutation. Their clinical applicability to patients with CRC from the general population requires evaluation. We studied a consecutive series of 214 patients with newly diagnosed CRC characterized for tumor microsatellite instability (MSI),

Antibacterial and antibiofilm activities of Zn2+ against Staphylococcus aureus and Streptococcus mutans.

STATEMENT OF PROBLEM: Staphylococcus aureus (S aureus) and Streptococcus mutans (S mutans) are pathogenic bacteria associated with denture-related stomatitis. Zinc (Zn), a widely used metallic antibacterial agent, can induce the production of reactive oxygen species (ROS) in cells, thereby inhibiting microbial growth and proliferation; studies, however, are lacking. PURPOSE: The purpose of this in vitro study was to determine the antibacterial and antibiofilm function of Zn2+ against S aureus and S mutans and assess gene expression changes to clarify its inhibitory mechanisms. Microsatellite status and its correlation with clinicopathological features in gastric carcinoma: insights from a retrospective study in Northern Pretoria.

INTRODUCTION/BACKGROUND: Gastric carcinomas (GC) are heterogeneous malignancies characterised by distinct histological and molecular subtypes. The microsatellite instability (MSI) molecular subtype, resulting from deficient DNA mismatch repair (dMMR), accounts for approximately 22% of global GC cases. Empirical evidence indicates differences in clinicopathological features, demographics, and treatment response in MSI GC compared to microsatellite stable (MSS) GC. MSI status has emerged as a potential biomarker for advanced GC, and this study aimed to determine the MSI prevalence of histopathol…

Msh3 and Pms1 Set Neuronal CAG-repeat Migration Rate to Drive Selective Striatal and Cortical Pathogenesis in HD Mice.

UNLABELLED: Modifiers of Huntington's disease (HD) include mismatch repair (MMR) genes; however, their underlying disease-altering mechanisms remain unresolved. Knockout (KO) alleles for 9 HD GWAS modifiers/MMR genes were crossed to the Q140 Huntingtin (mHtt) knock-in mice to probe such mechanisms. Four KO mice strongly ( Msh3 and Pms1 ) or moderately ( Msh2 and Mlh1 ) rescue a triad of adult-onset, striatal medium-spiny-neuron (MSN)-selective phenotypes: somatic Htt DNA CAG-repeat expansion, transcriptionopathy, and mHtt protein aggregation. Comparatively, Q140 cortex also exhibits an analogo

Primary Vulvar and Vaginal Adenocarcinomas of Intestinal Type Are Closer To Colorectal Adenocarcinomas Than To Carcinomas of Müllerian Origin.

Primary vulvar and vaginal adenocarcinomas of intestinal type (VVAIts) are very rare tumors, displaying morphologic and immunohistochemical overlap with colorectal adenocarcinomas. However, their immunoprofile and genomics are poorly studied, and their origin is still debated. Here, we studied a series of 8 VVAIts (4 vulvar and 4 vaginal) using a large panel of immunohistochemistry and DNA and RNA sequencing with clustering analyses. All tumors shared a similar morphology with colorectal adenocarcinomas and diffuse CK20 and CDX2 expression. SATB2 diffuse positivity was observed in 62.5% of tum
