Heat stress (HS), exacerbated by global climate change, severely limits crop productivity and quality. Although γ-aminobutyric acid (GABA) can alleviate heat stress, the mechanisms by which it maintains crop quality remain largely unclear. In this study, using pea (Pisum sativum L.), we demonstrated that exogenous GABA pretreatment significantly alleviated HS-induced inhibition of photosynthesis and growth, while effectively improving nutritional and edible quality by attenuating HS-mediated cellular damage. GABA enhanced plant thermotolerance by suppressing the heat-induced reactive oxygen sp

Glioma's metabolic reprogramming fortifies antioxidant defenses and fosters an immunosuppressive microenvironment, thus resulting in robust resistance to ferroptosis-immunotherapy. In this study, a biomimetic sequentially gated amorphous MOF-based nanotuner (ZB@HM) was constructed, which could cascade depletion of metabolic substrates of ferroptosis defense and redistribution of methionine for reprogramming metabolism-ferroptosis-immunity crosstalk. Specifically, we engineered a methionine-inhibitor-loaded amorphous ZIF-82, followed by coating with E. coli-glioma hybrid membrane to improve its

INTRODUCTION: Temperature plays a crucial role in embryonic development, particularly in oviparous species that experience natural fluctuations during incubation. Unlike viviparous embryos developing under stable maternal conditions, chick embryos are exposed to external temperature variability that can influence cellular regulatory processes. Notch signaling, a conserved pathway essential for maintaining the neural progenitor cell (NPC) pool, is regulated by endosomal recycling; however, the mechanism by which temperature modulates this process across species remains poorly understood.

Iron is indispensable for core physiological processes, and intracellular iron homeostasis is tightly governed by iron regulatory protein 1 (IRP1). Hydrogen sulfide (H2S), an endogenous gasotransmitter with well-established antioxidant and cytoprotective activities, remains poorly characterized for its regulatory roles in IRP1-dependent iron metabolism and fibroblast activation. Herein, we employed primary mouse embryonic fibroblasts (MEFs) to interrogate the functional crosstalk between NaHS (a H2S donor) and IRP1 governing iron balance and profibrotic responses. Our results reveal that IRP1

Natural sources of vitamin D (VD), have been proposed to represent an alternative to synthetic vitamin D in nutritional interventions, also holding therapeutic potential in non-alcoholic fatty liver disease (NAFLD). In this study lipidomics was used to comparatively investigate the molecular mechanisms behind the therapeutic effects of a natural VD formulation consisting of a Shitake mushroom extracts (NVD) and a synthetic cholecalciferol formulation (SVD) in HepaRG human hepatocytes exposed to free fatty acid (FFA)-induced lipotoxicity. The results demonstrate that the two VD formulations pre

Triacetic acid lactone (TAL) is a promising platform chemical to produce valuable compounds. The development of engineered microbial hosts to efficiently produce TAL from lipid-containing waste streams could be a cost-effective, sustainable and environmentally friendly approach to meet the industrial demand. In this study, we engineered the yeast Candida viswanathii, possessing robust fatty acid conversion capabilities, to develop an alternative route for TAL production from fatty acids that aims to maximize conversion of the acetyl-CoA pool generated by β-oxidation in the peroxisome. To do so

Lycopene, a potent carotenoid with high antioxidant capacity and extensive applications, holds significant potential for sustainable production via microbial engineering, particularly with the rising interest in methanol as an ideal non-grain feedstock for a carbon-negative economy. In this study, Komagataella phaffii was systematically engineered to enhance lycopene production using glucose and renewable methanol as alternative carbon sources. Firstly, we demonstrated that the cytoplasmic FPP could penetrate into the peroxisome, and thus achieved the dual-localized lycopene synthesis. Subsequ

Cholestatic liver diseases, including primary biliary cholangitis (PBC) and primary sclerosing cholangitis (PSC), are characterized by disrupted bile acid (BA) homeostasis and subsequent liver injury. Emerging evidence indicates that circadian rhythms significantly influence liver metabolism and the pathogenesis of liver diseases. CD36 has been identified as a regulator of the hepatic circadian clock and metabolic processes; however, the specific mechanisms by which CD36 links circadian rhythms to cholestatic liver disease remain unclear. In this study, we employed bile duct ligation (BDL) mic

Mammalian hematopoietic stem cells (HSCs) emerge from the hemogenic endothelium in the major embryonic arteries. HSCs undergo a complex journey first migrating to the fetal liver (FL) and from there to the fetal bone marrow (FBM), where they mostly remain during adult life. In this process, a pool of adult HSCs is produced, which sustains lifelong hematopoiesis. Multiple cellular components support HSC maturation and expansion and modulate their response to environmental and developmental cues. While the adult HSC niche has been extensively studied over the last two decades, the niches present

MicroRNAs (miRNAs) are short non-coding RNAs, that regulate gene-expression at post-transcriptional level. Unlike other RNA species, blood miRNAs circulate in a highly stable form, either within extracellular vesicles or bound to proteins. In recent years, circulatory miRNA profiles have been proposed as potential biomarkers for multitude of pathologies, including essential hypertension. However, the evidence of miRNA biomarker potential is limited, mainly due to the scarcity of profiling studies associating miRNA levels with hypertension. Furthermore, most of these studies have been performed

In lymphopenic environments, secondary lymphoid organs regulate the size of B and T cell compartments by supporting the homeostatic proliferation of mature lymphocytes. The molecular mechanisms underlying these responses and their functional consequences remain incompletely understood. To evaluate homeostasis of the mature B cell pool during lymphopenia, we turned to an adoptive transfer model of purified follicular B cells into Rag2-/- mouse recipients. Highly purified follicular B cells transdifferentiated into marginal zone-like B cells when transferred into Rag2-/- lymphopenic hosts but no

Fanconi anemia complementation group B (FANCB) protein is a major component of the Fanconi anemia (FA) core complex and plays an important role in hematopoiesis and germ cell development. Deletion of Fancb gene causes the defect of primordial germ cell (PGC) development and infertility in male mice. However, it remains unknown whether Fancb is required for female germ cell development. In this study, we found that the fertility of Fancb knockout male mice in C57/ICR mixed backgrounds was not affected. Female Fancb-/- mice were obtained by crossing Fancb+/- females with Fancb-/Y males. The numb

Small heterodimer partner (Shp) regulates several metabolic processes, including bile acid levels, but lacks the conserved DNA binding domain. Phylogenetic analysis revealed conserved genetic evolution of SHP, FXR, CYP7A1, and CYP8B1. Shp, although primarily studied as a downstream target of Farnesoid X Receptor (Fxr), has a distinct hepatic role that is poorly understood. Here, we report that liver-specific Shp knockout (LShpKO) mice have impaired negative feedback of Cyp7a1 and Cyp8b1 on bile acid challenge and demonstrate that a single copy of the Shp gene is sufficient to maintain this res

Obesity is a worldwide epidemic that predisposes individuals to many age-associated diseases, but its exact effects on organ dysfunction are largely unknown1. Hair follicles-mini-epithelial organs that grow hair-are miniaturized by ageing to cause hair loss through the depletion of hair follicle stem cells (HFSCs)2. Here we report that obesity-induced stress, such as that induced by a high-fat diet (HFD), targets HFSCs to accelerate hair thinning. Chronological gene expression analysis revealed that HFD feeding for four consecutive days in young mice directed activated HFSCs towards epidermal

UNLABELLED: Premalignant clonal expansion of human T-cell leukemia virus type-1 (HTLV-1)-infected cells occurs before viral carcinogenesis. Here we characterize premalignant cells and the multicellular ecosystem in HTLV-1 infection with and without adult T-cell leukemia/lymphoma (ATL) by genome sequencing and single-cell simultaneous transcriptome and T/B-cell receptor sequencing with surface protein analysis. We distinguish malignant phenotypes caused by HTLV-1 infection and leukemogenesis and dissect clonal evolution of malignant cells with different clinical behavior. Within HTLV-1-infected

BACKGROUND: Studies have recently shown that RHOA mutations play a crucial role in angioimmunoblastic T-cell lymphoma (AITL) pathogenesis. We aimed to pool data from these studies to provide a comparison of clinicopathological features between the RHOA mutant and RHOA wild-type groups in the AITL population.
METHODS: We searched PubMed and Web of Science for the keywords "RHOA AND lymphoma" and selected only studies reporting the clinical significance of RHOA mutations in AITL. We calculated the odds ratios (OR) or the mean difference with 95% CI using a random effect model.
RESULTS: Our poo

Several outbreaks of mosquito-borne diseases have taken place in Europe in recent years. In Spain, both active and passive surveillance have demonstrated that dengue and West Nile viruses are currently circulating, and seven autochthonous dengue cases have been reported in the last 2 yr. The effectiveness of vector control programs largely depends on the accuracy of the taxonomic identification of the species. However, in Spain, identification almost completely relies on the use of morphological keys to characterize the mosquito fauna. This study investigates the congruence between molecular a

Fragile X and fragile X-associated tremor-ataxia syndrome (FXTAS) are caused by mutations of the FMR1 gene. The mutations causing FXTAS can expand in a generation to a "full mutation" causing fragile X syndrome. The mutations causing FXTAS and the phenotype, fragile X-associated premature ovarian insufficiency (FXPOI), are referred to as the FMR1 premutation (PM). The objective of this paper was to formulate a theory to explain the Mechanism for FXPOI.Recent research on fragile X syndrome and FXTAS has led to sophisticated theories about the mechanisms underlying these diseases. It has been pr
