Pattern Recognition Receptor Ligands as an Emerging Therapeutic Agent for Latent HIV-1 Infection.

Toll-like receptors (TLRs) were first identified as molecular sensors that transduce signals from specific structural patterns derived from pathogens; their underlying molecular mechanisms of recognition and signal transduction are well-understood. To date, more than 20 pattern-recognition receptors (PRRs) have been reported in humans, some of which are membrane-bound, similar to TLRs, whereas others are cytosolic, including retinoic acid-inducible gene-I (RIG-I)-like receptors (RLRs), nucleotide-binding oligomerization domain (NOD)-like receptors (NLRs), and stimulator of interferon genes (ST

Hypereosinophilia and acute bilateral facial palsy: an unusual presentation of a common disease.

A 60-year-old man presented with an acute, pruritic, erythematous rash associated with marked hypereosinophilia (2.34×10(9)/l (0.04-0.40)). There was eosinophilic infiltration on hepatic, bone marrow and lymph node biopsies, with multiple lung nodules and mild splenomegaly. However, extensive investigation excluded parasitic or bacterial causes, specific allergens or the Fip1L1 mutation seen in myeloproliferative hypereosinophilia. Six months into the illness, he developed an acute, left, complete lower motor neurone facial palsy over hours, and an acute right lower motor neurone facial palsy

The Low-Cost Compound Lignosulfonic Acid (LA) Exhibits Broad-Spectrum Anti-HIV and Anti-HSV Activity and Has Potential for Microbicidal Applications.

OBJECTIVES: Lignosulfonic acid (LA), a low-cost lignin-derived polyanionic macromolecule, was extensively studied for its anti-HIV and anti-HSV activity in various cellular assays, its mechanism of viral inhibition and safety profile as potential microbicide. RESULTS: LA demonstrated potent inhibitory activity of HIV replication against a wide range of R5 and X4 HIV strains and prevented the uptake of HIV by bystander CD4+ T cells from persistently infected T cells in vitro (IC50: 0.07 - 0.34 μM). LA also inhibited HSV-2 replication in vitro in different cell types (IC50: 0.42 - 1.1 μM) and i

HIV risk reduction among African-American women who inject drugs: a randomized controlled trial.

A community-based HIV intervention for African-American women who are active injection drug users (IDUs) was evaluated. Seventy-one women (aged 20-54 years) were randomly assigned to one of two enhanced gender- and culturally specific intervention conditions or to the NIDA standard condition. Substantial decreases (p<.001) were found in the frequency of drug use and the frequency of drug injection as well as in the sharing of injection works or water and the number of injections. Trading sex for drugs or money, having sex while high, as well as other sexual risk behaviors were also reduced sig

Regular Testing of HIV and Sexually Transmitted Infections With Self-Collected Samples From Multiple Anatomic Sites to Monitor Sexual Health in Men Who Have Sex With Men: Longitudinal Study.

BACKGROUND: Regular HIV and sexually transmitted infection (STI) testing for men who have sex with men (MSM) is an important means of infection prevention, the adoption of which remains suboptimal in the community. OBJECTIVE: On the hypothesis that engagement plays an important role in sexual health monitoring, this study aimed to pilot-test internet-based HIV and STI testing with self-sampling to enhance engagement of MSM with regular testing. METHODS: This 1-year cohort study was conducted on HIV-negative MSM aged 18 years or older. A designated website was set up to enable participants to

Cutting-edge Bioinformatics strategies for synthesizing Cyclotriazadisulfonamide (CADA) analogs in next-Generation HIV therapies.

Cyclotriazadisulfonamide (CADA) is a macrocyclic compound known for its unique mechanism in inhibiting HIV infection by downregulating the CD4 T-cell receptor, a crucial entry point for the virus. Unlike other antiretrovirals, CADA exhibits activity against a wide range of HIV strains, as all HIV variants require CD4 binding for infection. Furthermore, CADA has shown a synergistic effect with clinically approved anti-HIV drugs, offering potential for enhanced therapeutic strategies (Vermeire & Schols, [65]). One proposed mechanism for CADA's inhibition of the CD4 receptor involves blocking the

Enablers and barriers to effective HIV self-testing in the private sector among sexually active youths in Nigeria: A qualitative study using journey map methodology.

BACKGROUND: HIV is a public health burden in Nigeria. HIV self-testing is one of the approaches to testing, which is the first of the 95:95:95 cascade of a coherent response to the epidemic. The ability to self-test HIV is influenced by various factors that can either serve as enablers or barriers. Exploring these enablers and barriers to the uptake of HIVST will help achieve optimal HIV self-testing and provide a deeper understanding of the HIVST kits users' journey. OBJECTIVE: The purpose of the study was to identify enablers and barriers to the uptake of HIV self-testing among sexually act

Efficacy of low doses of the polyethylene glycol derivative of interleukin-2 in modulating the immune response of patients with human immunodeficiency virus type 1 infection.

Interleukin-2 (IL-2) is a key cytokine in cellular immunity. Human immunodeficiency virus type 1 (HIV-1)-infected individuals lack IL-2 because of low CD4+ T lymphocyte numbers. In an attempt to enhance cellular immunity, low-dose recombinant human (rh) IL-2 at 10 micrograms or 180,000 units or its polyethylene glycol (PEG) derivative at 9 micrograms or 36,000 units was given by intracutaneous injection to 8 HIV-1-infected men for 30 days. Participants had no evidence of opportunistic infection and received concurrent zidovudine. IL-2 treatment was nontoxic and elicited a local cellular respon

Diagnostic value of miR-155, miR-29a and lncRNAs (NEAT1, GAS5 and COX2) in HIV, tuberculosis, and HIV/TB co-infection.

Tuberculosis (TB) and human immunodeficiency virus (HIV) co-infection remain leading causes of mortality, especially in source-limited settings where diagnostic challenges impede timely management. Non-coding RNAs (ncRNAs) are emerging as non-invasive biomarkers for infectious diseases due to their role in immune regulation. However, the diagnostic potential of ncRNAs in distinguishing TB monoinfection, HIV monoinfection, and TB/HIV co-infection remains unclear. In this study, we examined the expression of miR-155, miR-29a, and lncRNAs (lncRNA-COX2, lncRNA-NEAT1, lncRNA-GAS5) by qRT-PCR in per

Associations between HIV testing and multilevel stigmas among gay men and other men who have sex with men in nine urban centers across the United States.

BACKGROUND: Complex manifestation of stigma across personal, community, and structural levels and their effect on HIV outcomes are less understood than effects in isolation. Yet, multilevel approaches that jointly assesses HIV criminalization and personal sexual behavior stigma in relation to HIV testing have not been widely employed or have only focused on specific subpopulations. The current study assesses the association of three types of MSM-related sexual behavior-related stigma (family, healthcare, general social stigma) measured at both individual and site levels and the presence/absenc

Diffuse encephalitic toxoplasmosis in HIV.

This case demonstrates an atypical radiological presentation of cerebral toxoplasmosis in a 62-year-old HIV-positive patient. The diagnosis is discussed alongside MRI imaging, laboratory results and treatment. Central nervous system toxoplasmosis is typically associated with ring enhancing lesions on neuroimaging with contrast, but the radiology for this patient shows diffuse white matter changes and ependymal enhancement, which would normally suggest an alternative diagnosis.

Robotic-Assisted Total Knee Arthroplasty in HIV-Positive Patients: Precision Surgery for a Changing Epidemic.

BACKGROUND: Osteoarthritis(OA) and Human Immunodeficiency Virus(HIV) infection are two major global health burdens increasingly intersecting in orthopaedic practice. The global HIV-positive population is aging due to effective antiretroviral therapy, leading to a grwoing number of patients presenting with end-stage knee osteoarthritis requiring total knee arthroplasty(TKA). Conventional TKA, while successful in the general population, remains vulnerable to aseptic loosening and periprosthetic joint infection(PJI); complications that may be accelerated in immunocompromised hosts with altered bo

Immunomagnetic Sample Preparation Targeting Host- and Viral-Derived Antigens for HIV-1 Isolation from Limited Plasma Volumes.

Rapid and accurate HIV detection is crucial for early diagnosis and viral load (VL) monitoring in individuals receiving antiretroviral therapy (ART). To address the challenge of limited sample volume, we developed a novel immunomagnetic sample preparation method to isolate HIV-1 virions from 25 μL of plasma─about one-tenth of a small finger-prick sample. Immunomagnetic conjugates were produced by coupling antimouse IgG magnetic beads with mouse IgG antibodies targeting host- and virus-derived antigens on the HIV surface. In buffer spiked with inactivated HIV-1 grown in the HUT-78 cell line, th

Immunopathogenesis of TB-HIV Co-Infection: Mechanisms of Dual Immune Suppression and Viral-Bacterial Synergy.

Human immunodeficiency virus (HIV) and Mycobacterium tuberculosis (M. tuberculosis) co-infection represents one of the most devastating syndemics in global health, accounting for substantial morbidity and mortality worldwide. The interaction between these two pathogens is not merely additive but highly synergistic, driven by profound immune dysregulation. HIV-mediated depletion of CD4+ T cells facilitates tuberculosis (TB) progression, whereas TB-induced immune activation enhances HIV replication and accelerates disease progression. HIV preferentially infects CD4+ T cells and impairs the funct

The impact of the recent HIV non-nucleoside reverse transcriptase inhibitors and nucleoside reverse transcriptase inhibitors-based regimens on metabolic health outcomes: A narrative review.

The global prevalence of human immunodeficiency virus (HIV) has led to a significant rise in the chronic use of antiretroviral (ARV) drugs, both for HIV management and pre-exposure prophylaxis (PrEP), to meet the set Joint United Nations Programme on HIV/AIDS (UNAIDS) 95-95-95 targets. Although antiretroviral therapy (ART) has remarkably increased the life expectancy of people living with HIV (PLHIV), it has also been associated with metabolic complications, particularly in glucose and lipid metabolism. Notably, the development of type two diabetes mellitus (T2DM), accounting for 90-95 % of di

Strategies to achieve the UNAIDS 95% HIV diagnosis target in Australia: Insights from a multidisciplinary roundtable.

INTRODUCTION: In 2022, Australia met the global UNAIDS 95% targets for treatment and viral suppression. Achieving 95% of people knowing their HIV status requires implementation of a wider mix of HIV testing types and access points. METHODS: In October 2023, a community-led HIV testing roundtable meeting of 34 participants was convened to better understand current barriers and enablers to foster HIV testing equity. Delegates participated in data-informed discussions and a facilitated workshop. Outputs were collated and independently analysed to identify HIV testing barriers and potential actio

Dendritic cell-based vaccines, a promising prospect for ending AIDS and eradication of HIV worldwide: A narrative review on the latest updates.

Significant challenges are presented as a result of the ongoing HIV/AIDS pandemic, particularly given the ongoing burden of new infections and the lack of a proven treatment. Among other approaches, vaccinations based on Dendritic Cells (DCs) have become a viable tactic in the fight against HIV. These vaccines take advantage of DCs' special capacity to deliver antigens, which makes them crucial mediators between innate and adaptive immune responses. DC-based vaccines seek to offer a prolonged and effective protection against the virus by stimulating immunological responses against HIV-infected
