Risk of breast cancer in daughters of agricultural workers in Denmark.

OBJECTIVES: Agricultural workers face unique occupational hazards such as pesticide exposure, which has been associated with breast cancer. However, research considering the association between parental agricultural work and breast cancer in female offspring is lacking. Therefore, the aim of the present nested case-control study was to explore this association. METHODS: The Danish Cancer Registry was utilized to identify women diagnosed with primary breast cancer. A total of 5587 cases were included in the study, and for each case, 20 cancer-free female controls were selected, matched on year

Parental exposure to gamma radiation causes progressively altered transcriptomes linked to adverse effects in zebrafish offspring.

Ionizing radiation causes a variety of effects, including DNA damage associated to cancers. However, the effects in progeny from irradiated parents is not well documented. Using zebrafish as a model, we previously found that parental exposure to ionizing radiation is associated with effects in offspring, such as increased hatching rates, deformities, increased DNA damage and reactive oxygen species. Here, we assessed short (one month) and long term effects (one year) on gene expression in embryonic offspring (5.5 h post fertilization) from zebrafish exposed during gametogenesis to gamma radiat

A higher risk of congenital anomalies in the offspring of personnel who served aboard a Norwegian missile torpedo boat.

BACKGROUND: In the 1990s, congenital anomalies were reported among children whose fathers had served aboard a Norwegian missile torpedo boat (MTB). The Royal Norwegian Navy asked the University of Bergen to look into this problem as one part of a general health and work environment surveillance. AIMS: To estimate any increased risk of having children with congenital anomalies and having stillborn children among the offspring of workers that had served aboard the MTB and to investigate possible differences in exposure and other risk factors between these groups. METHODS: Data from a cross-sec

Maternal hyperglycemia during pregnancy is associated specifically with larger hypothalamic volumes at 12 years of age.

Maternal hyperglycemia is associated with higher risk of metabolic diseases in offspring. Despite various hypotheses, the exact mechanisms remain unclear and the neural implication is yet to be fully investigated. The hypothalamus plays a critical role in energy regulation. In utero exposure to maternal hyperglycemia might selectively affect the developing hypothalamus. To test this hypothesis, we investigated associations between in utero exposure to maternal hyperglycemia and hypothalamic volume at 10-12 years of age. We included 82 mother-child pairs from the Gen3G prospective birth cohort,

Suberoylanilide hydroxamic acid attenuates cognitive impairment in offspring caused by maternal surgery during mid-pregnancy.

Some pregnant women have to experience non-obstetric surgery during pregnancy under general anesthesia. Our previous studies showed that maternal exposure to sevoflurane, isoflurane, propofol, and ketamine causes cognitive deficits in offspring. Histone acetylation has been implicated in synaptic plasticity. Propofol is commonly used in non-obstetric procedures on pregnant women. Previous studies in our laboratory showed that maternal propofol exposure in pregnancy impairs learning and memory in offspring by disturbing histone acetylation. The present study aims to investigate whether HDAC inh…

In-utero exposure to bereavement and offspring IQ: a Danish national cohort study.

BACKGROUND: Intelligence is a life-long trait that has strong influences on lifestyle, adult morbidity and life expectancy. Hence, lower cognitive abilities are therefore of public health interest. Our primary aim was to examine if prenatal bereavement measured as exposure to death of a close family member is associated with the intelligence quotient (IQ) scores at 18-years of age of adult Danish males completing a military cognitive screening examination. METHODS: We extracted records for the Danish military screening test and found kinship links with biological parents, siblings, and matern

Newborn metabolomic perturbations associated with prenatal tobacco smoke exposure and early birth.

BACKGROUND: Exposure to tobacco smoke during pregnancy is an established risk factor for early birth including early term birth (ETB) and preterm birth (PTB). However, the underlying molecular mechanisms are minimally understood. In this study, we aimed to characterize the newborn metabolomic associations with early pregnancy maternal tobacco exposure biomarkers and early birth in 269 mother-child pairs in the Atlanta African American Maternal-Child Cohort (2016-2020). METHODS: Established tobacco exposure biomarkers, cotinine and trans-3'-hydroxycotinine (3HC), were measured in maternal urin

Impact of Maternal Aspirin Therapy on Neonatal Epigenetic Patterns.

BACKGROUND: Low-dose aspirin (LDA) is an intervention recommended to prevent the development of hypertensive disorders of pregnancy (HDP) in high-risk pregnancies. Maternal conditions such as HDP have been associated with cord blood epigenetic changes including those related to cardiovascular processes; however, it is unclear whether maternal aspirin therapy may impact neonatal epigenetics in otherwise healthy high-risk pregnancy. OBJECTIVE: This study aimed to determine if maternal LDA exposure in utero leads to altered DNA methylation in umbilical cord blood cells in term neonates compared

Prenatal and infant acetaminophen exposure, antioxidant gene polymorphisms, and childhood asthma.

BACKGROUND: Prenatal and infant acetaminophen exposure has been associated with an increased risk of childhood asthma phenotypes. Demonstration of biologically plausible interactions between these exposures and maternal and child antioxidant gene polymorphisms would strengthen causal inference. OBJECTIVE: To explore potential interactions between prenatal and infant acetaminophen exposure and antioxidant genotypes on childhood asthma. METHODS: In the Avon Longitudinal Study of Parents and Children, we typed a functional nuclear erythroid 2 p45-related factor 2 (Nrf2) polymorphism and glutath

Air pollution and very low birth weight infants: a target population?

OBJECTIVE: The goal was to examine systematically the association between maternal exposure to particulate matter of <10 microm and very low birth weight (<1500 g) delivery for evidence of an effect on duration of gestation and/or intrauterine growth restriction. METHODS: This case-control study took place between April 1, 1986, and March 30, 1988, in Georgia Health Care District 9 and included 128 mothers of very low birth weight infants, all of whom were preterm and were classified as either small for gestational age or appropriate for gestational age, and 197 mothers of term, appropriate-f…

Maternal antibody-associated fetal second-degree heart block and atrial flutter: case report and review.

Exposure to maternal anti-Ro (SS-A) and anti-La (SS-B) antibodies is a well-described risk factor for the development of fetal atrioventricular (AV) block. The role of maternal fluorinated steroids in the treatment and prevention of antibody-mediated fetal AV block is controversial. Fetal atrial flutter has rarely been described in association with maternal antibodies. This report describes a case of fetal exposure to maternal anti-Ro antibodies with associated second-degree AV block and atrial flutter. Interestingly, the reported patient had 2:1 AV conduction during both normal atrial rates (

Adverse childhood experiences and adverse community environments among maternal mortality cases in Arizona.

BACKGROUND: Adverse childhood experiences (ACE) and adverse community environments (ACoE) can have negative impacts on adult health and lead to premature death. Their association within the maternal mortality (MM) population remains unknown. OBJECTIVE: This study aimed to assess the association between ACEs and ACoEs with pregnancy outcomes and MM variables within the Arizona MM population. STUDY DESIGN: A secondary review of 146 maternal deaths in Arizona (2018-2019), originally reviewed by the Arizona Maternal Mortality Review Committee, was conducted to identify ACE and ACoE exposure, adv

Prenatal exposure to criteria air pollutants and associations with congenital anomalies: A Lebanese national study.

Maternal exposure to air pollution has been associated with a higher birth defect (BD) risk. Previous studies suffer from inaccurate exposure assessment methods, confounding individual-level variations, and classical analytical modelling. This study aimed to examine the association between maternal exposure to criteria air pollutants and BD risk. A total of 553 cases and 10,214 controls were identified from private and public databases. Two subgroups were then formed: one for a matched case-control design, and another for Feature Selection (FS) analysis. Exposure assessment was based on the me…

Intrauterine gestational diabetes mellitus, child anxiety, and internalizing symptoms: moderation by physical activity.

CONTEXT: Prenatal exposure to gestational diabetes mellitus (GDM) is linked to increased risk for offspring mental health disorders, yet few studies account for maternal mental health or potential protective factors. OBJECTIVE: This work aimed to examine whether prenatal GDM exposure is associated with child anxiety and depressive symptoms, independent of maternal depression, and whether child physical activity (PA) modifies these associations. METHODS: In this retrospective cohort study, data included 138 child-mother dyads (mean [SD] child age: 11.1 [2.5] years; 40% GDM-exposed; 56% female

Children's immunology, what can we learn from animal studies (2): Modulation of systemic Th1/Th2 immune response in infant mice after prenatal exposure to low-level toluene and toll-like receptor (TLR) 2 ligand.

It has been reported that the newborn immune system differs quantitatively and functionally from that of adults. Development of the immune system has important implications for childhood diseases. The immaturity of the immune system in the prenatal or suckling stage may contribute to susceptibility to environmental toxic chemical exposure. In the present study, to clarify the effect of low-level toluene exposure on immune functions during developmental stage, pregnant mice were exposed to 0, 5, and 50 ppm toluene from gestational day 14 to day 19 with or without stimulation by peptidoglycan (P

Maternal and cord blood manganese (Mn) levels and birth weight: The MIREC birth cohort study.

Epidemiological studies have hypothesized that both insufficient and excess blood manganese (Mn) levels during pregnancy are associated with reduced fetal growth. This literature is characterized by inconsistent results and a limited focus on women with exposures representative of the general North American population. We examined the relationship between maternal and cord blood Mn levels and fetal growth among women enrolled in the Maternal-Infant Research on Environmental Chemicals Study (MIREC). Mothers with singleton, term infants and complete maternal first and third trimester blood Mn da
