A Clinical and Pathological Analysis of Seven Cases of Salivary Gland Ductal Carcinoma.

Background and objective Salivary duct carcinoma (SDC) is a rare but highly aggressive malignancy. Due to its low incidence, most published data are derived from small retrospective series or case reports, and the optimal diagnostic work-up, prognostic stratification, and treatment algorithms remain poorly defined. This study aimed to investigate the clinical characteristics, pathological features, and treatment strategies for SDC by reviewing the relevant literature. Methods A retrospective analysis was conducted on the clinical and pathological data, histological morphology, immunophenotypic

Patient-derived 3D organotypic co-cultures as a long-term ex vivo model for pleomorphic adenoma of the parotid gland.

Pleomorphic adenoma is the most common benign salivary gland tumor and most frequently arises in the parotid gland. Although benign, it can recur and may undergo malignant transformation, underscoring the need for experimental models that preserve native tissue features over time. In this study, fresh tumor tissue from six patients with primary histologically benign pleomorphic adenoma was divided into fragments of approximately 3 × 3 mm in surface dimensions and approximately 3 mm in thickness and maintained in a patient-derived three-dimensional organotypic co-culture for up to 21 days. Late Recurrence/Metastasis of Acinic Cell Carcinoma After 10 Years of Resection: A Case Series and Literature Review.

BACKGROUND: Acinic cell carcinoma (AciCC) is a relatively rare malignancy of the salivary glands, predominantly affecting the parotid gland. Recent studies report that the local recurrence rate for AciCC ranges from 5% to 17%, while metastasis occurs in 2% to 12% of cases. Most recurrences or metastases are observed within the first 5 years following initial therapy. However, late recurrences or metastases occurring more than 10 years have been reported in rare cases. METHODS: This study presents 13 cases of AciCC in which patients experienced recurrence or metastasis 10 to 45 years after ini

Evolution of cervicofacial flaps: From classical reconstruction to Freestyle Multiperforator Island Flap.

BACKGROUND: This study presents a cost-effective and safe alternative for facial reconstruction, introducing the concept of the Freestyle Multiperforator Island Flap (FMIF) a subcutaneously based flap that can be customized in both location and size. Its versatility stems from the extensive vascular cutaneous territories supplied by the dermal and facial plexuses, which branch in an arborescent pattern to ensure reliable perfusion across multiple facial regions. METHODS: A retrospective case series of 34 patients (37 flaps) with cheek defects treated between 2012 and 2022 is presented. Demogr

Distribution and Frequency of Salivary Gland Tumours: An International Multicenter Study.

BACKGROUND: Salivary gland tumours (SGT) are a relatively rare group of neoplasms with a wide range of histopathological appearance and clinical features. To date, most of the epidemiological studies on salivary gland tumours are limited for a variety of reason including being out of date, extrapolated from either a single centre or country studies, or investigating either major or minor glands only. METHODS: This study aimed to mitigate these shortcomings by analysing epidemiological data including demographic, anatomical location and histological diagnoses of SGT from multiple centres acros

Comprehensive analysis of the transcriptome-wide m6A methylome in invasive malignant pleomorphic adenoma.

BACKGROUND: Invasive malignant pleomorphic adenoma (IMPA) is a highly invasive parotid gland tumor and lacks effective therapy. N6-Methyladenosine (m6A) is the most prevalent post-transcriptional modification of mRNAs in eukaryotes and plays an important role in the pathogenesis of multiple tumors. However, the significance of m6A-modified mRNAs in IMPA has not been elucidated to date. Hence, in this study, we attempted to profile the effect of IMPA in terms of m6A methylation in mRNA. METHODS: Methylated RNA immunoprecipitation with next-generation sequencing (MeRIP-seq) and RNA sequencing (

Clinicopathological characteristics and outcome of 31 patients with ETV6-NTRK3 fusion gene confirmed (mammary analogue) secretory carcinoma of salivary glands.

OBJECTIVES: In 2010, a new subtype of salivary gland cancer (SGC), (mammary analogue) secretory carcinoma (SC), was defined, characterized by the ETV6-NTRK3 fusion gene. As clinical behavior and outcome data of this histological subtype tumor are still sparse, we aimed to describe the clinicopathological course and outcome of a series of translocation positive SC patients. PATIENT AND METHODS: We re-evaluated the pathological diagnosis of a subset of SGCs, diagnosed in 4 of 8 Dutch head and neck centers. Subsequently, tumors with a morphological resemblance to SC were tested for the ETV6-NTRK

Delineation of the neck node levels for head and neck tumors: a 2013 update. DAHANCA, EORTC, HKNPCSG, NCIC CTG, NCRI, RTOG, TROG consensus guidelines.

In 2003, a panel of experts published a set of consensus guidelines for the delineation of the neck node levels in node negative patients (Radiother Oncol, 69: 227-36, 2003). In 2006, these guidelines were extended to include the characteristics of the node positive and the post-operative neck (Radiother Oncol, 79: 15-20, 2006). These guidelines did not fully address all nodal regions and some of the anatomic descriptions were ambiguous, thereby limiting consistent use of the recommendations. In this framework, a task force comprising opinion leaders in the field of head and neck radiation onc

Mass spectrometric identification of key proteolytic cleavage sites in statherin affecting mineral homeostasis and bacterial binding domains.

Human salivary statherin inhibits both primary and secondary calcium phosphate precipitation and, upon binding to hydroxyapatite, associates with a variety of oral bacteria. These functions, crucial in the maintenance of tooth enamel integrity, are located in defined regions within the statherin molecule. Proteases associated with saliva, however, cleave statherin effectively, and it is of importance to determine how statherin functional domains are affected by these events. Statherin was isolated from human parotid secretion by zinc precipitation and purified by reversed-phase high performanc

Role of Epstein-Barr virus in fine-needle aspirates of metastatic neck nodes in the diagnosis of nasopharyngeal carcinoma.

BACKGROUND: The patient with nasopharyngeal carcinoma (NPC) frequently is initially seen with regional node dissemination. Preliminary investigations suggest that the presence of Epstein-Barr virus (EBV) genomes in neck metastases from an occult primary may be diagnostic and predictive of NPC. The goal of this study was to test this proposition. METHODS: The polymerase chain reaction (PCR) was used to detect the presence of EBV DNA in fine-needle aspirate (FNA) samples obtained from malignant neck nodes. Control samples were obtained from other locations in the head and neck. PATIENTS: The p

The prognostic significance of the geriatric nutritional risk index in postoperative parotid gland carcinoma.

The impact of the Geriatric Nutritional Risk Index (GNRI) on the prognosis of postoperative parotid gland carcinoma (PGC) remains unclear. This study investigates the role of GNRI in predicting disease-free survival (DFS) and overall survival (OS) in postoperative PGC patients and develops a predictive model. We conducted a retrospective analysis of 389 postoperative PGC patients treated from May 2008 to June 2019 at two regional medical centers in China. The independent prognostic factors were identified by univariate and multivariate Cox regression analyses. Then, two nomograms were establis

Atypical Site of Presentation of a Rare Type of SMARCA4-Positive Cutaneous Squamous Cell Carcinoma of the Skin: Case Report and Review of the Literature.

Cutaneous squamous cell carcinoma (cSCC) comprises 20% of cases of nonmelanoma skin cancers in the United States. In total, 3% to 5% of squamous cell carcinoma (SCC) are metastatic at the time of presentation, associated with significant mortality due to a lack of standardized treatment options. In total, 95% of these tumors are amenable to the initial standard of treatment, which is surgical resection. However, a small percentage of them require systemic therapy as they are either locally advanced to regional lymph nodes or have distant metastasis. The common sites of presentation of cSCC are
