Expression of the CXCR3 ligand I-TAC by hepatocytes in chronic hepatitis C and its correlation with hepatic inflammation.

The factors that regulate lymphocyte traffic in chronic hepatitis C (CHC) are not completely defined. Interferon (IFN)-inducible T cell alpha chemoattractant (I-TAC) is a relatively new member of the CXCR3 chemokine ligand family that selectively recruits activated T cells to sites of inflammation. To determine if I-TAC plays a role in CHC, we investigated I-TAC expression in hepatitis C virus (HCV)-infected liver biopsy material. I-TAC messenger RNA (mRNA) levels were significantly increased in HCV-infected liver compared with normal liver, which correlated with both portal and lobular inflam

Missense mutation in the second RNA binding domain reveals a role for Prkra (PACT/RAX) during skull development.

Random chemical mutagenesis of the mouse genome can causally connect genes to specific phenotypes. Using this approach, reduced pinna (rep) or microtia, a defect in ear development, was mapped to a small region of mouse chromosome 2. Sequencing of this region established co-segregation of the phenotype (rep) with a mutation in the Prkra gene, which encodes the protein PACT/RAX. Mice homozygous for the mutant Prkra allele had defects not only in ear development but also growth, craniofacial development and ovarian structure. The rep mutation was identified as a missense mutation (Serine 130 to

SFTSV NSs interacts with AGO2 to regulate the RNAi pathway for viral replication.

RNA interference (RNAi) is a posttranscriptional gene silencing mechanism acting as an antiviral defense in eukaryotes. During viral replication, intermediate double-stranded RNAs are processed into virus-derived small interfering RNAs (vsiRNAs) by the host enzyme, DICER. These vsiRNAs are incorporated into the RNA-induced silencing complex (RISC), where AGO2 cleaves viral genomic RNAs. However, viruses have evolved mechanisms to suppress this pathway. Here, we report that the nonstructural protein (NSs) of severe fever with thrombocytopenia syndrome virus (SFTSV) interacts with RISC to suppre

Comparative Transcriptome Analyses of Schistosoma japonicum Derived From SCID Mice and BALB/c Mice: Clues to the Abnormality in Parasite Growth and Development.

Schistosomiasis, caused by the parasitic flatworms called schistosomes, remains one of the most prevailing parasitic diseases in the world. The prodigious oviposition of female worms after maturity is the main driver of pathology due to infection, yet our understanding about the regulation of development and reproduction of schistosomes is limited. Here, we comparatively profiled the transcriptome of Schistosoma japonicum recovered from SCID and BALB/c mice, which were collected 35 days post-infection, when prominent morphological abnormalities could be observed in schistosomes from SCID mice,

Inhibition of ocular angiogenesis by diced small interfering RNAs (siRNAs) specific to vascular endothelial growth factor (VEGF).

The effects of diced small interfering RNAs (siRNAs) designed for vascular endothelial growth factor (VEGF) on the expression of VEGF in human retinal pigment epithelial cell line ARPE-19 cells in vitro and on corneal angiogenesis in vivo were examined. The exposure to diced siRNAs significantly reduced the VEGF mRNA expression in ARPE-19 cells with minimal toxicity. In suture-induced corneal angiogenesis models, diced siRNAs minimized the severity of angiogenesis. Histological analysis displayed no particular tissue damage in the conjunctiva where siRNA was injected. The approach using diced

Molecular characterization and functional analysis of components of the TOR pathway of the salmon louse, Lepeophtheirus salmonis (Krøyer, 1838).

The salmon louse Lepeophtheirus salmonis (Copepods, Caligida) is a marine ectoparasite infecting salmonid fishes in the northern hemisphere. At present, salmon lice infections are the most severe disease problem in the salmon farming industry causing significant economic losses. Due to development of resistance towards available chemotherapeutants, it is clear that new chemotherapeutants or non-chemical control methods are essential to manage the parasite in the future. The TOR signaling pathway is present in all metazoans and is a major regulator of cellular activity according to nutrient ava

Compartmentation of enzymes in a microbody, the glycosome, is essential in Trypanosoma brucei.

All kinetoplastids contain membrane-bound microbodies known as glycosomes, in which several metabolic pathways including part of glycolysis are compartmentalized. Peroxin 2 is essential for the import of many proteins into the microbodies of yeasts and mammals. The PEX2 gene of Trypanosoma brucei was identified and its expression was silenced by means of tetracycline-inducible RNA interference. Bloodstream-form trypanosomes, which rely exclusively on glycolysis for ATP generation, died rapidly upon PEX2 depletion. Insect-form (procyclic) trypanosomes do not rely solely on glycolysis for ATP sy

Monkeypox virus induces the synthesis of less dsRNA than vaccinia virus, and is more resistant to the anti-poxvirus drug, IBT, than vaccinia virus.

Monkeypox virus (MPXV) infection fails to activate the host anti-viral protein, PKR, despite lacking a full-length homologue of the vaccinia virus (VACV) PKR inhibitor, E3. Since PKR can be activated by dsRNA produced during a viral infection, we have analyzed the accumulation of dsRNA in MPXV-infected cells. MPXV infection led to less accumulation of dsRNA than VACV infection. Because in VACV infections accumulation of abnormally low amounts of dsRNA is associated with mutations that lead to resistance to the anti-poxvirus drug isatin beta-thiosemicarbazone (IBT), we investigated the effects

Activation of cell membrane-localized Toll-like receptor 3 by siRNA.

Small interfering RNA molecules (siRNA) are short dsRNAs that are used for different therapeutic applications. On the other hand, dsRNAs can bind to and activate cell RNA sensors and consequently trigger inflammatory response. Here we show that siRNA activates primary human endothelial cells and human lymphatic endothelial cells and that this response is inhibited by antibodies against TLR3. In contrast, the activation of human lymphatic endothelial cells by poly(I:C) was inhibited by bafilomycin but not by anti-TLR3 antibodies. Bafilomycin also inhibited poly(I:C) but not siRNA cell stimulati

Mechanisms biomarkers and therapeutic strategies of human endogenous retroviruses in cancer.

Human endogenous retroviruses (HERVs), constituting roughly 8% of the human genome, have undergone a profound conceptual evolution from dismissed genomic “fossils” to critical, dualistic regulators in cancer biology. Their pathognomonic reactivation across malignancies orchestrates tumorigenesis through three interconnected molecular axes: (1) genomic destabilization via LTR-mediated insertional mutagenesis, disrupting key loci such as TP53 and MYC; (2) immune checkpoint subversion, driven by HERV-K envelope glycoprotein-induced PD-L1 upregulation (2.3-fold; p < 0.01); and (3) chronic inflamma

Poly I:C-induced monocyte-macrophages alleviate type 2 immunopathology in a murine asthma model.

RATIONALE: Pathogen-associated molecular patterns (PAMPs) are known to initiate pro-inflammatory responses, but emerging evidence has revealed their roles in inducing macrophage-dependent immunomodulatory pathways that dampen inflammation. OBJECTIVES: We explored the interactions between the asthma-associated type 2 immunopathology and immune response to poly I:C, a PAMP mimicking double-stranded RNA, in ovalbumin (OVA) sensitized and challenged mice. METHODS: Following intranasal co-administration of Poly I:C during OVA challenges, we performed immunophenotyping of the lung and measured air

Hochuekkito reversed poly(I:C)-induced anxiety-like behavior and reductions in hippocampal BDNF expression in mice.

We investigated the effects of hochuekkito (HET), a traditional Japanese herbal medicine, on anxiety-like behavior induced by the intraperitoneal administration of polyinosinic-polycytidylic acid (poly(I:C)), a synthetic double-stranded RNA that is widely used to mimic viral infection-associated inflammation, in mice. HET (1 g/kg) was orally administered to the mice once daily for two weeks prior to the injection of poly(I:C). Anxiety-like behavior was assessed for 24 h after the poly(I:C) injection using the light-dark box test. The repeated administration of HET significantly attenuated poly

Characterization of novel pesti- and astro-like viruses in hatchery-reared European seabass (Dicentrarchus labrax) associated with health disorders and low mortality.

European seabass is a major farmed species in Mediterranean aquaculture. Its early life stages are very sensitive to sanitary and environmental conditions and are therefore closely monitored. In this study, we characterized new viruses recently isolated from fingerlings in a hatchery showing whitish three-dimensional lesions on the fins and sides and distended abdomens and investigated their pathologic potential. A cytopathic effect was observed 17 days post-inoculation (dpi) of homogenized internal organs at 20 °C on a CHSE214 cell line. Viral presence was confirmed using the indirect fluores

Genomic and Transcriptomic Characterization of Umatilla Virus Isolated and Identified from Mosquitoes in Ningxia, China.

During the 2023 surveillance of mosquito-borne viruses in Ningxia Hui Autonomous Region, a strain of Umatilla virus (UMAV) was isolated from a pool of Culex pipiens pallens (NX23166) collected in Xiji County and cultured in C6/36 cells. Electron microscopy revealed that NX23166-infected mosquito cells showed approximately 70-nm virus particles, typical of the genus Orbivirus. Through next-generation sequencing, 10 double-stranded RNA (dsRNA) segments of the virus were obtained. Phylogenetic and homology analyses based on these sequences revealed that this strain was most closely related to the

PKR inhibition reduces high glucose and H2O2-induced injury in rat renal epithelial cells by suppressing the sonic hedgehog pathway.

Double-stranded RNA-dependent protein kinase (PKR) is involved in signaling pathways that mediate responses to oxidative and endoplasmic reticulum (ER) stress and has been linked to various cardiovascular diseases. However, the role of PKR in renal cells under chronic oxidative stress and the direct impact of PKR inhibitor treatment on such stress-induced damage remain unexplored. This study aimed to investigate the effect of PKR inhibition on oxidative stress-induced injury in renal epithelial cells using an in vitro model. Oxidative stress was induced in NRK-52E renal epithelial cells using

Racial Differences in Plasma Microbial Translocation and Plasma Microbiome, Implications in Systemic Lupus Erythematosus Disease Pathogenesis.

OBJECTIVE: Black groups have increased prevalence and accelerated pathogenicity of systemic lupus erythematosus (SLE) compared to other ethnic/racial groups. The microbiome and systemic microbial translocation are considered contributing factors to SLE disease pathogenesis. However, racial differences in the plasma microbiome and microbial translocation in lupus remain unknown. METHODS: In the current study, we investigated plasma levels of microbial translocation (lipopolysaccharide [LPS] and zonulin) and the plasma microbiome using microbial 16S RNA sequencing of Black and White patients wi

Maternal obstructive sleep apnea aggravates metabolic dysfunction-associated fatty liver disease via HMGB1-TLR4 signaling-mediated endoplasmic reticulum stress in male offspring rats.

AIMS/HYPOTHESIS: Maternal obstructive sleep apnea (MOSA) may inflict long-term metabolic effects on offspring. We hypothesize that MOSA increases the propensity for metabolic dysregulation in offspring and thus facilitates the development of metabolic dysfunction-associated fatty liver disease (MAFLD). This study aims to test the hypothesis and explore the underlying mechanism. METHODS: The MOSA rat model of upper airway obstruction was established and fecundated. The postweaning male offspring (n = 171) from both the control group and MOSA group were randomly fed the normal chow diet (NCD, n

Expression and Functional Analysis of CST1 in Intractable Nasal Polyps.

In this study, we found Cystatin SN (CST1), a type 2 cystatin subfamily member, to be highly expressed in nasal polyps from patients with intractable chronic rhinosinusitis (CRS) with nasal polyps, using a whole-transcript analysis with next-generation sequencing. Eosinophilic CRS (ECRS) involves nasal polyps that are refractory and recur immediately after endoscopic sinus surgery. We hypothesized that CST1 may contribute to the pathogenesis of ECRS. We examined the expression of CST1 in nasal polyps from patients with ECRS by assessing mRNA expression levels using real-time PCR and immunohist
