Evidence architecture of glaucoma-related biomaterials reveals an uneven transition toward smart materials, additive manufacturing, and functional tissue engineering.

BACKGROUND: Glaucoma-related biomaterial research has expanded from ocular drug delivery to responsive hydrogels, anti-fibrotic systems, gla … very platforms, but their transition toward smart materials, additive manufacturing, and functional tissue engineering remains uneven. Future studies should emphasize reproducible material design, disease-relevant functional endpoints, outflow-pathway models, neuroprotection, and engineered surgical interfaces.

Stimuli-responsive 4D-bioprinted constructs for musculoskeletal tissue regeneration: Shape-morphing mechanisms, cell-laden bioink engineering, and preclinical outcomes.

INTRODUCTION: Musculoskeletal disorders impose a substantial global disability burden. Conventional 3D bioprinting cannot replicate the dynamic, anisotropic architecture of bone, cartilage, skeletal muscle, and cardiac muscle. Four-dimensional (4D) bioprinting addresses this by integrating stimuli-responsive materials into constructs, enabling programmed shape transformation and adaptive behavior following implantation. METHODS: This narrative review examines primary experimental research on stimuli-responsive 4D-bioprinted musculoskeletal constructs, drawing on in vitro, in vivo, and combine

Engineering multifunctional chitosan-based hydrogels for integrated antimicrobial therapy and endogenous stem cell-driven tissue repair in infectious keratitis.

Bacterial keratitis is a major contributor to preventable blindness worldwide, with infections caused by Pseudomonas aeruginosa posing significant challenges due to the formation of biofilms and insufficient tissue regeneration following antimicrobial therapy. Conventional treatments predominantly target the infection while neglecting the promotion of healing processes, often resulting in corneal scarring and subsequent vision impairment. Therefore, therapeutic approaches for condition should extend beyond antibacterial measures to include strategies that facilitate early limbal stem cell part

Direct ink writing of collagen scaffolds functionalized with antimicrobial peptides: A proof-of-concept study for bone tissue engineering.

We developed a novel collagen-based biomaterial ink with an integrated antimicrobial peptide (AMP) intended for bone tissue engineering, enabling fabrication of a 3D-printed collagen-scaffold with antibacterial activity. A chemically synthesized derivative of the human AMP KR-12 was designed for enhanced stability and activity against Gram-positive bacteria that are key pathogens in fracture-related infections. KR-12 was covalently conjugated to collagen using both non-selective and maleimide-based coupling strategies. The choice of conjugation method significantly influenced peptide incorpora…

Potential of organ-on-a-chip in advancing synthetic extracellular matrix technology for bone tissue engineering in dentistry (Review).

Bone tissue engineering (BTE) is a cutting-edge approach within biomedical sciences, especially in regenerative medicine, addressing key challenges such as organ transplantation and complex tissue repair. At the core of BTE is the development of biomimetic scaffolds to replicate native tissue environments. However, conventional models often fall short in accurately mimicking the complexity of human tissue microenvironments. Organ-on-a-chip (OOAC) technology offers a transformative alternative. These microscale systems combine microfluidics, biomaterials and cell cultures to emulate the structu…

Human Omental Mesothelial Cells Exhibit a Corneal Endothelial-Like Cell Phenotype for Tissue Engineering of a Corneal Endothelium Biomimetic.

PURPOSE: To evaluate whether cultured human omental mesothelial cells (OMC) exhibit phenotypic and functional similarities to human corneal endothelial cells (CEC) and whether they can adhere to the corneal stroma and form a biomimetic corneal endothelium when grown on a human anterior lens capsule (HALC). METHODS: Human OMC were isolated from the greater omentum. Human B4G12 CEC were used as a reference for native and functional CEC phenotype whereas human mesenchymal stromal cells (MSC) served as a phenotypically distinct control group. OMC, CEC and MSC were compared using flow cytometry, i

Influence of Oligomeric Lactic Acid and Structural Design on Biodegradation and Absorption of PLA-PHB Blends for Tissue Engineering.

The advancing development in biomaterials and biology has enabled the extension of 3D printing technology to the bioadditive manufacturing of degradable hard tissue substitutes. One of the key advantages of bioadditive manufacturing is that it has much smaller design limitations than conventional manufacturing and is therefore capable of producing implants with complex geometries. In this study, three distinct blends of polylactic acid (PLA) and polyhydroxybutyrate (PHB) were produced using Fused Deposition Modeling (FDM) technology. Two of these blends were plasticized with oligomeric lactic

Iron oxide nanoparticle-stabilized Pickering emulsion-templated porous scaffolds loaded with polyunsaturated fatty acids (PUFAs) for bone tissue engineering.

Dietary intake of ω-3-polyunsaturated fatty acids (PUFAs) can significantly improve the expression levels of alkaline phosphatase (ALP) and osteocalcin. However, PUFAs are hydrophobic and highly sensitive to temperature, oxygen concentration, pH, and ionic strength. Hence, it is challenging to use PUFAs as bioactive compounds for bone tissue engineering. Here, we encapsulated PUFAs in liposomes to improve their stability. The hydrodynamic size of the PUFA-loaded liposomes was found to be 121.3 ± 35 nm. GC-MS analysis showed that the encapsulation efficiency of the PUFAs was 19.9 ± 3.4%. These …

Incorporation of montmorillonite into microfluidics-generated chitosan microfibers enhances neuron-like PC12 cells for application in neural tissue engineering.

The complexity in structure and function of the nervous system, as well as its slow rate of regeneration, makes it more difficult to treat it compared to other tissues. Neural tissue engineering aims to create an appropriate environment for nerve cell proliferation and differentiation. Fibrous scaffolds with suitable morphology and topography and better mimicry of the extracellular matrix have been promising for the alignment and migration of neural cells. On this premise, to improve the properties of the scaffold, we combined montmorillonite (MMT) with chitosan (CS) polymer and created microf…

Biomineralization in Three-Dimensional Scaffolds Based on Bacterial Nanocellulose for Bone Tissue Engineering: Feature Characterization and Stem Cell Differentiation.

Bacterial nanocellulose (BNC) has a negative surface charge in physiological environments, which allows the adsorption of calcium ions to initiate the nucleation of different calcium phosphate phases. The aim of this study was to investigate different methods of mineralization in three-dimensional microporous bacterial nanocellulose with the intention of mimicking the composition, structure, and biomechanical properties of natural bone. To generate the 3D microporous biomaterial, porogen particles were incorporated during BNC fermentation with the Komagataeibacter medellinensis strain. Calcium

Injectable, In Situ Self-cross-linking, Self-healing Poly(l-glutamic acid)/Polyethylene Glycol Hydrogels for Cartilage Tissue Engineering.

Injectable hydrogels have drawn much attention in the field of tissue engineering because of advantages such as simple operation, strong plasticity, and good biocompatibility and biodegradability. Herein, we propose the novel design of injectable hydrogels via a Schiff base cross-linking reaction between adipic dihydrazide (ADH)-modified poly(l-glutamic acid) (PLGA-ADH) and benzaldehyde-terminated poly(ethylene glycol) (PEG-CHO). The effects of the mass fraction and the molar ratio of -CHO/-NH2 on the gelation time, mechanical properties, equilibrium swelling, and in vitro degradation of the h…

Antibacterial Activity and Cell Responses of Vancomycin-Loaded Alginate Coating on ZSM-5 Scaffold for Bone Tissue Engineering Applications.

Despite the significant advancement in bone tissue engineering, it is still challenging to find a desired scaffold with suitable mechanical and biological properties, efficient bone formation in the defect area, and antibacterial resistivity. In this study, the zeolite (ZSM-5) scaffold was developed using the space holder method, and a novel vancomycin-loaded alginate coating was developed on it to promote their characteristics. Our results demonstrated the importance of alginate coating on the microstructure, mechanical, and cellular properties of the ZSM-5 scaffold. For instance, a three-fol…

Construction of spider silk protein small-caliber tissue engineering vascular grafts based on dynamic culture and its performance evaluation.

Tissue engineering is an alternative method for preparing small-caliber (<6 mm) vascular grafts. Dynamic mechanical conditioning is being researched as a method to improve mechanical properties of tissue engineered blood vessels. This method attempts to induce unique reaction in implanted cells that regenerate the matrix around them, thereby improving the overall mechanical stability of the grafts. In this study, we used a bioreactor to seed endothelial cells and smooth muscle cells into the inner and outer layers of the electrospun spider silk protein scaffold respectively to construct vascul…

Design and Characterization of a Bioinspired Polyvinyl Alcohol Matrix with Structural Foam-Wall Microarchitectures for Potential Tissue Engineering Applications.

Traditional medical soft matrix used in a surgical treatment or in wound management was not good enough in both the structural support and interconnectivity to be applied in tissue engineering as a scaffold. Avian skeleton and feather rachises might be good reference objects to mimic in designing a scaffold material with good structural support and high interconnectivity because of its structural foam-wall microarchitectures and structural pneumaticity. In this study, a biomimetic airstream pore-foaming process was built up and the corresponding new medical soft matrix derived from polyvinyl a…

Fabrication of Adipose-Derived Stem Cell-Based Self-Assembled Scaffold under Hypoxia and Mechanical Stimulation for Urethral Tissue Engineering.

Long urethral strictures are often treated with autologous genital skin and buccal mucosa grafts; however, risk of hair ingrowth and donor site morbidity, restrict their application. To overcome this, we introduced a tissue-engineered human urethra comprising adipose-derived stem cell (ASC)-based self-assembled scaffold, human urothelial cells (UCs) and smooth muscle cells (SMCs). ASCs were cultured with ascorbic acid to stimulate extracellular matrix (ECM) production. The scaffold (ECM) was stained with collagen type-I antibody and the thickness was measured under a confocal microscope. Resul

Photo-cross-linked Gelatin Glycidyl Methacrylate/N-Vinylpyrrolidone Copolymeric Hydrogel with Tunable Mechanical Properties for Ocular Tissue Engineering Applications.

Corneal transplantation is currently the primary treatment for corneal blindness. However, severe global scarcity of donor corneas is driving the scientific community to find novel solutions. One potential solution is to replace the damaged tissue with a biocompatible artificial cornea. Here, gelatin glycidyl methacrylate (GM) and N-vinylpyrrolidone (VP) were cocrosslinked to afford a hybrid bicomponent copolymeric hydrogel with excellent mechanical, structural, and biological properties. Our studies showed that the GM/VP ratio can be adjusted to generate a construct with high tensile modulus

Quality of Reporting in Preclinical Urethral Tissue Engineering Studies: A Systematic Review to Assess Adherence to the ARRIVE Guidelines.

Preclinical research within the area of urethral tissue engineering has not yet been successfully translated into an efficient therapeutic option for patients. This gap could be attributed, in part, to inadequate design and reporting of the studies employing laboratory animals. In this study, a systematic review was conducted to investigate the quality of reporting in preclinical studies utilizing tissue engineering approaches for urethral repair. The scope was on studies performed in rabbits, published between January 2014 and March 2020. Quality assessment of the data was conducted according…

Adipose-Derived Stromal Cells and Mineralized Extracellular Matrix Delivery by a Human Decellularized Amniotic Membrane in Periodontal Tissue Engineering.

Periodontitis is a prevalent disease characterized by the loss of periodontal supporting tissues, bone, periodontal ligament, and cementum. The application of a bone tissue engineering strategy with Decellularized Human Amniotic Membrane (DAM) with adipose-derived stromal cells (ASCs) has shown to be convenient and valuable. This study aims to investigate the treatments of a rat periodontal furcation defect model with DAM, ASCs, and a mineralized extracellular matrix (ECM). Rat ASCs were expanded, cultivated on DAM, and with a bone differentiation medium for four weeks, deposited ECM on DAM. P…

Effect of cell seeding density on matrix-forming capacity of meniscus fibrochondrocytes and nasal chondrocytes in meniscus tissue engineering.

The presence of intact menisci is imperative for the proper function of the knee joint. Meniscus injuries are often treated by the surgical removal of the damaged tissue, which increases the likelihood of post-traumatic osteoarthritis. Tissue engineering holds great promise in producing viable engineered meniscal tissue for implantation using the patient's own cells; however, the cell source for producing the engineered tissue is unclear. Nasal chondrocytes (NC) possess many attractive features for engineering meniscus. However, in order to validate the use of NC for engineering meniscus fibro…
