Central acoustic tract in an echolocating bat: an extralemniscal auditory pathway to the thalamus.

To determine the sources and targets of auditory pathways that bypass the inferior colliculus in the mustache bat, we injected WGA-HRP in the medial geniculate body and related auditory nuclei of the thalamus as well as in the lower brainstem. We used electrophysiological methods to verify that the injection electrode was in an area responsive to sound. The only thalamic injections that produced retrograde transport to cells in auditory nuclei caudal to the inferior colliculus were those that included the suprageniculate nucleus. These injections labeled a group of large multipolar cells lying

Demonstration of antigenic variation among rabies virus isolates by using monoclonal antibodies to nucleocapsid proteins.

Rabies virus isolates from terrestrial animals in six areas of the United States were examined with a panel of monoclonal antibodies to nucleocapsid proteins. Characteristic differences in immunofluorescence reactions permitted the formation of four antigenically distinct reaction groups from the 231 isolates tested. The geographic distribution of these groups corresponded well with separate rabies enzootic areas recognized by surveillance of sylvatic rabies in the United States. Distinctive reaction patterns were also identified for viral proteins from four infected bat species, and identical

Postexposure trial of a human diploid cell strain rabies vaccine.

A human diploid cell strain vaccine (HDCV) was evaluated in 90 persons treated after exposure to rabies, 21 of whom were bitten by proven rabid animals. Intramuscular doses of HDCV were given on days 0, 3, 7, 14, and 28, and human rabies immune globulin (HRIG) was given on day 0. Antibody to rabies virus was tested for by the rapid fluorescent focus inhibition test; adverse reactions were assessed from physician's forms. All 87 persons tested developed titers of greater than or equal to 0.5 international units (IU)/ml, with a maximum geometric mean titer of 15.0 IU/ml on day 42. One year after

A tale of two ears: development of binaural auditory processing in the big brown bat.

Big brown bat (Eptesicus fuscus) pups are born naked, blind, and with closed external ear canals. We examined the ears of newborn bat pups (n = 82; 42 females) and found that in a majority of pups (80%), the ear canals open asymmetrically. What remains unknown is whether this developmental asymmetry influences hearing. We recorded auditory brainstem response (ABR) waveforms in a subset of pups with both ears open by placing subdermal needle electrodes near the base of the auditory bullae and repeatedly presenting broadband acoustic clicks and averaging the evoked neural response. The resultant

Novel African Rhinolophus bat ACE2 sequences reveal the determinants of Afro-Eurasian sarbecovirus entry.

UNLABELLED: Sarbecoviruses, including SARS-CoV and SARS-CoV-2, are frequently linked to Rhinolophus bats as their putative natural reservoirs. Angiotensin-converting enzyme 2 (ACE2), a host carboxypeptidase widely expressed in mammalian tissues, plays a critical role in sarbecovirus infection by serving as the cellular receptor for the viral spike (S) protein. Given the recent human outbreaks and pandemics caused by members of sarbecoviruses and the wide distribution of Rhinolophus bats, it is essential to maintain surveillance of these viruses while improving our understanding of their intera

Synergistic activation of bat SARS-like coronaviruses spike protein by elastase and TMPRSS2.

Although numerous sarbecoviruses have been identified in bats, but most lack the ability to infect human cells. Some barriers limit coronavirus zoonosis, including susceptibility to host proteases. Here, we investigated whether exogenous protease treatment can circumvent host restrictions in two severe acute respiratory syndrome (SARS)-related bat coronaviruses. We found that the spike proteins of RaTG13 and Khosta-2, which are sarbecoviruses obtained from horseshoe bats in China and Russia, respectively, facilitated the ACE2-mediated entry of pseudotyped viruses into VeroE6/TMPRSS2 cells foll

Comparison of the biological properties of bat-derived filovirus envelope glycoproteins.

Although some filoviruses, such as Ebola virus (EBOV) and Marburg virus (MARV), are highly pathogenic in humans, novel filoviruses, including Lloviu virus (LLOV), Bombali virus (BOMV), Mengla virus (MLAV), and Dehong virus (DEHV), whose biological properties are poorly understood, have been found in bats. In this study, we characterized the envelope glycoproteins (GPs) of these bat-derived filoviruses (BatFiloVs). We first confirmed that virus-like particles consisting of their GPs, nucleoproteins, and matrix proteins were filamentous. Interestingly, although BatFiloVs were serologically disti

Specific binding of human P[28] rotavirus VP8* protein to blood group ABH antigens on type 1 chains.

Group A rotavirus (RV) has been the major cause of acute gastroenteritis in infants and young children. Among the five P genogroups almost all P genotype RVs in P[II], P[III] and P[IV] genogroups that infect humans can bind glycan histo-blood group antigens (HBGAs) as the receptors on the host cell surface to infect host through the viral spike protein VP8*. Although P[I] is the largest genogroup, P[28] and P[10] are the only two genotype RVs infecting humans in the group. It has recently been found that a P[28] strain is related to bat RV and considered a possible product of reassortment betw

Recombination alters the receptor binding and furin cleavage site in novel bat-borne HKU5-CoV-2 coronavirus.

UNLABELLED: HKU5-CoV-2 is a new bat-infecting coronavirus phylogenetically related to MERS-CoV. It has recently been confirmed that HKU5-CoV-2 can enter human cells and organoids in vitro via the ACE2 receptor, raising concerns about its pandemic potential due to zoonotic spillover. Whether recombination has an influence on HKU5-CoV-2 infectivity or biological fitness is completely unclear to date. Here, we report the first evidence that the HKU5-CoV-2 receptor-binding domain (RBD) and S1/S2 furin cleavage site (FCS) of the spike protein are recombination hotspots. Using linkage disequilibrium

Virological characteristics of a SARS-CoV-2-related bat coronavirus, BANAL-20-236.

BACKGROUND: Although several SARS-CoV-2-related coronaviruses (SC2r-CoVs) were discovered in bats and pangolins, the differences in virological characteristics between SARS-CoV-2 and SC2r-CoVs remain poorly understood. Recently, BANAL-20-236 (B236) was isolated from a rectal swab of Malayan horseshoe bat and was found to lack a furin cleavage site (FCS) in the spike (S) protein. The comparison of its virological characteristics with FCS-deleted SARS-CoV-2 (SC2ΔFCS) has not been conducted yet. METHODS: We prepared human induced pluripotent stem cell (iPSC)-derived airway and lung epithelial ce

Field testing Australian bat lyssavirus risk communication resources.

ISSUE ADDRESSED: Australian bat lyssavirus (ABLV) is a fatal zoonosis, which can be transmitted to humans through scratches or bites from infected bats. Currently, there is a lack of research evaluating risk communication resources about ABLV or the dangers from handling bats. The purpose of this study was to field test resources aimed at educating the public about risks to humans and bats from human-bat interaction, then update these resources based upon feedback to ensure they were relevant and appropriately targeted to the public. METHODS: Thirteen semi-structured interviews with a purposi

Trypsin Treatment Unlocks Barrier for Zoonotic Bat Coronavirus Infection.

Traditionally, the emergence of coronaviruses (CoVs) has been attributed to a gain in receptor binding in a new host. Our previous work with severe acute respiratory syndrome (SARS)-like viruses argued that bats already harbor CoVs with the ability to infect humans without adaptation. These results suggested that additional barriers limit the emergence of zoonotic CoV. In this work, we describe overcoming host restriction of two Middle East respiratory syndrome (MERS)-like bat CoVs using exogenous protease treatment. We found that the spike protein of PDF2180-CoV, a MERS-like virus found in a

Modification of sperm fatty acid composition during epididymal maturation in bats.

Biochemical properties of polyunsaturated fatty acids (PUFAs) are fundamental to sperm movements. Amongst all adjustments operated during epididymal maturation, sperm membrane lipid composition is remodelled. Specifically, the proportion of PUFAs usually increases from the caput towards the cauda epididymidis. In mammals, PUFAs are predominantly acquired through the diet, which can consequently impact male fertility. We aimed at analysing to what extent n-6 and n-3 PUFAs are incorporated into sperm in the Seba's short-tailed bat (Carollia perspicillata), and at demonstrating the effect of the

Nipah virus - the rising epidemic: a review.

The Nipah virus was discovered twenty years ago, and there is considerable information available regarding the specificities surrounding this virus such as transmission, pathogenesis and genome. Belonging to the Henipavirus genus, this virus can cause fever, encephalitis and respiratory disorders. The first cases were reported in Malaysia and Singapore in 1998, when affected individuals presented with severe febrile encephalitis. Since then, much has been identified about this virus. These single-stranded RNA viruses gain entry into target cells via a process known as macropinocytosis. The vir

Evaluating the public health response to a mass bat exposure-Wyoming, 2017.

Mass bat exposures (MBEs) occur when multiple people are exposed to a bat or a bat colony, often over an extended period. In August 2017, a public health investigation was started in response to an MBE that occurred during May-August 2017 at a national park research station in Wyoming. We identified 176 people who had slept primarily in two lodges (Lodges A and B) at the research station, and successfully contacted 165 (93.8%) of these individuals. Risk assessments (RAs) were administered to all 165 individuals to determine degree and type of exposures to bats (e.g., biting or scratching). Exp

Combination drug treatment prolongs survival of experimentally infected mice with silver-haired bat rabies virus.

Rabies is a lethal disease in humans and animals, killing approximately 60,000 people every year. Currently, there is no treatment available, except post-exposure prophylaxis (PEP) that can be administered whenever exposure to a rabid animal took place. Here we describe the beneficial effects of a combination treatment initiated at day 4 post infection, containing anti-viral drugs and immune modulators in infected mice. Combination therapy resulted in significant increase in survival time (P < 0.05) and significantly lowers viral RNA in the brain and spinal cord (P < 0.05). Furthermore, treatm

SYN023, a novel humanized monoclonal antibody cocktail, for post-exposure prophylaxis of rabies.

Rabies is a neglected zoonotic disease that is preventable in humans by appropriate post-exposure prophylaxis (PEP). However, current PEP relies on polyclonal immune globulin products purified from pooled human (HRIG) or equine (ERIG) plasma that are either in chronic shortage or in association with safety concerns. Here, we present the development of an antibody cocktail, SYN023, made of two novel monoclonal antibodies (MAb) CTB011 and CTB012 that could serve as safer and more cost-effective alternatives to the current RIG products. Both CTB011 and CTB012 are humanized MAbs that bind to non-o

Rabies: Knowledge and Practices Regarding Rabies in Rural Communities of the Brazilian Amazon Basin.

BACKGROUND: The occurrence of outbreaks of human rabies transmitted by Desmodus rotundus in Brazil in 2004 and 2005 reinforced the need for further research into this zoonosis. Studies of knowledge and practices related to the disease will help to define strategies for the avoidance of new cases, through the identification of gaps that may affect the preventive practices. METHODOLOGY/PRINCIPAL FINDINGS: A semi-structured questionnaire was applied to 681 residents of twelve communities of northeastern Pará state involved in the 2004 and 2005 outbreaks mentioned above. The objective was to eval
