The human brainstem is involved in the regulation of the sleep/waking cycle and normal sleep architectonics and is crucial for the performance of a variety of somatomotor, vital autonomic, oculomotor, vestibular, auditory, ingestive and somatosensory functions. It harbors the origins of the ascending dopaminergic, cholinergic, noradrenergic, serotonergic systems, as well the home base of the descending serotonergic system. In contrast to the cerebral cortex the affection of the brainstem in Alzheimer's disease (AD) by the neurofibrillary or tau cytoskeletal pathology was recognized only approx

Addiction is characterized by maladaptive decision-making, a loss of control over drug consumption and habit-like drug seeking despite adverse consequences. These cognitive changes may reflect the effects of drugs of abuse on prefrontal cortical neurobiology. Here, we review evidence that amphetamine and cocaine fundamentally remodel the structure of excitatory neurons in the prefrontal cortex. We summarize evidence in particular that these psychostimulants have opposing effects in the medial and orbital prefrontal cortices ('mPFC' and 'oPFC', respectively). For example, amphetamine and cocain

Cryptococcosis by the encapsulated yeast Cryptococcus neoformans affects mostly immunocompromised individuals and is a frequent neurological complication in AIDS patients. Recent studies support the idea that intracellular survival of Cryptococcus yeast cells is important for the pathogenesis of cryptococcosis. However, the initial steps of Cryptococcus internalization by host cells remain poorly understood. Here, we investigate the mechanism of Cryptococcus neoformans phagocytosis by peritoneal macrophages using confocal and electron microscopy techniques, as well as flow cytometry quantifica

The ER is a continuous membrane system consisting of the nuclear envelope, flat sheets often studded with ribosomes, and a polygonal network … of tubular ER proteins including some REEPs and the M1 spastin ATPase interact with the microtubule cytoskeleton. Flat ER sheets possess a different complement of proteins such as p180, CLIMP-63 and kinectin implicated in shaping, cisternal stacking and cytoskeletal interactions. The ER is also in constant motion, and numerous signaling pathways as well as int …

We have studied the structural properties of human erythrocytes under the action of antibacterial substances of different nature (macrolides, fluoroquinolones, tetracyclines, lincosamides). It has been established that antibiotics modify the cytoplasmic skeleton of erythrocytes and induce their hemolytic efficiency. The character of changes in the osmotic resistance of erythrocytes depends on the chemical structure of modifiers.

A case of leiomyosarcoma of the scrotum is reported for its rarity. The tumor measured 6.2 x 4 x 4 cm and presented as a gradually increasing painless mass of 1 month duration. Light microscopy showed the typical interlacing fascicles of neoplastic cells with mitotic count of 1-2 / 10 HPF in different areas. Immunoperoxidase stains of vimentin & desmin and electron microscopy confirmed its muscle origin.

The interaction of weak electromagnetic fields (EMF) with living cells is a most important but still unresolved biophysical problem. For this interaction, thermal and other types of noise appear to cause severe restrictions in the action of weak signals on relevant components of the cell. A recently presented general concept of regulation of ion and substrate pathways through microvilli provides a possible theoretical basis for the comprehension of physiological effects of even extremely low magnetic fields. The actin-based core of microfilaments in microvilli is proposed to represent a cellul

Troponin-tropomyosin complex from skeletal muscles was observed to regulate sliding movement of actin filaments on myosin molecules in a manner independent of their ATPase activity. When actin molecules were crosslinked with DSS (disuccinimidyl suberate), the myosin ATPase activity in the presence of the modified actin filaments complexed with both troponin and tropomyosin was only 10% less than that in the case of unmodified actin, and the ATPase activation was independent of calcium ions. In contrast, the sliding velocity of the modified actin filaments on myosin molecules decreased to zero

Open biopsies were carried out on the muscles of 23 clubfoot patients, ages 9 months to 4 years. Electron microscopic analyses of the so-called "clubfoot muscles" and the peroneal muscles were performed. The changes found were not present in every area of the muscles, but were surrounded by fields of normal structure. Fatty degeneration with fibrosis was observed as the consequence of immobilization. More marked loss was found in the contractile elements. The authors believe that neuromuscular atrophy is a primary cause of congenital clubfoot. The most seriously affected muscles in this study

A 79-year-old white man of Irish descent presented with lattice corneal dystrophy, blepharochalasis, and peripheral seventh cranial nerve palsies. Family studies revealed that his 23-year-old daughter had early lattice cornea dystrophy. The corneal button removed by penetrating keratoplasty exhibited characteristic amyloid accumulation by light and electron microscopy. Biopsy of the patient's normal appearing conjunctiva and skin of the lower lid revealed amyloid. Biopsy of the daughter's conjunctiva was negative for amyloid, but her lid skin had characteristic amyloid deposits by light and el

The fine structure of the epicyte of D. gigantea was investigated. The motility of the gregarine and the contractile elements are described. Four essential types of movements can be observed in this gregarine: (1) rolling up and pendular movements, (2) locomotion by gliding forward, (3) cytoplasmic streaming (Fig. 1), (4) peristaltic contractions (Fig. 2) which seem to be accompanied by the contraction of annular myonemes (Fig. 2). The epicyte is formed by the folding of the parasitic cell wall which is made from three membranes (Figs. 3 and 4). At the top of each fold one can see apical strut

Monoclonal antibodies that recognize an epitope within the triple helix of type III collagen have been used to examine the distribution of that collagen type in human skin, cornea, amnion, aorta, and tendon. Ultrastructural examination of those tissues indicates antibody binding to collagen fibrils in skin, amnion, aorta, and tendon regardless of the diameter of the fibril. The antibody distribution is unchanged with donor age, site of biopsy, or region of tissue examined. In contrast, antibody applied to adult human cornea localizes to isolated fibrils, which appear randomly throughout the ma

When migratory T cells encounter antigen-presenting cells (APCs), they arrest and form radially symmetric, stable intercellular junctions termed immunological synapses which facilitate exchange of crucial biochemical information and are critical for T-cell immunity. While the cellular processes underlying synapse formation have been well characterized, those that maintain the symmetry, and thereby the stability of the synapse, remain unknown. Here we identify an antigen-triggered mechanism that actively promotes T-cell synapse symmetry by generating cytoskeletal tension in the plane of the syn

The beta-thymosin/WH2 actin-binding module shows an amazing adaptation to multifunctionality. The beta-thymosins are genuine G-actin sequesterers of moderate affinity for G-actin, allowing an efficient regulation of the G-actin/F-actin ratio in cells by amplifying changes in the critical concentration for filament assembly. In contrast, the first beta-thymosin domain of the protein Ciboulot makes with G-actin a complex that supports filament growth, such as profilin-actin. We illustrate how the use of engineered chimeric proteins, actin-binding and polymerization assays, crystallographic, NMR,

Stem cell lineage determination is governed not only by biochemical cues but also by the organization and force-generating state of the cytoskeleton, which integrates extracellular mechanical cues into transcriptional and epigenetic responses. Cytoskeletal organization and remodeling play essential roles in stem cell fate determination, as well as other relevant biological processes including development, tissue regeneration, and disease progression. In this review, we first outline the different types of cytoskeletal structures-actin filaments, microtubules, and intermediate filaments-and the…

Leukocyte recruitment is critical for tissue homeostasis and inflammatory responses. Chemokine-induced β2 integrin-dependent adhesion is the first and most critical step of leukocyte recruitment. Previous studies have shown that crosstalk between integrins and the actin cytoskeleton is vital for the migration of adherent leukocytes. However, whether and how the actin cytoskeleton contributes to integrin-dependent leukocyte adhesion remains poorly defined. As we recently demonstrated, a defect in integrin clustering, even in the presence of normal integrin activation, is sufficient to cause an …

CAR-T therapy holds immense potential for eradicating residual tumors and restoring postoperative immune surveillance. However, nutrient deprivation and impaired immune contact within the postoperative tumor microenvironment (PTME) severely hinder the therapeutic efficacy of CAR-T cells. Herein, we developed a self-reinforcing hydrogel system that dually modulated metabolism and immune contact to reprogram the PTME. Specifically, the c-Myc inhibitor JQ1 and bovine serum albumin (BSA)-decorated nanoparticles loaded with the actin cytoskeleton inhibitor NP-G2-044 (BPG) were encapsulated into a r…
