Correlation between somatomedin A in serum and body height development in healthy children and children with certain growth disturbances.

The somatomedin A level in serum was determined in a series of 66 children in whom longitudinal data on body height growth and dental development were available. The series comprised normal children as well as children with some different types of growth disturbances. A high correlation was found between the somatomedin A level and the age at a certain stage of the dental maturity (r equals to minus 0.67) and between the somatomedrin A level and the growth rate at this age (r equals to 0.67). The different groups of patients ranging from pituitary dwarfs to unusually tall children demonstrated

[Key points of the International consensus guidelines on the implementation and monitoring of vosoritide therapy in individuals with Achondroplasia].

Achondroplasia (ACH) is a common inherited skeletal dysplasia (inherited dwarfism) that compromises quality of life across the lifespan. In 2021, vosoritide became the first approved precision therapy for ACH and is now available in more than 40 countries. Compared with prior symptomatic measures, vosoritide has demonstrated favorable efficacy and a reassuring safety profile. Nevertheless, existing international ACH guidelines largely emphasize complication management and symptomatic care, and there is no unified consensus on pharmacologic therapy. To address this gap, an international expert …

MtMAP3Kδ6 Modulates the Growth and Development through Sugar Metabolism Regulation in Medicago truncatula.

Plant growth and development are intricately regulated by molecular mechanisms, with the mitogen-activated protein kinase (MAPK) signaling cascade and its associated modules being pivotal. In this study, we identified and characterized a member of the MAPKKK family, MtMAP3Kδ6, from Medicago truncatula. This gene, classified within the B3 subgroup of the MAPKKK family, was expressed across various tissues during plant growth. The knockout mutant of MtMAP3Kδ6 displayed dwarfism, characterized by reduced branching and smaller leaf size, whereas overexpression of MtMAP3Kδ6 in Medicago truncatula l…

Rheb1 is required for limb growth through regulating chondrogenesis in growth plate.

Ras homology enriched in the brain (Rheb) is well established as a critical regulator of cell proliferation and differentiation in response to growth factors and nutrients. However, the role of Rheb1 in limb development remains unknown. Here, we found that Rheb1 was dynamically expressed during the proliferation and differentiation of chondrocytes in the growth plate. Given that Prrx1+ limb-bud-like mesenchymal cells are the source of limb chondrocytes and are essential for endochondral ossification, we conditionally deleted Rheb1 using Prrx1-Cre and found a limb dwarfism in Prrx1-Cre; Rheb1fl…

Thanatophoric Dysplasia With Concurrent Hydroureteronephrosis: A Rare Case Report From Rural Southern India.

Thanatophoric dysplasia (TD) is a severe and typically fatal skeletal disorder caused by mutations in the FGFR3 gene, often leading to perinatal death. It is characterized by extreme short-limb dwarfism and, occasionally, associated anomalies such as hydronephrosis. Prenatal diagnosis, usually made in the third trimester through ultrasound and genetic testing, is crucial for guiding management decisions. Here, we report a case of TD with hydronephrosis diagnosed at 24 weeks of gestation in a 24-year-old primigravida from rural southern India. Ultrasound findings included significantly short an…

Anesthetic management of parturients with achondroplasia: a case series.

BACKGROUND: Achondroplasia is the most common form of dwarfism, and cesarean delivery is often required in parturients with achondroplasia due to cephalopelvic disproportion. Given the challenges for both regional and general anesthetic techniques, there is no consensus on the optimal anesthetic management for cesarean delivery in these patients. METHOD: A search of our electronic medical records for all female patients who had a diagnosis of achondroplasia and had a delivery in our health system from January 1, 2001 through June 16, 2023 was performed. Institutional review board exemption wa…

Whole exome sequencing identifies a novel variant causing cockayne syndrome type I in a consanguineous Pakistani family.

BACKGROUND: Cockayne syndrome (CS) is a rare neurodegenerative disorder characterized by impaired neurological functions, cachectic dwarfism, microcephaly and photosensitivity. Complementation assays identify two groups of this disorder, CS type I (CSA) and CS type II (CSB), caused by mutations in ERCC8 and ERCC6, respectively. OBJECTIVES: This study aimed to investigate the genetic basis of a consanguineous Pakistani family with three affected individuals presenting with typical clinical symptoms of CS. METHODS: We employed whole exome sequencing of the proband and then Sanger sequenced all…

Megacolon as a Feature of Suspected Robinow Syndrome.

This study presents the routine prosection findings of a 73-year-old male cadaver, with the cause of death reported to be hypertension and respiratory failure. Deep thorax and abdomen dissection exposed profound external and internal anatomical abnormalities. Externally, the body exhibited the following: pectus excavatum; short-limbed dwarfism; and abnormalities of the head, face, and external genitalia. Most of these findings suggest that the donor had Robinow syndrome, a rare genetic disorder involving developmental delay and skeletal abnormalities akin to those found in this cadaver. The in…
