Desvenlafaxine prevents white matter injury and improves the decreased phosphorylation of the rate-limiting enzyme of cholesterol synthesis in a chronic mouse model of depression.

Serotonin/norepinephrine reuptake inhibitors antidepressants exert their effects by increasing serotonin and norepinephrine in the synaptic cleft. Studies show it takes 2-3 weeks for the mood-enhancing effects, which indicate other mechanisms may underlie their treatment effects. Here, we investigated the role of white matter in treatment and pathogenesis of depression using an unpredictable chronic mild stress (UCMS) mouse model. Desvenlafaxine (DVS) was orally administrated to UCMS mice at the dose of 10 mg/kg/day 1 week before they went through a 7-week stress procedure and lasted for over

Resilience and reduced c-Fos expression in P2X7 receptor knockout mice exposed to repeated forced swim test.

There is considerable evidence suggesting genetic factors play an important role in the pathophysiology of depression, possibly by increasing susceptibility to repeated environmental stressors. Recent linkage studies have associated a polymorphism of the gene coding for the P2X7 receptor (P2X7R) with both major depressive disorder and bipolar disorder. Here we assessed whether P2X7 deletion affected the behavioural and neural response to repeated stress. P2X7R knockout (P2X7-/-) mice were subjected to the forced swim test for three consecutive days and neuronal activation in response to the th

Anxiolytic effect of wogonin, a benzodiazepine receptor ligand isolated from Scutellaria baicalensis Georgi.

The search for novel anxiolytics devoid of undesirable side-effects typical of classical benzodiazepines (BDZs) has been intense, and flavonoids, as a relative new class of ligands, have been shown to possess anxiolytic effects in vivo. The present study evaluated the pharmacological properties of a naturally occurring monoflavonoid, 5,7-dihydroxy-8-methoxyflavone or wogonin. The affinity (K(i)) of wogonin for the benzodiazepine site (BZD-S) on the gamma-aminobutyric acid(A) (GABA(A)) receptor complex was 0.92 microM. Using electrophysiological techniques, we showed that wogonin enhanced the G

Neuroprotective efficacy of Nyctanthes arbor-tristis in a rotenone-induced rat model of Parkinson's disease.

ETHNOPHARMACOLOGICAL RELEVANCE: Nyctanthes arbor-tristis is widely used in traditional medicine systems for its anti-inflammatory, antioxidant, and neuroprotective properties. It has been traditionally employed in the management of neurological and inflammatory disorders, supporting its potential role in neurodegenerative conditions such as Parkinson's disease. These therapeutic effects are attributed to its rich phytochemical profile, particularly iridoid glycosides (e.g., Arbortristoside A, nyctanthoside) and flavonoids (e.g., quercetin, kaempferol), which are known to modulate oxidative str

A repeated low-dose regimen of MDMA has transient next-day effects on locomotor activity, anxiety-like behavior, and brain serotonin levels, with no effect on anhedonia-like behavior, in both female and male rats.

RATIONALE: MDMA (3–4 methylenedioxymethamphetamine) assisted psychotherapy has gained considerable attention as a potential adjuvant therapy for post-traumatic stress disorder (PTSD). Reports of the nonmedical use of MDMA suggest its adverse effects may include anxiety and anhedonia. However, the time course of these signs and symptoms and the potential alterations in neurotransmitter levels that may underlie these conditions are not well understood. METHODS: We subjected cohorts of male and female Sprague Dawley rats to MDMA (2.5 or 5 mg/kg) or saline (1 ml/kg), with each dose given three tim

The Combination of Lactobacillus reuteri RC-14® and Lactobacillus rhamnosus GR-1® Induces Anxiolytic-like and Antidepressant-like Effects via Estrogenic Receptors in Ovariectomized Rats.

Background/Objetives: Menopause increases anxiety and depression risk, linked to gut microbiota changes. Probiotics show psychobiotic effects that could therapeutically alleviate these symptoms in menopausal women. However, this potential and its mechanism of action involving estrogen receptors remain largely unexplored. To investigate whether Lactobacillus reuteri RC-14® and Lactobacillus rhamnosus GR-1® exert anxiolytic-like and antidepressant-like effects in ovariectomized rats via estrogenic receptor (ER) activation. Methods: Ovariectomized adult female Wistar rats were divided into four g

Mechanistic Study of Schisandrin B in Alleviating Anxiety-Like Behaviors in Rats via Enhancement of Hippocampal Synaptic Plasticity and Restoration of the Intestinal Barrier.

This study focused on examining the anxiolytic effects of Schisandrin B (SchB) in rats exhibiting anxiety-like behaviors and investigating the mechanisms involved. Behavioral assessments were conducted using the elevated plus-maze test and the open field test. The results showed that Schisandrin B at a dose of 10 mg/kg significantly alleviated anxiety-like behaviors in rats, with an efficacy comparable to that of the positive control drug diazepam. Mechanistic studies indicate that, at the central level, Schisandrin B effectively maintains the homeostasis of the glutamatergic system in the hip

Striatal injection of aminochrome in Wistar rats induces early-stage behavioral changes of Parkinson's disease and dopaminergic degeneration in the caudal and medial regions of the nigra pars compacta.

Most preclinical Parkinson's disease (PD) models use neurotoxic agents to cause rapid dopaminergic neuron degeneration, mimicking the late stage of PD. That creates a gap in understanding early-stage pathophysiology, critical for neuroprotective therapies and early diagnosis. To replicate the prodromal stage of PD, it is pivotal that preclinical study models promote a slow and selective death of dopaminergic neurons, triggering degenerative processes and early symptoms. In this context, we investigated behavioral and neuronal changes using a model of unilateral aminochrome injection (6 nmol/6

Loss of choroid plexus-derived insulin-like growth factor 2 (IGF2) leads to hyposmia, while retaining post-partum mood resilience in mice.

During the post-partum period, new mothers are vulnerable to mood disorders. In adults, impairments in neurogenesis commonly associate with anxiety and depressive behaviors. Insulin-like growth factor 2 (IGF2) is expressed in the choroid plexus (CP) within the subventricular zone (SVZ) neurogenic niche, and global loss of IGF2 leads to increased anxiety. Previously, we have shown that Igf2 expression in CP tissue increases 6-fold during lactation but returns to baseline on suppression of prolactin present in lactation, suggesting it is induced by high levels of prolactin. Peripherally administered amylin inhibits stress-like behaviors and enhances cognitive performance.

Amylin, a 37 amino acid peptide pancreatic hormone co-secreted with insulin, normalizes the altered eating patterns induced by chronic stress in the rat. Because these stress-induced changes are driven, in part, by brain corticotropin-releasing factor and corticosterone, and because alterations in the activity of these molecules and the stress system are commonly associated with neuropsychiatric diseases like anxiety, depression, and schizophrenia, we hypothesized that amylin might mitigate behavioral states associated with stress. Therefore, we tested the effects of rat amylin in rodent-based

Intra-accumbal orexin-1 receptor inhibition prevents the anxiolytic-like effect of ethanol and leads to increases in orexin-A content and receptor expression.

Alcohol use is frequently associated with mood disorders. Similarly, individuals suffering from these disorders have a higher risk of developing alcoholism. Several reports have implicated orexin signaling in different behaviors related to alcohol consumption, whereas antagonists block these actions. However, the involvement of orexin-1-receptor (Orx1R) in ethanol-induced anxiolysis remains relatively unexplored. The purpose of this study was to investigate whether intra-accumbal inhibition of Orx1R blocks the anxiolytic-like effect of ethanol and to determine if ethanol administration modifie

Weekday-only chronic oral methylphenidate self-administration in male rats: Reversibility of the behavioral and physiological effects.

Methylphenidate (MP) is a commonly prescribed psychostimulant for Attention Deficit Hyperactivity Disorder (ADHD). We recently reported behavioral and developmental effects of chronic MP use in healthy rats. The current study investigated how interrupting chronic MP treatment with weekend abstinence altered the behavioral and physiological consequences of chronic MP treatment, and if prolonged abstinence would reverse the observed effects. Male Sprague Dawley rats were assigned to one of three treatment groups: water (W); low dose (LD) MP; and high dose (HD) MP. For 13 weeks, rats had access t

Palmitoylethanolamide Ameliorates Hippocampal Damage and Behavioral Dysfunction After Perinatal Asphyxia in the Immature Rat Brain.

Perinatal asphyxia (PA) is an obstetric complication associated with an impaired gas exchange. This health problem continues to be a determinant of neonatal mortality and neurodevelopmental disorders. Palmitoylethanolamide (PEA) has exerted neuroprotection in several models of brain injury and neurodegeneration. We aimed at evaluating the potential neuroprotective role of PEA in an experimental model, which induces PA in the immature rat brain. PA was induced by placing Sprague Dawley newborn rats in a water bath at 37°C for 19 min. Once their physiological conditions improved, they were given
