A sodium channel variant in Aedes aegypti as a candidate pathogen sensor for viral-associated molecular patterns.

Recent work demonstrated that a splice variant of a human macrophage voltage-gated sodium channel expressed on endosomes acts as an intracellular sensor for dsRNA, a viral-associated molecular pattern. Here our goal was to identify a candidate gene in a clinically relevant invertebrate model with related cellular and pattern recognition properties. The para gene in drosophila and other insects encodes voltage-gated sodium channels with similar electrophysiological properties to those found in vertebrate excitable membranes. A database search revealed that the AAEL006019 gene in Aedes aegypti,

The challenge of treating orphan disease.

Uncommon or orphan diseases are less frequently addressed in mainstream medical journals and, as a consequence, their understanding and clinical recognition may rely on case series or anecdotal data with limited guidelines and management directions. The study of selected underrepresented autoimmune and allergy conditions is the subject of the present issue of Clinical Reviews in Allergy and Immunology to provide peculiar perspectives on common and rare themes. First, allergy remains a major concern for physicians worldwide despite the limited developments over the past years, particularly for

When less is more: primary immunodeficiency with an autoinflammatory kick.

PURPOSE OF REVIEW: Next-generation sequencing is revolutionizing the molecular taxonomy of human disease. Recent studies of patients with unexplained autoinflammatory disorders reveal germline genetic mutations that target important regulators of innate immunity. RECENT FINDINGS: Whole-exome analyses of previously undiagnosed patients have catalyzed the recognition of two new disease genes. First, a phenotypic spectrum, including livedo racemosa, fever with early-onset stroke, polyarteritis nodosa, and Sneddon syndrome, is caused by loss-of-function mutations in cat eye syndrome chromosome re

Susceptibility of mice genetically deficient in SP-A or SP-D gene to invasive pulmonary aspergillosis.

Pulmonary surfactant proteins, SP-A and SP-D, are carbohydrate pattern recognition molecules of innate immunity, which significantly enhance phagocytosis and killing of Aspergillus fumigatus, a pathogenic fungus, by neutrophils and macrophages. The present study examined the susceptibility of immunosuppressed SP-A gene deficient (SP-A(-/-)) or SP-D gene deficient (SP-D(-/-)) mice to A. fumigatus conidia challenge compared to wild-type (WT) mice. A. fumigatus-challenged SP-A(-/-) (SP-A(-/-) IPA) mice showed less mortality (40%) than the WT-IPA mice (100%) and increased mortality (60%) following

Alpha1,3-galactosyltransferase gene-knockout pigs for xenotransplantation: where do we go from here?

The ability to genetically engineer pigs that no longer express the Galalpha1,3Gal (Gal) oligosaccharide has been a significant step toward the clinical applicability of xenotransplantation. Using a chronic immunosuppressive regimen based on costimulatory blockade, hearts from these pigs have survived from 2 to 6 months in baboons. Graft failure was predominantly from the development of a thrombotic microangiopathy. Potential contributing factors include the presence of preformed anti-nonGal antibodies or the development of low levels of elicited antibodies to nonGal antigens, natural killer (

Unculturable bacteria exploit a secretory protein to antagonize insect melanization for persistent infection.

Phloem-inhabiting unculturable bacterial pathogens are persistently transmitted by insect vectors. However, how they evade insect immune responses to ensure persistent transmission remains unknown. The important melanization immune response in insects is triggered by cleavage of prophenoloxidase (PPO) into active phenoloxidase (PO) via clip-domain serine proteases (CLIPs). Here, we demonstrate that infection of Candidatus Liberibacter asiaticus (CLas) in psyllid vectors activates the peptidoglycan recognition protein (PGRP)-CLIP1-CLIP4-PPO-PO signaling cascade to induce a mild melanization res

Designing biomaterials for the modulation of allogeneic and autoimmune responses to cellular implants in Type 1 Diabetes.

The effective suppression of adaptive immune responses is essential for the success of allogeneic cell therapies. In islet transplantation for Type 1 Diabetes, pre-existing autoimmunity provides an additional hurdle, as memory autoimmune T cells mediate both an autoantigen-specific attack on the donor beta cells and an alloantigen-specific attack on the donor graft cells. Immunosuppressive agents used for islet transplantation are generally successful in suppressing alloimmune responses, but dramatically hinder the widespread adoption of this therapeutic approach and fail to control memory T c

Phosphoethanolamine-modified lipid A enables antimicrobial resistance and atypical signaling properties in the marine pathogen P. damselae subsp. damselae.

UNLABELLED: Lipid A, the immunostimulatory component of lipopolysaccharide (LPS), plays a multitude of roles in host-pathogen interactions. It functions as both a key antigen recognized by innate immune receptors and an essential structural element of the gram-negative bacterial outer membrane. Consequently, gram-negative bacteria exposed to stressful environments can modify their lipid A structure to preserve membrane integrity, evade immune cell recognition, and avoid disruption by antimicrobial agents. In this study, we isolated a strain (SF21) from a nurse shark stool, which was identified

Enterovirus D68 3C protease antagonizes type I interferon signaling by cleaving signal transducer and activator of transcription 1.

Following the successful control of poliovirus, the re-emergence of respiratory enterovirus D68 (EV-D68), a prominent non-polio enterovirus, has become a serious public health concern worldwide. Host innate immune responses are the primary defense against EV-D68 invasion; however, the mechanism underlying viral evasion of the antiviral activity of interferons (IFN) remains unclear. In this study, we found that EV-D68 inhibited type I IFN signaling by cleaving signal transducer and activator of transcription 1 (STAT1), a crucial factor in cellular responses to interferons and other cytokines. Microbiome-innate immune crosstalk in acute exacerbation of idiopathic pulmonary fibrosis: an amplification framework.

Acute exacerbation of idiopathic pulmonary fibrosis (AE-IPF) remains a major cause of short-term mortality, yet its biological drivers are incompletely understood. Increasing evidence indicates that the distal lung is not sterile and that microbial burden, ecological disruption, and host immune responses may interact within the fibrotic niche. Here, we summarize current evidence and propose a focused, hypothesis-generating framework for microbiome-immune crosstalk in AE-IPF. In this model, pulmonary dysbiosis and persistent microbial stimulation may converge with epithelial injury-derived dang

Neuraminidase A controls pneumococcal recognition and fate: deficiency enhances immune sensing and intracellular survival, while treatment promotes phagocytic clearance.

Streptococcus pneumoniae (pneumococcus) is a leading cause of bacterial meningitis worldwide and is associated with cerebrovascular complications and long-term neurological sequelae. These outcomes are largely driven by an excessive yet ineffective neuroinflammatory response. Although pneumococci express multiple virulence factors that enable immune evasion, how these factors modulate phagocytic responses in the central nervous system remains unclear. Here, we identify neuraminidase A (NanA) as a critical regulator of phagocytic activation in microglia and macrophages. Using murine and human m

Rhamnolipids elicit defense responses and induce disease resistance against biotrophic, hemibiotrophic, and necrotrophic pathogens that require different signaling pathways in Arabidopsis and highlight a central role for salicylic acid.

Plant resistance to phytopathogenic microorganisms mainly relies on the activation of an innate immune response usually launched after recognition by the plant cells of microbe-associated molecular patterns. The plant hormones, salicylic acid (SA), jasmonic acid, and ethylene have emerged as key players in the signaling networks involved in plant immunity. Rhamnolipids (RLs) are glycolipids produced by bacteria and are involved in surface motility and biofilm development. Here we report that RLs trigger an immune response in Arabidopsis (Arabidopsis thaliana) characterized by signaling molecul

Toll-like receptor agonists as adjuvants for HIV vaccines.

Toll-like receptors (TLRs) are pattern-recognition receptors responsible for detecting invading pathogens. About 13 TLRs are currently known to be expressed (see Table 1). TLR2 detects lipotechoic acid and bacterial lipoproteins, TLR4 recognizes LPS, TLR5, flagellin and TLR3 detects double-stranded RNA. The unmethylated CPG DNA of bacteria and viruses is detected by TLR9. TLR7 recognizes single-stranded RNA of viruses. TLR 11 in mice recognizes profillin from Toxoplasma gondii. Binding to TLRs expressed on dendritic cells (DCs) can trigger adaptive immune responses and DCs thus serve as a brid

Immunological and molecular signatures of carbamazepine-induced maculopapular exanthema.

Cutaneous adverse drug reactions to carbamazepine range from mild maculopapular exanthema to life-threatening syndromes such as Stevens-Johnson syndrome/toxic epidermal necrolysis. While the immunological basis for HLA-B*15:02-restricted carbamazepine-induced SJS/TEN has received greater attention, mechanisms underlying mild maculopapular exanthema remain poorly understood. In this study, we examined circulating T-cell immune recognition and transcriptomic signatures in the blood of patients with carbamazepine-induced maculopapular exanthema during active and resolved disease and compared thes

β-1,3-glucan-lacking Aspergillus fumigatus mediates an efficient antifungal immune response by activating complement and dendritic cells.

Complement system and dendritic cells (DCs) form - beside neutrophils and macrophages - the first line of defense to combat fungal infections. Therefore, we here studied interactions of these first immune elements with Aspergillus fumigatus lacking ß-1,3-glucans (fks1tetOnrep under repressed conditions) to mechanistically explain the mode of action of echinocandins in more detail. Echinocandins are cell wall active agents blocking β-glucan synthase, making the A. fumigatus fks1tetOn mutant a good model to study immune-modulatory actions of these drugs. We now demonstrate herein, that complemen

Bridging Autoimmunity and Hyperinflammation: Shared Immunological Pathways Among Autoimmune/Inflammatory Syndrome Induced by Adjuvants, Systemic Lupus Erythematosus, and Macrophage Activation Syndrome.

Autoimmune/inflammatory syndrome induced by adjuvants (ASIA) is characterized by immune dysregulation associated with exposure to exogenous substances with immunostimulatory properties, particularly in susceptible individuals. Systemic lupus erythematosus (SLE) is a complex and heterogeneous autoimmune disease characterized by loss of immune tolerance, autoantibody production, and immune complex deposition. Macrophage activation syndrome (MAS) is a life-threatening hyperinflammatory syndrome characterized by excessive activation of macrophages and T lymphocytes and may occur as a severe compli

Expression and immune recognition of SV40 Tag in transgenic mice that develop metastatic osteosarcomas.

Mature adult mice of the C57BL/6-TgN(Amy1TAg)501Knw transgenic mouse lineage, 501, containing a liver alpha-amylase promoted-SV40 Tag hybrid gene, routinely develop SV40 Tag-induced metastatic osteosarcomas. This form of alpha-amylase was known to be expressed in the liver, salivary glands, pancreas, and fat. Cells in the normal rib adjacent to the periosteum also express alpha-amylase suggesting that transgene expression is correctly targeted to generate osteosarcomas. 501 mice express SV40 Tag in the salivary glands but do not develop abnormalities in these organs by the time of their death

Rhino-orbito-cerebral mucormycosis in new-onset type 1 diabetes mellitus: a case-based short review.

Mucormycosis is an aggressive angioinvasive fungal infection that commonly involves the paranasal sinuses, orbit, lungs, and central nervous system (CNS). Patients with diabetic ketoacidosis (DKA) are uniquely susceptible because hyperglycemia and acidosis enhance fungal invasion and blunt innate immune defenses. We present a brief vignette of rhino-orbito-cerebral mucormycosis in a patient with recent diagnosis of type 1 diabetes mellitus (DM), and use it to frame a short review of epidemiology, pathogenesis, diagnosis, and management. Rapid recognition, urgent surgical debridement, prompt in

Adenovirus phagocytosis by neutrophils triggers a pro-inflammatory response.

Adenoviruses are common pathogens that have been engineered and used for medical purposes. While their recognition by innate immune cells such as macrophages and dendritic cells is well characterized, interactions with neutrophils remain poorly understood. Using cytometry, confocal and electron microscopy, we showed that neutrophils bind to antibody-coated adenoviruses and engulf them in a phagosome. Single-cell transcriptomic approach reveals that adenovirus phagocytosis activates a specific transcriptional program in neutrophils. It also triggers calcium entry, reactive oxygen species product

Spatial proteomics identifies a CRTC-dependent viral signaling pathway that stimulates production of interleukin-11.

Appropriate cellular recognition of viruses is essential for the generation of an effective innate and adaptive immune response. Viral sensors and their downstream signaling components thus provide a crucial first line of host defense. Many of them exhibit subcellular relocalization upon activation, resulting in the expression of interferon and antiviral genes. To comprehensively identify signaling factors, we analyzed protein relocalization on a global scale during viral infection. cAMP-responsive element-binding protein (CREB)-regulated transcription coactivators 2 and 3 (CRTC2/3) exhibited
