Outdoor air pollution exposures and micronuclei frequencies in lymphocytes from pregnant women and newborns in Crete, Greece (Rhea cohort).

BACKGROUND: Micronuclei (MN) are biomarkers of early genetic effects that have been used to investigate the association between environmental exposures and cancer. However, few studies have examined the association between environmental exposures during pregnancy and MN in mothers and newborns. OBJECTIVES: We examined MN frequency in maternal blood and in cord blood, in relation to maternal air pollution exposure, and the potential interaction with maternal vitamin C intake and maternal smoking. METHODS: We used the cytokinesis-block micronucleus assay to assess MN frequency per 1000 bi-nucl

Intrauterine exposure to HBeAg exerts modest effect on neonatal immune repertoire.

BACKGROUND: The paucity of information on fetal immune repertoire dynamics following intrauterine hepatitis B related antigens exposure precludes elucidation of its effects on fetal immune system. METHODS: Totally 34 cord blood (CB) samples from singleton full-term neonates and 29 paired maternal peripheral blood (PB) samples were collected at delivery and divided into four groups based on maternal hepatitis B condition and antiviral intervention history. Clonal information in complementary determining region 3 (CDR3) of T cell receptor β (TCRβ) chain and B cell receptor (BCR) heavy chain enc

Prenatal SSRI exposure alters neonatal corticosteroid binding globulin, infant cortisol levels, and emerging HPA function.

BACKGROUND: Serotonin influences the development of the hypothalamic-pituitary-adrenal (HPA) system; therefore prenatal exposure to selective serotonin reuptake inhibitor antidepressants (SSRIs) may alter HPA axis development and function. To address this, prenatal exposure to SSRIs and maternal mood were examined in relation to neonatal and infant levels of cortisol and its binding protein, corticosteroid-binding globulin (CBG). METHODS: Serum cortisol and CBG levels were assayed from SSRI-exposed and non-exposed mothers and their neonates at delivery. Maternal mood symptoms were documented

Mercury exposure, but not lead, increases risk of gestational diabetes mellitus: a meta-analysis.

Gestational diabetes mellitus (GDM) is a growing health problem causing a higher risk of delivering large infants and experiencing maternal and neonatal mortality. Studies have found a link between increased levels of mercury (Hg) and lead (Pb) in the blood and a higher risk of developing GDM. However, there are other reports contradicting the aforementioned findings. In this meta-analysis, we investigated the association between exposure to Hg and Pb and the elevated risk of being diagnosed with GDM. To that end, a systematic search was performed through Google Scholar, Web of Science, PubMed

Year 116 of the plastic age: a Pandora's box as a time bomb for pregnancy? Review of clinical and fundamental data on prenatal exposure to plastics.

Plastics constitute an area of interest within the context of the placental exposome. A growing body of evidence now indicates that various … etabolic and immune functions may contribute to abnormal development of the placenta and the fetus. Maternal exposure to these ubiquitous environmental pollutants may induce prenatal and neonatal disease states. In this review, we examine the current clinical, in vivo and in vitro data on the occurrence, distribution and impact of micro- and nanoplastics in the pla …

The effects of in utero exposure to teratogens on organ size: a prospective paediatric study.

In low-income countries, prospective data on combined effects of in utero teratogen exposure are lacking and necessitates new research. The aim of the present study was to explore the effect of in utero teratogen exposure on the size of the kidneys and pancreas 5 years after birth in a low-income paediatric population. Data was collected from 500 mother-child pairs from a low-income setting. Anthropometric measurements included body weight, (BW) body height, mid-upper arm and waist circumference (WC). Clinical measurements included blood pressure (BP), mean arterial pressure and heart rate. Ul

Air pollution and the fetal origin of disease: A systematic review of the molecular signatures of air pollution exposure in human placenta.

BACKGROUND: Fetal development is a crucial window of susceptibility in which exposure-related alterations can be induced on the molecular level, leading to potential changes in metabolism and development. The placenta serves as a gatekeeper between mother and fetus, and is in contact with environmental stressors throughout pregnancy. This makes the placenta as a temporary organ an informative non-invasive matrix suitable to investigate omics-related aberrations in association with in utero exposures such as ambient air pollution. OBJECTIVES: To summarize and discuss the current evidence and d

Folic acid fortification and the birth prevalence of congenital heart defect cases in Alberta, Canada.

BACKGROUND: Congenital heart defects (CHDs) are the most common type of congenital anomaly. The precise etiology is unknown and the development of successful primary prevention strategies is challenging. Folic acid may have a protective role; however published results have been inconsistent. This study examines the impact of mandatory folic acid fortification (FAF) on the prevalence of CHDs. METHODS: CHD cases were ascertained using the Alberta Congenital Anomalies Surveillance System, Pediatric Cardiology Clinics, Pathology, and hospital records. The birth prevalence and odds ratios (OR) of

Is maternal smoking more determinant than paternal smoking on the respiratory symptoms of young children?

BACKGROUND: It is generally accepted that maternal smoking has more detrimental effects than paternal smoking on the respiratory health of children. The objective of this study was to assess the effects of postnatal exposure due to the smoking behaviour by father and mother in the home, and prenatal exposure from maternal smoking during pregnancy, on the respiratory symptoms in children. METHODS: The parents of 484 children, aged 3-6 years, completed a questionnaire about smoking and respiratory symptoms in children. Bivariate and logistic regression analyses were used to examine the associat

Effects of early deprivation and maternal separation on pup-directed behavior and HPA axis measures in the juvenile female rat.

Juvenile female rats show maternal-like behavior toward pups. The purpose of the following experiment was to investigate whether the HPA axis, through the use of early separation manipulations that alter HPA functioning in rats, plays a role in the juvenile response to foster pups. Female rats were early deprived or maternally separated for 5 hours daily from PND 2 to 14 and compared to animal facility-reared rats. Deprivation or separation increased CRH-R1 IR in the juvenile PVN, but had no other effects on other HPA measures or on maternal behavior. Pup-exposure during the juvenile period bl

Estimating fetal exposure to the P-gp substrates, corticosteroids, by PBPK modeling to inform prevention of neonatal respiratory distress syndrome.

We have previously developed a maternal-fetal physiologically-based pharmacokinetic (m-f PBPK) model to dynamically predict (and verify) fetal-maternal exposure to drugs that passively diffuse across the placenta. Here, we extended the application of this model to dynamically predict fetal exposure to drugs which are effluxed by placental P-glycoprotein, namely the antenatal corticosteroids (ACS; dexamethasone [DEX], and betamethasone [BET]). To do so, we estimated both the placental P-gp mediated efflux clearance (CL) and the passive diffusion CL of the ACS. The efficacy and toxicity of the c…

Titanium dioxide nanoparticles oral exposure to pregnant rats and its distribution.

BACKGROUND: Titanium dioxide (TiO2) nanoparticles are among the most manufactured nanomaterials in the industry, and are used in food products, toothpastes, cosmetics and paints. Pregnant women as well as their conceptuses may be exposed to TiO2 nanoparticles; however, the potential effects of these nanoparticles during pregnancy are controversial, and their internal distribution has not been investigated. Therefore, in this study, we investigated the potential effects of oral exposure to TiO2 nanoparticles and their distribution during pregnancy. TiO2 nanoparticles were orally administered to

Developmental toxicity studies in Crl:CD (SD) rats following inhalation exposure to trichloroethylene and perchloroethylene.

The potential for trichloroethylene (TCE) and perchloroethylene (PERC) to induce developmental toxicity was investigated in Crl:CD (SD) rats whole-body exposed to target concentrations of 0, 50, 150 or 600 ppm TCE or 0, 75, 250 or 600 ppm PERC for six hours/day, seven days/week on gestation day (GD) 6-20 and 6-19, respectively. Actual chamber concentrations were essentially identical to target with the exception of the low PERC exposure level, which was 65 ppm. The highest exposure levels exceeded the limit concentration (2 mg/L) specified in the applicable test guidelines. Maternal necropsies

Association between prenatal exposure to indoor air pollution and autistic-like behaviors among preschool children.

Indoor air pollution is a recognized risk factor for a range of negative health outcomes. This study aimed to investigate the association between maternal prenatal exposure to indoor air pollution and the presence of autistic-like behaviors among preschool children. Data were obtained from the Longhua Child Cohort Study in 2017, in which we enrolled a total of 65 317 preschool children. Associations between maternal exposure to four sources of indoor air pollution (e.g., cooking, environmental tobacco smoke (ETS), mosquito coils, and home decoration) during pregnancy and preschool children's a…

Role of low-dose cadmium exposure to the pathogenesis of gestational diabetes mellitus.

Gestational diabetes mellitus (GDM) is caused by the imbalance between insulin-resistance hormones and insulin secretion. The association between prenatal cadmium (Cd) exposure and GDM remains unclear. Furthermore, how Cd contributes to GDM, especially in the perspective of placental development, is largely unknown. In this study, we aimed to define the association of blood Cd concentrations with GDM incidence based on a birth cohort (462 GDM cases and 924 matched controls), and unravel the mechanism of low-dose Cd in GDM development by applying trophoblast organoids and syncytiotrophoblasts (

[Prenatal exposure to organochlorine pesticides and cryptorchidism].

Fetuses and children are more susceptible to the effects of environmental toxins. The objective of this article is to determine the levels of organochlorine pesticides (HCB, ss-HCH, pp'DDT, op'DDT and pp'DDE) in the serum lipids of mothers of newborns with cryptorchidism and compare the levels to a control group of mothers of newborns with descended testicles. The cases were composed of newborns with cryptorchidism (n=41), and the controls (n=41) newborns with descended testicles. Blood samples from both groups of mothers were used to determine the organochlorine pesticide levels. Cryptorchidi
