Histaprodifens: synthesis, pharmacological in vitro evaluation, and molecular modeling of a new class of highly active and selective histamine H(1)-receptor agonists.

A new class of histamine analogues characterized by a 3, 3-diphenylpropyl substituent at the 2-position of the imidazole nucleus has been prepared outgoing from 4,4-diphenylbutyronitrile (4b) via cyclization of the corresponding methyl imidate 5b with 2-oxo-4-phthalimido-1-butyl acetate or 2-oxo-1,4-butandiol in liquid ammonia, followed by standard reactions. The title compounds displayed partial agonism on contractile H(1) receptors of the guinea-pig ileum and endothelium-denuded aorta, respectively, except 10 (histaprodifen; 2-[2-(3, 3-diphenylpropyl)-1H-imidazol-4-yl]ethanamine) which was a

Novel Insights on Benzo[b]thiophene Analogues for MAO-B Inhibition and Neuroprotection: Design, Synthesis, Molecular Modelling Studies and Biological Activity.

Neurodegenerative disorders (NDs), such as Alzheimer's disease and Parkinson's disease (PD), represent a significant challenge for ageing populations, with their prevalence increasing worldwide. Elevated human Monoamine Oxidase B (hMAO-B) activity has been related to neurodegenerative progression, where it contributes, among others, to oxidative stress and neuroinflammation. The identification and optimization of selective hMAO-B inhibitors is therefore pivotal in addressing the progression of NDs. In this work we introduced 2-aroylbenzothiophene analogues as promising agents to mitigate neuro

Palmitoylation-related lncRNAs link molecular signaling, immune remodeling, and tumor progression in glioma: prognostic modeling and functional validation of LYRM4-AS1.

BACKGROUND: Glioma is a molecularly heterogeneous tumor of the central nervous system with variable clinical behavior, immune contexture, and therapeutic response. Palmitoylation is a reversible lipid modification that regulates membrane localization, synaptic signaling, inflammatory pathways, and oncogenic signaling; however, the prognostic and functional relevance of palmitoylation-related long non-coding RNAs (PRlncRNAs) in glioma remains incompletely defined. METHODS: Transcriptomic and clinical data for 429 glioma patients were obtained from TCGA. Thirty palmitoylation-related genes were

Ex vivo and computational investigation of corneal iontophoresis to enhance penetration of high-molecular-weight compounds: a study using albumin as a model molecule.

Efficient ocular drug delivery remains a major challenge due to the strong barrier properties of the corneal epithelium and the rapid clearance of topically applied formulations. This limitation is particularly critical for hydrophilic and high-molecular-weight (HMW) therapeutics such as proteins and biologics. In this study, we investigated transcorneal iontophoresis as a strategy to enhance the delivery of HMW compounds using albumin (66 kDa) as a model macromolecule. Ex vivo rabbit corneas were exposed to iontophoretic currents ranging from 0.5 to 7 mA for clinically relevant conditions, wh

Machine learning-based construction of an oxidative stress-related model reveals molecular subtypes and tumor microenvironment infiltration signatures in colon cancer.

BACKGROUND: Colon cancer is a common malignant tumor of digestive system. More and more research has shown that oxidative stress plays an important role in the development and progression of colon cancer. Oxidative stress occurs when redox homeostasis in cells is broken which is usually accompanied by overproduction of reactive oxygen species (ROS). METHODS: In this study, we constructed a colon cancer prognostic model based on oxidative stress-related (OSR) genes. Lasso regression and multivariable cox proportional hazards were performed to identify the best gene signature. We established the

Kinetics of Polymorphic Phase Transformations of o-Aminobenzoic Acid: Application of a Dispersive Kinetic Model Plus Molecular Dynamics Simulation of Prenucleation Aggregates.

The specific rate at which one crystalline phase converts to another can vary as a function of time under isothermal conditions. This behavior gives rise to sigmoidal kinetic transients that are characteristic of nucleation and growth. Such curves are commonly fitted using the Johnson-Mehl-Avrami-Erofe'ev-Kolmogorov (JMAEK) equation. However, due to the ambiguity surrounding the time exponent in the JMAEK model, we present an alternative two-parameter dispersive kinetic model (DKM) and apply it to the study of solution-mediated polymorphic conversions of the prototypical molecule, o-aminobenzo

In vitro cell model to dilucidate the underlying molecular mechanism associated with ophthalmic manifestation of congenital disorders of glycosylation: studying an ALG2-CDG patient.

INTRODUCTION: Congenital Disorders of Glycosylation (CDG) are severe disruptions in the synthesis of glycoconjugates, resulting in inherited metabolic conditions. These multisystem diseases, typically inherited in an autosomal recessive manner, have an occurrence rate of approximately 1 in 20,000 to 1 in 50,000 live births. The clinical presentation of CDG is highly varied and complex, with neurological symptoms being predominant, affecting multiple organ systems. The process of glycosylation, a critical post-translational modification, is tightly controlled by proteins encoded by over 250 gen

Multi-omics analysis and single-cell sequencing revealed the lysosome associated molecular subtypes and prognostic model development of papillary thyroid carcinoma.

Papillary thyroid carcinoma (PTC) is the most common endocrine carcinoma in recent years, necessitating more precise risk stratification to accurately identify low-risk patients. Although preliminary evidence exists, studies on lysosomes in PTC are limited. This study utilized multi-omics data from the TCGA database to comprehensively investigate the genomic and biological characteristics of lysosomes in PTC patients and identify lysosome-associated genes (LAGs) linked to PTC prognosis. We developed a LAG scoring system for risk stratification based on the expression levels of risk coefficient

Integrative analysis of patient-derived tumoroids and ex vivo organoid modelling of ARID1A loss in bladder cancer reveals therapeutic molecular targets.

Somatic mutations in ARID1A (AT-rich interactive domain-containing protein 1A) are present in approximately 25 % of bladder cancers (BC) and are associated with poor prognosis. With a view to discover effective treatment options for ARID1A-deficient BC patients, we set out to identify targetable effectors dysregulated consequent to ARID1A deficiency. Integrative analyses of ARID1A depletion in normal organoids and data mining in publicly available datasets revealed upregulation of DNA repair and cell cycle-associated genes consequent to loss of ARID1A and identified CHEK1 (Checkpoint kinase 1)
