CDK8 phosphorylates SOX2 to maintain stemness of pituitary adenoma.

Pituitary adenomas (PAs) are intracranial tumours with severe clinical complications and increased morbidity. Stem cell-like characteristics play a crucial role in the initiation and progression of PAs. In this study, we identified CDK8 as a critical regulator of stemness in PA tumorigenesis. Immunohistochemical analysis demonstrated that CDK8 expression is elevated in clinical PA samples and correlates significantly with Knosp grades, indicating its potential role in parasellar invasion. Inhibition of CDK8 significantly impaired the self-renewal capacity of patient-derived PA stem-like cells

Prophylactic and therapeutic CSC-based vaccination reduced tumor growth, metastasis and enhanced survival in mouse model of breast cancer.

BACKGROUND: Immunotherapy is a promising cancer treatment, but its effectiveness is limited by the lack of specific strategies to target cancer stem cells (CSCs). CSCs a rare tumor cell subpopulation, are thought to drive tumor recurrence, metastasis, and therapy resistance and developing targeting CSCs is needed. This study investigates the efficacy of a CSC-based vaccine in preventing and treating breast tumor growth and metastasis in a mouse model. METHODS: 4T1-CSC was enriched by sphere formation and characterized by high expression of stemness genes using real-time PCR and higher in vivo

Chemical Stabilization and Transformation of Schiff Base Scaffold: A Path toward Superior Platinum-Based Anti-Cancer Stem Cell Drugs.

The clinical application of platinum-based anticancer agents is limited by poor aqueous stability, low solubility, and dose-limiting toxicities. Here, we report two Pt(II) complexes based on a Schiff base ligand and its reduced imine derivative, designed to overcome these limitations. We show that ligand stabilization through imine reduction markedly improves solution stability, which emerges as an important contributor to interaction, cellular responses, and antiproliferative activity. ICP-MS analysis demonstrated efficient intracellular platinum accumulation with preferential nuclear localiz

eNAMPT/Ac-STAT3/DIRAS2 Axis Promotes Development and Cancer Stemness in Triple-Negative Breast Cancer by Enhancing Cytokine Crosstalk Between Tumor-Associated Macrophages and Cancer Cells.

The intricate relationship between tumor-associated macrophages (TAMs) and cancer cells is pivotal for carcinogenesis, with TAMs being integral to the tumor microenvironment (TME). This study explores the novel mechanisms by which TAMs regulate the progression of triple-negative breast cancer (TNBC) within the TME. Using a co-culture system and methodologies such as cytokine arrays, proteomics, and CRISPR-Cas9, we investigated the crosstalk between TAMs and TNBC cells. We found that high levels of CD163+ TAMs in TNBC tissues correlate with poor prognosis. TNBC cell-conditioned medium induces m

Biomimetic Salivary Gland Cancer Spheroid Platform for In Vitro Recapitulation of Three-Dimensional Tumor-Stromal Interactions.

Salivary gland carcinomas (SGCs) are aggressive malignancies with limited treatment options, primarily due to the complexity of the tumor microenvironment (TME). Cancer-associated fibroblasts (CAFs) remodel the extracellular matrix (ECM), enhance cancer cell stemness, and drive drug resistance. This study introduces a decellularized CAF-derived spheroid system as a biomimetic platform to study tumor-stromal interactions in SGC. Multicellular spheroids were generated by co-culturing Medical Research Council cell strain 5 (MRC-5) fibroblasts (fetal lung-derived) with A253 salivary gland cancer c

Hyaluronic acid (HA)-coated naproxen-nanoparticles selectively target breast cancer stem cells through COX-independent pathways.

Cytotoxic chemotherapy continues to be the main therapeutic option for patients with metastatic breast cancer. Several studies have reported a significant association between chronic inflammation, carcinogenesis and the presence of cancer stem cells (CSC). We hypothesized that the use of non-steroidal anti-inflammatory drugs targeted to the CSC population could help reducing tumor progression and dissemination in otherwise hard to treat metastatic breast cancer. Within this study cationic naproxen (NAP)-bearing polymeric nanoparticles (NPs) were obtained by self-assembly and they were coated w

Dietary Turmeric Bisdemethoxycurcumin Suppresses Wilms' Tumor 1 and CD34 Protein Expressions in KG-1a Leukemic Stem Cells.

Leukemic cells remaining in the body is the main problem for cancer patients, and these cells are called Leukemic Stem Cells (LSCs). Many studies have revealed that the overexpression of the Wilms' tumor 1 (WT1) protein is related to leukemogenesis. Curcuminoids (curcumin, demethoxycurcumin, and bisdemethoxycurcumin) from Thai turmeric (Curcuma longa Linn.) are the focus of this study because they have been previously been reported to show potent antileukemic activity. This study aims to investigate the cytotoxic effect of in-house curcuminoids on the human leukemic stem cell line (KG-1a) when

Annexin A2-mediated cancer progression and therapeutic resistance in nasopharyngeal carcinoma.

Nasopharyngeal carcinoma (NPC) is a head and neck cancer with poor clinical outcomes and insufficient treatments in Southeast Asian populations. Although concurrent chemoradiotherapy has improved recovery rates of patients, poor overall survival and low efficacy are still critical problems. To improve the therapeutic efficacy, we focused on a tumor-associated protein called Annexin A2 (ANXA2). This review summarizes the mechanisms by which ANXA2 promotes cancer progression (e.g., proliferation, migration, the epithelial-mesenchymal transition, invasion, and cancer stem cell formation) and ther

Uncovering core stemness biomarkers in HBV-HCC through integrated multi-omics.

Chronic hepatitis B virus (HBV) infection is a major etiology of hepatocellular carcinoma (HCC), with cancer stem cells (CSCs) playing a critical role in tumor progression and therapeutic resistance. However, the specific mechanisms linking HBV infection to stemness features in HBV-associated HCC (HBV-HCC) remain incompletely defined. To identify potential regulators, we conducted an integrated in silico multi-omics analysis using single-cell and bulk transcriptomic datasets. Comparative stemness scoring across major cell populations prioritized the endothelial compartment for further analysis

Update on the treatment of primary immunodeficiencies.

A general review of advances in the treatment of Primary Immunodeficiencies (PID) has been performed. Treatment with immunoglobulins is indicated in cases of humoral immunodeficiencies and in selected cases of combined immunodeficiencies. The use of intramuscular immunoglobulins in the treatment of PID was abandoned after obtaining the intravenous immunoglobulins, since these are much more effective and have fewer adverse effects. Now subcutaneous immunoglobulins are also available. Immunoglobulins help to keep the patients free of symptoms and infections as these substances are able to neutra

CagA promotes gastric cancer stemness and metastasis via DNMT1-mediated epigenetic silencing of BEX1.

Helicobacter pylori (H. pylori) cytotoxin-associated gene A (CagA) critically contributes to gastric cancer (GC) pathogenesis, though its epigenetic control of cancer stemness is incompletely characterized. This study establishes that CagA enhances self-renewal capacity and metastatic potential in GC cells by elevating DNA methyltransferase 1 (DNMT1) expression. Upregulated DNMT1 catalyzes hypermethylation of the brain-expressed X-linked gene 1 (BEX1) promoter, resulting in transcriptional silencing of this tumor suppressor. Functional validation through tumorsphere/colony formation assays and

Merlin, the product of NF2 gene, is associated with aromatase expression and estrogen formation in human liver tissues and liver cancer cells.

The product of neurofibromatosis type 2 (NF2) gene, also known as Merlin/neurofibromin 2, homeostatically regulates liver stem cells by controlling abundance and signaling of epidermal growth factor receptor (EGFR), with a mechanism independent of the Hippo pathway. We have reported that locally elevated estrogen formation, driven by abnormally high expression and function of aromatase, may be implicated in development and progression of human hepatocellular carcinoma (HCC) through activation of a rapid signaling pathway mediated by amphiregulin (AREG) and EGFR. We have recently presented a mo

Tissue inhibitor of metalloproteinases-4 (TIMP-4) regulates stemness in cervical cancer cells.

Tissue inhibitor of metalloproteinase-4 (TIMP-4) belongs to a family of extracellular matrix (ECM) metalloproteinases inhibitors that are overexpressed in several cancers. However, the role of TIMP-4 during carcinogenesis is poorly understood. To evaluate TIMP-4 functions in carcinogenesis, stably transfected cells overexpressing this tissue inhibitor were used. Xenograft tumor growth, stem cell enrichment, colony formation, and gene regulation were investigated. Microarrays and in silico analysis were carried out to elucidate TIMP-4 molecular mechanisms. In the present report, we show that in

Unraveling the glioblastoma (GBM) tumor microenvironment: future perspective on targeted immunotherapy.

Glioblastoma (GBM) is one of the most aggressive, fast-growing, and therapeutically challenging brain tumors. The difficulty in managing GBM stems from its genetic instability and the intricately complex tumor microenvironment (TME). Within the TME, intricate interactions between neoplastic cells and signaling mediators drive tumor progression. Despite advances in current treatments, obstacles such as the blood-brain barrier (BBB) and pronounced inter- and intratumoral heterogeneity continue to hinder therapeutic success. Consequently, research efforts have increasingly focused on immunotherap

Progesterone from ovulatory menstrual cycles is an important cause of breast cancer.

Many factors, including reproductive hormones, have been linked to a woman's risk of developing breast cancer (BC). We reviewed the literature regarding the relationship between ovulatory menstrual cycles (MCs) and BC risk. Physiological variations in the frequency of MCs and interference with MCs through genetic variations, pathological conditions and or pharmaceutical interventions revealed a strong link between BC risk and the lifetime number of MCs. A substantial reduction in BC risk is observed in situations without MCs. In genetic or transgender situations with normal female breasts and

Advanced systemic mastocytosis: from molecular and genetic progress to clinical practice.

Systemic mastocytosis is a heterogeneous disease characterized by the accumulation of neoplastic mast cells in the bone marrow and other organ organs/tissues. Mutations in KIT, most frequently KIT D816V, are detected in over 80% of all systemic mastocytosis patients. While most systemic mastocytosis patients suffer from an indolent disease variant, some present with more aggressive variants, collectively called "advanced systemic mastocytosis", which include aggressive systemic mastocytosis, systemic mastocytosis with an associated hematologic, clonal non mast cell-lineage disease, and mast ce

Normal and malignant human plasma cells: proliferation, differentiation, and expansions in relation to CD45 expression.

In this study, we have evaluated the proliferation and the phenotype of human plasma cells of different origins, i.e., from tonsil, peripheral blood, bone marrow as well as plasma cells generated in vitro from memory B cells. We have demonstrated that plasma cells from tonsil, peripheral blood, as well as those generated in vitro, were highly proliferating and presented a homogeneous CD45bright phenotype. In contrast, bone marrow plasma cells were heterogeneous for CD45 expression but their proliferation was restricted to the CD45bright compartment. Subsequently, their CD45 expression decrease

Targeting the METTL14/Wnt9a/β-catenin pathway with pharmacological inhibitors enhances radiotherapy and immunotherapy efficacy in non-small cell lung cancer.

Non-small cell lung cancer (NSCLC) remains a leading cause of cancer mortality, with its molecular heterogeneity posing a therapeutic challenge. This study investigates the pharmacologically targetable METTL14-mediated pathway in modulating cancer stemness and response to combined radiotherapy and PD-1 blockade. We performed single-cell RNA sequencing and m6A methylation sequencing on matched untreated and radiotherapy/anti-PD-1-treated NSCLC samples. GANALYSIS: of 21,019 cells revealed that combined treatment reduced cancer stem cells (CSCs) and exhausted CD8+ T cells, while increasing naïve

Endothelial cells-derived SEMA3G suppresses glioblastoma stem cells by inducing c-Myc degradation.

The poor prognosis of glioblastoma (GBM) patients is attributed mainly to abundant neovascularization and presence of glioblastoma stem cells (GSCs). GSCs are preferentially localized to the perivascular niche to maintain stemness. However, the effect of abnormal communication between endothelial cells (ECs) and GSCs on GBM progression remains unknown. Here, we reveal that ECs-derived SEMA3G, which is aberrantly expressed in GBM patients, impairs GSCs by inducing c-Myc degradation. SEMA3G activates NRP2/PLXNA1 in a paracrine manner, subsequently inducing the inactivation of Cdc42 and dissociat

NIR irradiation enhances the apoptotic potentiality of quinacrine-gold hybrid nanoparticles by modulation of HSP-70 in oral cancer stem cells.

Cancer stem cells (CSCs) are the tumor cell subpopulations that can self-renew, differentiate, initiate and maintain tumor growth. CSCs are frequently drug-resistant, resulting in tumor recurrence, metastasis, and angiogenesis. Herein, using in vitro oral squamous cell carcinoma (OSCC) CSCs and in vivo xenograft mice model, we have systematically studied the apoptotic potentiality of quinacrine-gold hybrid nanoparticle (QAuNP) and its underlying mechanism after NIR irradiation. QAuNP + NIR caused DNA damage and induced apoptosis in SCC-9-CSCs by deregulating mitochondrial membrane potential (Δ
