Tunable bioactivity and mechanics of collagen-based tissue engineering constructs: A comparison of EDC-NHS, genipin and TG2 crosslinkers.

Due to its ubiquity and versatility in the human body, collagen is an ideal base material for tissue-engineering constructs. Chemical crosslinking treatments allow precise control of the biochemical and mechanical properties through macromolecular modifications to the structure of collagen. In this work, three key facets regarding the collagen crosslinking process are explored. Firstly, a comparison is drawn between the carbodiimide-succinimide (EDC-NHS) system and two emerging crosslinkers utilising alternate chemistries: genipin and tissue transglutaminase (TG2). By characterising the chemic

Design of Novel Perovskite-Based Polymeric Poly(l-Lactide-Co-Glycolide) Nanofibers with Anti-Microbial Properties for Tissue Engineering.

: There is a growing need for anti-microbial materials in several biomedical application areas, such are hernia, skin grafts as well as gynecological products, owing to the complications caused by infection due to surgical biomaterials. The anti-microbial effects of silver in the form of nanoparticles, although effective, can be toxic to surrounding cells. In this study, we report, for the first time, a novel biomedical application of Ag0.3Na1.7La2Ti3O10-layered perovskite particles, blended with poly(L-lactide-co-glycolide) (PLGA), aimed at designing anti-microbial and tissue engineering scaf…

Tissue Engineering Therapy for Atrophic Maxilla Using Minimally Manipulated CD90 and CD105 Bone Marrow Stem Cells: A Case Report.

Bone deficiencies in the jaws of edentulous patients limit the possibility of successful placement and osseointegration of endosseous implants. Bone resorption is defined as a vertical and horizontal loss of quality and quantity in the residual bone ridges; lack of bone structure requires bone grafting procedures to enable implant placement and to improve the function, predictability, and longevity of implants. The objective of this case study was to demonstrate the feasibility of a novel cell therapy treatment for localized jaw bone defects with tissue repair cells composed of a mixture of bo

Modified silk fibroin scaffolds with collagen/decellularized pulp for bone tissue engineering in cleft palate: Morphological structures and biofunctionalities.

Cleft palate is a congenital malformation that generates a maxillofacial bone defect around the mouth area. The creation of performance scaffolds for bone tissue engineering in cleft palate is an issue that was proposed in this research. Because of its good biocompatibility, high stability, and non-toxicity, silk fibroin was selected as the scaffold of choice in this research. Silk fibroin scaffolds were prepared by freeze-drying before immerging in a solution of collagen, decellularized pulp, and collagen/decellularized pulp. Then, the immersed scaffolds were freeze-dried. Structural organiza…

A rapid fabricated living dermal equivalent for skin tissue engineering: an in vivo evaluation in an acute wound model.

The encapsulation of both cells and a surgical mesh in a polymerizing collagen hydrogel followed by mechanical compression, after polymerization, results in the rapid formation of a living dermal equivalent (LDE) with physical properties suitable for in vivo application. It was found in the current study that the LDE supported the attachment, growth, and differentiation of keratinocytes, allowing for the formation of living skin equivalents (LSEs) with a monolayer epidermis (LSE-M) and a stratified epidermis (LSE-S). The utility of the LDE for the fabrication of living wound dressings was furt

Genetically modified canine Schwann cells--In vitro and in vivo evaluation of their suitability for peripheral nerve tissue engineering.

After peripheral nerve injury, Schwann cells (SC) guarantee for a regeneration-promoting milieu and are crucially involved in axonal regeneration. For extended nerve defects, bridging with an autologous nerve transplant is the gold standard therapy. Artificial biohybrid nerve transplants which combine a synthetic conduit with autologous SC genetically modified to express regeneration-promoting proteins may provide an alternative therapy to autotransplantation. The dog seems to be an ideal translational animal model for new treatment strategies. In the present study, utilizing a new transfectio

Applicability of the dyes CFSE, CM-DiI and PKH26 for tracking of human preadipocytes to evaluate adipose tissue engineering.

Adipose tissue engineering with preadipocyte-loaded scaffolds requires adequate tracking of preadipocytes to allow evaluation and quantification of cell proliferation, expansion and differentiation in three-dimensional systems. To differentiate between graft and host cells, labeling of preadipocytes before implantation and tracking of these cells until harvest would be useful. Immunohistochemistry enables the differentiation between cells of different species but is time-consuming, expensive, elaborate, and not applicable for autologous transplantation. So far, there is no published method to …

Engineering Multicellular Breast Cancer Spheroids in Decellularized Adipose Tissue Hydrogels Using a Microfluidic Platform to Recapitulate Tumor Microenvironment Complexity.

Three-dimensional (3D) cancer spheroids are critical for recapitulating tumor architecture and microenvironmental cues that govern disease progression and therapeutic response. However, most existing spheroid models lack biomimetic extracellular matrices, controlled fabrication, and stromal complexity representative of the native tumor microenvironment (TME). Here, we report a multifaceted engineering strategy that integrates a decellularized adipose tissue (DAT) hydrogel, a modified microfluidic T-junction platform, and multicellular coculture to generate physiologically relevant breast cance

Three-Dimensional-Printed GelMA-KerMA Composite Patches as an Innovative Platform for Potential Tissue Engineering of Tympanic Membrane Perforations.

Tympanic membrane (TM) perforations, primarily induced by middle ear infections, the introduction of foreign objects into the ear, and acoustic trauma, lead to hearing abnormalities and ear infections. We describe the design and fabrication of a novel composite patch containing photocrosslinkable gelatin methacryloyl (GelMA) and keratin methacryloyl (KerMA) hydrogels. GelMA-KerMA patches containing conical microneedles in their design were developed using the digital light processing (DLP) 3D printing approach. Following this, the patches were biofunctionalized by applying a coaxial coating wi

Development of cell adhesive and inherently antibacterial polyvinyl alcohol/polyethylene oxide nanofiber scaffolds via incorporating chitosan for tissue engineering.

Currently, polyvinyl alcohol (PVA) and polyethylene oxide (PEO), as tissue engineering scaffolds materials, had been widely studied, however the hard issues in cell adhesive and antimicrobial properties still seriously limited their application in biomedical respects. Herein, we solved both hard issues by incorporating chitosan (CHI) into the PVA/PEO system, and successfully prepared PVA/PEO/CHI nanofiber scaffolds via electrospinning technology. First, the hierarchical pore structure and elevated porosity stacked by nanofiber of the nanofiber scaffolds supplied suitable space for cell growth.…
