OBJECTIVE: To observe the effect of proliferating cell nuclear antigen (PCNA) antisense RNA on the growth characteristics of human gastric cancer cell.
METHODS: An eukaryotic expression vector pDR-PCNA with human PCNA gene insert in reverse direction was successfully constructed. The human gastric cancer cell line SGC-7901 was transfected with pDR-PCNA by lipofectamine.
RESULTS: Growth rate, protein and RNA biosynthesis and DNA contents in S-phase and G2/M-phase of the transfected SGC-7901 were significantly suppressed in comparison with those of the parental cell line.

Transfection of an Epstein-Barr virus (EBV)-encoded plasmid containing EBER caused a substantial decrease in the level of plasmid containing EBV in Akata and Mutu Burkitt's lymphoma (BL) lines, but failed to do so in other BL lines. The results suggest that EBER could replace the role of EBV, but other EBV products also play a role in the growth of BL.

PURPOSE: To investigate the effect of paired related homeobox1 (PRRX1) on autophagy of salivary adenoidcystic carcinoma (SACC) cells.
METHODS: PRRX1-overexpressed lentiviral vectors and their negative control lentiviral vectors were used to transfect SACC cells. The transfection effect was detected by Western blot. This study investigated the effect of transcriptional gene silencing of the heparanase gene on standard gallbladder carcinoma cells (GBC-SD). The miRNAs targeting the promoter region and coding region of the heparanase gene were designed and synthesized. We transfected four recombinant miRNA vectors into GBC-SD. AIM: To study the effects of CD74 gene on IFN-γR expression in myasthenia gravis thymic epithelial cell(TEC).
METHODS: PCMV-CD74 transiently transfect to primary cultured TEC mediated with lipofectamine, the result of transfection and some autoimmune related genes such as IFN-γR were detected by real-time RT-PCR.
