Detection, characterisation and purification of a murine liver factor capable of desensitising towards the lethal activity of tumour necrosis factor.

Tumour necrosis factor (TNF) is a major mediator in septic shock and several inflammatory diseases such as hepatitis. Galactosamine (GalN) sensitises experimental animals for TNF and the combination TNF/GalN leads to a lethal inflammatory hepatitis. We describe that a single injection of lipopolysaccharide (LPS), interleukin-1 (IL-1) or TNF can desensitise against the lethality induced by TNF/GalN, but also against changes in metabolic parameters such as hypothermia and transaminase release, in a dose responsive way. We also describe the desensitising capacity of a component present in Mouse L

[Allergic rhinitis].

Allergic rhinitis is one of the commonest diseases. The main allergens responsible are respiratory allergens. Pollinic or seasonal allergic rhinitis is currently distinguished from perannual or nonseasonal rhinitis, generally related to sensitization to allergens of the home environment. The diagnostic approach of allergic rhinitis is based on a careful clinical examination, in order to eliminate other causes of nasal dysfunction, and on complementary investigations, most importantly allergic skin tests. Several types of drugs, topically or systemically administered are currently available to

A comparative study of the clinical efficacy of immunotherapy and conventional pharmacological treatment for patients with perennial allergic rhinitis.

This study was designed to compare the clinical outcome of prolonged immunotherapy for perennial allergic rhinitis with that of pharmacological treatment. Patients with perennial allergic rhinitis due to Dermatophagoides farinae (D. farinae) were divided into two groups; a pharmacotherapy group and an immunotherapy group. The pharmacotherapy group was treated with conventional pharmacological treatment using antihistamine tablets and topical steroid sprays and the immunotherapy group was treated with D. farinae extracts for 5 successive years. None of symptom scores at enrollment differed sign

Immunoregulatory potential of exogenous Schistosoma mansoni soluble egg antigen in a model of experimental schistosomiasis--I. Regulation of granuloma formation in vivo.

This study was undertaken to assess the optimum conditions required to reduce the vigorous host granulomatous reaction around Schistosoma mansoni eggs. Soluble schistosomal egg antigen (SEA) at a concentration of 10 or 100 micrograms protein was administered i.p. or i.v. into unprimed C57BL/6 mice. SEA was injected either alone or in combination with cyclophosphamide (CY) 100 or 50 mg/kg via i.p. route. Seven or 14 days later viable eggs of S. mansoni were injected via the tail vein into treated groups and untreated normal controls. Mice were sacrificed 8, 16 and 24 days after the injection of

Rush immunotherapy with a standardized Bermuda grass pollen extract.

We carried out a double-blind clinical trial in 30 patients who were sensitized to Bermuda grass pollen. Before and after immunotherapy we performed in vivo tests (skin tests and standardized tests of specific and nonspecific bronchial hyperreactivity), in vitro tests (histamine release), and specific IgE and IgG antibodies to BGP. We found a significant decrease (P less than .001) in specific bronchial hyperreactivity and skin sensitivity to BGP in the group of patients treated with immunotherapy, a decrease in the delayed responses (P less than .05) in histamine release (P less than .01), an

Perioperative Desensitization Improves Outcomes Among Crossmatch Positive Recipients of Deceased Donor Renal Transplants.

CONTEXT: Graft failure due to chronic rejection is greater among renal transplant patients with donor-specific antibody (DSA) than among DSA-free patients. For patients dependent on deceased donor transplantation, preoperative desensitization to eliminate DSAs may be impractical. We speculated that perioperative desensitization might eliminate preexisting DSAs and prevent de novo DSAs and improve graft outcomes. We report that brief perioperative desensitization using either intravenous immunoglobulin (IVIG) or plasmapheresis/IVIG (PP/IVIG) treatment improves clinical outcomes among patients w

Kidney transplantation after desensitization in sensitized patients: a Korean National Audit.

INTRODUCTION: The number of end-stage renal disease (ESRD) patients with preformed antibodies waiting for a kidney transplant has been increasing lately. We conducted a nationwide study on the outcomes of kidney transplantation after desensitization in Korea. METHODS: Six transplant centers have run desensitization programs. The patients who underwent living donor kidney transplantation after desensitization from 2002 to 2010 were retrospectively analyzed. RESULTS: A total of 86 cases were enrolled. Thirty-five of these were cases of re-transplantation (40.7 %). Indications of desensitizatio

Tolerance induction in clinical transplantation.

Introduction of modern immunosuppressive agents has led to great success of allotransplantation in humans, and survival rates for all solid organs have been dramatically improved. However, a constant proportion of organs is lost every year due to chronic allograft rejection and immunosuppressive drug toxicity. This has led to a situation where, despite the of donor organ shortage, about one third of the patients on the kidney transplant waiting list are listed for a retransplant. The induction of donor-specific tolerance has the potential of at least partially resolving this problem, since it

[Asthma: prevalence, pathogenesis and perspectives on new treatments].

Asthma is a complex clinical syndrome with many phenotypes in both children and adults. Its major characteristics include airway inflammation and obstruction, bronchial hyper-responsiveness. The disease has its roots in infancy, and both genetic and environmental factors contribute to its inception and clinical evolution. The prevalence of asthma in increasing around the world. Hypotheses that attempt to explain the rising prevalence have been advanced but remain unproven. To understand its pathogenesis it is essential to identify factors that modulate or intensify the inflammatory response of

Outcomes of ABO-Incompatible Versus ABO-Compatible Liver Transplantation in Pediatric Recipients: An Updated Systematic Review and Meta-Analysis.

BACKGROUND AND OBJECTIVE: ABO-incompatible (ABO-i) liver transplantation (LT) in children is increasingly used to address organ scarcity, but concerns about immunological, vascular, and biliary complications remain. With evolving desensitization strategies and emerging pediatric data, an updated meta-analysis comparing ABO-i and ABO-compatible (ABO-c) LT is warranted. METHODS: PubMed, Embase, and Scopus were searched from inception to June 2026 to include studies evaluating ABO-i versus ABO-c LT in recipients ≤ 18 years of age. Primary outcomes were comparison of 1-, 3-, and 5-year patient an

Evaluation of desensitizing therapies for donor-specific antibodies in confirmed humoral allograft rejection in kidney transplantation: Experience of a center for 10 years.

UNLABELLED: Donor-specific antibodies (DSAs) in the recipient's serum are directed against donor-mismatched Human Leukocyte Antigen (HLA) antigens found on graft endothelial cells. These DSAs may be responsible for antibody-mediated rejection (AMR) in patients undergoing kidney transplantation. Once they are developed in the post-transplant period, a desensitizing treatment must be instituted to reduce these DSAs and not cause damage to the graft. The objectives were to analyze our cases of confirmed AMR in a series of kidney recipients over 10 years and to assess the impact of the anti-reject

Pilot study of intratonsillar immunotherapy for timothy grass pollen.

BACKGROUND: Allergen-specific immunotherapy (AIT) is a treatment for upper airway allergy that is resistant to optimal symptomatic management. Traditionally, it is administered via repeated subcutaneous injections (subcutaneous immunotherapy, SCIT) or daily sublingual tablets/drops (sublingual immunotherapy, SLIT) for 3–5 years. Direct allergen exposure has also been attempted through intralymphatic immunotherapy (ILIT) via injections into inguinal lymph nodes. OBJECTIVE: To evaluate the clinical and immunological effects of intratonsillar allergen immunotherapy for timothy grass pollen allerg

Targeting CD38 to reduce anti-HLA antibody levels: A new and effective option to be integrated into desensitization protocols?

Desensitization protocols are employed in patients with antibody-mediated organ rejection or hematopoietic stem cell (HSC) transplant recipients from ´HLA-mismatched´ donors with donor-specific antibodies (DSA). However, these protocols sometimes have limitations in effectively reducing anti-HLA antibody levels and maintaining this reduction over time. As a result, there is an ongoing search for new, effective therapies to implement current protocols, involving anti-CD20, bortezomib, and plasma exchange. This case report explores the impact of daratumumab, a well-known anti-CD38 antibody used

Neutralizing IgG4 antibodies are a biomarker of sustained efficacy after peanut oral immunotherapy.

BACKGROUND: Clinical efficacy of oral immunotherapy (OIT) has been associated with the induction of blocking antibodies, particularly those capable of disrupting IgE-allergen interactions. Previously, we identified mAbs to Ara h 2 and structurally characterized their epitopes. OBJECTIVE: We investigated longitudinal changes during OIT in antibody binding to conformational epitopes and correlated the results with isotype and clinical efficacy. METHODS: We developed an indirect inhibitory ELISA using mAbs to block conformational epitopes on immobilized Ara h 2 from binding to serum immunoglobu

Cellular and Humoral Response After Induction of Protection and After Finishing Hymenoptera Venom Immunotherapy.

Hymenoptera venom allergy (HVA) is an IgE-mediated hypersensitivity reaction caused by Hymenoptera species stings (honeybee, vespid, or ant). The only effective treatment is Hymenoptera venom immunotherapy (VIT). Our study aimed to evaluate whether humoral and cellular biomarkers measured before, during, and after honeybee VIT are associated with the success of VIT, which was assessed by the response to a sting challenge one year after finishing VIT. In this prospective study, blood biomarkers of 25 patients undergoing honeybee VIT at the referral center in Slovenia were evaluated. A controlle

Basophil Activation Test is a Relevant Biomarker of the Outcome of Rapid Desensitization in Platinum Compounds-Allergy.

BACKGROUND: Rapid drug desensitization (RDD) has become a cornerstone in the management of immediate drug hypersensitivity reactions (DHRs) to chemotherapeutic agents. Because of the inherent risk of anaphylaxis during RDD, biomarkers to predict patients at risk of developing such severe reactions are needed. The basophil activation test (BAT) has been used in DHRs as a diagnostic tool. OBJECTIVE: We evaluated basophil CD63 and CD203c expression (BAT) as a biomarker to assess the safety and effectiveness of RDD in platinum compounds-allergic patients. METHODS: Patients allergic to platinum c

[Allergen-specific immunotherapy in the treatment of pollen allergy].

Since its description by Noon in 1911, desensitization or allergen-specific immunotherapy (SIT) has been largely given by sub cutaneous injection in the treatment of allergic diseases. It remains the only treatment for allergic diseases aimed at the etiology. The development of sublingual route as an alternative to sub cutaneous injection, and of new forms of medication, has led to large-scale clinical trials, many of them performed with allergen tablets, particularly in the field of pollen allergy. These studies have confirmed that SIT is efficient in reducing allergic respiratory symptoms. D
