Pharmacological and biochemical studies on the protective effects of melatonin during stress-induced behavioral and immunological changes in relation to oxidative stress in rats.

Stress is known to precipitate neuropsychiatric diseases, and depending upon its nature and intensity it can also influence the functioning of the immune system. Melatonin (N-acetyl-5-methoxy tryptamine) a pineal gland hormone and potent antioxidant is known to protect against many diseases. Effect of melatonin in stress-induced neuro-immunomodulation is not well elucidated. Therefore in the present study, the protective effects of melatonin were evaluated in restraint stress (RS)-induced behavioral and immunological changes in rats. RS for 1 h significantly reduces (i) percentage of open-arm

Effects of deletion of mutant huntingtin in steroidogenic factor 1 neurons on the psychiatric and metabolic phenotype in the BACHD mouse model of Huntington disease.

Psychiatric and metabolic features appear several years before motor disturbances in the neurodegenerative Huntington's disease (HD), caused by an expanded CAG repeat in the huntingtin (HTT) gene. Although the mechanisms leading to these aspects are unknown, dysfunction in the hypothalamus, a brain region controlling emotion and metabolism, has been suggested. A direct link between the expression of the disease causing protein, huntingtin (HTT), in the hypothalamus and the development of metabolic and psychiatric-like features have been shown in the BACHD mouse model of HD. However, precisely

Neurotransmissions of antidepressant-like effects of neuromedin U-23 in mice.

Neuromedin U (NmU) is a widely distributed and multifunctional peptide in the central nervous system and the peripheral tissues. Little is know about the mechanisms of NmU on brain functions. The rodent isoform of the NmU, NmU-23, has been shown to have anxiolytic effects involved in the β-adrenergic and cholinergic nervous systems in elevated plus maze test. NmU-23 was tested for antidepressant-like effects in modified forced swimming test (FST) in mice and furthermore, the involvement of the adrenergic, serotonergic, cholinergic, dopaminergic or gaba-ergic receptors in the antidepressant-lik…

Dopaminergic system in CA1 modulates MK-801 induced anxiolytic-like responses.

RATIONALE: Today, there is relatively no debate on the notion that NMDA receptor antagonist agents in the hippocampus induce anxiolytic-like effects through distinct mechanisms. There is also a bulk of studies showing the involvement of the dopaminergic system in NMDA induced behaviors. Thus, on the basis of the involvement of dopaminergic system in anxiety-related behaviors, the present study aimed to investigate the involvement of the dorsal hippocampal (CA1) dopaminergic system in anxiolytic-like responses induced by MK801 (NMDA receptor antagonist) in male Wistar rats. METHODS: We used th

Not all 'predator odours' are equal: cat odour but not 2,4,5 trimethylthiazoline (TMT; fox odour) elicits specific defensive behaviours in rats.

The behavioural responses to two commonly used 'predator odours' were assessed in male Wistar rats. Cat odour was presented to rats in the form of a piece of collar that had been worn by a domestic cat. Fox odour was presented in an equivalent piece of (unworn) collar that had been impregnated with 2,4,5 Trimethylthiazoline (TMT)-an extract of fox faeces. Other rats were exposed to collars containing Triethylamine (TEA), a putrid fishy smell, or formaldehyde, which has an acrid irritating smell. Experiment 1 showed that rats approached cat odour, TMT and TEA significantly less than they did an

The effect of tert-butylhydroquinone on anxiolytic- and antidepressant-like behaviors induced by post-traumatic stress disorder: A behavioral and molecular study.

OBJECTIVES: Post-traumatic stress disorder (PTSD) is a crippling mental illness that commonly co-occurs with anxiety and depression. Recent studies have established a link between neuroinflammation and the development of PTSD. Studies have revealed that tert-butylhydroquinone (tBHQ) exhibits anti-inflammatory effects. This research investigated the impact of tBHQ on the amelioration of PTSD-induced depression- and anxiety-like behaviors with respect to amygdala and hippocampal MAO-A, MAO-B, IL-6, IL-10, and glucocorticoid receptor in rats. MATERIALS AND METHODS: PTSD was triggered through the

Curcumin Alleviates Methotrexate-Induced Neurobehavioral Deficits and Modulates the Redox-Inflammatory-Neurotrophic Axis in Mice.

Methotrexate (MTX), a widely used chemotherapeutic agent, is associated with central nervous system toxicity characterized by oxidative stress, neuroinflammation, and behavioral disturbances. The present study investigated whether curcumin mitigates MTX-induced neurotoxicity by modulating redox balance and inflammatory-neurotrophic signaling pathways. Twenty-four male BALB/c mice were randomly assigned to four groups: control, MTX (40 mg/kg, single intraperitoneal dose), MTX+curcumin (50 mg/kg oral pretreatment), and curcumin alone. Behavioral alterations were assessed using the open field, el

Increased c-Fos immunoreactivity in anxiety-related brain regions following paroxetine discontinuation.

Selective serotonin reuptake inhibitor (SSRI) therapy cessation often induces a disabling discontinuation syndrome, including increased anxiety. We recently reported that SSRI discontinuation induced behavioural changes in mice, which we hypothesise arose from activated anxiety circuitry. Here, we investigated the effect of discontinuation from the SSRI paroxetine on the expression of the activity-dependent gene c-fos in selected anxiety-related midbrain and forebrain regions. Male mice were injected daily with paroxetine (10 mg/kg) or saline for 12 days, then treatment was either continued or

Enhanced sympathetic nerve activity induced by neonatal colon inflammation induces gastric hypersensitivity and anxiety-like behavior in adult rats.

Gastric hypersensitivity (GHS) and anxiety are prevalent in functional dyspepsia patients; their underlying mechanisms remain unknown largely because of lack of availability of live visceral tissues from human subjects. Recently, we demonstrated in a preclinical model that rats subjected to neonatal colon inflammation show increased basal plasma norepinephrine (NE), which contributes to GHS through the upregulation of nerve growth factor (NGF) expression in the gastric fundus. We tested the hypothesis that neonatal colon inflammation increases anxiety-like behavior and sympathetic nervous syst
