Endocrine disruptors and prostate cancer risk.

There is increasing evidence both from epidemiology studies and animal models that specific endocrine-disrupting compounds may influence the development or progression of prostate cancer. In large part, these effects appear to be linked to interference with estrogen signaling, either through interacting with ERs or by influencing steroid metabolism and altering estrogen levels within the body. In humans, epidemiologic evidence links specific pesticides, PCBs and inorganic arsenic exposures to elevated prostate cancer risk. Studies in animal models also show augmentation of prostate carcinogene

Deep learning-based predictions and analysis of Norovirus dynamics using a fractional-order mathematical model.

Norovirus, which is extremely contagious and has a significant impact on the gastrointestinal tract, has also been one of the main potential threats to health in the population. The use of fractional calculus has received much interest in the past few years with regard to the modeling of infectious diseases, especially because it allows modeling the memory and nonlocal processes in a dynamic system. In this study, a mathematical model is developed to investigate the transmission dynamics of norovirus. The proposed model consists of five epidemiological compartments: susceptible, exposed, infec

Spatiotemporal diffusion of influenza A (H1N1): Starting point and risk factors.

Influenza constitutes a major challenge to world health authorities due to high transmissibility and the capacity to generate large epidemics. This study aimed to characterize the diffusion process of influenza A (H1N1) by identifying the starting point of the epidemic as well as climatic and sociodemographic factors associated with the occurrence and intensity of transmission of the disease. The study was carried out in the Brazilian state of Paraná, where H1N1 caused the largest impact. The units of spatial and temporal analysis were the municipality of residence of the cases and the epidemi

Formulation and in vitro evaluation of niacin-loaded nanoparticles to reduce prostaglandin mediated vasodilatory flushing.

The ability to explicitly represent infectious disease distributions and their risk factors over massive geographical and temporal scales has transformed how we investigate how environment impacts health. While landscape epidemiology studies have shed light on many aspects of disease distribution and risk differentials across geographies, new computational methods combined with new data sources such as citizen sensors, global spatial datasets, sensor networks, and growing availability and variety of satellite imagery offer opportunities for a more integrated approach to understanding these rel

The multistep pathogenic hypothesis of amyotrophic lateral sclerosis is incompatible with the epidemiological data.

Amyotrophic lateral sclerosis (ALS) is a multifactorial neurodegenerative disease whose incidence increases with age. According to the gene-time-environment hypothesis, ALS onset occurs through the interaction between genes and environmental exposures during ageing, which may involve a continuous accumulation process. Alternatively, the multistep pathogenic hypothesis, based on the Armitage-Doll multistep model from cancer research, posits that a discrete number of specific sequential "hits" are necessary to trigger ALS. Here we analyzed three large population-based epidemiological datasets of

Modeling the implementation of population-level isoniazid preventive therapy for tuberculosis control in a high HIV-prevalence setting.

BACKGROUND: We model the epidemiological impact of providing isoniazid preventive therapy (IPT) to South African adolescents, among whom HIV prevalence is low, latent tuberculosis (TB) prevalence is high, and school-based programs may enable population-level coverage. METHODS: We simulate a dynamic compartmental model of age-structured HIV and TB coepidemics in South Africa. HIV dynamics are modeled by infection status, CD4 cell count, and antiretroviral therapy; TB dynamics are modeled by disease stage, diagnosis, treatment, and IPT status. We analyze the effects of continuous IPT coverage a

New paradigms for Salmonella source attribution based on microbial subtyping.

Microbial subtyping is the most common approach for Salmonella source attribution. Typically, attributions are computed using frequency-matching models like the Dutch and Danish models based on phenotyping data (serotyping, phage-typing, and antimicrobial resistance profiling). Herewith, we critically review three major paradigms facing Salmonella source attribution today: (i) the use of genotyping data, particularly Multi-Locus Variable Number of Tandem Repeats Analysis (MLVA), which is replacing traditional Salmonella phenotyping beyond serotyping; (ii) the integration of case-control data i

[Development of a porcine experimental model of alcoholic intoxication].

Despite important epidemiologic suggestions that alcohol causes a variety of cardiovascular abnormalitíes, and particularly cardiac arrhythmias, the demonstration of this causal effect in animal models is still lacking. Previous open-chest models, heavily instrumented and using epicardial stimulation have failed to demonstrate such causal effect. We developed a porcine, closed-chest model with sedation, a rigorous control of venous alcohol concentration and endocardial electrical stimulation, in a multidisciplinary framework. In this model, we observed that the infusion of alcohol facilitated

To poly(I:C) or not to poly(I:C): advancing preclinical schizophrenia research through the use of prenatal immune activation models.

The neurodevelopmental hypothesis of schizophrenia has been highly influential in shaping our current thinking about modeling the disease in animals. Based on the findings provided by human epidemiological studies, a great deal of recent interest has been centered upon the establishment of neurodevelopmental rodent models in which the basic experimental manipulation takes the form of prenatal exposure to infection and/or immune activation. One such model is based on prenatal treatment with the inflammatory agent poly(I:C) (=polyriboinosinic-polyribocytidilic acid), a synthetic analog of double

Wastewater-Based Epidemiology for Infectious Diseases: A New Trick for an Old Threat.

Wastewater-based epidemiology (WBE) is an innovative approach to epidemiology that offers unique opportunities for public health surveillance. Its potential had been recognized in various applications over the years, but it was the global scale of the response to the SARS-CoV-2 pandemic that truly brought WBE to the fore. In this perspective paper we explore the untapped potential of WBE as a catalyst for infectious disease surveillance and as a One Health epidemiological tool, and the future horizons and innovative applications of WBE. It is clear that WBE will address a growing number of pat

Behavioral mechanisms in HIV epidemiology and prevention: past, present, and future roles.

In the 1980s, behavioral variations across geographically and socially defined populations were the central focus of AIDS research, and behavior change was seen as the primary means of controlling HIV epidemics. Today, biological mechanisms--especially other sexually transmitted infections, antiretroviral therapy, and male circumcision--predominate in HIV epidemiology and prevention. We describe several reasons for this shift in emphasis. Although the shift is understandable, we argue for a sustained focus on behavioral mechanisms in HIV research in order to realize the theoretical promise of

Air quality measurements for the aerosol research and inhalation epidemiology study.

Measurements of pollutant gases, airborne particulate matter mass and composition, and meteorology have been made at a core site near downtown Atlanta, GA, since August 1998 in support of the Aerosol Research and Inhalation Epidemiology Study (ARIES). This site is one of eight in the Southeastern Aerosol Research and Characterization network. The measurement objective is to provide a long-term, multivariate dataset suitable for investigating statistical associations of respiratory and cardiovascular disease with airborne particulate matter composition, meteorology, and copollutant gases throug

Post-pandemic forecasting of pediatric acute respiratory infections with deep learning: a multi-pathogen, multi-time-horizon study.

BACKGROUND: Acute respiratory infections (ARIs) remain a major cause of morbidity and hospitalization in children worldwide. In the post-COVID-19 era, the epidemiological patterns of respiratory pathogens have changed considerably, posing new challenges to conventional forecasting approaches. This study aimed to compare the performance of traditional statistical and deep learning models in forecasting pediatric ARIs and to improve prediction accuracy for clinical and public health preparedness. METHODS: A total of 29,260 pediatric hospitalization records with multiplex polymerase chain reacti

Revisiting the impact of Schistosoma mansoni regulating mechanisms on transmission dynamics using SchiSTOP, a novel modelling framework.

BACKGROUND: The transmission cycle of Schistosoma is remarkably complex, including sexual reproduction in human hosts and asexual reproduction in the intermediate host (freshwater snails). Patterns of rapid recrudescence after treatment and stable low transmission are often observed, hampering the achievement of control targets. Current mathematical models commonly assume regulation of transmission to occur at worm level through density-dependent egg production. However, conclusive evidence on this regulating mechanism is weak, especially for S. mansoni. In this study, we explore the interplay

A risk prediction algorithm based on family history and common genetic variants: application to prostate cancer with potential clinical impact.

Genome wide association studies have identified several single nucleotide polymorphisms (SNPs) that are independently associated with small increments in risk of prostate cancer, opening up the possibility for using such variants in risk prediction. Using segregation analysis of population-based samples of 4,390 families of prostate cancer patients from the UK and Australia, and assuming all familial aggregation has genetic causes, we previously found that the best model for the genetic susceptibility to prostate cancer was a mixed model of inheritance that included both a recessive major gene

Modeling repeated ordinal responses using a family of power transformations: application to neonatal hypothermia data.

BACKGROUND: For analyzing a repeated ordinal response, it is common to use a multivariate cumulative logit model. This model may fit poorly, especially when a nonsymmetric response is available. In these cases, alternative strategies should be utilized. METHODS: In this paper, we present a family of power transformations for the cumulative probabilities to model asymmetric departures from the random-intercept cumulative logit model. To illustrate this method, we analyze the data from an epidemiologic study to identify risk factors of hypothermia among newly born infants in some referral unive

A One-Health integrated approach to control fascioliasis in the Cajamarca valley of Peru.

Fasciola hepatica infection is reported from many Latin American countries, with very high prevalence rates in both humans and livestock in the Andean countries. Due to its environmental characteristics, particularly suitable for liver fluke infection, the Cajamarca valley of Peru has often been chosen as a model to study the epidemiology of liver fluke infection in the Andes. In this paper we describe the profile of a project aimed at a multidisciplinary and integrated approach for the control of fascioliasis in animals and humans in this valley. The One-Health integrated approach applied her
