Hypothalamic-pituitary-thyroid (HPT) axis functioning in anxiety disorders. A systematic review.

Depression has repeatedly been linked to subclinical hypothyroidism, and thyroid hormones have successfully been used to augment antidepressant treatment. By contrast, the extent of thyroid dysfunction in anxiety disorders remains less clear. This is surprising, given that anxiety-related symptoms (e.g., nervousness, palpitations, increased perspiration) are highly prevalent in hyperthyroidism. The present study was undertaken to synthesize the literature on hypothalamic-pituitary-thyroid (HPT) axis functioning in anxiety disorders. The PubMed and PsycINFO databases were systematically searche

Role of the Orexin System on the Hypothalamus-Pituitary-Thyroid Axis.

Hypocretin/orexin (ORX) are two hypothalamic neuropeptides discovered in 1998. Since their discovery, they have been one of the most studied neuropeptide systems because of their projecting fields innervating various brain areas. The orexinergic system is tied to sleep-wakefulness cycle, and narcolepsy is a consequence of their system hypofunction. Orexinergic system is also involved in many other autonomic functions such as feeding, thermoregulation, cardiovascular and neuroendocrine regulation. The main aim of this mini review article is to investigate the relationship between ORX and thyroi

Hypothalamic-pituitary-adrenocortical axis dysfunction in epilepsy.

Epilepsy is a common neurological disease, affecting 2.4million people in the US. Among the many different forms of the disease, temporal lobe epilepsy (TLE) is one of the most frequent in adults. Recent studies indicate the presence of a hyperactive hypothalamopituitary- adrenocortical (HPA) axis and elevated levels of glucocorticoids in TLE patients. Moreover, in these patients, stress is a commonly reported trigger of seizures, and stress-related psychopathologies, including depression and anxiety, are highly prevalent. Elevated glucocorticoids have been implicated in the development of str

Adrenal insufficiency secondary to lymphocytic panhypophysitis in a cat.

CASE REPORT: A 13-year-old male castrated Domestic Shorthair cat was presented for investigation of lethargy, vomiting, polydipsia and polyu … atrophy and the changes in the pituitary gland were suggestive of a T-cell-rich immune-mediated panhypophysitis. CLINICAL SIGNIFICANCE: Secondary adrenal insufficiency and panhypophysitis have not been previously reported in the cat. This report should raise awareness of this rare but potentially treatable disease process.

Metabolic syndrome, activity of the hypothalamic-pituitary-adrenal axis and inflammatory mediators in depressive disorder.

Depression has been associated with various cardiovascular risk factors such as hypertension, obesity, atherogenic dyslipidemia and hyperglycemia. In depressive disorder, hyperactivity of the hypothalamic-pituitary-adrenal (HPA) axis and changes in the immune system have been observed. On the other hand, somatic diseases such as obesity, hyperlipidemia, hypertension and diabetes mellitus type 2 are now perceived as important comorbid conditions in patients with depression. The pathogenesis of the metabolic syndrome and depression is complex and poorly researched; however, it is considered that

A minimal physiologically based model of the HPA axis under influence of the sleep-wake cycles.

The hypothalamic-pituitary-adrenal axis (also called the HPA or stress axis) exhibits distinct circadian and ultradian rhythms in cortisol release that cannot be explained solely by the feedback loops from cortisol to the control systems in the paraventricular nucleus (PVN) and pituitary gland. The HPA axis is intimately connected with other brain functions. In particular, it is strongly affected by the sleep-wake cycles via direct and indirect effects of the circadian and homeostatic mechanisms. For example, the HPA axis has direct inputs from the master circadian clock in the suprachiasmatic

Recent advances in reproductive neuroendocrinology: a role for RFamide peptides in seasonal reproduction?

Most temperate-zone species use photoperiod to coordinate breeding and ensure that offspring are born during favourable conditions. Although photoperiodic influences on the reproductive axis have been well characterized, the precise mechanisms by which photoperiodic information and other seasonal cues are integrated to regulate reproductive function remain less well specified. Two recently discovered neuropeptides, kisspeptin and gonadotropin-inhibitory hormone, have pronounced opposing influences on reproductive function. This paper will review recent evidence for a role of these peptides in

Innovative approaches for the treatment of depression: targeting the HPA axis.

Altered activity of the hypothalamic pituitary adrenal (HPA) axis is one of the most commonly observed neuroendocrine abnormalities in patients suffering from major depressive disorder (MDD). Altered cortisol secretion can be found in as many as 80% of depressed patients. This observation has led to intensive clinical and preclinical research aiming to better understand the molecular mechanisms which underlie the alteration of the HPA axis responsiveness in depressive illness. Dysfunctional glucocorticoid receptor (GR) mediated negative feedback regulation of cortisol levels and changes in arg

Hypopituitarism oddities: congenital causes.

BACKGROUND: The last 15 years have witnessed an explosion in our knowledge of hypothalamo-pituitary development, acquired mainly from naturally-occurring and transgenic animal models. A complex genetic cascade dictates organ commitment, cell differentiation and cell proliferation within the anterior pituitary. Mutations in genes encoding both signalling molecules and transcription factors have been implicated in the aetiology of hypopituitarism, with or without other syndromic features, in mice and humans. These include HESX1, LHX3, LHX4, PROP1, POU1F1 and, more recently, SOX3 and SOX2. Phenot

Hormonal changes in aging men: a therapeutic indication?

The rise in male life expectancy is paralleled by increased age-related clinical signs and symptoms such as muscle weakness, osteoporosis, benign prostatic hyperplasia, changes in body composition, fatigue, decreased sexual interest and activity, and increased prevalence of erectile dysfunction, all of which limit the quality of life. Many of these symptoms are similar to those of clinically well-defined hormone deficiencies, e.g. Kallman syndrome, Prader--Labhart--Willi syndrome or deficiencies due to treatment of pituitary tumors. Three male endocrine axes are characterized by age-related ch

[Neonatal detection of central hypothyroidism].

BACKGROUND: Congenital hypothyroidism is very rare compared to primary hypothyroidism. Its early diagnosis may escape neonatal mass screening using TSH assay. CASE REPORT: Anthony was born at 37 weeks, weighing 3,060 g. He presented with hypotony, jaundice, tongue protrusion evoking congenital hypothyroidism. Thyroid function tests favored hypothyroidism central in origin, while the systematic neonatal screening was normal. CONCLUSION: Clinical signs of congenital hypothyroidism must lead to more specific tests when neonatal screening is normal.

Perturbations of hypothalamic-pituitary-gonadal (HPG) axis and adrenal androgen (AA) functions in rheumatoid arthritis.

The available evidence reviewed does not allow definitive response to the question of a primary versus secondary role of sex hormone perturbations in RA. However, this conclusion should not be discouraging in view of the relatively recent focus upon this facet of the physiopathogenesis of RA and the enormous complexities of sex hormone biology and this disease. Specifically, data on the incidence of RA as well as life cycle changes in serum androgenic-anabolic (A-A) and sex hormone levels suggest important risk correlations. Furthermore, HLA-susceptibility markers for RA, gender, menopause and

Hypothalamo-pituitary effects of RU486: inhibition of progesterone-induced hyperprolactinaemia.

Monkeys given oestrogen priming at physiological levels for at least 1 week become hyperprolactinaemic upon the addition of physiological progesterone administration. Here, using RU486, we test whether that this oestrogen/progestin-induced hyperprolactinaemia results from classical progesterone actions at the hypothalamo-pituitary level. Blood samples were collected daily from study day 1-67. Each monkey (n = 2) received daily injections of 25 micrograms/kg oestradiol benzoate, i.m., on study days 5-60. Progesterone-filled silastic capsules (3 cm) were inserted on study day 14 and removed on d

[Functional anatomy of the hypothalamic-hypophysial system: neuroendocrine control mechanism].

The hypothalamic neuroendocrine mechanisms involved in the regulation of the anterior pituitary functions were reviewed on the basis of the findings obtained mainly in rats. The hypothalamic-hypophysial system has long been considered as a hormonary associated organ system. Recent development of immunohistochemistry and molecular biology, however, has localized the hormone-synthesizing neurons in the hypothalamus, and disclosed the synaptic junctions between them, between those neurons and non-hormonal neurons in the hypothalamus, and further, between neurons in the hypothalamus and other brai

Neuroendocrinology of pyridoxine deficiency.

Dihydroxyphenylalanine decarboxylase and 5-hydroxytryptophan decarboxylase respectively have high and low affinities for pyridoxal phosphate. In the pyridoxine-deficient animal, hypothalamic serotonin content is significantly reduced without any change in catecholamine levels. Hypothalamic neurotransmitters affect the hypothalamo-pituitary-end organ axes. Specifically, the decrease in hypothalamic serotonin in the pyridoxine-deficient rat results in tertiary hypothyroidism. In addition, pineal function is affected in deficient animals due to decreased synthesis of melatonin.

The hypothalamic neurosecretory system of the marine teleost fish, Mugil auratus risso.

The hypothalamic neurosecretory system of the marinefish, Mugil auratus, consists of two nuclei, viz., the nucleus praeopticus and the nucleus laterlis tuberis. Both are paired, and while those of the nucleus praeopticus are vertically arranged as L-shaped bodies, their strand-like counterparts in the nucleus lateralis tuberis extend in an antero-posterior direction. The two constituent bodies of the nucleus praeopticus lie on both sides of the third cerebral ventricle. Each is differentiated into a dorsal pars magnocellularis and a ventral pars parvocellularis. A nervous tract, the hypothalam
