In the absence of acceptable cures for ocular diseases, stem cells and their paracrine agents, including exosomes, have become a promising therapeutic approaches. Exosomes are lipid spherical bilayer-encased particles and the smallest subtype of extracellular vehicles (EVs) that are secreted by cells to facilitate intercellular communication in the form of lipids, proteins, mRNAs, and small RNAs. Recent advances in exosome-based therapeutics have witnessed unique opportunities for treating ocular diseases, including autoimmune diseases, traumatic diseases, chorioretinal diseases, and uveal mel

Nano-sized outer membrane vesicles (OMVs) are lipid-bilayered structures that primarily encapsulate periplasmic components, with minor inclusion of cytoplasmic materials. Rather than passive byproducts of cellular damage, OMVs are now understood as active mediators of bacterial physiology, environmental adaptation, and host interaction. Recent evidence identifies envelope instability as a key mechanistic driver of OMV biogenesis. Disruptions in outer membrane-peptidoglycan connectivity, imbalances in periplasmic homeostasis, and alterations in lipid asymmetry collectively promote vesicle forma

Plasma proteins (human serum albumin, immunoglobulin gamma-1, complement C3, and fibrinogen) are simulated with 26 and 36 nm-sized cationic, anionic, and zwitterionic liposomes in water using coarse-grained models. Proteins, initially placed randomly around a liposome, adsorb more significantly onto charged liposomes than onto zwitterionic liposomes through electrostatic interactions with lipid headgroups and hydrophobic interactions with lipid tails, with the greatest adsorption observed on cationic liposomes, in agreement with experimental observations. For zwitterionic liposomes, adsorption

Extracellular vesicles (EVs), including exosomes, microvesicles, and apoptotic bodies, are lipid bilayer-bound nanoparticles secreted by various cell types. EVs mediate intercellular communication by selectively transporting bioactive molecules including proteins, lipids, and nucleic acids. This review comprehensively analyzes EVs biogenesis, classification, and cargo diversity, emphasizing their multifaceted roles in physiological processes and cancer. EVs exhibit intrinsic dual regulatory roles in tumor biology. On the one hand, they promote malignant tumor progression by delivering pro-tumo

Chemotherapy-induced cardiotoxicity (CIC) is a major limitation of modern anticancer therapy, particularly with anthracyclines and targeted agents, and contributes to long-term cardiovascular complications in cancer survivors. Extracellular vesicles (EVs) are nanoscale lipid-bilayer particles that mediate intercellular communication through the transfer of proteins, nucleic acids, and lipids. Emerging evidence indicates that EVs play a dual role in CIC. On one hand, EVs released from tumor cells or stressed cardiomyocytes can propagate cardiotoxic signals by modulating pathways related to oxid

Secondary iron overload disorders demand effective strategies for removing excess iron, yet conventional chelators such as deferoxamine (DFO) are limited by poor pharmacokinetics, low bioavailability, and dose-dependent toxicities. Here we introduce DFOsomes, a first reported lipidized DFO nanomedicine. Developed through hydrophobic modification and self-assembly into highly stable bilayer nanostructures, DFOsomes combine high chelator density with markedly improved cellular uptake and prolonged circulation. Leveraging DFO's metal-binding affinity, DFOsomes were radiolabeled with 89Zr to enabl

In eukaryotic cells, the endoplasmic reticulum (ER) utilizes membrane fusion to form interconnected networks. GTP hydrolysis-triggered conformational changes in ER-anchored atlastins (ATLs) have been shown to be responsible for this process, which is facilitated by the amphipathic helix within its C-terminal tail (AHATL-C). However, how the multiple ATL molecules cooperate to complete membrane tethering and subsequent fusion is still not known. Here, we used a programmable DNA origami ring to generate uniformly sized proteoliposomes containing a controlled maximum number of Drosophila ATL (dmA

Extracellular vesicles (EVs), particularly small extracellular vesicles (sEVs), are lipid bilayer-delimited particles involved in intercellular communication through the transfer of proteins, lipids, and nucleic acids; many products and studies in aesthetic medicine refer to these preparations as exosomes, although endosomal origin is not always demonstrated. This review examines current evidence on the mechanisms, clinical effectiveness, safety, therapeutic applications, and regulatory challenges of EV- and sEV-based interventions, complemented by an exploratory qualitative assessment of phys

Placenta-derived extracellular vesicles (EVs) are emerging as critical regulators of maternal-fetal communication during pregnancy. These lipid bilayer-enclosed particles, primarily secreted by trophoblasts, transport bioactive cargos-including RNAs, proteins, lipids and neurotransmitters-that influence a wide range of developmental and immunological processes. While much attention has been given to their roles in maternal adaptation and health outcomes, recent studies highlight their direct impact on fetal development, particularly fetal brain development. Emerging evidence suggests that plac

Extracellular vesicles (EVs) are lipid bilayer nano- to micro-sized particles that carry biomolecules, such as proteins, lipids, and genetic material. Their composition depends on the cellular microenvironment and the health status of tissues. EVs are released by different cell types under distinct circumstances, mediating intercellular communication in both physiological and pathological contexts. In Alzheimer's disease (AD), EVs have been shown to influence pathological events, carrying neurotoxins, such as neuroinflammatory factors, pathogenic forms of amyloid-β, and phosphorylated tau into

Exosomes are small vesicles covered by a lipid bilayer, ranging in size from 50 nm to 90 nm, secreted by different cell types in the body under normal and pathological conditions. They are surrounded by cell-segregated membrane complexes and play a role in the pathological and physiological environments of target cells by transfer of different molecules such as microRNA (miRNA). Exosomes have been detected in many body fluids, such as in the amniotic fluid, urine, breast milk, blood, saliva, ascites, semen, and bile. They include proteins, lipids, and nucleic acids such as DNA, RNA, and miRNA,

pH responsive drug delivery systems are one of the new strategies to address the spread of bacterial resistance to currently used antibiotics. The aim of this study was to formulate liposomes with 'On' and 'Off'' pH responsive switches for infection site targeting. The vancomycin (VCM) loaded liposomes had sizes below 100 nm, at pH 7.4. The QL-liposomes had a negative zeta potential at pH 7.4 that switched to a positive charge at acidic pH. VCM release from the liposome was quicker at pH 6 than pH 7.4. The OA-QL-liposome showed 4-fold lower MIC at pH 7.4 and 8- and 16-fold lower at pH 6.0 agai

Islet Amyloid Polypeptide (IAPP) is a 37-residue hormone cosecreted with insulin by the β-cells of the pancreas. Amyloid fiber aggregation of IAPP has been correlated with the dysfunction and death of these cells in type II diabetics. The likely mechanisms by which IAPP gains toxic function include energy independent cell membrane penetration and induction of membrane depolarization. These processes have been correlated with solution biophysical observations of lipid bilayer catalyzed acceleration of amyloid formation. Although the relationship between amyloid formation and toxicity is poorly

Micropipet aspiration of phase-separated lipid bilayer vesicles can elucidate physicochemical aspects of membrane fluid phase coexistence. Recently, we investigated the composition dependence of line tension at the boundary between liquid-ordered and liquid-disordered phases of giant unilamellar vesicles obtained from ternary lipid mixtures using this approach. Here we examine mechanical equilibria and stability of dumbbell-shaped vesicles deformed by line tension. We present a relationship between the pipet aspiration pressure and the aspiration length in vesicles with two coexisting phases.

OpcA is an integral outer membrane adhesin protein from Neisseria meningitidis, the causative agent of meningococcal meningitis and septicemia. It binds to sialic acid (SA)-containing polysaccharides on the surface of epithelial cells. The crystal structure of OpcA showed that the protein adopts a 10-stranded beta-barrel structure, with five extensive loop regions on the extracellular side of the membrane. These form a crevice structure, lined with basic residues, which was hypothesized to act as the binding site for polysaccharide ligands. In the current study, a distinctly different OpcA str

In this study we developed emulsomes, a novel lipoidal vesicular system with an internal solid fat core surrounded by a phospholipid bilayer. Plain emulsomal formulations composed of solid lipid (trilaurin or tristearin), cholesterol and soya phosphatidylcholine and stearylamine containing cationic emulsomes loaded with an antiviral drug (zidovudine) were prepared by a simple cast film method followed by sonication to produce emulsomes of nanometric size range. All different types of formulations were optimized for lipid ratios and characterized in-vitro for shape, morphology, size and in-vitr

The overall picture of platelet abnormalities in diabetes mellitus (DM), including altered adhesion and aggregation, is hypersensitivity of diabetic platelets to agonists. "Primed" diabetic platelets respond more frequently even to subthreshold stimuli, sooner become exhausted, consumed and finally hyposensitive, thus contributing to accelerated thrombopoiesis and release of 'fresh' hyperreactive platelets. In diabetes disturbed carbohydrate and lipid metabolism may lead to physicochemical changes in cell membrane dynamics, and consequently result in altered exposure of surface membrane recept

Many different amphiphilic compounds cause an increase in the fluid-phase endocytosis rates of cells in parallel with a decrease in membrane-cytoskeleton adhesion. These compounds, however, do not share a common chemical structure, which leaves the mechanism and even site of action unknown. One possible mechanism of action is through an alteration of inositol lipid metabolism by modifying the cytoplasmic surface of the plasma membrane bilayer. By comparing permeable amphiphilic amines used as local anesthetics with their impermeable analogs, we find that access to the cytoplasmic surface is ne

N-Acylphosphatidylethanolamines and N-acylphosphatidylserines have been isolated from mammalian cells and have been associated with some tissue degenerative changes, although the relationship between their synthesis and the uncontrolled sequence of events that ends in irreversible tissue damage is not completely established. Our results show that monovalent and divalent cations induce aggregation and fusion of liposomes constituted by N-palmitoylphosphatidylethanolamine (NPPE) and N-palmitoylphosphatidylserine (NPPS). The effectiveness of cations to induce the aggregation of NPPE and NPPS lipo

Coughing, sneezing, talking, bed-making, turning pages of books, etc. all generate microbial aerosols which are carried and dispersed by air movements. Inhalation of these particles may cause allergic responses but whether or not infectious disease ensues depends in part on the viability and infectivity of the inhaled microbes and their landing sites. Desiccation is experienced by all airborne microbes; gram-negative bacteria and lipid-containing viruses demonstrate phase changes in their outer phospholipid bilayer membranes owing to concomitant changes in water content and/or temperature.
