Negative selection by IgM superantigen defines a B cell central tolerance compartment and reveals mutations allowing escape.

To analyze B lymphocyte central tolerance in a polyclonal immune system, mice were engineered to express a superantigen reactive to IgM of allotype b (IgM(b)). IgM(b/b) mice carrying superantigen were severely B cell lymphopenic, but small numbers of B cells matured. Their sera contained low levels of IgG and occasionally high levels of IgA. In bone marrow, immature B cells were normal in number, but internalized IgM and had a unique gene expression profile, compared with those expressing high levels of surface IgM, including elevated recombinase activator gene expression. A comparable B cell

Impaired glucose tolerance and predisposition to the fasted state in liver glycogen synthase knock-out mice.

Conversion to glycogen is a major fate of ingested glucose in the body. A rate-limiting enzyme in the synthesis of glycogen is glycogen synthase encoded by two genes, GYS1, expressed in muscle and other tissues, and GYS2, primarily expressed in liver (liver glycogen synthase). Defects in GYS2 cause the inherited monogenic disease glycogen storage disease 0. We have generated mice with a liver-specific disruption of the Gys2 gene (liver glycogen synthase knock-out (LGSKO) mice), using Lox-P/Cre technology. Conditional mice carrying floxed Gys2 were crossed with mice expressing Cre recombinase u

Peroxiredoxin 4 knockout results in elevated spermatogenic cell death via oxidative stress.

Prx (peroxiredoxin) is a multifunctional redox protein with thioredoxin-dependent peroxidase activity. Prx4 is present as a secretory protein in most tissues, whereas in sexually mature testes it is anchored in the ER (endoplasmic reticulum) membrane of spermatogenic cells via an uncleaved N-terminal hydrophobic peptide. We generated a Prx4 knockout mouse to investigate the function of Prx4 in vivo. Prx4(-/y) mice lacking Prx4 expression in all cells were obtained by mating Prx4(flox/+) female mice with Cre-transgenic male mice that ubiquitously expressed Cre recombinase. The resulting Prx4(-/

Recombination between ccrC genes in a type V (5C2&5) staphylococcal cassette chromosome mec (SCCmec) of Staphylococcus aureus ST398 leads to conversion from methicillin resistance to methicillin susceptibility in vivo.

Various types of the staphylococcal cassette chromosome mec (SCCmec) are known to confer methicillin resistance on the human pathogen Staphylococcus aureus. Such cassettes are not always stably maintained. The present studies were aimed at identifying the mechanism underlying the in vivo conversion of methicillin-resistant S. aureus (MRSA) to methicillin-susceptible S. aureus (MSSA) derivatives as encountered in two patients suffering from pneumonia and an umbilicus infection, respectively. All MRSA and MSSA isolates identified belong to multilocus sequence type (MLST) 398, have spa type t034,

Triggering genetically-expressed transneuronal tracers by peripheral axotomy reveals convergent and segregated sensory neuron-spinal cord connectivity.

To better understand the mechanisms through which non-painful and painful stimuli evoke behavior, new resources to dissect the complex circuits engaged by subsets of primary afferent neurons are required. This is especially true to understand the consequences of injury, when reorganization of central nervous system circuits likely contributes to the persistence of pain. Here we describe a transgenic mouse line (ZWX) in which there is Cre-recombinase-dependent expression of a transneuronal tracer, wheat germ agglutinin (WGA), in primary somatic or visceral afferent neurons, but only after trans

Heparan sulfate deficiency in periocular mesenchyme causes microphthalmia and ciliary body dysgenesis.

The heparan sulfate (HS) is a component of proteoglycans in the extracellular matrix and on cell surfaces, modulating developmental processes. The aim of this study is to investigate whether the defect of HS in the periocular mesenchyme impairs ocular morphogenesis. First, using Protein 0-Cre transgenic mice, we ablated Ext1, which encodes an indispensable enzyme for HS synthesis, in the developing periocular mesenchyme. The expression of Ext1 messenger RNA (mRNA) and HS were observed by RT-PCR and immunohistochemistry, respectively. The phenotypes in the mutant were evaluated by light microsc

Skeletal abnormalities and extra-skeletal ossification in mice with restricted Gsalpha deletion caused by a renin promoter-Cre transgene.

We have recently generated a transgenic mouse line (termed hRen-Cre) that expresses Cre-recombinase under the control of a 12.2-kb fragment of the human renin promoter. In the present study, we have crossed hRen-Cre mice with a mouse strain in which exon 1 of the Gnas gene is flanked by loxP sites. Gnas encodes the alpha-subunit of the stimulatory G protein (Gs alpha). Our aim has been to generate a mouse model with locally restricted inactivation of Gs alpha to extend studies of the role of Gs alpha function in vivo. Mice with local Cre-mediated inactivation of Gs alpha (rCre-Gs alpha) are vi

Brain-derived neurotrophic factor supports facial motoneuron survival after facial nerve transection in immunodeficient mice.

Numerous studies have shown that motoneuron survival can be facilitated by neurotrophic factors (NTF) after injury. However, the ability of specific NTF to rescue facial motoneurons (FMN) from axotomy-induced death in immunodeficient mice has not been tested. Therefore, one goal of this study was to determine if brain-derived neurotrophic factor (BDNF), an NTF with a known ability to rescue FMN from axotomy-induced death, supports FMN from axotomy-induced death in recombinase activating gene-2 knockout (RAG-2 KO) mice that lack functional T and B lymphocytes. Neisseria gonorrhoeae secretes chromosomal DNA via a novel type IV secretion system.

The process of DNA donation for natural transformation of bacteria is poorly understood and has been assumed to involve bacterial cell death. Recently in Neisseria gonorrhoeae we found that mutations in three genes in the gonococcal genetic island (GGI) reduced the ability of a strain to act as a donor in transformation and to release DNA into the culture. To better characterize the GGI and the process of DNA donation, the 57 kb genetic island was cloned, sequenced and subjected to insertional mutagenesis. DNA sequencing revealed that the GGI has characteristics of a horizontally acquired geno

Novel non-mecA-containing staphylococcal chromosomal cassette composite island containing pbp4 and tagF genes in a commensal staphylococcal species: a possible reservoir for antibiotic resistance islands in Staphylococcus aureus.

Among methicillin-resistant Staphylococcus aureus isolates, a staphylococcal chromosomal cassette containing the mecA gene (SCCmec) is integrated into the chromosome at a unique site. SCCmec also contains unique ccrAB recombinase genes mediating its integration and excision from the genome and is flanked by characteristic left and right direct- and inverted-repeat sequences. A few non-mecA-containing SCC elements that have the other molecular features described above have recently been described. The origin of these cassettes is not clear. We have identified two new members of the SCC family i

Antineutrophil cytoplasmic autoantibodies specific for myeloperoxidase cause glomerulonephritis and vasculitis in mice.

Antineutrophil cytoplasmic autoantibodies (ANCAs) are identified in the circulation of approximately 80% of patients with pauci-immune necrotizing and crescentic glomerulonephritis and systemic small vessel vasculitis, such as microscopic polyangiitis and Wegener granulomatosis. The most common antigen target for ANCAs is myeloperoxidase (MPO), which is found in neutrophils and monocytes. We report definitive experimental animal evidence that ANCAs are pathogenic. MPO knockout (Mpo(-/-)) mice were immunized with mouse MPO. Splenocytes from these mice or from control mice were injected intraven

Immunoglobulin heavy chain variable region gene replacement As a mechanism for receptor revision in rheumatoid arthritis synovial tissue B lymphocytes.

Mature B cells can alter their antibody repertoires by several mechanisms, including immunoglobulin heavy chain variable region (V(H)) replacement. This process changes the antigen combining site by replacing a portion of the original V(H)/diversity/heavy chain joining region (V(H)DJ(H)) rearrangement with a corresponding portion of a new V(H) segment. This exchange can involve cryptic heptamer-like sequences embedded in the coding regions of V(H) genes. While studying the B lymphocytes that expand in the synovial tissues of patients with rheumatoid arthritis (RA), clones with V(H)DJ(H) varian

Development of a rapid RPA-CRISPR/Cas12b assay for dual detection of Hantaan virus and Seoul virus.

Hantaan virus (HTNV) and Seoul virus (SEOV) are the primary causative agents of hemorrhagic fever with renal syndrome (HFRS), presenting a persistent challenge to global public health. Prompt and precise diagnostic tools are essential for clinical investigation and the containment of HFRS outbreaks. Leveraging the programmable precision of CRISPR/Cas12b system, which has significantly advanced the field of molecular diagnostics, we developed a rapid, high-sensitivity assay for the dual detection of HTNV and SEOV. Each virus was detected in a separate, target-specific single-tube reaction. The

Primer-programmed circular template generation enables padlock-free rolling circle amplification for low-background CRISPR-based pathogen detection.

Rolling circle amplification (RCA) is a powerful nucleic acid amplification strategy, yet conventional RCA-based biosensing often relies on target-specific padlock probes for circular template construction, which increases probe-design complexity and may introduce nonspecific ligation and background amplification. Here, we develop a padlock-free RCA strategy for CRISPR-based pathogen detection through primer-programmed circular template generation (PPCTG) strategy. Asymmetric recombinase polymerase amplification (RPA) with rationally designed phosphorylated primers generates target-derived sin

Nanoporous gold electrode-assisted CRISPR/Cas12a electrochemical detection of synthetic methylated DNA models for breast cancer liquid-biopsy development.

Circulating tumor DNA (ctDNA) methylation is a promising liquid-biopsy signal for early breast cancer, but low target abundance, fragmented cell-free DNA (cfDNA), and matrix fouling remain major barriers to electrochemical implementation. Here, a nanoporous gold (NPG) electrode-assisted CRISPR/Cas12a electrochemical assay was developed for synthetic methylated DNA models that mimic breast-cancer-associated cfDNA promoter fragments. The assay integrates HpaII methylation-sensitive restriction-enzyme (MSRE) digestion, recombinase polymerase amplification (RPA), Cas12a trans-cleavage, and methyle

Cut-and-Bind Colorimetric Detection of Viral and Non-Coding RNAs Using CRISPR/Cas12a with Zinc Finger-Facilitated Gold Nanoparticle Assembly.

Accurate and rapid detection of RNA biomarkers is crucial for early disease diagnosis and infectious disease management. Although RT-qPCR remains the gold standard, its utility for point-of-care testing is limited by run-to-run variability and dependence on specialized instrumentation. Conversely, colorimetric assays offer operational simplicity but generally fall short of the sensitivity and specificity demanded for reliable applications. To bridge this gap, we present an innovative cut-and-bind colorimetric RNA detection platform that synergistically integrates the sequence-specific programm

Leptin alleviates obesity hypoventilation via serotonergic pathways.

RATIONALE: There is no effective pharmacotherapy for obesity hypoventilation syndrome (OHS). Intranasal leptin augments the hypercapnic ventilatory response (HCVR), attenuates upper airway obstruction, and increases ventilation during sleep in diet-induced obese (DIO) mice. Respiratory effects of leptin can be attenuated by serotonergic antagonists. OBJECTIVES: To establish if serotonergic innervation of the hypoglossal motoneurons (XII MN) mediates effects of leptin on OHS. METHODS: We examined effects of intranasal leptin on the HCVR, sleep architecture, arousal latency, flow limited (obst

Direct Comparison of Constitutive Rax-Cre Transgenic Drivers That Activate in the Mouse Embryonic Eye Field.

PURPOSE: Multiple mouse lines using constitutive or inducible Cre recombinase expression have taken advantage of the early optic field expression of the Rax/Rx gene and its subsequent, progressive restriction to retinal progenitors, RPE, and the optic stalk. Among these, the constitutive transgenic lines, Rx3-Cre and Rax-Cre BAC Tg, are currently used by vision researchers. Here we directly compare their prenatal ocular and extraocular Cre activities. METHODS: Rx3-Cre or Rax-Cre BAC transgenic male mice were mated to the Cre-dependent lineage tracer Ai9/tdTomato. The resulting live and fixed

Development of an hAPE1-based RT-RAA method for rapid detection of Sweet potato chlorotic stunt virus.

Sweet potato chlorotic stunt virus (SPCSV) is a major pathogen that threatens sweet potato production. It frequently acts synergistically with other viruses, causing severe yield losses and quality decline. A rapid and efficient detection method is therefore essential for preventing and controlling the spread of this virus. Here, we developed a reverse transcription real-time fluorescence recombinase-aided amplification (RT-RAA) assay using human apurinic/apyrimidinic endonuclease 1 (hAPE1, designated as tool enzyme E10). This enzyme exhibits high AP endonuclease activity but extremely low exo
