Vascular involvement in a patient with mitochondrial myopathy, encephalopathy, lactic acidosis, and stroke-like episodes.

A 26-year-old man with mitochondrial myopathy, encephalopathy, lactic acidosis, and stroke-like episodes (MELAS) was admitted to our hospital for further cardiovascular examination. A muscle biopsy demonstrated strongly succinate dehydrogenase-reactive blood vessels. Pulse wave contour analysis revealed that both capacitive and oscillatory compliance were markedly reduced in this patient compared with 45 normal age-matched control subjects. Hepatocyte growth factor was remarkably elevated in this patient over that of 10 normal control subjects. These findings suggest that a MELAS patient has n

Mitochondrial dysfunction and rod-like lesions associated with administration of beta2 adrenoceptor agonist formoterol.

Formoterol, a selective beta 2-adrenoceptor agonist, has been introduced recently in the treatment of poorly controlled asthma. A patient is presented who developed myalgia and muscle weakness, associated with an elevation of creatine kinase (CK) during treatment with formoterol. Subsequent muscle biopsy demonstrated atrophic fibres lacking cytochrome C-oxidase and succinate dehydrogenase, suggestive of mitochondrial dysfunction. There were no inflammatory changes. Immunocytochemical analysis using antibodies to alpha-actinin-2 and alpha-actinin-3 demonstrated positive staining of 'rod-like' b

Brugia malayi: status of host during different stages of infection.

The status of glycogen, protein, lipid components, lipid peroxides and a few enzymes of energy metabolism was studied in liver of Mastomys natalensis during the development of Brugia malayi infection. Glycogen and lipid contents were decreased during the patent phase of infection while total protein was slightly altered in latent animals. Phospholipid and triglyceride contents declined at prepatent and patent phase of infection. The levels of lactate and malate dehydrogenases, as well as of adenosine triphosphatases (Na+K+, Mg2+, Ca2+), were significantly elevated and monoamine oxidase activit

Multiple cytochrome deficiency and deteriorated mitochondrial polypeptide composition in fatal infantile mitochondrial myopathy and renal dysfunction.

Mitochondria isolated from the skeletal muscle of an infant with mitochondrial myopathy and renal dysfunction were analyzed. Activities of NADH dehydrogenase, succinate dehydrogenase, ubiquinol-cytochrome c oxidoreductase, and cytochrome c oxidase were severely decreased. Cytochromes aa3 and b were not detected in patient mitochondria, and the cytochrome c+c1 content was 14% of control. Immunoblotting demonstrated that the amount of cytochrome c oxidase subunits were markedly decreased in patient mitochondria. The polypeptide profile of patient mitochondria was quite different from that of con

[Histoenzymologic characteristics of the myocardium in sudden death in patients with alcoholic cardiomyopathy].

Study of the activity of some myocardial enzymes in sudden death cases with alcoholic cardiomyopathy (ACMP) was made by quantitative histochemical methods. The decrease of dehydrogenase activity of succinate, lactate, beta-hydroxybutyrate, alpha-glycerophosphate and NAD-diaphorase was found in line with the increase of the activity of glucose-6-phosphate dehydrogenase, alcohol dehydrogenase and catalase versus control (myocardium of those who died of trauma). Disorders of major metabolic processes in the myocardium may occasionally lead to electrical instability resulting in ventricular fibril

Schistosoma haematobium infection in the white mouse--histopathological and histochemical studies.

Mice were infected with 1000 Schistosoma haematobium cercariae (Egyptian strain). Histopathological and histochemical studies were performed on the different organs, during the first four weeks after infection and on the fourth week after oviposition. Pathological changes during early prepatency matched with those in S. mansoni infection. Eggs were laid in aggregates in the colon and liver. They initiated the development of typical granulomatous lesions. Abundant bilharzial pigment and areas of sclerosis were present in both liver and spleen. The heart, kidneys and urinary bladder were patholo

Histochemical observations on muscle from normal and malignant hyperthermia-susceptible swine.

A comparison was made of muscle from two locations in both the longissimus and the semitendinous muscles of normal and malignant hyperthermia-susceptible swine. Serial frozen sections were stained for alkali-stable adenosine triphosphatase (ATPase), phosphorylase, and the oxidative enzymes succinate dehydrogenase and reduced nicotinamide adenine dinucleotide (NADH)-diaphorase. Myofiber types were identified on the basis of these staining reactions. There was no consistent statistically significant difference between muscle from normal and muscle from susceptible swine with any system of fiber

[Iron, copper, zinc and cobalt content and activity of respiratory metalloenzymes in animal tissues under toxic hypoxia].

The rabbits being repeatedly poisoned with small doses of sodium cyanide, the activity of succinic dehydrogenase in the tissues does not essentially change. The activity of NAD.H2-cytochrome-c-reductase and NAD.H2-diaphorase in the brain, myocardium and kidneys increases. Under histotoxic hypoxia the level of iron in the tissues increases by 52-93%, that of copper--by 28-36%, of zinc--by 21-74% and of cobalt by 28-40%. There existed a positive correlation between the content of iron and the activity of NAD-dependent enzymes. In nonlethal form of histotoxic hypoxia the content of nonhemin iron

[The blocking action of snake venom polypeptides on the cholinergic mechanisms of leech dorsal muscle].

The blocking action of alpha-polypeptides from the venoms of elapid snakes (Bungarus multicinctus and Naja naja siamensis) was studied on leech dorsal muscle. It was shown that these neurotoxins failed to inhibit the responses to mononitrogen cholinomimetics (acetylcholine, nicotine, carbamylcholine, etc.). The capacity of neurotoxins to inhibit the responses to the bisquaternary cholinommimetics depended upon the length of the molecule. The neurotoxins were active against the "shorter" cholinomimetics, such as decamethonium, and bischolinic esters of malonic, succinic and glutaric acids. The

miR-669a-5p promotes adipogenic differentiation and induces browning in preadipocytes.

Obesity is a major global health issue that contributes to the occurrence of metabolic disorders. Based on this fact, understanding the underlying mechanisms and to uncover promising therapeutic approaches for obesity have attracted intense investigation. Brown adipose tissue (BAT) can help burns excess calories. Therefore, promoting White adipose tissue (WAT) browning and BAT activation is an attractive strategy for obesity treatment. MicroRNAs (miRNAs) are small, non-coding RNAs, which are involved in regulation of adipogenic processes and metabolic functions. Evidence is accumulating that m

Eye muscle antibodies in patients with ocular myasthenia gravis: possible mechanism for eye muscle inflammation in acetylcholine-receptor antibody-negative patients.

Myasthenia gravis is an organ-specific autoimmune disorder generally thought to be caused by an antibody-mediated attack against the skeletal muscle nicotinic acetylcholine (Ach) receptor (AchR) at the neuromuscular junction. Extraocular muscle weakness and double vision are present in about 90% of patients with myasthenia gravis and are the predominant complaints in about 20% of patients, when the condition is called ocular myasthenia gravis (OMG). While serum antibodies against the AchR are detected in most patients with generalized myasthenia gravis (GMG), they are not found in about one-th

Developmental Toxicity and Mechanistic Insights into the Novel Fungicide Fluindapyr in Zebrafish Embryos.

Succinate dehydrogenase inhibitor (SDHI) fungicides pose potential risks to nontarget aquatic organisms. Fluindapyr, a newly registered SDHI fungicide, is likely to enter aquatic environments, yet its toxicity remains largely unexplored. Here, zebrafish (Danio rerio) embryos were used to assess their developmental toxicity and underlying mechanisms. Fluindapyr exhibited high toxicity, with a 96 h LC50 of 0.36 mg/L, and induced multiple developmental defects including edema, growth retardation, reduced locomotion, and bradycardia. Transcriptomic analysis revealed that differentially expressed g

Macrophage-to-myofibroblast transition-derived itaconate promotes bone metastasis in lung cancer through targeting of HSPA8.

Lung cancer bone metastasis carries a poor prognosis, yet the metabolic determinants driving tumour-stroma crosstalk remain largely elusive. Despite extensive investigations into itaconate, a prominent immunometabolite implicated in macrophage polarization, the precise mechanisms by which it modulates bone metastasis remain unresolved. Integrating metabolomics and transcriptomics profiling, the molecular landscape of lung cancer bone metastasis is delineated and the mechanistic role of itaconate is uncovered. Further ubiquitination proteomics of tumour cells and CRISPR-Cas9-mediated knockout o

A novel germline (c.314T>A) SDHB variant in metastatic paraganglioma: case report and literature review.

INTRODUCTION: most sympathetic paragangliomas are driven by germline pathogenic variants. Identifying germline succinate dehydrogenase B (SDHB) pathogenic variant has important management implications. Here we report a novel germline variant in the SDHB gene in a patient with metastatic paraganglioma and his response to available treatments. CASE PRESENTATION: a 37-year-old Serbian man was admitted to hospital due to hypertension, tachycardia and hyperhidrosis. Screening for secondary hypertension revealed elevated 24-h urinary normetanephrine. A CT scan showed the presence of a 54 x 76 mm re

In Vivo Immunomodulatory Programming of Circulating Myeloid Cells via Itaconic Acid Liposomes to Mitigate Myocarditis.

Myocarditis is fundamentally characterized by the excessive infiltration of inflammatory cells (monocytes and macrophages) within the myocardium, posing a substantial risk of progressive cardiac dysfunction. Blocking the C-C motif chemokine receptor 2 (CCR2) pathway to curtail the infiltration of CCR2+ monocytes can suppress the immune response; nevertheless, these approaches may unwittingly impede the normal migration and activation of myeloid cells, potentially undermining the body's immune defenses. To address this backdrop, we designed itaconic acid liposomes (ITA-Lipo) to modulate the imm

Clinical, genetic, radiological characteristics and management of mediastinal paragangliomas: a literature review and case series.

Paragangliomas (PGLs) are neuroendocrine tumours (NETs) that arise from neural crest-derived cells. Up to 40% of cases occur due to the presence of a pathogenic germline variant (PGV) in a known gene. Mediastinal PGLs are rare but are being diagnosed with increasing frequency. Treatment generally involves surgery but is complicated in mediastinal PGLs due to their anatomy. Here, we will perform a literature review and discuss our experience with 18 such cases. Cases were identified via the Guy's and St Thomas' NHS Foundation Trust NET multidisciplinary team database. Tumours ranged in size fro

Research progress on the pathogenesis of the SDHB mutation and related diseases.

With the improvement of genetic testing technology in diseases in recent years, researchers have a more detailed and clear understanding of the source of cancers. Succinate dehydrogenase B (SDHB), a mitochondrial gene, is related to the metabolic activities of cells and tissues throughout the body. The mutations of SDHB have been found in pheochromocytoma, paraganglioma and other cancers, and is proved to affect the occurrence and progress of those cancers due to the important structural functions. The importance of SDHB is attracting more and more attention of researchers, however, reviews on
