Afferent connections of the interstitial nucleus of the posterior limb of the anterior commissure and adjacent amygdalostriatal transition area in the rat.

The interstitial nucleus of the posterior limb of the anterior commissure is, like the striatum, very rich in tyrosine hydroxylase and acetylcholinesterase, but on the basis of most other neurochemical criteria displays features that are typical of the extended amygdala (Alheid, de Olmos and Beltramino, 1995). Its afferent connections were examined in the rat with retrograde (cholera toxin B subunit) and anterograde (Phaseolus vulgaris leucoagglutinin) tracers and compared to those of the neighboring amygdalostriatal transition area and central amygdaloid nucleus. Deposits of cholera toxin B s

Influence of surfactant on the activity of slowly adapting stretch receptors in the lung.

Since surfactant has a major role in lung mechanics, this study was designed to determine whether it influences afferent neural feedback to the brainstem. Rats were anaesthetised, paralysed, artificially ventilated and unilaterally vagotomised before recording neural action potentials responding to ventilation stroke (NAPvS) from single units of slowly adapting receptors. The mean (+/-SEM) NAPvs decreased by 7.4% (+/-2.3%) at post-instillation of Tween-20 (a commercial surfactant previously employed in pulmonary studies) from their pre-instillation levels, whereas, by contrast, NAPvs increased

The late blink reflex response abnormality due to lesion of the lateral tegmental field.

We report on a blink reflex abnormality observed in two patients, which provides additional information on the central pathways mediating this reflex. Autopsy was performed in one patient and MRI in the other. In the first patient there was a small lesion at the dorsal middle third of the lateral tegmental field and in the second patient at the level of the dorsal lower third of the medulla oblongata. In both patients the common finding was the absence of the late response (R2) ipsilateral to the side of the lesion, while the R2 response on the unaffected side was normal regardless of the side

Ascending pathways from the nucleus of the solitary tract to the subfornical organ in the rat.

Electrical stimulation of the nucleus of the solitary tract (NTS) produced orthodromic excitation (n = 28, 15%) and inhibition (n = 6, 4%) of the activity of neurons in the subfornical organ (SFO) in male rats under urethane anesthesia. Almost all (n = 26) of the excitatory responses (n = 28) were blocked by microiontophoretically applied phentolamine, an alpha-adrenergic antagonist, but not by timolol, a beta-adrenergic antagonist. In contrast, the inhibitory response of all the neurons (n = 6) tested was not affected by either phentolamine or timolol. Approximately two-third (n = 19) of SFO

Serial kinematic analysis of the canine hindlimb joints after deafferentation and anterior cruciate ligament transection.

OBJECTIVE AND DESIGN: Transection of the anterior cruciate ligament 2 weeks after ipsilateral hindlimb deafferentation leads to osteoarthritis of the knee joint within 3 weeks. We analyzed the gait of six dogs that underwent this procedure in order to identify kinematic changes that could account for this rapid joint degeneration. All animals were video taped, 1, 3, 6, 9 and 13 weeks after surgery while they trotted on a treadmill. RESULTS: In each dog, extension of the hip, knee and ankle joints of the unstable limb was increased, and the yield phase of the unstable knee was delayed or atten

Spontaneous and mechanically evoked afferent activity originating from myelinated fibres in ferret inferior alveolar nerve neuromas.

The inferior alveolar nerve is a predominantly sensory branch of the trigeminal nerve which runs within a bony canal, and is frequently damaged in patients. A small proportion of these patients develop neuropathic pain, and this may result from neural activity generated at the injury site. To investigate this abnormality we have used electrophysiological techniques in an animal model to determine the level of spontaneous activity and mechanical sensitivity of myelinated fibres ending in a neuroma of the inferior alveolar nerve. In 20 anaesthetised adult male ferrets the left inferior alveolar

Trigeminal structure-function relationships: a reevaluation based on long-range staining of a large sample of brainstem a beta fibers.

Prior studies suggest that some classes of thickly myelinated (A beta) afferents have distinct morphologies in the trigeminal (V) brainstem complex, and that single fibers have collaterals with different shapes in the four V subnuclei. However, these conclusions are based upon relatively few and incompletely stained fibers and limited statistical rigor. In the present study, 104 fibers were stained more completely with neurobiotin in rats to provide within-fiber intersubnucleus comparisons, and between-fiber intrasubnucleus comparisons, of collaterals associated with a vibrissa, guard hairs, h

Functional magnetic resonance imaging of the human motor cortex.

Functional magnetic resonance (MR) imaging of the brain was performed during motor task activation in five normal subjects and a patient with meningioma using conventional fast low-angle shot sequences and a 2.0 T system. A high intensity area in the motor cortex was observed in all normal subjects. Single-slice studies showed the right-sided finger task produced an increase of 1.9-23.5% (6.67 +/- 4.36%) in the signal intensity of the left motor cortex, while the left-sided finger task increased the signal by 1.5-18.2% (6.09 +/- 3.34%) in the right motor cortex. There was no significant differ

Morphological characterization of substance P receptor-immunoreactive neurons in the rat spinal cord and trigeminal nucleus caudalis.

Although there is considerable evidence that primary afferent-derived substance P contributes to the transmission of nociceptive messages at the spinal cord level, the population of neurons that expresses the substance P receptor, and thus are likely to respond to substance P, has not been completely characterized. To address this question, we used an antibody directed against the C-terminal portion of the rat substance P receptor to examine the cellular distribution of the receptor in spinal cord neurons. In a previous study, we reported that the substance P receptor decorates almost the enti

Plasticity in human somatosensory thalamus as a result of deafferentation.

Experimental studies indicate that deafferentation results in reorganization of the somatosensory map at various levels of the CNS, such that the representation of a body part adjacent to a region that is denervated expands into the deafferented area. Recent data suggest that in the human this occurs at the cortical level, but subcortical structures have not been systematically investigated. To test the hypothesis that the human thalamus is capable of significant reorganization as a result of changes in afferent input, microelectrode recording and stimulating techniques were used to define tha

Splanchnic and somatic afferent convergence on cervical spinal neurons of the rat.

The rostral cervical spinal cord is increasingly being considered the source of important propriospinal regulation. To better understand the substrate for this function, we investigated the effects of stimulation of the greater splanchnic nerve (GSN) and both thoracic and cervical somatic afferents on the activity of cervical spinal neurons. Extracellular single-neuron recordings were made in the C2-C5 spinal segments of chloralose-anesthetized, paralyzed, and artificially ventilated rats. Neurons were classified according to their responses to GSN stimulation. Neurons were inhibited by this s

[Intravesical electrostimulation in detrusor hypocontractility].

Intravesical electrostimulation activates bladder mechanoreceptors and can induce detrusor contraction. The latency of afferent impulses indicates that myelinated A-delta fibres are used for impulse transmission. Twenty patients, of whom twelve showed acontractile, and eight hypocontractile detrusor muscles, were treated by means of intravesical electrostimulation. Prior to treatment, the average residual urine was 218 ml. Eleven out of the 20 patients on completion of the stimulation programme had a residual urine of less than 100 ml. Only four patients remain on intermittent clean catheteriz

[Clinical significance of cervico-ocular reactions].

Cervically induced eye movements consist of a nystagmus and a deviation of the mean eye position (shift). They show in relation to different neck torsion velocities maximum reactions at slow velocities. The clinical significance of these cervico-ocular reactions is discussed controversially. Therefore we investigated 40 healthy subjects without any neurootological findings, who, in addition, underwent a manual examination. The neck torsion test was performed automatically with quantifiable stimulus parameters and a complete head fixation by means of individual dental casts. It could be shown t

Influence of halothane, enflurane, and isoflurane on motor evoked potentials.

The influence of the inhalational anesthetics halothane, enflurane, and isoflurane on motor evoked potentials was studied in a total of 10 rabbits. Motor evoked potentials were recorded from the extremity muscles as well as from the epidural space of the spinal cord and cauda equina in response to electrical stimulation of the motor cortex at baseline conditions and equianesthetic concentrations (0.25 to 1.5 minimal alveolar concentration). Our results show a dose-dependent suppression of the electromyographic responses, which was similar with all anesthetics. Beyond 0.5 minimal alveolar conce

Ventilatory compensation for lactacidosis in ponies: role of carotid chemoreceptors and lung afferents.

We investigated changes in arterial PCO2 (PaCO2) and pulmonary ventilation (VE) in normal, carotid chemoreceptor-denervated, and hilar nerve-denervated ponies during intravenous lactic acid infusion at rest and treadmill exercise at 1.8 mph-5% grade (mild) and 1.8 mph-15% grade (moderate). Lactic acid, (0.5 M) infusion of 0.10, 0.13, and 0.20 ml.min-1.kg-1 at rest and mild and moderate exercise increased arterial [H+] linearly throughout the 10 min of acid infusion. At 10 min of infusion, arterial [H+] had increased approximately 20 nmol/l (0.2 pH units) for each condition and group. Under mos

Pain relief achieved by transcutaneous electrical nerve stimulation and/or vibratory stimulation in a case of painful legs and moving toes.

A patient is described with painful legs and moving toes. The pain had been occurring for more than 15 years, and a variety of therapies had been attempted with only partial, if any, success. Only morphine had succeeded in relieving the pain, but it had to be discontinued to avoid tolerance and dependence. We devised a treatment consisting of transcutaneous electrical nerve stimulation (TENS), vibratory stimulation (VS), and a combination of the two methods (TENS + VS). TENS brought about partial pain relief, but was less effective than VS; dual stimulation (TENS + VS) led to complete alleviat

Relative afferent pupillary defects in multiple sclerosis.

Relative afferent pupillary defects were found in 69 of 386 patients (18%) seen by the author at the University of British Columbia Multiple Sclerosis Clinic between Feb. 1, 1985, and Apr. 30, 1987. Pupil defects were more common in patients with clinically definite multiple sclerosis, those with recent or unilateral optic neuritis and those with unilateral or asymmetric optic atrophy. A relative afferent pupillary defect was found in 15 patients with no history of optic neuritis, 5 patients with no optic atrophy and 2 patients with normal visual evoked potentials. The frequency of relative af

Reflex responses to myocardial ischemia and reperfusion. Role of prostaglandins.

Powerful vasodepressor and cardioinhibitory reflexes are activated in humans during inferior wall myocardial ischemia or infarction and during restoration of flow to the ischemic region. Experiments in dogs have demonstrated that these responses are due to stimulation of afferent vagal fibers that are located mainly in the inferior wall of the heart. Prostaglandins are released during myocardial ischemia and possibly during reperfusion. Prostaglandins stimulate chemosensitive but not mechanosensitive endings in the ventricles. Our studies determined whether blockade of prostaglandin synthesis

Electrophysiological characteristics of sensory mechanisms in the kidney.

R2 chemoreceptors are excited by backflow of urine or isotonic KCl into the renal pelvis, they do not respond to backflow of isotonic saline at the same intrapelvic pressure but are excited during periods of renal ischemia. R1 chemoreceptors are excited during complete renal ischemia but otherwise exhibit no activity. The responses of multiunit afferent renal nerve activity (ARNA) to these stimuli in Sprague Dawley rats follow the same patterns exhibited by R2 chemoreceptors, and the data do not support the presence of mechanoreceptive nerves which are excited by increases in intrapelvic press
