An immunomodulatory encapsulation system to deliver human iPSC-derived dopaminergic neuron progenitors for Parkinson's disease treatment.

Parkinson's disease is a neurodegenerative condition associated with the progressive loss of dopaminergic neurons. This leads to neurological impairments with heightening severity and is globally increasing in prevalence due to population ageing. Cell transplantation has demonstrated significant promise in altering the disease course in the clinic, and stem cell-derived grafts are being investigated. Current clinical protocols involve systemic immunosuppression to prevent graft rejection, which could potentially be avoided by encapsulating the therapeutic cells in a locally immunosuppressive b

Carboxylated Nanoparticle Surfaces Enhance Association with Mucoid Pseudomonas aeruginosa Biofilms.

Pseudomonas aeruginosa biofilms comprise three main polysaccharides: alginate, psl, and pel, which all imbue tolerance against exogenous antimicrobials. Nanoparticles (NPs) are an exciting new strategy to overcome the biofilm matrix for therapeutic delivery applications; however, zero existing FDA approvals for biofilm-specific NP formulations can be attributed to the complex interplay of physiochemical forces at the biofilm-NP interface. Here, we leverage a set of inducible, polysaccharide-specific, expressing isogenic P. aeruginosa mutants coupled with an assembled layer-by-layer NP (LbL NP)

Designing an antimicrobial film for wound applications incorporating bacteriophages and ε-poly-l-lysine.

Alginate-based dressings have been shown to promote wound healing, leveraging the unique properties of alginate. This work aimed to develop and characterize flexible individual and bilayered films to deliver bacteriophages (phages) and ε-Poly-l-lysine (ε-PLL). Films varied in different properties. The moisture content, swelling and solubility increased with higher alginate concentrations. The water vapour permeability, crucial in biomedical films to balance moisture levels for effective wound healing, reached optimal levels in bilayer films, indicating these will be able to sustain an ideal mo

Biomimetic mineralization of hydrated magnesium carbonate for hydrogel reinforcement and heavy metal adsorption.

With excessive Mn(Ⅱ) and Cu(Ⅱ) pollution in aquatic environments posing potential health risks to inhabitants, the emergence of carbon capture, utilization and storage (CCUS) technology has promoted the improvement of heavy metal remediation technologies. Using hydrothermal sediment as a crystal seed, rhamnolipid was used to mediate biomimetic mineralization to prepare hydrated magnesium carbonate (HMC) composites to enhance the Mn(Ⅱ)/Cu(Ⅱ) adsorption performance of alginate hydrogels. Hydrothermal sediment is beneficial for accelerating biomimetic mineralization, while rhamnolipid can induce

Novel approach to water-efficient bulk industrial textile printing production of cotton fabric.

Water scarcity poses a global threat in climate change era, and regrettably, the textile processing industry is squandering a significant volume of water during bulk production. This research focused on a sustainable water-saving approach in the printing of cotton fabric by modifying the reactive printing recipe and methodology. Three modified recipes (X, Y, Z) and one controlled recipe (C) were tested using reactive dyes. The conventional reactive printing recipe (Control) includes sodium alginate, urea, mild oxidizing agent, and sodium bicarbonate. In contrast, the modified recipe trials inc

Evaluation of polymeric adjuvants associated with cadidate vaccine strain Brucella ovis ∆abcBA in a murine model of Brucella ovis infection.

Brucellosis is an infectious disease caused by facultative intracellular Gram-negative bacteria, of great importance in animal and human health. An ideal vaccine against brucellosis should induce protection, not cause disease in animals or humans, and not interfere with serological diagnosis. Vaccine adjuvants can improve the immune response, leading to a more intense and prolonged protection, improving its effectiveness. The Brucella ovis ΔabcBA strain encapsulated in alginate provides an experimental vaccine formulation that protects against Brucella spp. infection. However, the investigatio

Enhancing composite emulsion stability through electrostatic modulation of polysaccharides-whey protein isolate interactions for probiotic encapsulation.

This study investigates how electrostatic interactions between whey protein isolate (WPI) and polysaccharides (konjac glucomannan, guar gum, pectin, sodium alginate, chitosan) affect the stability and encapsulation of high-internal-phase emulsions (HIPEs, oil fraction >74%), revealing charge-driven structural and network stabilization mechanisms. The findings demonstrate that anionic WPI (pH 7.0) alone was more effective than cationic WPI (pH 4.0) in stabilizing oil-in-water HIPEs. Neutral polysaccharides enhanced emulsifying performance through viscosity-induced stabilization. While electrost

Bacteriophage and Nitric Oxide Combined Release from a Dual Hydrogel Matrix for Wound Healing Applications.

Infection sites and open wounds provide a prime environment for the growth of opportunistic pathogens, leading to persistent infections caused by bacteria that contain antimicrobial-resistant phenotypes. Mistreatment of these wound infections often increases antimicrobial resistance (AMR), thereby decreasing the effectiveness of antibiotics. With the increase in AMR, new antimicrobial therapeutics that target these hard-to-kill pathogens are needed. Herein, naturally harvested bacteriophages (ECΦ) were combined with another established antimicrobial molecule, nitric oxide (NO). This combinatio

Beyond the matrix: rethinking antibiotic tolerance in CF biofilms using 3D models.

Chronic lung infections in cystic fibrosis (CF) patients are associated with Pseudomonas aeruginosa biofilms exhibiting high antibiotic tolerance with no clear explanation. We investigate the role of the biofilm matrix in this antibiotic tolerance using 3D biofilm models based on acetylated alginate and DNA, mimicking mucoid biofilms. Printed from these bioinks seeded with P. aeruginosa (PAO1), these models support robust microcolony formation as observed in vivo and enable high-throughput assessment of antibiotic diffusion and efficacy. Surprisingly, antibiotic diffusion is not significantly

Mechanically reinforced hydrogel vehicle delivering angiogenic factor for beta cell therapy.

Type 1 diabetes mellitus (T1DM) is a chronic disease affecting millions worldwide. Insulin therapy is currently the golden standard for treating T1DM; however, it does not restore the normal glycaemic balance entirely, which increases the risk of secondary complications. Beta-cell therapy may be a possible way of curing T1DM and has already shown promising results in the clinic. However, low retention rates, poor cell survival, and limited therapeutic potential are ongoing challenges, thus increasing the need for better cell encapsulation devices. This study aimed to develop a mechanically rei

Factors affecting on in vitro release of miconazole from in situ ocular gel.

The reason for conducting this study is to prolong release of miconazole in the ocular site of action by ocular-based gels (OBGs) formulations. The formulation factors affecting on the release from OBG should be studied using various gelling agents in various concentrations to achieve the improvement in retention and residence time in response to prolonged release. In this study, the formulations were prepared using carbopol 940, pectin, sodium alginate, poloxamer 407, and poly(methacrylic acid) at 0.5%, 1%, and 1.5% w/v, respectively. Hydroxypropyl methylcellulose E5 (HPMC E5) 1% was added as

Improving the 3D Printability of Sugar Glass to Engineer Sacrificial Vascular Templates.

A prominent obstacle in scaling up tissue engineering technologies for human applications is engineering an adequate supply of oxygen and nutrients throughout artificial tissues. Sugar glass has emerged as a promising 3D-printable, sacrificial material that can be used to embed perfusable networks within cell-laden matrices to improve mass transfer. To characterize and optimize a previously published sugar ink, we investigated the effects of sucrose, glucose, and dextran concentration on the glass transition temperature (Tg), printability, and stability of 3D-printed sugar glass constructs. Fabrication and characterization of bioprints with Lactobacillus crispatus for vaginal application.

Bacterial vaginosis (BV) is characterized by low levels of lactobacilli and overgrowth of potential pathogens in the female genital tract. Current antibiotic treatments often fail to treat BV in a sustained manner, and > 50% of women experience recurrence within 6 months post-treatment. Recently, lactobacilli have shown promise for acting as probiotics by offering health benefits in BV. However, as with other active agents, probiotics often require intensive administration schedules incurring difficult user adherence. Three-dimensional (3D)-bioprinting enables fabrication of well-defined archi

The marine sponge-derived inorganic polymers, biosilica and polyphosphate, as morphogenetically active matrices/scaffolds for the differentiation of human multipotent stromal cells: potential application in 3D printing and distraction osteogenesis.

The two marine inorganic polymers, biosilica (BS), enzymatically synthesized from ortho-silicate, and polyphosphate (polyP), a likewise enzymatically synthesized polymer consisting of 10 to >100 phosphate residues linked by high-energy phosphoanhydride bonds, have previously been shown to display a morphogenetic effect on osteoblasts. In the present study, the effect of these polymers on the differential differentiation of human multipotent stromal cells (hMSC), mesenchymal stem cells, that had been encapsulated into beads of the biocompatible plant polymer alginate, was studied. The different
