Sordarins are a new class of antifungal agents which selectively inhibit fungal protein synthesis (FPS) by impairing the function of elongation factor 2. The present study investigates possible correlations between sordarin pharmacokinetic (PK) properties and therapeutic efficacy, based on a murine model of invasive systemic candidiasis, and provides a rationale for dose selection in the first study of efficacy in humans. A significant correlation between PK parameters and the in vivo activity of GM 237354, taken as a representative FPS inhibitor, was demonstrated in a murine model of lethal s

BACKGROUND: The incidence of invasive fungal disease (IFD) has increased significantly, and IFD is a major cause of mortality among those with hematological malignancies. As a novel second-generation triazole antifungal drug offering both efficacy and safety, isavuconazole (ISA) is recommended by various guidelines internationally for the first-line treatment of invasive aspergillosis (IA) and invasive mucormycosis (IM) infecting adults. Given that it was only approved in China at the end of 2021, there is currently a lack of statistical data regarding its usage in the Chinese population. The

UNLABELLED: Invasive fungal infections are a major cause of human mortality due in part to a very limited antifungal drug arsenal. The identification of fungal-specific pathogenic mechanisms is considered a crucial step to current antifungal drug development and represents a significant goal to increase the efficacy and reduce host toxicity. Although the overall architecture of F1FO-ATP synthase is largely conserved in both fungi and mammals, the subunit i/j (Su i/j, Atp18) and subunit k (Su k, Atp19) are proteins not found in mammals and specific to fungi. Here, the role of Su i/j and Su k in

PURPOSE: Autosomal recessive (AR) CARD9 deficiency is an inherited immune disorder which results in impaired innate immunity against various fungi. Superficial and invasive fungal infections, mainly caused by Candida or Trichophyton species, are the hallmark of CARD9 deficiency. Together with the increasing number of CARD9-deficient patients reported, different pathogenic fungal species have been described such as Phialophora, Exophiala, Corynespora, Aureobasidium, and Ochroconis. Saprochaete capitata is an opportunistic infectious agent in immunocompromised patients and is a common cause of i

BACKGROUND: There is increasing evidence that invasive fungal disease (IFD) is more likely to occur in non-neutropenic patients in critical care units. A number of randomised controlled trials (RCTs) have evaluated antifungal prophylaxis in non-neutropenic, critically ill patients, demonstrating a reduction in the risk of proven IFD and suggesting a reduction in mortality. It is necessary to establish a method to identify and target antifungal prophylaxis at those patients at highest risk of IFD, who stand to benefit most from any antifungal prophylaxis strategy.
OBJECTIVES: To develop and va

Natural killer (NK) cells are critical components of the innate immune system. Their primary role is to induce apoptosis in target cells, such as cancerous or virally infected cells. These targets are recognized through interactions between activating or inhibitory receptors on the NK cell surface. Among the activating receptors is the natural cytotoxicity receptor NKp46. Several ligands for this receptor have been identified, including the epithelial adhesin Epa1 from the yeast Candida glabrata. Invasive candidiasis caused by this yeast is a significant complication for patients with hematolo

Candida glabrata is the second most common cause of invasive candidiasis and is widely known to have reduced susceptibility to fluconazole relative to many other Candida spp. Upc2A is a transcription factor that regulates ergosterol biosynthesis gene expression under conditions of sterol stress such as azole drug treatment or hypoxia. Through an in vitro microevolution experiment, we found that loss-of-function mutants of the ATF/CREB transcription factor CST6 suppresses the fluconazole hyper-susceptibility of the upc2A∆ mutant. Here, we confirm that the cst6∆ upc2A∆ mutants are resistant to f

This study assessed the development of disseminated candidiasis within Galleria mellonella larvae and characterized the proteomic responses of Candida albicans to larval hemolymph. Infection of larvae with an inoculum of 1 × 10⁶ yeast cells reduced larval viability 24 (53.33 ± 3.33%), 48 (33.33 ± 3.33%) and 72 (6.66 ± 3.33%) h post infection. C. albicans infection quickly disseminated from the site of inoculation and the presence of yeast and hyphal forms were found in nodules extracted from infected larvae at 6 and 24 h. A range of proteins secreted during infection of G. mellonella by C. alb

We report the association of an undescribed, reversible metaphyseal dysplasia (RMD) with autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED) in two patients, one homozygous and one heterozygous for a 13-bp deletion in exon 8 of the autoimmune regulator (AIRE) gene. One patient also had a novel deletion in exon 6, resulting in a frameshift mutation and introduction of a STOP codon in exon 10. Their APECED phenotypes differed, but both patients developed progressive skeletal deformities and growth failure from early childhood. Radiological examination suggested a generalized

Modern therapeutic procedures that are based on advanced chemotherapy and modern medical technique have led to a prolongation of chronic patient's survival, but have also created conditions for the development of till now little known infectious complications. Among these are, no doubt, mycotic diseases caused by opportunistic microorganisms. Primarily apathogenic fungi, living as a rule in the external environment as saprophytes or colonizing as harmless epiphytes the integumental localizations of healthy humans, start to behave in the subjects with reduced resistance as invasive pathogens. Candida glabrata is the second leading cause of mortality in immunocompromised patients hospitalized for invasive candidiasis (IC). Several drugs have been available to treat this disease for decades, such as polyenes, azoles, echinocandins, flucytosine, and, in critical cases, amphotericin B. However, these antifungals' constant and routine use have led to the development of resistance mechanisms, making the design and development of new drugs indispensable. The first step for the design and subsequent synthesis of a new chemical molecule as a potential antifungal is the identification of new

Namibia is a sub-Saharan country with one of the highest HIV infection rates in the world. Although care and support services are available that cater for opportunistic infections related to HIV, the main focus is narrow and predominantly aimed at tuberculosis. We aimed to estimate the burden of serious fungal infections in Namibia, currently unknown, based on the size of the population at risk and available epidemiological data. Data were obtained from the World Health Organization (WHO), Joint United Nations Programme on HIV/AIDS (UNAIDS), and published reports. When no data existed, risk po

Mice lacking expression of the homologous phosphatases Sts-1 and Sts-2 (Sts-/- mice) are resistant to disseminated candidiasis caused by the fungal pathogen Candida albicans To better understand the immunological mechanisms underlying the enhanced resistance of Sts-/- mice, we examined the kinetics of fungal clearance at early time points. In contrast to the rapid C. albicans growth seen in normal kidneys during the first 24 h postinfection, we observed a reduction in kidney fungal CFU within Sts-/- mice beginning at 12 to 18 h postinfection. This corresponds to the time period when large numb

Vulvovaginal candidiasis (VVC) caused by non-albicans Candida species, particularly Candida glabrata, is increasingly associated with treatment failure due to intrinsic or acquired resistance to antifungal drugs. In this study, we explored the anticandidal activity of a postbiotic cell-free supernatant (CFS) obtained from a microbial consortium isolated from homemade kefir (Lactococcus lactis subsp. hordniae, Lactococcus lactis subsp. lactis, and Pichia kudriavzevii) against one reference strain and two multidrug-resistant clinical isolates of C. glabrata. Antifungal susceptibility assays show

Candida species resistant to fluconazole have raised concern in the scientific medical community due to high mortality in patients with invasive disease. In developing countries, such as Brazil, fluconazole is the most commonly used antifungal, and alternative treatments are expensive or not readily available. Furthermore, the occurrence of biofilms is common, coupled with their inherent resistance to antifungal therapies and the host's immune system, these microbial communities have contributed to making infections caused by these yeasts an enormous clinical challenge. Therefore, there is an

Nakaseomyces glabratus (formerly Candida glabrata) (N. glabratus) is a commensal yeast that is increasingly associated with invasive candidiasis. The rapid acquisition of echinocandin resistance by N. glabratus has raised public health concerns and led the WHO to classify it as a high-priority fungal pathogen in 2022. However, the impact of antifungal resistance on the host immune response remains poorly understood. Since echinocandin resistance often involves alterations to the fungal cell wall, which is critical for host immune recognition, such mutations may affect fungal fitness and immune

Invasive fungal infections are a leading cause of mortality and morbidity in patients with severely impaired host defenses, while treatment options remain limited. Fatty acid synthase (FASN), the key enzyme regulating de novo biosynthesis of fatty acids, is crucial for the lethal infection of fungi; however, its pathogenic mechanism is still far from clear. Here, we identified the α subunit of FASN as a potential immunotherapeutic target against systemic Candida albicans infection. The avirulence of the encoded gene (FAS2) -deleted mutant in a mouse model of systemic candidiasis is not due to

Candida auris was listed as a critical fungal priority group pathogen by the World Health Organization (WHO) in 2022. It has become a leading cause of invasive candidiasis in serious nosocomial infections globally. While Candida species, particularly C. albicans, are linked to cancer development, the role of C. auris in carcinogenesis remains unexplored. This scoping review aimed to evaluate the existing evidence on the role of C. auris infection in carcinogenesis and its associated risk factors. Following the PRISMA-ScR guidelines, a comprehensive search of three databases was conducted from

Among the millions of species in the fungal kingdom, fewer than 300 cause disease in humans, compared to the tens of thousands that cause disease in plants, insects, and ectothermic vertebrates. While the number of 'pathogenic' species is relatively small, fungal infections remain one of the leading threats to human health. Vulnerable populations with compromised immunity face the highest risk of serious infection, with over 2.5 million individuals succumbing to systemic fungal infections annually. Invasive candidiasis caused by Candida species is the leading cause of mycotic death worldwide,

The ATP-binding cassette (ABC) transporter Cdr1 is a key mediator of antifungal drug resistance in Candida albicans, a major pathogen responsible for invasive candidiasis. Cdr1 belongs to the pleiotropic drug resistance (PDR) ABC transporter subfamily, exhibits broad substrate specificity, and plays a central role in protecting C. albicans cells from azole drugs and many other xenobiotics. Although high-resolution structures of Cdr1 and its homologs are now available, the transport cycle of Cdr1 remains incompletely understood. Furthermore, although these structures provide valuable informatio
