Integrated Cathode Design for Polysulfide Limitation and Kinetic Enhancement Enabled by In Situ Intermittently Encapsulating Sulfur with Poly(ionic liquid).

Although lithium-sulfur (Li-S) batteries are promising next-generation energy storage devices, polysulfide shuttling and electrode volume deformation remain challenges for their practical applications. Herein, a groundbreaking modification strategy is proposed that leverages an integrated cathode design to synchronously restrict polysulfide shuttling and enhance electrochemical kinetics. Specifically, the poly(ionic liquid) termed VIBM with high ionic conductivity is in situ intermittently encapsulated on S surface, which effectively facilitates Li+ transport and ensures free electron transfer

Intelligent α-Cyclodextrin-Based Nanomachines Co-loading Lenalidomide and Au@TPE for NIR-I-Light Programmed Combinatorial Breast Cancer Therapy.

Malignant tumors have emerged as a major threat to human health. Micro/nanomachines offer minimally invasive advantages over traditional cancer therapies, making them promising for medical applications. Herein, we designed an α-cyclodextrin-based nanomachine (αALN) encapsulating Au@TPE as a photothermal agent, leveraging cyclodextrins' encapsulation and carrier properties to achieve high Au@TPE loading and targeted delivery to tumor sites for effective photothermal therapy (PTT). Furthermore, the immunomodulatory drug lenalidomide was integrated into this system. Post-administration, αALN deli

Recent progress in cancer detection and therapeutic applications of calixarene- and pillararene-based systems.

Macrocycles, such as calixarenes and pillararenes, have received more focus as potential therapeutic agents and drug carriers. They can solubilize/encapsulate different anti-cancer drugs by host-guest interactions or self-assembly. Good chemical stability, good compatibility, responsiveness to external stimuli and low toxicity to normal cells are properties that make these macrocycles good candidates for cancer detection and therapy. This review focuses on strategies developed for cancer detection and therapeutics by calixarenes and pillararenes over the past decade (from 2015 to 2025). Cone c

Starch nanoparticle platform for oral delivery of sinigrin in colitis therapy.

Sinigrin, a glucosinolate known for its potential therapeutic effects on ulcerative colitis (UC), suffers from rapid gastric clearance and low bioavailability when administered orally. To address these limitations, we developed a novel delivery system using starch nanoparticles (SNPs) to encapsulate sinigrin via a sinigrin-lecithin complex (SNG-L). This approach significantly improved sinigrin's encapsulation efficiency, stability against acidic degradation, and controlled release in simulated intestinal conditions. In vivo studies using a preclinical UC mouse model demonstrated that SNG-L@SNP

Guest-Mediated Modulation of Photophysical Pathways in a Coronene Bisimide Cyclophane.

The properties and functions of chromophores utilized by nature are strongly affected by the environment formed by the protein structure in the cells surrounding them. This concept is transferred here to host-guest complexes with the encapsulated guests acting as an environmental stimulus. A new cyclophane host based on coronene bisimide is presented that can encapsulate a wide variety of planar guest molecules with binding constants up to (4.29 ± 0.32) × 1010 M-1 in chloroform. Depending on the properties of the chosen guest, the excited state deactivation of the coronene bisimide chromophore

Periodic temperature changes drive the proliferation of self-replicating RNAs in vesicle populations.

Growth and division of biological cells are based on the complex orchestration of spatiotemporally controlled reactions driven by highly evolved proteins. In contrast, it remains unknown how their primordial predecessors could achieve a stable inheritance of cytosolic components before the advent of translation. An attractive scenario assumes that periodic changes of environmental conditions acted as pacemakers for the proliferation of early protocells. Using catalytic RNA (ribozymes) as models for primitive biocatalytic molecules, we demonstrate that the repeated freezing and thawing of aqueo

Nanogel Particles Based on Modified Nucleosides and Oligosaccharides as Advanced Delivery System.

This work addresses the challenge of delivering bioactive molecules by designing biocompatible nanogel particles (NGPs) utilizing rationally modified nature-sourced building blocks: capryl-oligochitosan and oxidized inosine. Capryl substituents endowed the resultant NGPs with membrane-penetration capabilities, while purine-containing inosine allowed H-bond/π-π/π-cation interactions. The prepared NGPs were complexed with carboxyfluorescein-labeled single-stranded oligonucleotide (FAM-oligo) and DsRed-encoding plasmid DNA. The successful delivery of FAM-oligo to the cell cytoplasm of the Nicotia

Confinement of Dyes inside Boron Nitride Nanotubes: Photostable and Shifted Fluorescence down to the Near Infrared.

Fluorescence is ubiquitous in life science and used in many fields of research ranging from ecology to medicine. Among the most common fluorogenic compounds, dyes are being exploited in bioimaging for their outstanding optical properties from UV down to the near IR (NIR). However, dye molecules are often toxic to living organisms and photodegradable, which limits the time window for in vivo experiments. Here, it is demonstrated that organic dye molecules are passivated and photostable when they are encapsulated inside a boron nitride nanotube (dyes@BNNT). The results show that the BNNTs drive

Use of mPEG-PLGA nanoparticles to improve bioactivity and hemocompatibility of streptokinase: In-vitro and in-vivo studies.

Streptokinase, a clot-dissolving agent, is widely used in treatment of cardiovascular diseases such as blood clots and deep thrombosis. Streptokinase is a cost-effective drug with a short biological half-life (i.e. 15 to 30 min). In addition, due to its prokaryotic source, the immune response quickly reacts to the drug. Despite these limitations, streptokinase is still the first choice for diseases associated with thrombosis. In this work, streptokinase was encapsulated in mPEG-PLGA nanoparticles to improve its pharmacokinetic properties. The nanoparticles containing the enzyme were prepared b

The remarkable role of emulsifier and chitosan, dextran and PEG as capping agents in the enhanced delivery of curcumin by nanoparticles in breast cancer cells.

Curcumin has been found to be a powerful anti-cancer agent. However, its efficacy is limited by its poor solubility in aqueous medium. The bioavailability of curcumin was increased by poly (lactic-co-glycolic) acid [PLGA (60/40)] nanoparticles with different capping agents such as Chitosan, Dextran and PEG and emulsifier (Tocopherol Poly (Ethylene Glycol)1000 Succinate: TPGS) with good drug loading and delivery performance. The preparation of nanoparticles derived from PLGA was achieved by emulsion solvent evaporation method and the resultant nanoparticles were characterized. The encapsulation

Co-administration of paclitaxel and 2-methoxyestradiol using folate-conjugated human serum albumin nanoparticles for improving drug resistance and antitumor efficacy.

The use of chemotherapeutic drug paclitaxel (PTX) for the treatment of tumors has several limitations, including multidrug resistance (MDR) and serious adverse reactions. This research aims to co-encapsulate PTX and the chemosensitizer 2-methoxyestradiol (2-ME) into folate-conjugated human serum albumin nanoparticles (FA-HSANPs) to reduce multiple drug resistance and improve antitumor efficiency. The results show PTX/2-ME@FA-HSANPs had uniform particle size (180 ± 12.31 nm) and high encapsulation efficacy. It also exhibited highly potent cytotoxicity and apoptosis-inducing activities in the G2

Adrenal Incidentaloma: Challenges in Diagnosing Adrenal Myelolipoma.

Adrenal myelolipomas (AMLs) are rare benign adrenal tumors, containing adipose and hematopoietic tissue, a result of reticuloendothelial cell metaplasia. Incidence on autopsy has been reported from 0.08% to 0.4%. AMLs are generally considered nonsecretory. The functional aspect of adrenal incidentaloma should be evaluated. In this article, we report a case of a 40-year-old male, who presented with uncontrolled hypertension and renal failure, with imaging revealing an adrenal incidentaloma. He was started on dialysis for acute fluid overload, and workup for pheochromocytoma revealed an elevated

MMP-sensitive PEG hydrogel modified with RGD promotes bFGF, VEGF and EPC-mediated angiogenesis.

Traumatic soft tissue defects such as bedsores, chronic skin ulcers, limb necrosis, osteonecrosis and other ischemic orthopedic diseases are the most clinically intractable and common problems in orthopedics due to unsatisfactory conventional treatments. The present study designed poly(ethylene glycol; PEG) hydrogels with covalently binded arginylglycylaspartic acid (RGD). Endothelial progenitor cells (EPCs) were encapsulated in the modified hydrogel along with vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF). Results demonstrated that the modified hydrogel d

Effect of Surface Chemical Functionalities on Collagen Deposition by Primary Human Dermal Fibroblasts.

Surface modification has been identified as an important technique that could improve the response of the body to implanted medical devices. Collagen production by fibroblasts is known to play a vital role in wound healing and device fibrous encapsulation. However, how surface chemistry affects collagen I and III deposition by these cells has not been systematically studied. Here, we report how surface chemistry influences the deposition of collagen I and III by primary human dermal fibroblasts. Amine (NH3), carboxyl acid (COOH), and hydrocarbon (CH3) surfaces were generated by plasma depositi

Controlling the in vivo activity of Wnt liposomes.

Liposomes offer a method of delivering small molecules, nucleic acids, and proteins to sites within the body. Typically, bioactive materials are encapsulated within the liposomal aqueous core and liposomal phase transition is elicited by pH or temperature changes. We developed a new class of liposomes for the in vivo delivery of lipid-modified proteins. First, we show that the inclusion of a chromophore into the liposomal or vesosomal membrane renders these lipid vesicles extremely sensitive to very small (muJ) changes in energy. Next, we demonstrate that the lipid-modified Wnt protein is not

Conjugation of a Cryptococcus neoformans-derived metalloprotease to antifungal-loaded PLGA nanoparticles treats neural cryptococcosis in an in vitro model.

Overcoming the blood-brain barrier to deliver therapeutics is a major hurdle in treating diseases of the central nervous system. We engineered 4-arm carboxyl terminated poly(D,L-lactide-co-glycolide) nanoparticles with the fungal metalloprotease Mpr1, an enzyme utilized by the neurotropic pathogen Cryptococcus neoformans (Cn) to cross the blood-brain barrier. Nanoparticles were prepared using a modified single emulsion solvent evaporation technique, and characterized in terms of shape, size, zeta potential, encapsulation efficiency, and toxicity to brain microvascular endothelial cells. Hyaluronic Acid-Functionalized Alginate Microspheres via Electrospraying for CD44-Mediated Targeted Delivery of Probiotics.

Probiotic delivery faces challenges in maintaining bacterial viability and achieving precise targeting of the gastrointestinal tract. In this study, alginate microspheres (ALG MSs) encapsulating Clostridium butyricum (CB) were fabricated using electrospray technology. Hyaluronic acid (HA) was used to modify both the microspheres and the CB surface to enable targeted delivery through the receptor-ligand interaction between HA and CD44. The resulting CBHA@ALGHA MSs exhibited an encapsulation efficiency of 69.23 ± 7.30% and a postdigestion survival rate of 94.56 ± 4.87%. HA modification enhanced

Co-localization of lymph node therapeutics enhances T cell activation and anti-tumor response.

Lymph nodes (LNs) are essential tissues in generating and modulating effective immune responses. In this study, we investigated the intra-lymphatic delivery of immunotherapy cues encapsulated in degradable microparticles (MPs). We demonstrate that co-localization of antigens and adjuvants within the same lymph node generates robust antigen-specific T cell responses and significant anti-tumor protection, while spatial separation of cargos across different lymph nodes diminishes immune efficacy. While co-encapsulation of signals in the same particle within a lymph node ensures efficient delivery
