Molecular genetic analysis of familial hypercholesterolemia: spectrum and regional difference of LDL receptor gene mutations in Japanese population.

To determine the molecular basis of familial hypercholesterolemia (FH) in Japan, 200 unrelated patients with clinically diagnosed heterozygous FH were screened for mutations in coding and promoter region of the low density lipoprotein (LDL) receptor gene using denaturing gradient-gel electrophoresis (DGGE), DNA sequencing and Southern blotting analysis. About 37 different mutations in the LDL receptor gene were identified in 125 (62.5%) of the patients, 22 of these mutations have not been described before. The most common mutations were K790X (19.5%), P664L (6.0%), FH-Tonami-1 (6.0%), IVS15-3C

Interactions between genetic and reproductive factors in breast cancer risk in a population-based sample of African-American families.

Incidence of breast cancer (BC) varies among ethnic groups, with higher rates in white than in African-American women. Until now, most epidemiological and genetic studies have been carried out in white women. To investigate whether interactions between genetic and reproductive risk factors may explain part of the ethnic disparity in BC incidence, a genetic epidemiology study was conducted, between 1989 and 1994, at the Howard University Cancer Center (Washington, DC), which led to the recruitment of 245 African-American families. Segregation analysis of BC was performed by use of the class D r

Angiotensin converting enzyme genetic polymorphism is not associated with hypertension in a cross-sectional sample of a Japanese population: the Shibata Study.

BACKGROUND: The studies on the association of deletion/ insertion (D/I) polymorphism of angiotensin converting enzyme (ACE) gene with blood pressure and hypertension reported contradictory results. Because there was no population-based study in Japan, we examine the hypothesized association in a cross-sectional sample of a Japanese cohort. METHODS AND RESULTS: The blood pressure of 464 men and 876 women aged 40-80 years was measured, and their DNA was analyzed for ACE D/I genotypes. The prevalence of the D allele was 38.7 and 39.2% in men and women, respectively (overall 39%). There was a ten

Enzyme polymorphism and genetic variability of laboratory populations of Phlebotomus papatasi, P. bergeroti, P. langeroni, and P. perniciosus.

Enzyme allomorph frequency and electrophoretic mobility were evaluated to clearly separate between morphologically similar sandfly species and to yield quantitative measures of their genetic similarity. Twelve enzyme systems were studied in laboratory colonies of Phlebotomus perniciosus, P. langeroni, P. papatasi, and P. bergeroti. Phlebotomus perniciosus and P. langeroni confirmed vectors of visceral leishmaniasis, were found to have fixed diagnostic allomorph differences at 3 enzyme loci (ICD-2, FUM, XDH). Phlebotomus papatasi, a known vector of cutaneous leishmaniasis and phleboviruses and

[Genetic diagnosis of phenylketonuria. IV. Mutations of phenylalanine hydroxylase gene in Caucasian and Gypsy populations in Czech and Slovakia Republics].

Direct sequencing was conducted on the regions of the exon 7 and 12 in the phenylalanine hydroxylase (PAH) gene amplified by the polymerase chain reaction, using solid-phase technology involving the biotin streptavidin system. A novel mutation and seven previously known mutations were identified in the PAH genes among 15 Caucasians and 10 Gypsies in the Czech and Slovakia republics, affected with classical phenylketonuria (PKU). Two of these substitutions (R243X and G272X) resulted in the generation of a premature stop codon, and a single base transition of G to A at codon 261 resulted in the

Novel promoter and splice junction defects add to the genetic, clinical or geographic heterogeneity of beta-thalassaemia in the Portuguese population.

In order to delineate the spectrum and the relative abundance of beta-globin gene defects causing thalassaemia in the Portuguese population, a representative sample was analysed including 51 beta-thalassaemia carriers along with 26 patients representing different clinical phenotypes. Seven mutations were identified, four of which [codon 39 (C----T), 39%; intervening sequence (IVS) 1 nucleotide (nt) 1 (G----A), 26%; IVS 1 nt 110 (G----A), 17%; IVS 1 nt 6 (T----C), 15%] account for 97% of 93 beta-thalassaemia chromosomes. Population Genetics of the Greater Grison (Galictis vittata; Mustelidae, Carnivora) and How Changes Since Miocene-Pliocene Have Shaped the Phylogenetic Relationships Between Grison Species and Other Neotropical Mustelid Genera.

Mustelids are a very diverse carnivoran group, boasting the largest number of carnivoran species today, with the most extreme ecological adaptations. Nevertheless, Neotropical Lyncodontini, one Neotropical mustelid subfamily, is one of the least studied from a genetic and evolutionary perspective. The use of small fragments of three mitochondrial (mt) markers (NADH5, 12S rRNA, and D-loop) proved to be very useful for recovering relevant information on the evolutionary history of Neotropical Lyncodontini species of the genera Galictis and Lyncodon. This study used the largest population sample

A genetic and clinical risk factor algorithm to aid in identifying new cases of chronic kidney disease from the general population.

PURPOSE: The purpose of this study is to develop a genetic test to aid in diagnosing chronic kidney disease (CKD). One challenge in treating CKD is that 80%-90% of people with it are undiagnosed and thus do not access healthcare promptly. The problem arises because early-stage CKD has no overt symptoms, and the current policy is to perform diagnostic tests only when accompanied by risk factors such as old age, hypertension, and diabetes. METHODS: This study describes the development of the RICK (RIsk for Chronic Kidney disease) algorithm that employs a polygenic risk score for CKD plus clinic

SNP-Based KASP Markers Reveal Genetic Diversity and Population Structure Among African Sorghum Breeding Lines and Hybrids.

Sorghum is a strategic food-security and livestock-feed crop in Africa, and molecular characterization of its germplasm underpins genetic diversity assessment and hybrid parent selection. This study evaluated 106 sorghum accessions (23 A-lines, 28 B-lines, 32 R-lines, and 23 hybrids) from ICRISAT and Seed Co gene banks using 79 single nucleotide polymorphism (SNP)-based Kompetitive Allele-Specific PCR (KASP) markers. Genetic diversity, population structure, cluster analysis, and analysis of molecular variance (AMOVA) were assessed to support heterotic grouping. Gene diversity ranged from 0.23

Assessment of genomic prediction and genetic gain in multi‑population half-sib families in the perennial grass crop intermediate wheatgrass.

The University of Minnesota has been domesticating the perennial forage intermediate wheatgrass (IWG) since 2011 using a combination of conventional methods and modern breeding tools such as genomic selection. Globally, most IWG selection nurseries are spaced-planted individuals of several hundred genotypes whereas commercial fields established for grain production are row-planted panmictic populations. This study evaluated genomic prediction models and estimated genetic gain in yield and agronomic performance of row-planted IWG half-sib families assessed over 3 years and 2 locations, Lamberto

Unravelling genetic diversity, population connectivity, and evolutionary relationships of Indian mealybugs through mtCO1-based phylogenetic and haplotype analyses.

BACKGROUND: Mealybugs (Hemiptera: Pseudococcidae) are economically important invasive pests that cause substantial losses in agricultural and horticultural crops worldwide. However, information on their diversity, genetic variation, and population structure in India remains limited. This study investigated the diversity, phylogenetic relationships, and population genetic structure of economically important mealybugs across different agroclimatic regions of India using mitochondrial cytochrome c oxidase subunit I (mtCOI) sequences. METHODS: A total of 134 mealybug specimens were analysed using

Life's Essential 8, genetic susceptibility, and risk of cognitive impairment: results from an elder population-based cohort study.

INTRODUCTION: The American Heart Association's Life's Essential 8 (LE8) framework has been linked to cognitive impairment, but evidence in Chinese older adults remains limited. METHODS: This cross-sectional study included 7469 adults aged ≥60 years from the West China Health and Aging Cohort. Modified Poisson regression with robust error variance was used to examine the associations of LE8 scores and genetic risk, as well as their joint associations, with cognitive impairment. RESULTS: Compared with the low LE8 score group, the high LE8 score group had a prevalence ratio (PR) of 0.65 (95% co

Genetically diverse influenza antibodies highlight the role of IG germline gene variation and inform population-comprehensive vaccine strategies.

The regular emergence of influenza strains with pandemic potential necessitates vaccines that elicit protective immune responses across genetically diverse human populations. A critical but understudied factor is how germline-encoded variation in immunoglobulin genes shapes the development of neutralizing antibodies. Here, by combining personalized immunoglobulin genotyping with high-throughput paired-chain antibody sequencing from influenza A hemagglutinin (HA)-binding B cells across four donors, using a technique we developed called individualized single-cell analysis of paired expressed ant

Genetic architecture of externalizing and internalizing traits in the general population and its link to psychiatric conditions and well-being: a multivariate approach.

Externalizing and internalizing behaviours capture heritable, transdiagnostic dimensions of psychopathology, but their genetic architecture and links to societal outcomes remain incompletely understood. This study applied a multivariate framework to population-based, non-clinical traits to explore simultaneous genetic associations across externalizing and internalizing behaviours to elucidate their biological underpinnings and genetic relation to clinical diagnoses. Using genomic structural equation modelling, we built a two-factor model for externalizing (EXT) and internalizing (INT) behaviou

High Polyunsaturated Fatty Acid Intake Attenuates the Genetic Risk of Higher Waist Circumference in a Sri Lankan Adult Population.

BACKGROUND: Metabolic diseases, like type 2 diabetes mellitus and obesity, show a growing public health concern in Sri Lanka. Genetic predisposition and diet contribute to metabolic disease risk, but there are limited investigations into the impact of gene-diet interactions on metabolic disease risk in the Sri Lankan population. In this study, we examined whether a metabolic genetic risk score (GRS), constructed from 10 single nucleotide polymorphisms (SNPs), interacts with dietary factors to influence metabolic health indicators in Sri Lankan adults. METHODS: This cross-sectional study inclu
