Regulation of gas exchange and haemolymph pH in the cockroach Nauphoeta cinerea.

Ventilatory control of internal CO(2) plays an important role in regulating extracellular acid-base balance in terrestrial animals. While this phenomenon is well understood among vertebrates, the role that respiration plays in the acid-base balance of insects is in need of much further study. To measure changes in insect haemolymph pH, we implanted micro pH optodes into the haemocoel of cockroaches (Nauphoeta cinerea). They were then exposed to normoxic, hypoxic, hyperoxic and hypercapnic atmospheres while their haemolymph pH, VCO(2) and abdominal ventilation frequency were measured simultaneo

The angiotensinogen AGT p.M235T gene polymorphism may be responsible for the development of severe anaphylactic reactions to insect venom allergens.

BACKGROUND: Insect venom allergy (IVA) is present in 1-3% of the population. A group of patients with high specific IgE do not react to stings. In contrast, a proportion of patients with IVA have low specific IgE levels. These findings indicate that factors other than specific IgE may also be involved in IVA. Dysfunction of the renin-angiotensin system (RAS) has been described as a potential factor in IVA. The objective of this study was to determine the prevalence of angiotensin AGT p.M235T and angiotensin-converting enzyme ACE I/D, I/I, D/D gene polymorphisms in patients with IVA and to rela

Green Management Strategies for Cochliomyia hominivorax Myiasis: Potential of Essential Oils and Nanodelivery Systems.

Cochliomyia hominivorax, the main etiological agent of traumatic myiasis in the New World, continues to have significant health and economic impacts due to its effects on animal health, livestock production and public health in affected regions. Although conventional control strategies, including the Sterile Insect Technique and chemical insecticides, have substantially reduced its distribution, concerns regarding insecticide resistance, environmental impacts, and recent re-emergence events underscore the need for sustainable alternative control approaches. This review summarizes current advan

Evaluation of the protective efficacy of a spatial repellent to reduce malaria incidence in children in Mali compared to placebo: study protocol for a cluster-randomized double-blinded control trial (the AEGIS program).

BACKGROUND: Spatial repellents have been widely used for the prevention of mosquito bites but their efficacy in reducing mosquito-borne diseases has never been evaluated in Africa. Additionally, spatial repellents have the potential of being critical tools in the prevention of mosquito-borne diseases in contexts where typical vectors control efforts such as insecticide-treated nets (ITNs) and indoor residual spray (IRS) are inaccessible or underutilized such as among displaced populations or in emergency relief settings. To address this knowledge gap, Kolondieba District, Sikasso Region, Mali

A fat body transcriptome analysis of the immune responses of Rhodnius prolixus to artificial infections with bacteria.

BACKGROUND: Rhodnius prolixus is an important vector of Trypanosoma cruzi, the causal agent of Chagas disease in humans. Despite the medical importance of this and other triatomine vectors, the study of their immune responses has been limited to a few molecular pathways and processes. Insect immunity studies were first described for holometabolous insects such as Drosophila melanogaster, and it was assumed that their immune responses were conserved in all insects. However, study of the immune responses of triatomines and other hemimetabolous insects has revealed discrepancies between these and

Controlled Transmission of a Fijivirus Under Field Conditions Using Mass-Reared Planthoppers.

Mal de Río Cuarto disease, caused by a Fijivirus, is a major constraint for maize production in Argentina. The traditional evaluation of resistant hybrids is limited by the low efficiency of natural virus transmission and the lack of standardized field inoculation methods. We developed a protocol that combines laboratory mass-rearing of the planthopper vector Delphacodes kuscheli with a controlled field transmission system. The method involves the synchronized production of large insect populations, acquisition of viruliferous vectors under controlled conditions, and their safe transport to th

Identification of Specific Virulence Factors of Pseudomonas Strains in the Biocontrol of the Potato Pest Tecia solanivora.

Tecia solanivora (The Guatemalan potato tuber moth) is a major potato pest, responsible for up to 20% of crop losses and a significant economic impact. Certain Pseudomonas exhibit insecticidal activity and produce virulence factors with cytotoxic and antimicrobial properties, positioning them as promising candidates for biological control. This study evaluated seven Pseudomonas strains with insecticidal activity and identified key virulence factors involved. The strains demonstrated varying degrees of insecticidal activity, with Pseudomonas protegens strains CHA0 and 59C being the most lethal,

Decoding cis-regulatory elements in the germline of the human malaria vector Anopheles gambiae.

Cis-regulatory elements (CREs) drive tissue- and cell-specific gene expression and are essential for safe, sustainable genetic control strategies in pest and vector insects, including the engineering of gene drives in the primary human-malaria vector Anopheles gambiae. Yet CREs remain poorly defined in mosquitoes due to limited computational tools and practical methods for identification and validation. We present a systematic in silico approach for CRE discovery, correlating targeted DNA-motif searches with gene expression, followed by frequency and distribution analysis within putative promo

Extract of Tangerine Peel as a Botanical Insecticide Candidate for Smallholder Potato Cultivation.

BACKGROUND: Contemporary agriculture heavily relies on synthetic chemicals to ensure high yields and food security; however, their overuse has led to health issues and the development of pesticide resistance in pests. Researchers are now exploring natural, eco-friendly alternatives for pest control. METHODS: This study evaluated two ethanol-based formulations (1.25% and 2.50%, v/v) derived from the tangerine peel (Citrus reticulata L. var. Clementina) against conventional chemical treatments and an untreated control group in the cultivation of potatoes (Solanum tuberosum L. var. Capiro). A ra

A review of the potential impacts of coastal mosquito control programs on Australian Stingless Bees (Apidae, Meliponini)-likely exposure pathways and lessons learned from studies on honey bees.

The impact of the programmatic use of larvicides for mosquito control on native stingless bees (e.g., Apidae, Meliponini) is a growing concern in Australia due to heightened conservation awareness and the growth of hobbyist stingless bee keeping. In Australia, the two most widely used mosquito larvicides are the bacterium Bacillus thuringiensis var. israelensis (Bti) and the insect hormone mimic methoprene (as S-methoprene). Each has a unique mode of action that could present a risk to stingless bees and other pollinators. Herein, we review the potential impacts of these larvicides on native A

wMel Wolbachia alters female post-mating behaviors and physiology in the dengue vector mosquito Aedes aegypti.

Globally invasive Aedes aegypti disseminate numerous arboviruses that impact human health. One promising method to control Ae. aegypti populations is transinfection with Wolbachia pipientis, which naturally infects ~40-52% of insects but not Ae. aegypti. Transinfection of Ae. aegypti with the wMel Wolbachia strain induces cytoplasmic incompatibility (CI), allows infected individuals to invade native populations, and inhibits transmission of medically relevant arboviruses by females. Female insects undergo post-mating physiological and behavioral changes-referred to as the female post-mating re

Novel molecular approaches to combat vectors and vector-borne viruses: Special focus on RNA interference (RNAi) mechanisms.

Vector-borne diseases, such as dengue, chikungunya, zika, yellow fever etc pose significant burden among the infectious diseases globally, especially in tropical and sub-tropical regions. Globalization, deforestation, urbanization, climate change, uncontrolled population growth, inadequate waste management and poor vector-management infrastructure have all contributed to the expansion of vector habitats and subsequent increase in vector-borne diseases throughout the world. Conventional vector control methods, such as use of insecticides, have significant negative environmental repercussions in

Chagas Disease Vectors of Paraguay: Entomoepidemiological Aspects of Triatoma sordida (Stål, 1859) and Development of an Identification Key for Paraguayan Triatomines Based on Cytogenetics Data.

Approximately 150,000 people are living with Chagas disease in Paraguay. Although the country has been since 2008 considered as one of the countries that succeeded in interrupted the vector transmission of Chagas by Triatoma infestans in houses of the eastern region, there are nine other species notified in the country that are potential vectors and also deserve attention from vector control programs. Thus, we carried out an entomoepidemiological study of T. sordida in the eastern and western regions of the country and we developed an identification key for Paraguay's triatomines based on cyto

Evaluation of Chilean Boldo Essential Oil as a Natural Insecticide Against Chrysomya megacephala (Diptera: Calliphoridae).

Chrysomya megacephala (Fabricius, 1794) is a vector of enteric bacteria, protozoa, helminths, and viruses. These Diptera can also be responsible for secondary myiasis in several animal species. Therefore, it is easy to understand the relevance of studies focusing on C. megacephala dissemination control. The employment of essential oils as natural insecticides must be considered as a promising alternative for the replacement of synthetic insecticides. In this context, the essential oil obtained from Chilean boldo leaves should be highlighted. The aim of the present work was to assess the insect
