omega-agatoxin IVA and excitotoxicity in cortical neuronal cultures.

Because the P- and Q-type Ca2+ channel antagonist omega-agatoxin IVA inhibits glutamate release, agatoxin-related drugs could have a role in the treatment of excitotoxic neurologic disorders such as stroke. We examined the effect of omega-agatoxin IVA on excitotoxicity (lactate dehydrogenase release) induced by depolarization with veratridine or ouabain, or by N-methyl-D-aspartate, in neuron-enriched cerebral cortical cultures. omega-Agatoxin IVA (< or = 300 nM) failed to reduce excitotoxicity, as did the L-type Ca2+ channel antagonist nimodipine and the N-type antagonist omega-conotoxin GVIA.

[Basedow's disease].

Graves' disease has been shown to be an autoimmune disorder. Circulating antibodies to the thyrotropin receptor stimulate thyroid hormone synthesis and hence lead to hyperthyroidism. The diagnosis is made mainly on clinical grounds. Measurements of thyroid hormone concentrations and titers of thyroid-stimulating immunoglobulins confirm the diagnosis. Further diagnostic procedures are rarely needed. There still exists some controversy about the appropriate treatment for Graves' disease. Nevertheless some therapeutic guidelines are proposed. Advantages and disadvantages of the various methods ar

Hypoglycemia: a pathophysiologic approach.

An exploration of the factors that sustain glucose levels in the normal fasting subject reveals that the single major component is conservation of glucose rather than gluconeogenesis. Conservation is achieved by recycling of glucose carbon as lactate, pyruvate and alanine, and a profound decrease in the oxidation of glucose by the brain brought about by the provision and use of ketones. What glucose continues to be oxidized is for the most part formed from glycerol. Gluconeogenesis from protein plays little part in the process. Fasting hypoglycemia results from disorders affecting either one o

Peripubertal Physical Activity in Male Wistar Rats Helps to Prove the Concept of Developmental Origins of Health and Disease.

INTRODUCTION: The global rise in obesity, driven by sedentary lifestyles and poor eating habits, presents significant public health and economic challenges. Obesity is a risk factor for several non-communicable diseases; moreover, it impacts childhood and puberty, critical windows for metabolic programming, which increases the risk of developing metabolic disorders in adulthood. Physical exercise has emerged as an effective strategy to prevent and manage obesity, promoting chronic adaptations that enhance quality of life. Considering the developmental plasticity of the peripubertal phase, we h

Maternal Programming with a Cafeteria Diet Leads to Glucose Intolerance, Increased Insulinemia, Inflammation, and Intestinal Dysbiosis in Rat Offspring.

The intake of diets high in fat and sugar but low in protein, fiber, and vitamins during pregnancy can contribute to metabolic imbalance, immunological disorders, and intestinal barrier disruption in the offspring. This study investigated the effects of a cafeteria (CAF) diet, high in fat and sugar, consumed throughout pregnancy and lactation, on metabolism, inflammation-related markers, and intestinal microbiota in rat offspring. Maternal programming was conducted for more than 9 weeks in 16 female Wistar rats aged 8-10 weeks. They were divided into two diet groups: control and CAF, each with

Postpartum sacral stress fractures and pubic symphysis diastasis as under-recognized structural causes of low back and pelvic girdle pain.

Pelvic girdle pain (PGP) is common after pregnancy, but clinical attention mainly focuses on benign, self-limiting ligamentous and muscular disorders. This narrative review highlights postpartum sacral stress fractures and postpartum pubic symphysis diastasis (PPSD) as under-recognized structural causes of severe postpartum low back and PGP and situates them within a postpartum pelvic ring instability spectrum (bony stress fractures and ligamentous disruption, with or without posterior ring injury). We narratively reviewed the English-language literature from 1980 to 2025 on sacral stress or i

Early-Life Melatonin Supplementation Reduces the Long-Term Behavioral, Morphological, and Molecular Alterations in a Rat Model of Autism Spectrum Disorder.

Autism spectrum disorder (ASD) is a complex neurodevelopmental condition characterized by significant challenges in social interactions and repetitive behaviors. Its prevalence is high, with males affected at a rate four times higher than females. Among various comorbidities associated with ASD, sleep disorders and abnormal melatonin levels have emerged as critical areas of concern. Melatonin, a potent endogenous hormone, plays essential roles as a circadian regulator, anti-inflammatory, and antioxidant agent through the release of pro- and anti-inflammatory cytokines and stimulation of endoge

The effect of N-acetylcysteine use in high-fat diet-exposed pregnant rats on glucose metabolism and oxidative status in dams and their offspring.

Maternal obesity has been associated with an increased risk of oxidative and metabolic disorders in offspring later in life. This study investigates the therapeutic effects of N-acetylcysteine (NAC), a potent antioxidant, on obesity-induced inflammation, oxidative stress and impaired glucose metabolism in an experimental rat model. Maternal obesity was induced in female rats by administering a high-fat diet (HFD) (60% kcal from fat). NAC was given via intragastric gavage at a dose of 150 mg/kg. Female rats were mated at 12 weeks of age, and the respective diets were maintained throughout gesta

Prolactin effects on the pathogenesis of diabetes mellitus.

BACKGROUND: Prolactin (PRL) is a pituitary hormone promoting lactation in response to the suckling reflex. Beyond its well-known effects, novel tissue-specific and metabolic functions of PRL are emerging. AIMS: To dissect PRL as a critical mediator of whole-body gluco-insulinemic sensitivity. METHODS: PubMed-based search with the following terms 'prolactin', 'glucose metabolism', 'type 2 diabetes mellitus', 'type 1 diabetes mellitus', 'gestational diabetes mellitus' was performed. DISCUSSION: The identification of the PRL-glucose metabolism network poses the basis for unprecedented avenues

Maternal Di-(2-ethylhexyl)-Phthalate exposure during pregnancy altered energy metabolism in immature offspring and caused hyperglycemia.

Di-(2-ethylhexyl)-phthalate (DEHP), as distinctive endocrine disrupting chemicals, has become a global environmental pollutant harmful to human and animal health. However, the impacts on offspring and mothers with maternal DEHP exposure are largely unknown and the mechanism remains elusive. We established DEHP-exposed maternal mice to investigate the impacts on mother and offspring and illustrate the mechanism from multiple perspectives. Pregnant mice were administered with different doses of DEHP, respectively. Metagenomic sequencing used fecal and transcriptome sequencing using placentas and

Effects of maternal exposure to procymidone on hepatic metabolism in the offspring of mice.

As an effective fungicide widely used in agricultural production, the excessive procymidone (PRO) residue has been detected in the environment and food. Our previous study demonstrated that PRO could destroy the intestinal barrier in mice and has a joint toxic effect. To explore the cross-generational impact of maternal exposure, 10-week-old C57BL/6 female mice were orally administrated to 10 and 100 mg/kg body weight/day of PRO during pregnancy and lactation. The offspring obtained nutrients from the maternal through the placenta and breast milk, and PRO residues were detected in the liver, i

Perinatal oxycodone exposure causes long-term sex-dependent changes in weight trajectory and sensory processing in adult mice.

RATIONALE: In utero opioid exposure is associated with lower weight and a neonatal opioid withdrawal syndrome (NOWS) at birth, along with longer-term adverse neurodevelopmental outcomes and mood disorders. While NOWS is sometimes treated with continued opioids, clinical studies have not addressed if long-term neurobehavioral outcomes are worsened with continued postnatal exposure to opioids. In addition, pre-clinical studies comparing in utero only opioid exposure to continued post-natal opioid administration for withdrawal mitigation are lacking. OBJECTIVES: Here, we sought to understand the

Melatonin on hypothyroidism and gonadal development in rats: a review.

We evaluated the evidence in research on the effects of melatonin on hypothyroidism and gonadal development. According to the World Health Organization, thyroid disorders due to iodine deficiency affect about 740 million people worldwide. Hypothyroidism is a thyroid dysfunction characterized by hypometabolism of the gland, with reduced or physiologically normal T3 and T4 serum levels, and high TSH level. This disorder occurs mainly in adult women in the reproductive phase, with a prevalence of 2% among the world's female population, with profound repercussions on gestation and fetal formation.

Inflammation and LPS-Binding Protein Enable the Stimulatory Effect of Endotoxin on Prolactin Secretion in the Ovine Anterior Pituitary: Ex Vivo Study.

Prolactin is a hormone that plays an important role in the regulation of many physiological processes including lactation, reproduction, fat metabolism, and immune response. The secretion of prolactin could be disturbed by an immune stress commonly accompanying infection. This study was designed to determine the influence of bacterial endotoxin-lipopolysaccharide (LPS)-on prolactin gene (PRL) expression and prolactin release from the ovine anterior pituitary (AP) explants collected from saline- and LPS-treated ewes in the follicular phase. The expressions of toll-like receptor 4 (TLR4) and pro

Long-term valproic acid exposure increases the number of neocortical neurons in the developing rat brain. A possible new animal model of autism.

The aim of this study was to test the hypothesis that long-term fetal valproic acid (VPA) exposure at doses relevant to the human clinic interferes with normal brain development. Pregnant rats were given intraperitoneal injections of VPA (20mg/kg or 100mg/kg) continuously during the last 9-12 days of pregnancy and during the lactation period until sacrifice on the 23rd postnatal day. Total number of neocortical neurons was estimated using the optical fractionator and frontal cortical thicknesses were sampled in VPA exposed pups compared with an unexposed control group. We found that pups expos

Male tuberculous mastitis: a rare entity.

A 28-year old male presented to our clinic complaining of a recent onset of a painful right breast lump with redness and nipple discharge. Fine-needle aspiration biopsy revealed caseating granulomas, with a culture positive for Mycobacterium tuberculosis. He was found to have a positive PPD, but no other site of pulmonary or extra-pulmonary tuberculosis was identified. Treatment with anti-tuberculous drugs lead to complete clinical resolution of the breast lesion. The breast is a rare site of extra-pulmonary tuberculosis (TB), comprising only 0.1% of all cases. TB is re-emerging in the Western

[Hyperprolactinemia].

Hyperprolactinemia is frequent in clinical endocrinology. Its commonest causes are, besides pregnancy and lactation, drugs, mainly involving the generally used psychopharmaca and the equally ubiquitously prescribed estrogens. The single most important cause is a pituitary tumor, the prolactinoma, but lesions of the hypothalamus or pituitary stalk, primary hypothyroidism, liver cirrhosis and chronic renal failure, among others, may also provoke hyperprolactinemia. The clinical features of hyperprolactinemia in women are mainly amenorrhea, or irregular menses, galactorrhea, hirsutism, infertilit
