We report surprising results for the self-assembly of lecithin (a common phospholipid) in water-ethanol mixtures. Lecithin forms vesicles (∼ … plex trends to the dual role played by alcohols: (a) first, alcohols reduce the propensity for flat lipid bilayers to bend and form closed spherical vesicles; and (b) second, alcohols diminish the tendency of lipids to self-assemble in the solvent mixture. At low alcohol fractions, (a) dominates, causing the initially unilamellar vesicles to grow into multilamel …

UNLABELLED: Chlamydia trachomatis is an intracellular bacterial pathogen that undergoes a biphasic developmental cycle, consisting of intrac … zyme responsible for activating fatty acids from the host cell or synthesis to be incorporated into lipid bilayers. The isolates had distinct lipidomes with varied fatty acid compositions. A reduction in the lipid compositions that HtrA prefers to bind to was detected, yet HtrA and MOMP (a key outer membrane protein) were present at higher levels in the variants …

While the roles of intrinsically disordered protein domains in driving interprotein interactions are increasingly well-appreciated, the mech … β1-9, is toxic by itself. The ordered part (Aβ10-40) is the major determinant of how Aβ attaches to lipid bilayers, enters neuronal cells, and localizes primarily in the late endosomal compartments. However, once Aβ enters the cell, it is the disordered part (only when it is connected to the rest of the peptide) that has a strong and stereospecific interaction w …

The increasing prevalence of antimicrobial resistance in bacteria highlights the need for alternative membrane-active compounds with favorable safety profiles. β-Caryophyllene (BCP), a bicyclic sesquiterpene, has demonstrated antimicrobial effects; however, its biological responses in Gram-negative bacteria and associated membrane interactions remain insufficiently characterized. In this study, the growth inhibitory effect of BCP against E. coli ATCC 25922 was evaluated through OD595 growth kinetics, while hemocompatibility was assessed using sheep erythrocytes, and cannabidiol (CBD) was inclu

Alzheimer's disease is an age-related pathology associated with accumulation of amyloid-β peptides, products of enzymatic cleavage of amyloid-β precursor protein (APP) by secretases. Several familial mutations causing early onset of the disease have been identified in the APP transmembrane (TM) domain. The mutations influence production of amyloid-β, but the molecular mechanisms of this effect are unclear. The "Australian" (L723P) mutation located in the C-termini of APP TM domain is associated with autosomal-dominant, early onset Alzheimer's disease. Herein, we describe the impact of familial

Multidrug-resistant Gram-negative bacteria have increased the prevalence of a variety of serious diseases in modern times. Polymyxins are used as the last-line therapeutic options for the treatment of infections. However, the mechanism of action of polymyxins remains in dispute. In this work, we used a coarse-grained molecular dynamics simulation to investigate the mechanism of the cationic antimicrobial peptide polymyxin B (PmB) interacting with both the inner and outer membrane models of bacteria. Our results show that the binding of PmB disturbs the outer membrane by displacing the counteri

Many drugs are available for the treatment of systemic or superficial mycoses, but only a limited number of them are effective antifungal drugs, devoid of toxic and undesirable side effects. Furthermore, resistance development and fungistatic rather than fungicidal activities represent limitations of current antifungal therapy. Therefore an urgent need for a new generation of antifungal agents remains. We recently synthesised a set of linear and cyclic peptides characterized by sequences typical of membrane-active antimicrobial peptides (AMP). AMT2, cyclo-AMT2, AMT3 and cyclo-AMT3 (Scheme 1) w

Changes of membrane organization upon cross-linking of its components trigger cell signaling response to various exogenous factors. Cross-linking of raft gangliosides GM1 with cholera toxin (CTxB) was shown to cause microscopic phase separation in model membranes, and the CTxB-GM1 complexes forming a minimal lipid raft unit are the subject of ongoing cell membrane research. Yet, those subdiffraction sized rafts have never been described in terms of size and dynamics. By means of two-color z-scan fluorescence correlation spectroscopy, we show that the nanosized domains are formed in model membr

The susceptibility of the membranous Ca2+-ATPase of sarcoplasmic reticulum to enzymatic inactivation at hyperthermic temperatures was investigated. Inactivation produced a break in the Arrhenius plot at 45-46 degrees C and was accompanied by an increased mobility of spin label, covalently attached to the Ca2+-ATPase. MgADP and MgATP exerted a markedly stabilizing effect on inactivation, both at pH 7.0 and in acidic media. By contrast, high-affinity Ca2+ or Mg2+ binding only moderately stabilized Ca2+-ATPase (inactivation rates were decreased 2-3 times), and this effect was non-additive, i.e.,

Charybdotoxin (CTX), a small, basic protein from scorpion venom, strongly inhibits the conduction of K ions through high-conductance, Ca2+-activated K+ channels. The interaction of CTX with Ca2+-activated K+ channels from rat skeletal muscle plasma membranes was studied by inserting single channels into uncharged planar phospholipid bilayers. CTX blocks K+ conduction by binding to the external side of the channel, with an apparent dissociation constant of approximately 10 nM at physiological ionic strength. The dwell-time distributions of both blocked and unblocked states are single-exponentia…

The macrolide polyene antibiotic amphotericin B (AmB), remains a valuable drug to treat systemic mycoses due to its wide antifungal activity and low probability of developing resistance. The high toxicity of AmB, expressed in nephropathy and hemolysis, could be partially resolved by lowering therapeutic AmB concentration while maintaining efficacy. This work discusses the possibility of using plant polyphenols and alkaloids to enhance the pore-forming and consequently antifungal activity of AmB. We demonstrated that phloretin, phlorizin, naringenin, taxifolin, quercetin, biochanin A, genistein

Liposomes, nanosized vesicles consisting of a lipid bilayer membrane, have been widely investigated as versatile drug delivery systems. Clinically, several liposomal formulations have been used as anticancer drug carriers owing to their enhanced permeability and retention (EPR) effects. Nevertheless, the accumulation of conventional liposomes in tumors remains insufficient to achieve optimal therapeutic efficacy. Therefore, the development of innovative liposomes with improved tumor selectivity and delivery efficiency is required. In this study, we designed and evaluated novel liposomes prepar

Mycobacteria are responsible for causing severe illnesses like tuberculosis and leprosy in humans. Studying the mycobacteria cell envelope presents a significant challenge due to its intricate lipid compositions and structural variations and also its harmful nature in a typical experiment setting. In this study, we use all-atom molecular dynamics simulations to study mycobacterial inner membranes (MIMs). By incorporating different types of phosphatidyl-myo-inositol-mannosides (PIMs) and their glycoconjugates such as lipomannan (LM) and lipoarabinomannan (LAM) lipoglycans, we have constructed b

PURPOSE: To elucidate the mechanical properties of the bovine lens cortical membrane (CM), the nuclear membrane (NM) containing cholesterol bilayer domains (CBDs), and whole bovine lenses.
METHODS: The total lipids (lipids plus cholesterol) from the cortex and nucleus of a single bovine lens were isolated using the monophasic methanol extraction method. Supported CMs and NMs were prepared from total lipids extracted from the cortex and nucleus, respectively, using a rapid solvent exchange method and probe-tip sonication, followed by the fusion of unilamellar vesicles on a flat, freshly cleave

Voltage-activated ion channels are essential for electrical signaling, yet the mechanism of voltage sensing remains under intense investigation. The voltage-sensor paddle is a crucial structural motif in voltage-activated potassium (K(v)) channels that has been proposed to move at the protein-lipid interface in response to changes in membrane voltage. Here we explore whether tarantula toxins like hanatoxin and SGTx1 inhibit K(v) channels by interacting with paddle motifs within the membrane. We find that these toxins can partition into membranes under physiologically relevant conditions, but t
